Antioxidant and Inhibitory Activities on Angiotensin Converting Enzyme in Lysimachia clethroides Duby (큰까치수영의 항산화 및 안지오텐신 전환 효소 저해 활성)

  • This study was conducted to develop physiologically active plant materials from medicinal plants. Crude extracts and solvent fractions prepared from Lysimachia cletroides Duby were tested for their antioxidant and antihypertensive activities. For ellucidating antioxidant potential, inhibition rate on linoleic acid peroxidation, as well as scavenging activities on superoxide anion and 1,1-dipicrylphenylhydrazyl (DPPH) radical were evaluated. For analyzing antihypertensive effect, inhibitory activity on angiotensin converting enzyme (ACE) was done. Methanol extract of L. cletroides showed potent inhibition activity of 83% on linoleic acid peroxidation, which was more effective than -2% of ${\alpha}-tocopherol$ at $25\;{\mu}g/ml$. Methanol and water extracts exhibited strong scavenging activities of $86{\sim}109%$ and $96{\sim}122%$ on superoxide anion radical which was higher than $-4{\sim}69%$ of ascorbic acid at $5{\sim}200\;{\mu}g/ml$. Hexane, ether and ethylacetate fractions possessed 133, 100 and 88% inhibitory activities on ACE at $4,000\;{\mu}g/ml$, respectively. From the results, it was expected that Lysimachia cletroides could be a new antioxidant and antihypertensive resource.

  • The angiotensin converting enzyme (ACE) inhibitory activity in peptic hydrolysate of raw anchovy cooking discards was 51.3% at 1 mg of protein per $100\;{\mu}L$ sample solution. While, after the treatment of pepsin for 4 h, was 65.8%. The crude peptides fractionated through Bio-gel P-2 column chromatography consisted of five fractions $({P-1}{\sim}{P-5})$ and had maximum inhibitory activity in the fraction P-2 ($IC_{50}$=0.319 mg protein/mL). The fraction P-2 was rich in aspartic acid, glutamic acid, and glycine.

Physiological Activities of Hot Water Extract from Ailanthus altissima (가죽나무(Ailanthus altissima) 열수 추출물의 생리활성)

  • In this study, extract of Ailanthus altissimawere prepared using hot water under high pressure. The extract were examined for election donating ability (EDA), superoxide dismuase (SOD)-like activity, nitrite scavenging ability (NSA), xanthine oxidase inhibition levels, and tyrosinase inhibition ability. The EDA, using the 1,1-diphenyl-2-picrylhydrazyl method of root extract was 91.25% at 1.0mg/mL. The SOD-like activity of leaf extract was highest at 49.07% and the NSA was 93.33% at pH 1.2, and 85.40% at pH 3.0. The xanthine oxidase inhibitory levels of extracts of A. altissima roots, stems, and leaves were 92.09 97.44% when the extract were tested at 2.0mg/mL. The highest tyrosinase inhibition levels obtained from loot extract were 67.38% at 2.0mg/mL and 63.97% at 0.1mg/mL.

Production of Anti-dementia Acetylcholinesterase Inhibitor from Pleurotus ostreatus (Heuktari) and Inhibitory Effect on PC12 Neuron Apoptosis (흑타리버섯으로부터 항치매성 Acetylcholinesterase 저해물질의 생산 및 PC12 신경세포사 저해 효과)

  • To develop a new antidementia acetycholinesterase (AChE) inhibitor from edible mushrooms, the inhibitory effects on AChE of water and ethanol extracts from various edible mushrooms were measured. Among the tested compounds, 70% ethanol extracts from Tremella fuciformis showed the highest AChE inhibitory activity, at 25.3% (IC50: 9.9 mg). Water extracts from the fruiting body of Pleurotus ostreatus (Heuktari) showed AChE inhibitory activity of 20.2% (IC50: 12.4 mg). However, the yield (40.8%) from Pleurotus ostreatus (Heuktari) was higher than that from Tremella fuciformis (5.0%). Therefore, we selected Pleurotus ostreatus (Heuktari) as the most promising candidate for a mushroom containing anti-dementia AChE inhibitors. The AChE inhibitor from Pleurotus ostreatus (Heuktari) was optimally extracted when its fruiting body was treated with water for 6 h at 30℃. The anti-dementia effects of the partially purified AChE inhibitor from Pleurotus ostreatus (Heuktari) were observed in PC12 nerve cells.

