Single Dose Oral Toxicity Test of Water Extract of Corni Fructus in ICR Mice (ICR 마우스를 이용한 산수유 건피 추출물의 단회 경구투여 독성시험)

  • Herbal medicines are widely used as therapeutic products in many countries. Corni fructus (CF), the dried ripe sarcocarp of Cornus officinalis Sieb. et Zucc (Cornaceae), has been used for thousands of years in traditional medicine and has been reported to be effective for the prevention and treatment of various diseases, such as kidney diseases and diabetes. Recent research on CF has documented a wide spectrum of therapeutic properties, which include anti-inflammatory, ant-oxidative, immunomodulatory, and anti-cancer effects. However, there is no information on its safety. Therefore, in this study, the toxicity of water extract of CF to ICR mice was investigated. The mice received a single dose of water extract of CF (1,000, 2,000, and 5,000 mg/kg of body weight) via the oral route. Mortality, clinical signs, body weight changes, gross findings, and weights of the principal organs after 14 d were then assessed. The results revealed no adverse effects of CF as determined by clinical signs, body weights, or organ weights and no gross pathological findings in any of the treatment groups. These results suggest that the 50% lethal dose and approximated lethal dose of CF extract is over 5,000 mg/kg. The findings provide scientific evidence for the safety of CFs.

  • 우리나라 전역에 자생하는 소나무의 화분은 건강 식품의 하나로 당질, 단백질, 지질 중 PUFA와 식물성 sterol류, 무기질뿐만 아니라 자연의 항생물질, 효소, 호르몬, 성장물질 및 미지의 유효물질들을 다량 함유하고 있으며, 특히 강장작용, 심장병, 신경장애, 급만성전립선염, 동맥경화, 빈혈 및 간장해독 효과가 밝혀짐에 따라 송화가루에 대한 관심이 증대되고 있다 본 실험에서는 우리밀에 송화가루를 1%,2%,3% 첨가했을 때 제빵적성에 미치는 효과를 알아보기 위하여 반죽의 물성, 호화점도변화, 수분함량, 1차 발효손실률, 굽기 손실률, 색도, 부피 측정, 조직감, 노화도, 주사현미경 관찰, mold-free shelf-life 및 관능검사를 실시해서 최적 배합조건을 찾고자 하였다.

  • 원자력발전소는 지진과 같은 자연재해나 극한 운전조건에서 방사능물질이 외부로 누출되는 것을 방지하도록 설계 되어야한다. 따라서 이와 관련된 안전등급기기는 원전설비의 정상운전조건뿐 아니라 원전설계기준 사고조건(DBE, Design Basis Events)에서도 그 안전성 관련 기능이 검증되어야 한다. 본 연구에서는 국내 원자력 발전소의 다양한 환경조건을 만족하며 엄격한 기기검증요건에 따라 당사가 수행한 안전등급(Class 1E) 저압 유도전동기의 개발사례를 중심으로 방사능노출시험, 가속열노화해석 및 시험, 내진해석 및 시험으로 구성되는 기기검증의 절차와 방법을 제시하였다.

태양광 발전용 축전지의 연구 개발 현황

  • 태양광발전 시스템에서 축전지는 일조시간에 태양 전지에 의해 충전된 전력을 일몰 후와 같이 태양전지로 부터의 출력이 없을때나 흐린날, 우천시와 같이 태양 전지의 출력이 부족할 때 방전하여 부하에 전력을 공급하게 된다. 이와같이 축전지는 태양 광발전 시스템이라는 특별한 용도에 사용되기 때문에 일반적인 용도와는 달리 매우 가혹한 사용 조건하에서 가동된다. 즉, 축전지의 충전량은 기후 변화에 의해서 좌우되므로 과충전, 부족충전, 과방전이 진행되면 축전지의 노화 즉, 용량 감소는 급격히 일어나게 된다. 물론 최적의 충전 상태를 유지하기 위해서 충전기를 개량하거나, 부조일을 감안하면 축전지의 용량을 설계하거나, 디젤 보조발전기를 사용하여 일사량이 적은 경우에 충전 전기량을 보충하는 등 여러가지 개선책이 있지만, 불규칙적이며 가혹한 사용조건은 자연 현상에 의한 것이므로 근본적인 해결책은 될 수가 없다.

