Differences in Free Amino Acids between Korean Ginsengs and Mountain Ginsengs (고려인삼과 장뇌삼의 유리 아미노산 비교)

  • Free amino acids were extracted and analyzed from Korean mountain ginsengs. Chinese mountain ginsengs and Korean white ginsengs by HPLC. The highest total free amino acid content was 12.46 mg/g in Korean white ginseg(P) and the lowest total free amino acid content was 6.86 mg/g in Korean mountain ginseng (Kㅡ6) The content of arginine in a Korean white ginseng(Y) was 8.77 mg/g Arginine was 77.80% of total free amino acids in a Korean mountin ginseng (KM2) The amount of histidine and methionine in Korean mountain ginsengs were higher than any other ginsengs. The highest amount of threonine and lysine were observed in Korean white ginseng and Chinese mountain ginseng respectively, The contents of glycine in Korean mountain ginseng and korean white ginseng were higher than Chinses mountain ginseng. There is no significant difference between two mountain ginsengs and Korean white ginseng.

Ginsenoside Compositions and Antioxidant Activity of Cultured and Mountain Ginseng (장뇌삼과 재배삼의 ginsenoside 함량과 항산화활성 추정)

  • This study was conducted to investigate the antioxidant activities and ginsenoside compositions of 4-year-old cultured ginseng roots (4CGR), 4-year-old mountain ginseng roots (4MGR) and leaves (4MGL), and 8-year-old mountain ginseng roots (MGR) and leaves (8MGL). Ginseng root and leaves were extracted with water and 80% ethanol. Crude saponin content of 4CGR was 3.85% (d.b.) and the contents of 4MGR, 4MGL, 8MGR and 8MGL were 6.75, 8.57, 6.53 and 7.54% (d.b.), respectively. 4CGR showed the highest content of ginsenoside-$Rh_1$ (6.07 mg/g), 4MGR showed the highest content of ginsenoside-$Rb_1$ (11.63 mg/g), 4MGL showed the highest content of ginsenoside-Re (24.35 mg/g), 8MGR showed the highest content of ginsenoside-$Rh_1$ (19.77 mg/g), and 8MGL showed the highest content of ginsenoside-Re (20.43 mg/g). Total antioxidant activity (AEAC) was ranged from 5.56 at 4MGR to 20.67 mg AA eq/g at 8MGL.

  • 현재 국내외의 인삼시장에 각종 인삼이 종래에 없든 대규모의 집산이 활발 하게 진행되고 있다. 그러나 기술 관리의 부당으로 각종 인삼에 대한 품종 감별이 제대로 되지않아 시장의 정상적 교역에 막대한 영향을 주고 있는 실정이다. 다년간 대량의 산삼, 장뇌삼, 재배인삼의 상품 경영에서 얻은 실지 경험과 각종 인삼 특성에 대한 관찰 연구 결과를 보고 한다.

Identification of Korean Mountain Cultivated Ginseng by RAPD (RAPD(Random Amplified Polymorphic DNA)를 이용한 장뇌삼의 지역별 품종 구분)

  • This study was conducted to examine the genetic variations and intraspecific relationships between 9 individuals of Panax ginseng C.A Meyer by using RAPD (Randomly Amplified Polymorphic DNA) analysis. The 34 primers out of 40 random primers were amplified for all tested plants. The 48 (40%) among 244 bands derived from 34 primers shown polymorphism, and the 72 (64%) rest of bands showed similar forms. By regional groups Sangju and Andong samples located in Kyungsang buk-do showed a high similarity. However, Punggi located in Kyungsang buk-do showed higher similarity with Jinan's of Junla buk-do. In this way, it did not show that Panax ginseng from the same area has similarities. In future study we need to more specific molecular phylogenetic analysis such as AFLP technology and gene sequencing with nuclear chloroplast DNA in all samples.

  • Mineral that is included to wild ginseng rhizomes that is assumed that is born at X-ray diffraction(XRD) analysis result 20 appears as is different according to each part by whewellite. Whewellite is accumulated much on rhizomes in wild ginseng, but do not exist hardly from mail body of root. The 6 years that in rhizomes of ginseng yet whewellite decision unobserved. This is considered by thing associated with ingredient of growth period of mineral and soil of plantation. For from, may present possibility of years distinction if analyze ginseng, wild ginseng etc. variously. Whewellite's chemical formula seems to be decision that combine with carbon(C) of air by photosynthesis function that Ca because is CaC$_2$O$_4$(H$_2$O) in rises by rhizomes through radices and is formed.

  • Differences in phenolic $\infty$mpounds were observed between cultured and mountain ginsengs. Cinnamic acid and p-hydroxy­b benzoic acid in Korean mountain ginseng and Chinese mountain ginseng were much higher than those in Korean ginseng. C Contents of the esculetin in Korean cultured ginseng and Korean mountain ginseng were higher than that in Chinese m mountain ginseng. The highest contents of esculetin in Korean mountain ginseng was$47.2\mu\textrm{g}/g$. Contents of the ferulic acid a and caffeic acid in red $\infty$lored Korean mountain ginseng were higher than any other ginseng.

Genetic Analysis of 5 Mountain Cultivated Ginseng and Wild Ginseng in Korea (국내 5개 지역의 장뇌삼과 산삼의 유전 분석)

  • ISSR PCR technique was applied to investigate genetic relationship among 5 Mountain cultivated ginseng populations (Jinan, Hongcheon, Punggi, Andong and Yeongju) and cDNA libraries of wild ginseng roots were constructed and analyzed functional genes related to morphogenesis via EST. Twenty four ISSR markers tested produced 127 polymorphic loci from 5 regional Mountain cultivated ginseng. Among the regional samples, Yeongju was made 18 polymorphic loci that were the highest level of variations among the cultivated regions. The range of similarity coefficient was 0.46~0.58 and the regional samples of Punggi and Hongcheon, Jinan and Andong were classified to similar groups respectively, whereas Yeongju was shown to be separate group with high level of genetic variation in UPGMA cluster analysis. As a result, there was no relationship according to geographical distance and genetic similarity. Eleven cDNA clones were consisted of 9 known genes and 2 unknown genes analyzed by BLAST program of NCBI. To recognize expression pattern of Homeodomain transcription factor related genes, Northern Blot analysis was performed for wild ginseng's leaf and root. As a result, the gene was only expressed by Mountain wild ginseng root.
