Fecal Microbiota Profiling of Holstein and Jersey, in South Korea : A Comparative Study (국내에서 사육되는 Holstein 젖소과 Jersey 젖소의 대변 미생물 분석 : 비교연구)

  • In light of the complex interactions between the host animal and its resident gut microbiomes, studies of these microbial communities as a means to improve cattle production are important. This study was conducted to analyze the intestinal microorganisms of Holstein (HT) and Jersey (JS), raised in Korea and to clarify the differences in microbial structures according to cattle species through next-generation sequencing. The alpha-diversity analysis revealed that most species richness and diversity indices were significantly higher in JS than in HT whereas phylogenetic diversity, which is the sum of taxonomic distances, is not significant. Microbial composition analysis showed that the intestinal microbial community structure of the two groups differed. In the both groups, a significant correlation was observed among the distribution of several microbes at the family level. In particular, a highly significant correlation (p<0.0001) among a variety of microbial distributions was found in JS. Beta-diversity analyis was to performed to statistically verify whether a difference exists in the intestinal microbial community structure of the two groups. Principal coordinate analysis and unweighted pair group method with arithmetic mean (UPGMA) clustering analysis showed separation between the HT and JS clusters. Meanwhile, permutational multivariate analysis of variance (PERMANOVA) revealed that their microbial structures are significantly different (p<0.0001). LEfSe biomarker analysis was performed to discover the differenc microbial features between the two groups. We found that several microbes, such as Firmicutes, Bacilli, Moraxellaceae and Pseudomonadales account for most of the difference in intestinal microbial community structure between the two groups.

Early Vegetation Succession in Abandoned Field in Cheju Island (제주도 저지대 묵밭 식물군락의 2차 천이)

  • In order to clarify the successional trend and its characteristics of plant community in abandoned field in Cheju Island, we investigated the seral changes of vegetation height, coverage, growth form, species richness, diversity and dominance index along with the time lapse, and then compared with those of mainland researches. And in order to seek the overall trend of vegetation changes, we used cluster, TWINSPAN and PCA ordination techniques. The succession was characterized by the lower species number, lower vegetation height, longer period of herb dominant and later invasion of tree species. These results may come from that ecological traits of Cheju Island itself, and ecosystem fragmented influences that block a seed (propagule) transport. Sere of the dominant species was shown as follows: Mazus japonicus(0.5∼1 years)→Conyza sumatrensis (2 years)→Artemisia princeps var. orientalis(4 years)→Artemisia princeps var. orientalis, indigofera pseudotinctoria(5 years)→Rosa multiflora, Miscanthus sinensis, etc.(8 years)→Miscanthus sinensis(12 years)→Boehmeria nivea, Pueraria thunbergiana etc.(15 years)→Mallotus japonicus(20 years)→Litsea japonica, Machilus thunbergii (20 years<). Abandoned fields were classified into three groups according to time lapse; earlier stage(0∼1 years), middle stage(2∼8 years) and later stage(8∼20 years).

  • This study was performed to develop low-fat comminuted sausages (LFSs, < 3%) manufactured with 1% single (Konjac flour, KF; kappa-carrageenan, k-CN and Locust bean gum, LBG) or mixed hydrocolloids and to select the best combination which had similar textural characteristics to those with regular-fat (~25% fat) control. In experiment 1, LFSs were formulated with each 1% hydrocolloid, smoked and cooked to an internal temperature of 71.7$^{\circ}C$. The pH range of LFSs was 6.29 to 6.34 and approximately 23~24% of fat was removed in the final products, resulting in the higher moisture and protein contents (%) in LFSs, as compared to regular-fat control. No differences (p>0.05) in cooking loss (CL, %), expressible moisture (EM, %), and hunter color values (L, a, b) were observed with the addition of each 1% hydrocolloid. However, LFSs containing 1% k-CN had textural hardness values similar to those with low-/regular-fat controls, whereas LFSs having either KF or LBG had similar cohesiveness values to those with regular-fat counterpart. Tn experiment 2, two or three mixed hydrocolloids were added to the low-fat sausage formulation. The addition of mixed KF+LBG (KLL) and KF+CN+LBG(KCL) reduced EM and textural hardness values, as compared to low-fat control. Among the treatments, LFSs containing two or three combinations of CN with KF or/and LBG had similar textural characteristics to those with regular-fat control. These results suggested that multiple addition of CN with other hydrocolloids (KF or LBG) for the replacement of fats in LFSs would be recommended for the proper functional and textural properties.

