A Study on Multiplication Response of "Tricholoma matsutake" (Pine Mushroom) Conidio to Cultural Media Environment (송이균(松茸菌) (Tricholoma matsutake)의 배양환경(培養環境)에 대한 증식반응(增殖反應)에 관한 연구(硏究))

  • This study was conducted to examine the physiology of pine mushroom mycelia cultured with various media for artificial culture of pine mushroom. The results obtained were as follows: 1) Among the various media, the medium composed of honey, boiled pine mushroom and soil extract fluid, fibrous root extract fluid, dry yeast, $KH_2PO_4$ inositol, folic acid, and biotin was the best for the growth of pine mushroom mycelium. 2) The optimum temperature for germinating pine mushroom spore and for culturing pine mushroom mycelium, was $24^{\circ}C$ and the optimum pH was 4.5. 3) There was no significant difference in growth between the mycelium separated from the tissue of pine mushroom sporophore and that separated from the spore. 4) No noticeable effect was found on the growth if such salts as $ZnSO_4$, $MnSO_4$, $MgSO_4$, $CaCl_2$ and ferric citrate were added to the Hamada's medium. 5) The addition of fibrous root extract promoted the growth of pine mushroom mycelium. 6) As a carbon source of artificial media, honey was more effective than glucose. 7) The culture infiltration of Mortierlla growing often in Fairy Ring was good for the growth of mycelium compared with the control. 8) The addition of fibrous root extract, inositol, biotin, and folic acid to artificial culture media was greatly effective in growth. When the temperature was lowered $19^{\circ}C$ after mycelium has appeared, the formation of primordium was observed.

  • Wasong(Orostachys japonicus A. Berger) has been used as anti-cancer medicinal plants from ancient time. This experiment was conducted to obtain some fundamental informations concerning anatomical study, germination test, soil analysis of indigenious districks, adaptaion of soil for wasong. The leaf of wasong was xerotype with spine, mesophyl chloroplast is dense surrounded vascular bundle, with a many large water storage tissue without chloroplast, and was bifacial stomata leaf type. The stem of wasong was amphicribral vascular bundle, central cylinder was cylinder type and protostele. Root was polarch type, potoxylem of it's was arranged in ring shape of annulation and metaxylem. The structure of flower was cymose, was caylyx of 5 sepals, corolla of 5 petals, roecium of 10 stames, ovules of five, placentation parietal, ovary superior, axile placenta. The optimum temperature for seed germination was $25^{\circ}C$ under light and germination percentage was 22.5%. Dormancy breaking was effective for 6-8 days at 5$^{\circ}C$ and at 100 ppm level of $GA_3$ The soil analysis of wasong local districks, content of available $P_2O_5$, Ca were higer than optimal level of upland and C.E.C., exchangeable k was not significantly different of that, but exchangeable magnesium was very low. The growth of wasong was affected draining regardless soil combination treatment.

