  • For the development of a sporeless strain of P. ostreatus we used sporeless strain ASI 2069. We have recovered both nuclear types of strain ASI 2069 as monokaryons of nh9, nh15, nh26 and nh36 (here after referred to as neohaplonts) by protoplasting the mycelium. Crosses between neohaplonts and SSI's(single spore isolates) obtained from a sporulating commercial strain ASI 2180. Five excellent strains are selected from 30 bred strains by quality of fruitbodies and spore number. To development of molecular markers linked to sporeless strain of P. ostreatus, we are screened helicase, recombinase(DMC1) and topoisomerase(Spo11) genes related meiosis by PCR and sequencing. Among three genes, Spo11 gene was identified into molecular marker of sporeless from neohaplonts and bred strains of P. ostreatus.

  • The Green Infrastructure Framework refers to an interconnected network formed by greenways that links gardens, parks, green spaces, streams, wetlands, agricultural lands, and green belts. Green infrastructure supports diverse functions to environment, provides various benefits to people, and helps in the community's health and viability. It can store stormwater runoff and abate its non-point source pollutants. Due to its advantages and profits, advanced countries in environment policies have adopted green infrastructure in planning and implementing urban and regional development. The Korean government and municipalities have focused upon grey infrastructure investment in the past, which causes occurrence of natural disasters such as draught, flood, and landslides, degradation of water and air quality, decline of biodiversity, and even inhibition of economic activities. In order to alleviate these problems, it is requested to formulate and implement policies for green infrastructure at the national government level. USA and Korean situation of green infrastructure were investigated; forty components of green infrastructure were drawn. Nine policies utilized in the USA cases were identified, which are applicable to Korea. Among them, five policies can be implemented in public sector and four in private one. The green infrastructure law needed in Korea was suggested. The amendments of laws regarding green infrastructure and alternatives expending it were proposed.

  • This study was to investigate the sanitary microbiological aspects of Cheonho Reservoir and its antibiotic resistance in the natural environment to contribute to the public healthcare improvement. Groups of coliforms, were counted at three sites of the Cheonho Reservoir in July and September 2013, and the isolates were identified according to the water pollution process test standards. Antibiotic resistance was evaluated using the Korea National Institute of Health's standard antibiotic susceptibility test. The average coliform counts in Cheonho Reservoir were $2.0{\times}10^3CFU/mL$, 41% of which were identified as Escherichia coli(E. coli). Antibiotic resistance rate in E. coli isolated from Cheonho reservoir was highest in Ampicillin 31.3%, Ticarcillin 25.0%, Cefaxolin and Cefoxitin 18.8% respectively. Convergence multiple resistance patterns, 38,5% presented resistance to 4 durgs, 6 drugs resistance were 7.7%. The more than 2 drugs resistance were 92.3%. Cheonho Reservoir is a public park that accessible to the citizens, further convergence studies are needed to develop sanitary microbiological management practices and study antibiotic resistance of the reservoir.

  • Zoysiagrasses are important turf plants used for school playgrounds, parks, golf courses, and sports fields. The two most popular zoysiagrass species are Zoysia japonica and Zoysia sinica. These are widely distributed across different growing zones and are morphologically distinguishable from each other; however, it is phenotypically difficult to differentiate those that grow along the coastal line from those in beach area habitats. A combination of morphological and molecular approaches is desirable to efficiently identify these two plant cultivars. In this study, we used a rapid identification system based on DNA barcoding of the nrDNA-internal transcribed spacer (ITS) regions. The nrDNA-ITS regions of ITS1, 5.8S nrDNA, and ITS2 from Z. japonica, Z. sinica, Agrostis stolonifera, and Poa pratensis were DNA barcoded to classify these grasses according to their molecular identities. The nrDNA-ITS sequences of these species were found at 686 bp, 687 bp, 683 bp, and 681 bp, respectively. The size of ITS1 ranged from 248 to 249 bp, while ITS2 ranged from 270 to 274 bp. The 5.8S coding region ranged from 163 - 164bp. Between Z. japonica and Z. sinica, nineteen (2.8%) nucleotide sites were variable, and the G+C content of the ITS region ranged from 55.4 to 63.3%. Substitutions and insert/deletion (indel) sites in the nrDNA-ITS sequence of Z. japonica and Z. sinica were converted to cleaved amplified polymorphic sequence (CAPS) markers, and applied to the Zoysia grasses sampled to verify the presence of these markers. Among the 62 control and collected grass samples, we classified three groups: 36 Z. japonica, 22 Z. sinica, and 4 Z. japonica/Z. sinica hybrids. Morphological classification revealed only two groups; Z. japonica and Z. sinica. Our results suggest that used of the nrDNA-ITS barcode region and CAPS markers can be used to distinguish between Z. japonica and Z. sinica at the species level.

  • A new baekji(Angelica dahurica Bentham et Hooker) variety, Baekji l, was developed through a pure line selection at the Gyeongbug Provincial RDA during the period of 1990 to 1995. The variety was characterized to have high plant height. Long root length and large root diameter as compared with a check varicty of Bonghwa baekji but emergence date, flowering date and leaf number of Baekji l was similar to that of the check variety, and it was also more tolerant to nematode and heat stress. Peeled root color of Backji l was yellowbrown but number of the lateral root of the variety was greater. The dried-root yield of Baekji l in yield trial, regional adaptation trial and farmer's field trial was always $21\sim31%$ higher than that of the check variety.