In vitro Inhibitory Activity on Rat Intestinal Mucosa ${\alpha}-glucosidase$ by Rice Hull Extract (왕겨추출물의 쥐소장점막 ${\alpha}-glucosidase$에 대한 in vitro에서의 저해효과)

  • In order to search for the way to utilize rice hull as a renewable resource, the inhibition on ${\alpha}-glucosidase$ and the fractionation of rice hull extract was investigated. An ethanol extract of rice hull from Japonica-type rice seeds exhibited 30% inhibitory activity on rat intestinal brush border ${\alpha}-glucosidase$ (1.4 mU/mL) in vitro at the concentration of 0.8 mg/mL using 6 mM p-nitrophenyl ${\alpha}-D-glucopyranoside$ as a substrate $(IC_{50}\;162\;mg/mL)$. Among the fractions obtained by partitioning the ethanol extract successively with solvents, the ethyl acetate fraction at the concentration of 0.8 mg/mL was found to exhibit the most potent inhibitory activity i.e. 65% inhibition of ${\alpha}-glucosidase\;(IC_{50}\;0.14\;mg/mL)$. Silica gel column chromatography of the ethyl acetate fraction exhibited slightly higher (90%) inhibitory activity, and its subsequent fractionation by Sephadex LH-20 column chromatography did not improve inhibitory activity. Considering the inhibitory activity and yield, the ethyl acetate fraction obtained by the solvent-partitioning process would be a candidate for the hypoglycemic food if it has in vivo effectiveness.

Quality Changes in Fresh-Cut Potato (Solanum tuberosum var. Romano) after Low-Temperature Blanching and Treatment with Anti-Browning Agents (저온 blanching 및 갈변저해물질의 처리에 따른 최소가공 감자의 저장 중 품질변화)

  • Quality changes in fresh-cut potatoes during storage at $4^{\circ}C$ after treatment with low-temperature blanching and antibrowning agents were studied. Fresh-cut potatoes were treated by dipping for 1.5 min in a browning inhibitor solution containing 0.5% (w/v) ascorbic acid, 0.5% (w/v) citric acid, 0.5% (w/v) sodium chloride, 0.1% (w/v) trehalose, and 0.005% (w/v) biotin, at $60^{\circ}C$, with subsequent cooling for 1.5 min and storage at $4^{\circ}C$. The browning properties of fresh-cut potatoes were examined by measurement of polyphenol oxidase (PPO) activity and total phenolic content. Changes in quality attributes over a 14-day period were assessed in terms of titratable acidity, pH, water-soluble solid level, and gas analysis at $4^{\circ}C$. During storage, PPO activity increased, with the lowest activity seen after about 7 days of storage. Treatment with antibrowning solution at $4^{\circ}C$ increased visual sensory attributes during storage. Low-temperature blanching in distilled water more effectively inhibited browning compared with exposure to browning inhibitor solution, as assessed after 7 days of storage. Fresh-cut potatoes respired aerobically after different treatments during storage at $4^{\circ}C$.

