To improve Bacillus strains producing biopolymer, conditions for protoplast formation and regeneration were investigated in biopolymer producing Bacillus subtilis K-1 and lactose utilizing Bacillus coagulans. Bacillus subtilis K-1 mutant (SM-2) and Bacillus coagulans mutants (CM-12) were marked auxotrophic and antibiotics-resistant (SM-2) and an antibiotics-resistant mutants, respectively. To formate protoplasts derived from the mutants, conditions were established as follows. For B. subtilis mutant SM-2, its culture in mid-logarithmic phase was added with penicillin G (1.0 unit/ml) and further reacted for 1.5 hr. Cells were collected and then treated in lysis fluid (pH 7.0) containing 0.4 M sucrose and lysozyme $25\;{\mu}g/ml$ for 40 min at $37^{\circ}$. Protoplast formation was very successful (99.6%) and the ratio of cell wall regeneration was 2.4%. For Bacillus coagulans mutant CM-12, its mid-logarithmic phase culture was treated with penicillin G (0.3 unit/ml) and glycine (0.5%) for 1hr. Cells were collected and then resuspended in lysis buffer (pH 7.0) containing 0.6 M lactose and lysozyme $(300\;{\mu}g/ml)$ for 30 min at $37^{\circ}$. Protoplast formation was also successful (90.8%) and cell wall regeneration ratio was similar to SM-2 (2.2%). To improve regeneration frequency, regeneration medium was obtained as followed condition,. Cell wall regeneration was improved 2-4 folds with 5.1% for B. subtilis SM-2 and 10.3% for B. coagulans CM-12 when protoplasts mixed with soft top agar(0.4%) was overlaid onto trypticase soy broth medium containing 0.4 M sucrose, 0.7% casamino acid, 1% PVP, 25 mM $MgCl_2,\;25\;mM\;CaCl_₂$ and 1.5% agar.

한반도 국가바람지도(김현구, 2009)는 한국에너지기술연구원에서 지식경제부의 부처임무사업으로 구축되었으며 현재 웹서비스(http://www.kier-wind.org)를 통하여 정보를 제공하고 있다. 국가바람지도는 수치기상예측(NWP; Numerical Weather Prediction) 모델을 이용하여 영토, 영해에 대해 $1km{\times}1km$의 고해상도로 작성한 뒤(이순환 등, 2009) 풍력자원 정보로 재가공되었다. 한반도 국가바람지도는 5년의 장기간에 대한 시계열 수치기상예측에 의하여 구축되었기 때문에 데이터베이스(DB; database)의 효율적 관리가 필연적으로 요구된다. MM5 또는 WRF 모델의 고유 출력포맷의 자료구조는 풍력자원분석에 필요한 기상요소 외에도 대기과학자에게 필요한 수많은 기상인자를 종합적으로 포함하고 있다. 따라서 2차원 층(layer) 또는 3차원 공간분포 분석 및 계산격자인 셀(cell)에서의 1차원 시계열 분석 등 다양한 자료축출에는 비효율적인 자료구조가 된다. 이러한 자료구조의 불편을 해소하기 위해서는 기상요소별로 독립적이고 빈번한 시계열 자료 추출에 효율성을 가지며 어떤 프로그래밍 언어를 사용하든지 직관적으로 쉽게 사용할 수 있는 바람지도 데이터베이스의 재구성이 요구된다. 이에 대용량 수치자료의 처리 측면에서 장점을 가지는 과학기술 프로그래밍 언어인 IDL을 기반으로 국가바람지도의 자료구조를 효율화하여 데이터베이스화 하였으며 IDL에 내재된 그래픽 기능을 활용하여 가시화를 구현함으로써 연구개발자의 입장에서 국가바람지도의 활용성 및 효율성을 향상시키고자 하였다.

To achieve safe and high-speed navigation of a mobile service robot, velocity of dynamic obstacles should be considered while planning the trajectory of a mobile robot. Trajectory planning schemes without considering the velocity of the dynamic obstacles may collide due to the relative velocities or dynamic constraints. However, the general planning schemes that considers the dynamic obstacle velocities requires long computational times. This paper proposes a velocity control scheme by scaling the time step of trajectory to deal with dynamic obstacle avoidance problem using the RNLVO (Robot Nonlinear Velocity Obstacles). The RNLVO computes the collision conditions on the basis of the NLVO (Nonlinear Velocity Obstacles). The simulation results show that the proposed scheme can deal with collision state in a short period time. Furthermore, the RNLVO computes the collisions using the trajectory of the robot. As a result, accurate prediction of the moving obstacles trajectory does not required.

