Comparison of Effective Constituents of Korean Paeony Roots(Paeoniae radix) Cultivated in Different Regions (한국산 작약의 산지별 유효성분 비교)

  • To compare the contents of well-known effective constituents, paeoniflorin, albiflorin and benzoic acid, of Korean paeony roots cultivated at different regions, this study was to provide the basic information for the quality evaluation of paeony roots. HPLC analysis showed that the paeoniflorin contents of white paeony roots of 3 years grown at Eui-seong gun, Kyungpook was the highest of 2.06% and red paeony roots of 3 years grown at Im-sil gun, Junpook was the lowest of 1.33%. The highest level of albiflorin contents was 2.04% of red paeony root grown at Ul-jin gun, Kyungpook, while the lowest levels were 0.05% of Eui-seong gun, Kyungpook and 0.06% of Im-shil gun, Junpook. The highest level of benzoic acid contents was 0.12% of red paony roots grown at Choung-do gun, Kyungpook and that grown at Ul-jin gun, Kyungpook was the lowest. Based on the average of three available constituents, the paeony roots cultivated at Kyungpook province was the highest contents of 2.03%, while 1.47% at Junpook province. These results suggest that Kyungpook province is the best region for the cultivation of white and red paeony roots, the combining of three available constituents would be more accurate than use of paeoniflorin only.

Antimicrobial Characteristics of Paeonia lactiflora Pall. Extract Tested against Food-putrefactive Microorganisms (작약추출물의 식품변패미생물에 대한 항균특성)

  • Paeonia lactiflora Pall. was extracted with water and the Paeonia lactiflora Pall. extract (PLE) was tested for antimicrobial activities against Corynebacterium xerosis, Candida albicans, Listeria monocytogenes and Pseudomonas syringae. PLE showed pronounced antimicrobial effects at concentrations at or above 50 ${\mu}g$/mL. The activities were stable at $100^{\circ}C$ and over the pH range of 3-11. PLE may serve as a natural antimicrobial agent in food preservation. It is suggested that hydrophillic components in the extract synergistically perturb microbial membrane functions.

Effect of Peeling Degrees and Drying Methods on the Quality of Paeonia lactiflora Pallas (작약근(芍藥根)의 박피정도(剝皮程度)와 건조방법(乾燥方法)에 따른 품질(品質) 특성(特性))

  • Peony is commonly used as roots that peeled and dried. But intact root contains higher medicinal constituents than peeled root (machine peeling). The yield of roots was also higher in intact roots. Use of intact root saved the labor time for peeling and 15% of root from yield loss by peeling. Paeoniflorin content was different with each zones of root. The content of Paeoniflorin was 5.11% in epidermis and 3.28% in cortex. When peeled peony roots were dried at hot air after briquet fire drying, root color was good for commercialization. But color of intact root was good at hot air drying without briquet fire drying. The required time for drying was longer in drying of intact roots than peeled roots drying.

Convergence Study on the Effects of Using Peony Drinking with Combined Exercise on Serum Immunoglobulin and DHEAs in Middle Aged Women (작약섭취와 복합운동 중재가 중년여성의 혈청 면역글로불린 및 DHEAs에 미치는 효과에 관한 융합 연구)

  • The purpose of this study was to investigate the changes of serum immunoblobulin and DHEAs when 8 weeks of combined exercise and peony intake have been held to women in their 40s and 50s. 15 participants were chosen, divided into three groups, and they had undergone combined exercise with the level of 1RM 60~70%, HRmax 60~70%, three times a week for 60 minutes each. For the proper intake, 6g of peony were warmed in a double boiler with water 107ml, and the crude liquid was ingested 100ml each by the participants, everyday before breakfast, for 8 weeks. The following conclusions were obtained through these objectives and procedures. First, the amount of immunoglobulin has increased in two groups, the 8-week combined exercise group and the group which undergone both the peony intake and the combined exercise, but did not show the significant difference(p>.05). Second, the results of DHEAs showed an increas in two groups, the 8-week combined exercise group and the group which undergone both the peony intake and the combined exercise, but did not show the significant difference(p>.05). Therefore, the consumption of peony and complex exercise in this study did not positively affect the immune function and dementia prevention.

The Improving Effect of Paeoniae Radix on Dextran Sulfate Sodium-induced Colitis in Mice (Dextran Sulfate Sodium으로 유도된 궤양성 대장염에 대한 작약의 개선 효과)

  • Paeoniae Radix has been used as a traditional medicine for various diseases including hepatic disease. However, the inhibitory effect of Paeoniae Radix on intestinal inflammation has not been fully understood yet. The aim of this study was to evaluate the effect of Paeoniae Radix on colitis induced by dextran sulfate sodium in mice. To investigate the protective effects of Paeoniae Radix, the colitis mice were induced by drinking water containing 5% dextran sulfate sodium for 7 days. Mice were randomized into groups receiving Paeoniae Radix (100 mg/kg), sulfasalazine (150 mg/kg) as a positive control, or water as a negative control. We evaluated the effects of Paeoniae Radix on clinical signs induced by dextran sulfate sodium, measuring weight loss, colon length, and disease activity index. Additionally, to find a possible explanation for the anti-inflammatory effects of Paeoniae Radix, we evaluated the effects of Paeoniae Radix on the interleukin-6 and cyclooxygenase-2 levels in colitis tissue. The results indicated that mice treated with dextran sulfate sodium showed measurable clinical signs, including weight loss and reduced colon length. However, Paeoniae Radix treatment significantly improved the weight loss and disease activity index as clinical symptoms. Moreover, Paeoniae Radix inhibited the interleukin-6 and cyclooxygenase-2 expression levels in colon tissues treated with dextran sulfate sodium. Taken together, the findings of this study suggest that Paeoniae Radix may be useful for treating intestinal inflammation, including ulcerative colitis.