  • 비고사리과(Lindsaeaceae) 버들일엽[Laxqgramme saliciblia (Makino) Makino]은 잎이 녹색의 단엽으로 버들처럼 보여 관상가치가 높아 조경소재로 이용되는 착생식물이다. 또한 식물구계학적 특정식물종 V급, 적색자료집 VU급, 국외반출 승인대상종에 수록된 식물로 생태학적 보전이 요구되며, 개체수도 적은 편으로 알려져 있다. 본 연구는 보전이 요구되는 버들일엽의 전엽체 증식 및 포자체 형성을 위한 기내 외 번식조건을 구명하고자 수행되었다. 서귀포시 상효동 일대에서 식물을 획득하였으며, 기내에서 포자를 발아시켜 획득한 전엽체를 8주 간격으로 계대배양하면서 실험재료를 확보하였다. 전엽체 증식에 적합한 배양배지를 선발하고자, 전엽체 300mg을 다진 후 배지종류와 농도를 달리한 배지(Knop, 1/4, 1/2 및 1MS)에 8주간 배양하였다. 배양실은 온도 $25{\pm}1.0^{\circ}C$, 광도 $30{\pm}1.0{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$(16/8h)로 조절되었다. 연구의 결과, 1/2MS배지에서 전엽체의 생체중이 5.8g으로 가장 많이 증가하였으며, 형태형성의 발달도 주걱형의 전엽체로 정상적으로 발달하였다. 기외 포자체 형성에 적합한 토양조건을 확인하고자, 인공토양(원예상토, 피트모스, 펄라이트 및 마사토)의 비율을 달리하여 5종류의 혼합토양을 조성하였다. 준비된 토양을 사각분($7.5{\times}7.5{\times}7.5cm$)에 충진하고, 전엽체 1g을 10초간 분쇄한 다음 토양표면에 분주하여 16주간 재배하였다. 재배환경은 온도 $25{\pm}1.0^{\circ}C$, 광도 $43{\pm}2.0{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$(16/8h) 및 습도 $72{\pm}2.0%$로 조절되었다. 연구의 결과, 원예상토가 많은 비율로 혼합된 처리에서 포자체의 형성이 확인되었다. 원예상토와 펄라이트를 2:1(v:v)로 혼용한 토양, 원예상토와 마사토를 2:1(v:v)로 혼용한 토양, 원예상토 단용한 토양에서 각 28.5, 23.0, 22.3개로 조사되었다.

A Study on the Collection and Utilization of Myanmar Plants as Garden Plants (정원소재로서의 미얀마 유용 및 관상식물 수집·활용에 관한 연구)

  • Studies on the preservation and propagation of tropical and subtropical plants are required, but the studies are insufficient in Korea. Since 2013, the Tropical Plant Resources Research Center has been collaborating on field research and publication to preserve plant diversity in Myanmar protected areas and Sagging areas in cooperation with Ministry of Natural Resources and Environment Conservation and the Myanmar Forest Research Institute. This study is to provide information on cooperative projects. As a result of the joint research between the Korea National Arboretum and Myanmar, in December 2018, the Korea National Arboretum Tropical Plant Resources Research Center is on display for conservation and education purposes with the 65 Myanmar useful materials and specimens of 326 kinds of plants and specimens that Myanmar Forest Research Institute donated. This donation is the first case of Myanmar's export of plant organisms to a foreign country, and as for the Korea National Arboretum, it is the second official case of introducing a mass plant from foreign governments. The study will also accelerate preservation biodiversity and propagation methods of tropical and subtropical plants as well as research on biodiversity conservation and industrial use.

Assessment of New Algicide Thiazolidinedione (TD49) for the Control of Marine Red Tide Organisms (해양적조생물제어를 위한 살조물질 Thiazolidinedione 유도체(TD49) 평가)

  • Worldwide development of harmful algal blooms causes serious problem for public health and fisheries industries. To evaluate the algicidal impact on the harmful algae bloom species in aquatic ecosystems of coast, a new algicide thiazolidinedione derivative (TD49) were tentatively examined in the growth stages (i.e., lag, logarithmic and stationary phase) of rapidophyceae $Heterosigma$ $akashiwo$, $Chattonella$ $marina$ and $Chattonella$ sp..Three strains could easily destroy in the lag phase due to relatively weak cell walls than those of the logarithmic and stationary phase. It is thought that inoculation of TD49 substances into initial or developmental natural blooms with a threshold concentration ($2{\mu}M$) can maximize the algicidal activity. Also, bio-chemical assays revealed that the algicidal substances from all culture strains were likely to be extracellular substances because those cells have easily destroyed in cell walls. On the other hand, natural zooplankton communities were influenced within the exposure experiments of $2{\mu}M$, which is showed the maximum algcidal activity of tested organisms. These results indicate that although the TD49 substance is potential agents for the control of $H.$ $akashiwo$, $C.$ $marina$ and $Chattonella$ sp. in the enclosed eutrophic bay and coastal water, more detailed research of acute toxicity effect on high trophic organism in marine ecosystems need to be conducted.