Comparison of aging fingerprint enhancement by pre-treatment of various initiator powders in the cyanoacrylate fuming method (다양한 개시제(initiator)분말 전처리(pre-treatment)를 통한 노화된 지문의 Cyanoacrylate Fuming법 현출 증강 비교)

  • We tested the pre-treatment of alumina, leucine, alumina mixed with leucine at ratios of 9:1 (Al-Leu (9:1)) and 5:1 (Al-Leu (5:1)), and alumina mixed with sodium lactate at a ratio of 5:1 (Al-Lac (5:1)) on aged fingerprints (1, 7, 14, and 30 days after natural fingerprinting) before cyanoacrylate (CA) fuming to improve the development efficacy. As a result of the experiment, fingerprints with pre-treatment of alumina, leucine, Al-Leu (9:1), Al-Leu (5:1), and Al-Lac (5:1) showed better development efficacy (area and minutiae) than fingerprints without pre-treatment. Therefore, this modified CA fuming method improved the development efficacy with five pretreatments.

Effect of Combined Environmental Factors on Degradation Behavior of Carbon Fiber/Epoxy Composites (복합적인 환경인자의 영향에 따른 탄소섬유/에폭시 복합재의 열화 특성)

  • Thermal analysis properties and chemical structure of carbon fiber/epoxy composites under environmental exposure were examined using an accelerated aging tester which can simulate real weather conditions such as temperature, moisture and ultraviolet. The composite specimens were exposed to combined environmental factors up to 3000 hours. Thermal analysis properties and chemical structure of the composites were evaluated with various exposure times through Modulated DSC and FTIR. According to the results of Modulated DSC, the glass transition temperature increased as exposure time increased due to the formation of network structures in the composites. Also endotherm peaks of enthalpy relaxation related to physical aging that can affect the properties of the composites were observed as exposure time increased. From the results of FTIR, it was found that the location of the peaks was little affected by exposure time, but the intensity of the peaks slightly decreased as exposure time increased due to the curing reaction in the epoxy group.

The Role of ROS-NF-κB Signaling Pathway in Enhancement of Inflammatory Response by Particulate Matter 2.5 in Lipopolysaccharide-stimulated RAW 264.7 Macrophages (RAW 264.7 대식세포에서 지질 다당류에 의한 미세먼지(PM2.5) 유발 염증 반응 증진에 미치는 ROS-NF-κB 신호 전달 경로의 역할)

  • The purpose of this study was to investigate whether the inflammatory response in lipopolysaccharide (LPS)-treated RAW 264.7 macrophages could be promoted by particulate matter 2.5 (PM2.5) stimulation. To this end, the levels of inflammatory parameters, reactive oxygen species (ROS) and inflammation-regulating genes were investigated in RAW 264.7 cells treated with PM2.5 in the presence or absence of LPS. Our results showed that the production levels of pro-inflammatory mediators (nitric oxide and prostaglandin E2) and cytokines (interleukin-6 and -1β) were significantly increased by PM2.5 stimulation in LPS-treated RAW 264.7 cells, which was correlated with increased expression genes involved in their production. In addition, when LPS-treated RAW 264.7 cells were exposed to PM2.5, nuclear factor-kappaB (NF-κB) expression was further increased in the nucleus, and the expression of inhibitor of NF-κB as well as NF-κB in the cytoplasm was decreased. These results suggest that the co-treatment of PM2.5 and LPS further increases the activation of the NF-κB signaling pathway compared to each treatment alone, thereby contributing to the promotion of transcriptional activity of inflammatory genes. Furthermore, although the generation of ROS was greatly increased by PM2.5 in LPS-treated RAW 264.7 cells, the NF-κB inhibitor did not reduce the generation of ROS. In addition, when the generation of ROS was artificially suppressed, the production of inflammatory mediators and the activation of NF-κB were both abolished. Therefore, our results suggest that the increase in the NF-κB-mediated inflammatory response induced by PM2.5 in LPS-treated RAW 264.7 macrophages was a ROS generation-dependent phenomenon.