본 논문에서는 제작한 가상현실 게임의 소개와 적용기술에 대해여 논한다. HTC VIVE를 이용하여 강력한 주요 게임 개발엔진 중 하나인 Unreal engine4를 이용하여 쉽게 접근 할 수 있는 VR 리듬게임 구현을 목적으로 한다. 플랫폼의 한계의 벗어나 HUD로 좀 더 현실감과 몰입을 요구하는 게임개발을 목표로 리듬감 증진 혹은 운동효과도 기대할 수 있다. 사운드 플러그를 이용하여 주파수별 스펙트럼을 시각화하여 재생되는 음원중 스펙트럼 값이 조건을 만족하면, 노트가 자동으로 생성되고 일정 시간이 경과한 후 사라지거나, 플레이어가 타격하여 점수를 획득 하는 방식으로 진행되며, 플레이어가 노트를 맞을 경우 체력값이 단계별로 낮아져 게임이 종료된다. 더 많은 곡과 맵을 추가하여 흥미소요를 늘릴 수 있으며, 단순타격이 아닌 조건이나 임무를 부여 다양성과 복잡함을 추가해 낮은 접근성과 높은 정복성을 가질 수 있다.
This paper reviews the engineering approach needed to support humans during their long-term missions in space. This approach includes closed plant production systems under microgravity or low pressure, mass recycling, air revitalization, water purification, waste management, elimination of trace contaminants, lighting, and nutrient delivery systems in controlled ecological life support system (CELSS). Requirements of crops f3r space use are high production, edibility, digestibility, many culinary uses, capability of automation, short stems, and high transpiration. Low pressure on Mars is considered to be a major obstacle for the design of greenhouses fer crop production. However interest in Mars inflatable greenhouse applicable to planetary surface has increased. Structure, internal pressure, material, method of lighting, and shielding are principal design parameters for the inflatable greenhouse. The inflatable greenhouse operating at low pressure can reduce the structural mass and atmosphere leakage rate. Plants growing at reduced pressure show an increasing transpiration rates and a high water loss. Vapor pressure increases as moisture is added to the air through transpiration or evaporation from leaks in the hydroponic system. Fluctuations in vapor pressure will significantly influence total pressure in a closed system. Thus hydroponic systems should be as tight as possible to reduce the quantity of water that evaporates from leaks. And the environmental control system to maintain high relative humidity at low pressure should be developed. The essence of technologies associated with CELSS can support human lift even at extremely harsh conditions such as in deserts, polar regions, and under the ocean on Earth as well as in space.

Solar powered aircraft are becoming more and more interesting for future long endurance missions at hight altitudes, because they could provide surveillance, earth monitoring, telecommunications, etc. without any atmospheric pollution and hopefully in the near future with competitive costs compared with satellites. However, traditional aircraft sizing methods currently employed in the conceptual design phase are not immediately applicable to solar powered aircraft. Hence, energy balance and constraint analyses were performed to determine how various power system components effect the sizing of a solar powered long endurance aircraft. The primary power system components considered in this study were photovoltaic (PV) modules for power generation and regenerative fuel cells for energy storage. To verify current research results, these new sizing methods were applied to HALE aircraft and results were presented.
To improve biopolymer productivity and properties of Bacillus strains, protoplast fusion was performed between biopolymer producing Bacillus subtilis K-1 and lactose utilizing Bacillus coagulans. The results were as follows; Protoplasts mixed in fusion fluid containing 33% PEG 6000, 1% PVP and 10 mM $CaCl_2$ were reacted for 5 min at $37^{\circ}C$ and then centrifused protoplasts were directly overlaid on the selective media containing $100\;{\mu}g/ml$ antibiotics and incubated for 3 days. At this conditions, the frequency of protoplast fusion was generally in the range of $4.6{\times}10^{-5}\;to\;1.8{\times}10^{-7}$ in ratio. Segregation ratio was observed between 1 to 6% indicating genetic stability of all the fusants. Fusants growth were also observed on the media contained amino acid and antibiotics as required marked materials. DNA contents of the selected fusants were 1.6 to 2.7 times more than that of parent strains. With observation by TEM microscopy, spherical protoplasts were first released from the swollen parental cells and then contracted to fuse in the process of fusion. And fused cells were observed representative vesicle. Originally, the parental cells were observed as in the morphology of thick-walled and double membrane-surrounded rod shape with TEM microscopy.