A nationwide analysis of mammalian biodiversity hotspots in South Korea (전국단위의 포유류 생물다양성우수지역 분석 연구)

  • Hotspots are top sites in terms of species diversity as the most threatened and most diverse sites which have been used to select priority areas for reserves. The purpose of this paper is to identify biodiversity hotspots through analyzing nationwide spatial patterns of species richness and rarity of Korean mammals. Four endangered mammals and eleven common mammals were selected as target species. Environmental variables as model input data were consisted of topography, distance, and vegetation structure etc. and Maxent was used to develop species distribution models for target species. Species richness and rarity were used as index of biodiversity. The results of this study were as follows. Firstly, hotspots of species richness for endangered mammals were in high elevation and steep mountain areas. However, species richness for whole mammals were high in low elevation of mountains. Secondly, distribution pattern of species rarity for endangered mammals were similar as richness. However, hotspots of species rarity for whole mammals were a little different from species richness. Species rarity was high in both low and high elevation of mountain areas. This study will provide the useful information for a biodiversity assessment, a habitat conservation, a national ecological network plan, and the management of protected areas.

Control Efficacy of Fungicides on Pepper Bacterial Wilt (고추 풋마름병에 대한 살균제의 방제 효과)

  • Control efficacy was investigated with fungicides as 3 copper compound, 3 antibiotic fungicides and one fungicide containing to quinolone against the growth of Ralstonia solanacearum on NA medium and the disease occurrence on pepper seedlings. Among 7 fungicides, oxytetracycline was shown the highest activity against a growth of the pathogen in the agar diffusion method, but validamycin showed no activity against the pathogen. With $1000{\mu}g\;mL^{-1}$ of each copper fungicide as copper hydroxide, copper oxychloride+ dithianone and copper sulfate, 2.2, 1.3 and 1.5 mm in size of clear zone only could be found, respectively. In pepper seedling test, oxytetracycline showed a perfect activity in all treatments 7 days after inoculation. However, its activity was decreased from $500{\mu}g\;mL^{-1}$ of treatment over the time. Copper fungicides showed the control efficacy lower than antibiotic fungicides except for validamycin. Based on the results, it was suggested that it would be better to use antibiotic fungicides than copper fungicides to control pepper bacterial wilt in the fields.

  • 우울증은 동기, 의욕, 관심, 주의력, 정신기능 및 식욕의 감소를 특징으로 하는 일종의 기분 장애이다. 우울증은 유전적, 내분비 및 환경적 스트레스를 포함한 다양한 원인에 의해 발생하지만 가벼운 우울증은 식이요법으로 개선되는 것으로 보고되었다. 따라서 우울증 환자를 치료하기 위해서는 기능성 및 영양 보충제를 포함한 다양한 식품 공급원이 필요하다. 치즈에는 숙주 건강에 유익한 영향을 미치는 생리 활성 펩타이드가 포함되어 있다. 특히 저지(Jersey) 우유는 홀스타인(Holstein) 우유보다 고형분 함량이 높은 것으로 보고되었다. 이 연구는 저지(Jersey) 와 홀스타인(Holstein) 우유의 가우다 치즈(Gouda cheese)가 만성 스트레스(CUMS, chronic unpredictable mild stress)에 미치는 영향을 조사했다. 치즈를 먹인 만성 스트레스 마우스 모델의 개선적 변화는 젖소 종에 관계없이 통계적으로 유의미하게 효과적으로 나타났다. 흥미롭게도 PCR을 통한 분변 미생물 균총 분석에서 저지 치즈를 섭취함으로써 Bacteroidetes가 증가하고 Firmicutes가 유의적으로 감소하는 것으로 나타났다. 종합하면, 본 연구는 치즈 섭취가 스트레스 개선 작용이 있음을 제시하며, 특히 장내 미생물 균총의 유익한 방향으로의 변화가 관찰되는데, 치즈의 생리활성물질 혹은 장내미생물 균총의 대사물질들이 이러한 행동·정신학적 개선 작용과의 연관성이 있음을 시사한다.

