장내 가수분해조건에 의한 Casein 가수분해물의 Angiotensin Converting Enzyme 저해 효과

  • 장내 단백질 가수분해효소에 의한 casein 분해산물들의 ACE 저해 활성을 비교하기 위하여 인공위액, 인공장액 및 인공위액과 장액 연속처리를 각각 반응조건으로 일정시간 casein을 가수분해한 후 각 효소 가수분해물이 혈압상승 peptide 생성효소인 Angiotensin converting enzyme(ACE)에 대한 저해활성을 측정하였다. 각 소화액에 따른 가수분해물은 모두 ACE 저해활성을 나타내었으며, 2시간 효소처리를 하였을 때, 인공위액, 인공장액, 인공위액과 장액 가수분해물은 각각 69%, 80% 및 88%로 최대 ACE 저해활성을 보였다. 그리고 casein의 가수분해 정도를 확인하기 위한 SDS-PAGE에서는 인공위액으로 처리한 군보다 인공장액으로 처리한 군이 더 작은 분자량으로 분해되는 것을 확인할 수 있었고, 인공위액과 장액을 함께 처리한 군에서는 1시간 안에 대부분의 분해가 이루지는 것을 관찰할 수 있었다.

  • In the course of studies for anti-plaque agents, novel procyanidin structure isolated from Artocarpus heterophyllus folium was established by thiolysis and spectroscopic analysis. The chemical structure was identified for $(-)-epiafzelecin-(4{\beta}{\rightarrow}8)-afzelecin-(4{\alpha}-8)-catechin$ containing the trimeric flavan-3-ols and molecular weight was 833[M-H] by FAB-MS negative ion method. The inhibitory effect on the glucosyltransferase activity was investigated, novel compound showed complete inhibition at 1.0 mM and inhibited on the glucosyltransferase noncompetitively.

Inhibitory Effects on the Enzymes Involved in the Inflammation by the Ethanol Extracts of Plant Foodstuffs (식물성 일반식품 자원의 에탄올 추출물이 염증 효소계에 미치는 영향)

  • Inflammation is a complex process resulting from a variety of mechanisms. Combined inhibition of the activities of enzymes involved in the process may therefore be considered more important in anti-inflammatory property of plant extracts than any single contribution. In this study, the inhibitory effects of the ethanol extracts of thirty plant foods on the activities of secretory phospholipase $A_{2}$ ($sPLA_{2}$), cyclooxygenase-1 (COX-1), cyclooxygenase-2 (COX-2), and 12-lipoxygenase (12-LOX) were examined. Several legumes, mungbean sprout and some leaf vegetables inhibited the activity of $sPLA_2$, upstream enzyme of inflammation pathway. Only soybean sprout and mungbean sprout significantly inhibited 12-LOX activity. Although most of extracts inhibited the activities of both COX-1 and COX-2, water dropwort and amaranth showed selectivity for the inhibition of COX-2 over COX-1. Especially, mungbean showed anti-inflammatory property at both upstream and downstream of inflammation pathway with relatively low $IC_{50}$ values for $sPLA_{2}$ and COX-2 enzymes. Mungbean sprout exhibited inhibitory effects on all enzymes related to early and late inflammation and soybean sprout suppressed 12-LOX and COX-2 simultaneously, although the activities of these plants were showed at relatively high concentration. Therefore, mungbean, mungbean sprout, and soybean sprout appear to exhibit anti-inflammatory effects by combined inhibition of inflammatory enzymes.

Biological Activity of Chamomile (Matricaria chamomilla L.) Extracts (캐모마일(Martricaria chamomilla L.)의 생리활성)

  • The biological activity of water and ethanol extracts from Chamomaile for functional food source were examined. Total phenol contents in the $60\%$ ethanol extracts $(24.98\pm0.20\;mg/g)$ from Chamomaile leaf was higher than those of water extracts $(23.64\pm0.35\;mg/g)$ The major phenolic compound by HPLC were rosemarinic acid and Quercetin. $60\%$ ethanol extracts had higher content of these phenolics than water extacts. Electron donating ability showed $91.05\%$ in the water extracts and $95.49\%$ in the $60\%$ ethanol extracts. Antioxidant protection factor (PF) showed $0.71\pm0.02 $ in the water extracts and $1.48\pm0.03 $ in the $60\%$ ethanol extracts. The water extracts of Chamomaile leaves did not have antimicrobial activity against H. pylori, but the $60\%$ ethanol extracts revealed the slight antimicrobial activity as 9.42 mm of clear zone. Angiotensin converting enzyme inhibition was $57.98\%$ in water extracts and $91.36\%$ in $60\%$ ethanol extracts. Xanthine oxidase activity was $73.48\%$ in water extracts and $81.96\%$ in $60\%$ ethanol extracts. The results suggest that Chamomailes extract may be useful as potential source as antioxidant, angiotensin converting enzyme and xanthine oxidase inhibitors.

lnhibitory Effect o fVarious Cruciferous Vegetable on the Growth of Human Cancer Calls (인체암세포증식에 있어 십자화과 채소의 억제효과)

  • The anticarcingenic effect of methanol extracts from such cruciferous vegetables as cabbage, red cabbage, Korean cabbage, kale, cauliflower, broccoli, radish root, leafy radish, rape leaves and shepherd’s purse on the growth of human K-562 leukemia cells, MG-63 osteosarcoma cells, HT-29 colon cancer cells and AGS gastric cancer cells were studied. All of cruciferous vegetables inhibited more than 70% of the growth of K-52 leukemia cells and more than50% fo rhe growth fo AGS gastric cancer cells. Particularly, kale, broccoli and shepherd’s purse showed inhibition rates of 93.5%, 93,5% and 96.3% on the growth of AGS gastric cancer cells, respectively. In case of HT-29 colon cancer cells, the methanol extracts of cabbage, kale and shepherd’purse exhibited 82.4%, 72.15, 79.4% and 95.6% of inhibitory effects, respectively. The cabbage, kale, cauliflower and shepherd’s purse extracts also highly suppressed the proliferation of MG-63 cells. Generally the 10 cruciferous vegetable we studied strongly decreased the growth of various human cancer cells in vitro, however, kale and shepherd’s showed the most effective vegetable among them.