Some Kinetic Properties of an Extracellular Chitinase from Streptomyces sp, 115-5 (Streptomyces속 115-5 균주로부터 생성된 Chitinase의 저해작용기작)

  • An extracellular chitinase was purified from the culture fluid of Streptomyces sp. 115-5, and its inhibition mechanism by end product was studied. The activity of chitinase was suppressed by the reducing sugar as the reaction proceeded, and the activity was inhibited by the addition of D-glucose. Besides D-glucose, the rate of chitin hydrolysis was inhibited by D-glucuronic acid, D-sorbitol and D-xylose in the reaction system of the enzyme. it was found that the hydroxyl groups at the C-2, C-3 and C-4 position of D-glucose molecule play an important role in the inhibition of the chitinase activity. D-glucose was found to inhibit the enzyme activity by mixed type of competitive and non-competitive mode.

Effect of Salt Type and Concentration on the Growth of Lactic Acid Bacteria Isolated from Kimchi (소금의 종류와 농도가 배추김치에서 분리한 유산균의 생육에 미치는 영향)

  • Tests show that the growth of lactic bacteria in kimchi varies according to the type and concentration of salt used. Weissella confusa, the early stage bacteria in kimchi fermentation, increased sharply after 5 hr of induction with 3% light salt and refined salt. However, the induction period lengthened to 12 hr with 3% sea salt and bamboo salt. Lactobacillus delbrueckii ss lactis and L. pentosus which grow in the middle stage of fermentation, and L. hamsteri, which grows at the end stage of fermentation, were found after 12 hr of induction with 3% salt of all kinds. When 5% light salt was added to the culture medium, the induction period of bacteria other than W. confusa lengthens to 12 hr. The trend is similar for sea salt, bamboo salt, and refined salt, with a higher NaCl concentration resulting in less growth. W. confusa showed salt tolerance, but L. hamsteri was affected by the type and concentration of salt. With 5% sea salt, bamboo salt, and refined salt, the growth of bacteria was inhibited by up to 24 hr.

Inhibitory Effect on Rotavirus by Exopolysaccharides Extracted from Kefir (Kefir에서 추출한 Exopolysaccharide의 Rotavirus의 저해효과)

  • This study was carried out to investigate the toxicity of exopolysaccharides (EPS) from kefir toward MA104 cells and evaluate the inhibitory effects of kefir EPS on rotavirus infection. The results obtained are summarized as follows: Lactic acid bacteria (Lactobacillus fermentum, L. acidophilus, L. brevis) and yeasts (Candida kefyr, Cryptococcus albidus, Pichia ohmeri) were isolated and identified from kefir grain and culture. At 1% EPS, the inhibitory effects of EPS on the infection of MA-104 cells using the MTT assay were $72.52{\pm}6.48%$ for human rotavirus (KU), $36.06{\pm}7.63%$ for bovine rotavirus (NCDV), and $81.66{\pm}1.11%$ for porcine rotavirus (OSU). At 1/128% EPS, the effects were $24.98{\pm}4.58%$ for human rotavirus (KU), $4.71{\pm}6.16%$ for bovine rotavirus (NCDV), and $4.05{\pm}14.90%$ forporcine rotavirus (OSU). EPS isolated from kefir have inhibitory effects on rotaviruses of various serotypes and rotaviruses from different animals.

Effect of Antioxidation and Inhibition of Matrix Metalloproteinase-1 from Ligularia fischeri (곰취의 항산화와 UVA에 의한 MMP-1 발현 저해효과)

  • In this study, we examined the effects of antioxidant, lipid peroxidation inhibition and suppression of UVA-induced MMP-1 expression in human dermal fibroblasts. Ligularia fischeri extract showed free radical scavenging effect by 82.3% at 5 mg/mL and superoxide radical scavenging effect by 79.3% at 5 mg/mL in the kanthine/xanthine oxidase system, respectively. At the concentration of $500{\mu}g/mL$, L. fischeri extract showed 97% inhibition on lipid peroxidation of linoleic acid. UVA induced MMP expression in human dermal fibroblasts was reduced 35% by treatment with $100{\mu}g/mL$ of L. fischeri extract. These results suggest that L. fischeri extract can be used for an anti-aging agent by antioxidation, lipid peroxidation inhibition and reducing UVA-induced MMP-1 production.