Effects of Leaf and Pod Removal on Photosynthesis and Assimilate Partition in Soybean (적엽ㆍ제협처리가 콩의 광합성과 동화물질 배분에 미치는 영향)

  • To clarify the effects of sink demand for assimilate on leaf photosynthetic rate, tissue composition, and leaf senescence of soybean [Glycine max (L.) Merr.] J plants, pod and leaf tissues were removed at growth stage $R^3$. Plant responses were measured every 10days from 2 through 42days following treatment. Leaves of depodded plants exhibited increased starch and chlorophyll contents and specific leaf weight. Stomatal resistance was also increased and leaf photosynthetic rate was reduced. Dry weight of vegetative tissues except leaves was increased by pod removal. Leaf removal resulted in a decreased starch content of leaves from 22 to 42days after treatment and that of roots at all sampling times. Specific leaf weight was decreased while leaf photosynthetic rate was increased. Stomatal resistance and chlorophyll content were little affected. Weight per seed was decreased 3.0% by leaf removal. Except for the seed, tissue protein contents were increased by pod removal but decreased by leaf removal, however, seed protein content was not affected by either. Apparent senescence was delayed by depodding. Both apparent and functional senescence were accelerated by leaf removal.

Analysis and Verification of Ancient DNA (고대 DNA의 분석과 검증)

  • The analysis of ancient DNA (aDNA) has become increasingly considerable anthropological, archaeological, biological and public interest. Although this approach is complicated by the natural damage and exogenous contamination of a DNA, archaeologists and biologists have attempted to understand issues such as human evolutionary history, migration and social organization, funeral custom and disease, and even evolutionary phylogeny of extinct animals. Polymerase chain reaction(PCR) is powerful technique that analyzes DNA sequences from a little extract of an ancient specimen. However, deamination and fragmentation are common molecular damages of aDNA and cause enzymatic inhibition in PCR for DNA amplification. Besides, the deamination of a cytosine residue yielded an uracil residue in the ancient template, and results in the misincorporation of an adenine residue in PCR. This promotes a consistent substitution (cytosine thymine, guanine adenine) to original nucleotide sequences. Contamination with exogenous DNA is a major problem in aDNA analysis, and causes oversight as erroneous conclusion. This report represents serious problems that DNA modification and contamination are the main issues in result validation of aDNA analysis. Now, we introduce several criterions suggested to authenticate reliance of aDNA analysis by many researchers in this field.

New Functional Properties of Passion Fruit Extract on Skin (패션 프룻 추출물이 피부에 미치는 새로운 기능적 효과)

  • In this research, the anti-inflammatory, antioxidant, antiaging, and skin whitening properties of pulp and seed extracts of passion fruit were studied. The result of the primary skin irritation test using a skin-attached patch determined the skin irritation index to be 0.00 for the passion fruit extract. In addition, RAW 264.7 macrophages produce NO by stimulation of lipopolysaccharides, and the application of extracts to this resulted in significantly lower NOs, confirming the excellent anti-inflammatory properties of passion fruit extracts. The 2,2-diphenyl-1-picrylhydrazyl test further confirmed that the passion fruit extract exhibits a good 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonate radical scavenging ability of 5.11% and strong antioxidant properties. The presence of collagen type I in the skin is a measure of aging and various skin diseases. The results obtained from the analysis of the activity of human procollagen I alpha 1 confirmed that the passion fruit extract reduces the synthesis of procollagen. In addition, the skin whitening property of the passion fruit extract was confirmed by the melanin inhibition test, and a sample was obtained that contained more than 2% of arbutin, a whitening agent approved by the Ministry of Food and Drug Safety, which is generally present in the form of a white powder and is used as a functional ingredient. This confirms that the whitening efficacy of the passion fruit extract obtained from nature contributes to the development of functional raw materials for cosmetics and food.
