인공피부 개발을 위한 생채 적합성 지지체에 관한 연구

  • Chitosan scaffold is widely applied to drug delivery and tissue engineering. We have developed chitosan scaffolds, with various pore size, by differing freezing temperature and duration of ultraviolet (UV) irradiation, for reconstructing skin equivalent. Chitosan scaffold was coated with type I collagen, fibronectin and basic fibroblast growth factor (bFGF) in various combinations and concentrations, to evaluate the effect of extracellular matrix (ECM) and bFGF on cell adhesion, growth and differentiation of dermal fibroblasts. Human dermal fibroblasts, isolated from newborn foreskin and passaged between 3 and 5, were seeded on the top of scaffolds and cultivated for 2 weeks. We examined the morphology and the secretion of ECM of fibroblasts using scanning electron microsopy (SEM) and histochemistry. A stellate morphology of fibroblasts were seen in all groups. The scaffold coated with either type I collagen and bFGF or type I collagen and fibronectin, however, showed the best condtion of dermal fibroblasts, in that the highest cell number and ECM secretion were seen. On the contrary, scaffolds coated with all three factors, type I collagen, bFGF and fibronectin, showed lower number of cells and ECM secretion than scaffolds with two factors. There was a tendency of dose-dependence in all three factors for fibroblast growth and ECM secretion. In conclusion, we may suggest that chitosan scaffold coated with either type I collagen/bFGF or type I collagen/fibronectin could provide more favorable environment for the growth and differentiation of dermal fibroblasts.

Development of Dermal Transduction Epidermal Growth Factor (EGF) Using A Skin Penetrating Functional Peptide (피부투과 기능성 펩타이드를 이용한 경피투과성 상피세포성장인자의 개발)

  • The epidermal growth factor (EGF) has a intrinsic function of inducing growth and proliferation of cells through interacting with cell membrane receptors in human epidermis and dermis layer. These functions of EGF are used as a main ingredient for wound healing medicines and anti-aging cosmetics. As a cosmetic ingredient, the EGF has a problem in exhibiting its natural efficacy due to the lack of the ability to penetrate through the stratum corneum, which is known as the skin barrier. In this study, a recombinant human epidermal growth factor ($MTD_{151}-EGF$) fused with the macromolecule transduction domain $(MTD)_{151}$ with the skin penetration ability was developed to improve the skin penetration efficiency of the EGF. Expression of $MTD_{151}-EGF$ was performed in E. coli transformed with a vector encoding the $MTD_{151}-EGF$ gene and then purified. The purified $MTD_{151}-EGF$ was evaluated using cell proliferation assay, cytotoxicity test and skin penetration test by franz diffusion cell assay and artificial skin. Cell proliferation activity of $MTD_{151}-EGF$ purified to high purity of 99% or above was equivalent to the EGF or better, and cytotoxicity was not observed. In addition, the $MTD_{151}-EGF$ showed an excellent penetration efficiency compared to the EGF in the skin penetration test with EGF and $MTD_{151}-EGF$ labeled by FITC in an artificial skin penetration model. Based on the quantitative analysis of the penetrating substance using franz diffusion cell assay, the amount of penetration was about 16 times more than that of EGF. These results can be regarded as an effective alternative to improve the existing physical transdermal penetration method related to the use of various active ingredients for cosmetics.

Skin Hydration Effect of Jeju Lava Sea Water (제주용암해수의 피부 보습 효과 연구)

  • Many minerals and nutrient salts are abundant in Jeju lava sea water. The objective of this study was to evaluate the skin hydration effects of Jeju lava sea water. The skin barrier serves as a protective barrier that prevents the loss of moisture. The water holding capacity and water transport of the epidermis have been proposed to be important determinants of skin hydration. Jeju lava sea water increased the mRNA expression of filaggrin and caspase-14 which is related to natural moisturizing factor (NMF) formation. Aquaporins 3 (AQP3) are proteins that facilitate the transport of water across cell membranes. Jeju lava sea water increased the mRNA expression and protein expression of AQP3. We employed a skin equivalent model to assess the efficacy of Jeju lava sea water. In a skin equivalent model, Jeju lava sea water increased the CD44 (hyaluronic acid receptor) which is related to skin hydration. From these results, we found out Jeju lava sea water maybe help to skin hydration.

