Effects of Oral Intake of Gromwell Water Fraction on Ceramides Content and the Development of Atopic Dermatitis in NC/Nga Mice (자초 추출물 극성 성분의 피부 보습 증진 및 아토피 피부염 호전 효과)

  • Gromwell (LE, Lithospermum erythrorhizon), a perennial herbal plant, has been used for the treatment of various problems associated with atopic dermatitis of the skin, such as water loss, epidermal hyperproliferation, and severe inflammation. Previously, it was shown that oral supplementation with a 70% ethanol extract of gromwell prevented the development of atopic dermatitis in NC/Nga mice. In this study, in order to identify the fraction that mediates gromwell's efficacy, the dietary effects of water and ethyl acetate fractions from the ethanol extract of gromwell were assessed in the development of dermatitis using NC/Nga mice. Dietary supplementation of the hot water fraction significantly reduced scores for epidermal hyperproliferation in parallel with a marked increase of ceramides. Supplementation of the gromwell hot water fraction also decreased scratching behavior, which was accompanied by a decrease in plasma levels of IgE. These results showed that the hot water fraction of the gromwell ethanolic extract prevented the development of atopic dermatitis by increasing ceramides in NC/Nga mice.

Protective Effect of Mulberry and Lithospermum erythrorhizon Extracts on Anti-aging against Photodamage (오디 및 자초추출물의 피부 광노화 보호효과)

  • We investigated the protective effect of UVB inducing photodamage from mulberry extract (ME) and Lithospermum erythrorhizon extract (LE). The contents of total anthocyanin and shikonin as a color compound of ME and LE were 4.92 mg/g and 9.58 mg/g, respectively. The electron donating ability and superoxide radical scavenging activity of ME were 84.32% and 76.34%, respectively. The oxygen radical absorbance capacity of the ME ($545.37{\mu}moles$ TE/g) was higher than LE ($427.18{\mu}moles$ TE/g). MMP-1 production in the HS68 cells were exposed to UVB suppressed by treatment with $200{\mu}g/mL$ of ME (68.6%) and LE (32.7%). ME and LE were applied to a skin aging mouse model, which was induced by the irradiation of UVB to the backs of hairless mice. The value of skin erythema index, wrinkle depth and thickness, epidermis thickness, and collagenous fiber damage in the experiment groups (MEL: ME 3%, MEM: ME 5%, MEH: ME 7%, LEL: LE 3%, LEM: LE 5%, LEH: LE 7%) were remarkably reduced than in the control group (only UVB exposure group), while water capacity increased. The level of total wrinkles depth in the skin was decreased to be 30% of the control group by MEH and LEM. These results suggest that ME and LE are useful cosmetic materials for skin protection against UVB-inducing.

Isolation and Identification of Red Color Pigments from the Korean Lithospermum erythrorhizon (한국산 자초 적색소의 분리 및 확인)

  • Conditions of isolation for the red color pigments from the Korean Lithospermum erythrorhizon were investigated and identification of the red pigment was analysed. Non-polar solvents were more effective than water. Especially, 95% ethanol was observed as optimum solvent for the pigments extraction. About 20 minutes at 40$^{\circ}C$ with 95% ethanol was enough for the extraction of the red pigments. The major pigment was analysed as acetylshikonin by TLC, IR, NMR and GC/MS.

Protective Effect of Lithospermum erythrorhizon on Galactosamine Induced Liver Injury (자초(Lithospermum erythrorhizon)추출물의 투여가 Galactosamine으로 유도된 간손상에 미치는 보호 효과)

  • The present study was carried out to investigate the hepatoprotective effect of water extract of Lithospermum erythrorhizon on acute hepatotoxicity induced in Sprague-Dawley (SD) rats by a single dose of galactosamine (400 mg/kg, i.p). The animals were divided into four groups. The animals in the Con group were fed basal diet. GalN group were administered with galactosamine. LE200 and LE500 groups treated with water extract of Lithospermum erythrorhizon (such as 200 and 500 mg/kg/day, p.o) for 7 days before galactosamine injection. In the change of AST, ALT, ALP, GGT and LDH contents, as compared with GalN group, LE200 group were significantly decreased. According to the electron microscopical observation, liver cells were increased the lipid droplet, change of mitochondria in the GalN compared with LE200. These results suggest that administration of water extract of Lithospermum erythrorhizon suppress or retard galactosamine induced acute liver injury.

  • Gromwell colorants were extracted with methanol and dried. Four fractions were obtained by silica gel adsorption column chromatography using step-wise elution method. Relative ratio of four fraction is 1.00:0.07:0.22:0.30(Fl:F2:F3:F4) and gromwell colorants mainly consist of Fl, F3 and F4. IR analysis shows that each fraction has similar structure. Main component of gromwell extracts is acetyl derivative of naphthoquinone, and the rest are isobutyl derivative and isovaleryl derivative etc., in order. Gromwell colorants exhibit relatively good affinity to protein and polyamide fibers, but low affinity to cellulose and regenerated cellulose fibers.

A Study of Effectiveness of Lithospermum erythrorhizon Extracts on the Skin by UVB-Irradiation C57BL/6 Mouse (UVB로 조사된 C57BL/6 마우스의 피부에서 자초(Lithospermum erythrorhizon)추출물의 유효성에 관한 연구)

  • This study was intended to identify the effectiveness of Lithospermum erythrorhizon in the UVB-damaged mouse skin. The C57BL/6 mouse that weighted about 18 g were divided into three groups; the control group (A group), the UVB irradiated group (B group), and the group treated with Lithospermum erythrorhizon extracts after UVB irradiation (C group). In the results of superoxidase dismutant(SOD) activities, the C group was meaningfully lower at the 48hrs, 120 hrs, and 168 hrs group than B group (p<0.05). In the result of catalase(CAT) activities, the C group was decreased at the 120 hrs and 168 hrs group, but meaningless (p>0.05). In the result of light micrograph observation, the B group observed sunburn cells in the epidermis and acute inflammation in the dermis at the 24 hrs and 48 hrs. And proliferation of the epidermis and the stratum granulosum, and found the hyperkertosis at the 72 hrs, 120 hrs, and 168 hrs group than C group. The C group alleviated inflammation in the dermis than B group at the 24 hrs and 48 hrs. And inhibited the proliferation of the epidermis at the 72 hrs, 120 hrs, and 168 hrs groups than B group. In conclusion, Lithospermum erythrorhizon water extracts may protect the UVB-damaged mouse skin.

Analysis on Red-colored Dyeing by using Non-destructive UV-visible and Fluorescence Spectrophotometry (비파괴 자외-가시 및 형광 분광 분석법을 이용한 적색계 전통 염료 분석)

  • This study examined non-destructive UV-Vis spectrophotometry as well as 3-D fluorescence spectrophotometry of textile that made use of red dye such as Sappan wood, madder, Safflower, Gromwell. The authors produced two textile specimen that were dyed by not only two kinds of textile (cotton and silk) but also three kinds of mordanting (no-mordanting, alumen and iron), and they investigated effects of each dye material upon investigation results. At analysis with UV-Vis spectrophotometry of dyed textile specimen, dyeing made by sappan wood, madder and gromwell had significant difference depending upon mardant regardless of kinds of textile, and safflower had no significant difference depending upon textile and mordant. At analysis with 3D-fluorescence spectrophotometry, specimen dyed with sappan wood had difference with mordants, and with madder, there were difference with textiles, and safflower had inherent fluorescence spectrum regardless of textiles and mordants, while gromwell had no fluorescence spectrum.