Antimutagenic and Cytotoxic Effects of Hovenia dulcis Thumb Leaves Extracts (헛개나무 잎 추출물의 항돌연변이원성 및 암세포 성장억제효과)

  • The effect of Hovenia dulcis Thumb leaves on the mutagenicity in salmonella assay and inhibitory effects on the growth of cancer cells were studied. On antimutagenicity as evaluated by Ames test, the extract and fractions of Hovenia dulcis Thumb leaves had no effect on the mutagenicity by themselves. However, methanol extract and fractions from Hovenia dulcis Thumb showed strong inhibitory effect on the mutagenesis induced by N-methyl-N#-nitro-N-nitroso-guanidine (MNNG) and benzo(a)pyrene (B(a)P). Among the solvent fractions of methanol extract, the hexane, chloroform and butanol fraction exhibited stronger inhibitory activity against MNNG and B(a)P induced mutagenesis than water fraction. For anticancer effects, Hovenia dulcis Thumb loaves extract and fractions against cancer cell lines including HepG2 and HT29 were investigated. The methanol extract, the hexane fraction and the chloroform fraction of Hovenia dulcis Thumb leaves inhibited growth of cancer cells but they had no effect on the cytotoxicity of normal human liver cells under the same conditions.

Inhibitory Effect of Extracts of Platycodon grandiflorum (the Ballon Flower) on Oxidation and Nitric Oxide Production (도라지 부탄올 추출물의 항산화 및 nitric oxide 생성 저해 효과)

  • We explored the effect of extracts of dried Platycodon grandiflorum on production of reactive oxygen species (ROS), glutathione (GSH) and nitric oxide (NO). To determine antioxidant activity in the presence of $H_2O_2$-induced oxidative stress, DCFH-DA (dichlorodihydrofluorescin diacetate) assay was employed. Acetone/methylene chloride (A+M) and methanolic (MeOH) extracts of P. grandiflorum reduced intracellular ROS levels. Of the various tested fractions, n-BuOH fraction showed the highest protective effect in terms of lipid peroxide production. Total GSH levels were measured after treatment of HT1080 cells with the A+M and MeOH extracts, and other solvent fractions, at various concentration. The A+M extacts and 85% (v/v) aqueous MeOH fraction significantly increased GSH levels (p<0.05). When lipopolysaccharide (LPS)-induced NO production was evaluated, all tested crude extracts, and fractions thereof, significantly reduced NO production (p<0.05), and the n-BuOH and 85% (v/v) aqueous MeOH fractions (at 0.05 mg/mL) showed the strongest inhibitory effects. The results showed that the n-BuOH fraction inhibited both cellular oxidation and NO production, and this fraction may thus contain valuable active compounds.

Comparison of Biological Activity Between Soybean Pastes Adding Sword Bean and General Soybean Pastes (작두콩 첨가 된장과 일반 된장의 생리활성 특성 비교)

  • This study was carried out to analyse the biological activity in soybean pastes with the sword beans and general soybean pastes. The electron donating activity of soybean pastes with the sword beans(J2, J4) was higher than other soybean pastes with value of $81.5{\pm}2.2%$ and $88.3{\pm}0.7%$, respectively. And the content of total phenol compounds was higher in J2 and J4 compared to the others with value of $1773.8{\pm}2.9\;mg/kg$ and $1785.7{\pm}3.9\;mg/kg$ respectively. ACE inhibitory effect was higher in J2 and J4 than other soybean pastes with value of $63.1{\pm}1.0%$, $62.1{\pm}1.4%$, respectively. And aglycone type(daidzein, genistein) isoflavone content was higher than glucoside type(daidzin, genistin). But tyrosinase inhibitory effect was not different among test samples. From the results, soybean pastes with the sword beans(J2, J4) have higher anti-oxidant activity and ACE inhibitory effect than other soybean pastes, therefore, it may have potential to prevent hypertension.

Biological And Antimicrobial Activity of Portulaca oleracea (Portulaca oleracea의 생리활성과 항균활성)

  • The concentration of total phenolic compounds of the water extracts and 80% ethanol extracts form Portulaca oleracea were 3.05 ${\mu}g/ml$ and 6.33 ${\mu}g/ml$, respectively. The total antioxidant activities of water extracts and 80% ethanol extracts of Portulaca oleracea were 89.2% and 72.9% in DPPH assay, 69.0% and 96.5% in ABTS assay, antioxidant protection factor of the water and 80% ethanol extracts were each 2.73 PF and 3.63 PF. Tyrosinase inhibitory activities were water extracts and 80% ethanol extracts of Portulaca oleracea were 20.2% and 38.7%. Portulaca oleracea showed high antimicrobial activites against Helicobater pylori, Staphylococcus epidermidis, Staphylococcus aureus, Eschericia coli and Streptococcus mutans. Minimum inhibitory concentrations (MICs) on Helicobacter pylori, Staphylococcus epidermidis, Staphylococcus aureus, Escheichia coli and Streptococcus mutans were 200, 50, 100, 100 and 150 ${\mu}g/ml$, respectively. The result suggest that Portulaca oleracea extracts may be useful as potential source as antioxidant and antimicrobials.