In Vitro Culture of Entomopathogenic Nematode with Its Symbiont for Biopesticide (생물살충제를 위한 곤충병원선충 및 공생박테리아의 in vitro 배양)

  • An in vitro culture method for entomopathogenic nematode Steinernema glaseri was developed. A symbiotic bacterium was isolated from Steinernema glaseri and identified as Xenorhabdus nematophilus. Phase variation that differed in some biochemical characteristics of symbiotic bacterium was observed. Entomopathogenic nematodes carried only phase I bacterium in their guts. Phase I bacterium could be converted into phase II form in in vitro culture medium consisting of 5% yeast extract, 0.5% NaCl, 0.05% $K_2HPO_4$, $0.02% MgSO_4$.$7H_2O$. The optimum temperature for bacterial growth was $28^{\circ}C$. The pH of the culture medium increased up to 9.0-9.5 during the exponential growth period of the culture, regardless of initial pH 6-7. Various culture media such as chicken offal, dog food, bovine liver, peanut, and so on were tested for in vitro culture of the nematodes. The best medium for Steinernema glaseri production was obtained from concentrated homogenate of bovine liver and the nematode growth was highest at 80% bovine liver. In the co-culture of entomopathogenic nematode with its symbiont, the growth rate of nematodes was 2 times faster than that without its symbiont and the nematode concentration reached about $5.5\times10^4$/mL within 15 days.

Multiple Shoot Induction from Radicle-derived Callus and in Vitro Propagation of Silene Acaulis Subsp. Arctics (극지식물 Silene acaulis subsp. arctica의 유근 유래 캘러스로부터 다신초 유도와 기내 증식)

  • We describe here an efficient in vitro propagation method of Silene acaulis subsp. arctica (Caryophyllaceae), one of the higher arctic angiosperms, through the multiple shoot regeneration after callus induction from the radicle. The seeds of S. acaulis subsp. arctica collected from Svalbard, the Norwegian Arctic, were germinated and calli were induced from the radicle on solid MS media supplemented with 0.25mg/L 2,4-D and 1mg/L $GA_3$ at both $10{\pm}1^{\circ}C\;and\;23{\pm}1^{\circ}C$ Two weeks after callus induction, the multiple shoots were efficiently regenerated on the MS media supplemented with 0.25 g/L BA and 0.05mg/L HPh. The total biomass increment of regenerated shoots increased most efficiently of S. acaulis subsp. afctica was showed the maximum efficiency in at $23{\pm}1^{\circ}C$ on 1/2 MS salt strength. The multiple regenerated plantlets of S. acaulis subsp. arctics were grown to normal plants on soil.

  • Dogs were divided into 3 groups of two each; Bacillie Calmette-Guerin(BCG) pretreatment, M bovis only treatment and uninfected control group. BCG were vaccinated intradermally with 0.2ml before 3weeks of M bovis intraperitoneal infection. Infection at necropsy 4months later was readily in the both treated dogs. Histopathologically, the BCG pretreated dogs produce the moderate accumulation of macrophages and focal granuloma formation in the lung, whereas the M bovis only treared dogs produce the accumulation of predominantly macrophages, occasionaly polymorphonuclear cells and the more larger granuloma Bronchoalveolar lavage(BAL) was obtained and total and differential cell counts were examined. Total number of BAL cells harvested from uninfected dogs is lower compared with those of the both treated groups. The total cell number of M bovis only treated dogs were singificantly higher 1.8 times than that of the BCG pretreated dogs. The Fe receptor activity and the growth of organism in alveolar macrophages obtained from BCG pretreated dogs were compared with that in macrophages from M bovis only treated dogs. BCG vaccination resulted in substantial macrophage activation, measured as increased Fc receptor mediated phagocytosis and rosette formation, as wells as the inhibition of intracellular mycobacteria multiplication. However, actibated macrophages taken from BCG pretreated dogs are incapable of killing the M bovis. Thus, these results suggest that BCG pretrearment in the dog may produce a protective effect against tuberculosis because active alveolar macrophages have acquired antituberculous immunity, although few mycobacteria within the lung remain in a metabolically active state.