  • Two acardic acids showing antimicrobial activity against Propionibacterium acnes ATCC 6919, were isolated from Galla Rhois. They were identified as 6-pentadec-8-enyl salicylic acid and 6-pentadecyl aslicylic acid on the base of spectroscopic evidence.

On the Kinds of Ants(Hymenoptera: Formicidae) and Vertical Distribution in Jiri Mountain (지리산지역(智異山地域)의 개미의 종류(種類)와 수직분포(垂直分布))

  • Vertical distribution of ants were studied in Jiri mountain by collecting 15 times at 42 sites for 3 years since 1984. 31 species of 16 genera in 3 subfamilies were collected during the survey. The following 6 species, i.e., Formica japonica, F. lemani, Lasius niger, L. alienus, Pheidole fervida and Myrmica ruginodis were dorminant one, but 9 species such as Leptothorax congruus, Pheidole nodus, Aphanogaster japonica, Jollenhovia emeryi, Camponotus tokioensis, Polyrhachis lamellidens, Phagiolepsis mandjurica, myrmica yessensis and M. sulcinodis were rare species. The ant of Jiri mountain consisted of 16 species in Formicinae, 13 species in Myrmicinae and 2 species in Ponerinae. Ponerinae was distbriuted as far as 1,800m, but Myrmicinae and Formicinae to 1,915m. Only 8 species-Myrmica ruginosis, Myrmica sp., M. yessensis, M. sulcinodis, Leptothorax acevorum, Formica japonica, F. lemani and Camponotus japonicus-were collected at the submit of Jiri mountain, Cheonwangbong. The vertical distribution of C. atrox was ranged from 800m to 1,800m.

Altitudinal Vegetation Structure of Sunginbong in Ullungdo(Island) (울릉도 성인봉지역의 해발고별 식생구조)

  • Altitudinal changes in the vegetation of Sunginbong(984m) in UIlungdo (Island) were investigated by sample plots(forty l00$\m^2$) along elevation and analyzed by TWINSPAN and DCA techniques. In the results from the analysis of both techniques, altitudinal zonations were divided into 3 groups such as lower 300m area, 400~700m area, and above 800m area. DCA and similarity index analyses of elevational ranges showed discontinuities between lower and middle elevation areas. But there was a vegetation continuum between middle and high elevation areas. In the analysis of species diversity, there was no significant difference due to altitude except for maximum species diversity which was decreased with altitude. The mean tree density of canopy and understory layers in the middle elevation area showed the highest value, and the highest basal area was recorded at the lower elevation area.

Habenaria iyoensis (Orchidaceae): First Report for Korea (해오라비난초속(난과)의 미기록종: 애기방울난초(Harbenaria iyoensis))

  • Habenaria iyoensis Ohwi (Orchidaceae), previously known only from southern Japan and Taiwan, was found in the lowland of Jeju-do. This species is distinguished from the related taxa of Korean Habenaria by 3-nerved sepals, short-cylindrical spurs, and rosette leaves. The local name Ae-gi-bang-ul-nan-cho implies a smaller plant than H. flagellifera. The morphological characters and illustrations of the species are provided with photographs at the habitat.