Study on the Correlation between the Soil Properties and Albiflorin, Paeoniflorin Contents of Paeonia lactiflora Pall. (작약의 Albiflorin, Paeoniflorin 함량과 토양특성 간의 상관관계 연구)

  • The aim of this study was to investigate the correlation between soil properties and marker compounds contents of Paeonia lactiflora. The methods of determining marker compounds were validated by measuring the linearity, limit of detection (LOD), limit of quantification (LOQ), precision, accuracy and recovery using UPLC analysis. P. lactiflora contained albiflorin at 0.04 ± 0.00 ~ 2.79 ± 0.21%, paeoniflorin at 1.98 ± 0.14 ~ 6.67 ± 0.84%. The root dry weight (RDW) of P. lactiflora was 0.06 ± 0.02 ~ 1.27 ± 0.28 kg. The soil properties analysis such as soil pH, electric conductivity (EC), organic matter (OM), total nitrogen (TN), available phosphate (Avail. P2O5), exchangeable cation and cation exchange capacity (CEC) were performed following standard analysis manual. The results of correlation analysis between soil properties and growth characteristics, available P2O5 was positively coreelated with the RDW of P. lactiflora. On the other hand, the RDW of P. lactiflora showed significantly negative correlation with contents of albiflorin and paeoniflorin. The results of this study was might be help to provide useful information on the establish of standard cultivation by the investigate correlation analysis between growth characteristics and marker compound contents of P. lactiflora.

Analysis of Constituents Related to Five Sensory Test in Peony Root (작약근의 오관평가와 관련된 성분 검정)

  • Experiment was conducted to establish the standard of quality evaluation in Korean cultivated peony roots. free sugars, free amino acids, organic acids and aromatic components, which were generally considered to be information components of five sensory test in peony roots, were examined. As free sugars, sucrose, glucose and fructose were identified in peony roots. $\gamma-aminoisobuturic$ acid, arginine and other 16 kinds of free amino acids were found in peony roots. The major organic acids of peony roots were oxalic acid, citric acid and malic acid. Eugenol and other 10 aromatic components were identified in peony roots by GC/MSD.

Isolation and Determination of Paeoniflorin and Albiflorin in Korean Peony(Paeonia lactiflora Pall) Root (한국산(韓國産) 작약근(芍藥根) 함유(含有) Paeoniflorin 과 Albiflorin의 분리(分離) 및 분석(分析))

  • From Korean cultivated peony root, paeoniflorin and albiflorin, which are generally considered to be principal components of peony root, were isolated by silicagel column chromatography. Their structures were identified by spectroscopic methods $(UV,\;FT-IR,\;^1H{\cdot}^{13}C-NMR)$ and their purities were 98% and 93%, respectively. The concentrations of paeoniflorin and albiflorin in ten Korean cultivated peony lines were determined by reverse phase HPLC. The concentrations of paeoniflorin ranged from 1. 56 to 4.04% and those of albiflorin ranged from 0.04 to 1. 98% in ten Korean cultivated lines. In the ten cultivated lines, the concentrations of albiflorin in Punggi lines were higher than other lines.

Comparison on the Extract Content by Different Processing Method in Peony (Paeonia lactiflora Pall.) Root (작약 품종의 가공방법에 따른 엑스 함량 비교)

  • This experiment was conducted to establish the standard of quality evaluation in peony root (Paeonia lactiflora Pall.) cultivated in Korea. The contents of extract and changes of extract pH in peony root with different root ages, cultivars and drying method were investigated. The contents of extract and changes of extract pH in peony root with the removed and the unremoved cork layer showed no difference among different root ages. On the other hand, the contents of extract in the root with the unremoved cork layer which was two- to four-year-old, were higher by 3.7 to 9.2% than those in the root with removed cork layer. This suggests that cork layer might be a good source of extracts. The contents of extract in root of Youngchonjakyak in both the removed and the unremoved cork layer were 36% and 30%, respectively and were higher than of Euisungjakyak and Jomjakyak, but the extract pH was not significantly different among three cultivars which were four-year-old. It showed that the contents of extract and the changes of extract pH in peony root with the removed and the unremoved cork layer of Euisungjakyak, which being four-year-old, showed clear difference at various drying methods. Among the different drying methods, it showed that the contents of extract of that with unremoved cork layer in the room temperature drying method was 32.8%, and that of root with the removed cork layer in the $80^{\circ}C$ hot water treatment drying method was 28.1% which were the highest values, respectively. The pH of extract in freeze drying was the highest (about 5.1), and the $80^{\circ}C$ hot water treatment drying showed the lowest (about 3.7).