Spatio-temporal Variation of Mesozooplankton in Asan Bay (아산만 해역 중형동물플랑크톤의 시공간적 변동)

  • Previous studies on zooplankton in Asan Bay were mostly based on samples collected seasonally with three months intervals. Present study was aimed to know the temporal variation of meso-zooplankton distribution using the data collected monthly. Relationships between zooplankton abundances and environmental factors such as seawater temperatures, salinities and chlorophyll-a contents were also studied. Seawater temperature showed typical pattern of seasonal variation found in temperate waters. The fluctuations of environmental factors ranged relatively wider In the inner part of the bay than those in outer part of the bay. Salinity was very low right after the summer rainy period due to the sporadic outflow of freshwater from the adjacent artificial lakes. Sudden changes in salinity seemed to have significant impact on zooplankton assemblages. Chlorophyll-a contents were increased in general when compared with previous reports probably due to the recent human exploitations in the coastal zone, which might enhance the nutrients level . The timing and duration of spring bloom showed geographical differences. In the inner part of the bay it began earlier (February) and last longer (three months) while in the outer part of the bay it began late (April) and last just one month. Zooplankton abundance, especially most abundant taxon Acartia hongi, showed weak but significant positive correlation with chlorophyll-a contents. The difference in temporal variation found with two different sampling intervals indicated the necessity of shorter time interval samplings.

Conditions for Artificial Culture of Lemna Paucicostata and Potentiality as an Alternative Biomass Source (바이오매스 자원으로서의 Lemna Paucicostata의 인공배양조건과 이화학적 특성에 관한 연구)

  • Conditions for artificial culture of Lemna Paucicostata and its nutritional values were examined in this study. Lemna P. was cultured using artificial wastewater and a bioreactor (total volume $2,630\;cm^3$, working volume $2,240\;cm^3$) was operated at conditions of 6,250 lux and $28^{\circ}C$. Water flow affected the growth of Lemna P.: growth rate was very high (more than $1.1\;d^{-1}$) at a condition of no-water movement, but it was very low (less than $0.15\;d^{-1}$) when water moved slowly. The growth of Lemna P. was higher in $16h\;d^{-1}$ light cycle than in Sand $24h\;d^{-1}$, and it was also severely affected by the initial $NH_4$-N levels of wastewater. The growth rate of Lemna P. was high in lower $NH_4$-N level, indicating that the growth rate is in inverse proportion to $NH_4$-N concentration in wastewater. However, the contents of crude protein (CP) of Lemna P. were proportional to the initial $NH_4$-N concentration. The CP contents of Lemna P. cultured at 2, 10, 50 and 100 $NH_4$-N mg $L^{-1}$ was 18, 24, 37, 43%, respectively, showing the Lemna P. cultured at 50 and $100\;mg\;L^{-1}$ had similar protein contents to linseed (CP 35%), cottonseed (CP 38%) and soybean (CP 45%). Fat, protein, fiber, NDF and ADF contents of Lemna P. harvested at conditions of $16h\;d^{-1}$ light cycle and less than $2\;mg\;L^{-1}$ of $NH_4$-N level was 2.8, 18, 27, 20, 41 and 65.7%, respectively. Since the growth rate of Lemna P. was very high (more than $1.1\;d^{-1}$) at those conditions, it was convinced that mass production of valuable protein and fiber sources are feasible. In particular, since the Lemna P. has unsaturated fatty acids found mainly in animal fat as well as beneficial fatty acids to health such as C18:ln9c, C18:2n6c, C20:5n3 and C22:2, the Lemna P. biomass would be a highly valuable alternative feed source to grains.