Evaluating the Micronucleus Induction Potential for the Genotoxicity Assay Using the Human Skin Model, KeraSkinTM (인공피부모델 KeraSkinTM을 이용한 유전독성 평가)

  • Micronucleus test is genotoxicity assay for detection of micronuclei in the cytoplasm of interphase cells. The reduction and replacement of in vivo toxicity testing on animals require the development of in vitro models to predict the genotoxicity or other tests for cosmetic products. In this study, we evaluated a genotoxicity assay for topically applied chemicals using a three-dimensional human reconstructed skin model, KeraSkin$^{TM}$. Two genotoxins, mitomycin C (MMC) and methyl methanesulfonate (MMS), induced significant dose-related increases in cytotoxicity and micronuclei induction in the skin model. In contrast, two non-genotoxins, 4-nitrophenol (4-NP) and trichloroethylene (TCE), induced cytotoxicity but not micronucleus formation. In conclusion, micronucleus test using human skin model may be useful for predicting in vitro genotoxic potentials of cosmetic products.

  • Deficiency of collagen regeneration, denaturalization of fibers, increased melanin synthesis and reactive oxygen species are important factors inducing deterioration of healthy skin function. They induce freckles and decrease in skin elasticity. Vitamin C, vitamin A and their derivatives have been used as cosmetic ingredients for improvement of these problems but they have various problems. We have been developing the various derivatives of these ingredients. In this study, we investigated whitening effect of 3-o-cetyl-L-ascorbic acid (VCCE), a new vitamin C derivative. The VCCE inhibited melanogenesis in a dose dependent manner(44 % at $20\;{\mu}g/mL $) and tyrosinase expression. For 8 weeks, we also investigated skin brightening effect of VCCE on pigmented spots in UV-irradiated human skin. In results, VCCE showed a statistically significant skin whitening effect by mechanical and visual evaluation. Taken together, our findings suggest that the VCCE has potential benefits as an active ingredient for whitening cosmetics.

A case of congenital neurocutaneous melanosis (선천성 신경피부멜라닌증 1례)

  • Neurocutaneous melanosis is a rare congenital syndrome characterized by the presence of large or multiple congenital melanocytic nevi and benign pigment cell tumors of the leptomeninges. Neurocutaneous melanosis is thought to represent an error in the morphogenesis of embryonal neuroectoderm. We experienced a neonate who presented with giant, dark colored pigmented nevi covering chest, abdomen, neck and arms, with satellite lesions. Magnetic resonance image showed a nodular hyperintense lesion in the amygdala of the right temporal lobe, and T1-weighted images showed hyperintensities in the adjacent leptomeninges. We report a rare case of neurocutaneous melanosis with a brief review of related literature.

Antimicrobial activities and skin barrier improvement effect of Eruca sativa extract (루꼴라(Eruca sativa) 추출물의 항균활성과 피부장벽 개선 효과)

  • Eruca sativa is a rocket plant and a member of the Brassicaceae, which is considered to be an important chemo-preventive plant family. Although Eruca sativa has positive biological effects, the effect of Eruca sativa extract (ES) on improvement of skin barrier function has not been reported. In this study, we investigated the applicability of functional materials by examining a variety of physiological activities of Eruca sativa extract. ES showed anti-microbial activities against Bacillus subtilis, Escherichia coli, and Candida albicans. In particular, antimicrobial activities of ES against B. subtilis was the highest. Additionally, immunohistochemical analysis of protein marker related to keratinocyte differentiation was determined. The treatment by ES (50 mg/L) showed a significant increase of involucrin expression compared with treatment by 0.1% DMSO as a control in skin equivalents, the ES-treated group showed similar level in the expression of involucrin compared to the group treated with the same concentration of WY14643 in $EpiDerm^{TM}$, a three-dimensional model of skin equivalents. These results indicate that ES promotes the expression of protein related to barrier properties of the skin. Therefore, ES may be an effective ingredient for skin barrier improvement.

  • The skin of fishes is also the important external barrier against a variety of the environmental detrimental agents, allowing normal internal physiological function, so its condition is very important in many disease processes. Since fishes are aquatic, morphologically and physiologically they posses specific characteristics lacking in terrestial animals. Thus, it is necessary to keep in mind various specific histologic characteristics for a precise interpretation of the histopathological lesions. In the normal environment the fish skin is more subtly adapted for its physiological requirements as a limiting barrier than the skin of higher terrestial vertebrates. However, its delicacy makes it high vulnerable to damage in fish culture facilities or polluted waters. Although changes in the skin of fish such as discoloration, ulcerative lesions, are the the most readily observed clinical features of fish, there is a wide variety of possible responses, which are best considered in terms of their site within the skin. Up to now, there are relatively few sytematically described information on the histological structure of the teleost skin as well as integrated reviews of the range of pathological processes that can occur in teleost skin. Therefore it was tried in this article to rearrange the information already described by other investigators on the histological structure and histopathological changes of teleost skin.

Histopathogenic Characteristics of Haemorrhagic Ulcer in Cultivated Snakehead Channa argus Artificially Infected with Aeromonas veronii (Aeromonas veronii 인공감염에 의한 양식 가물치 궤양증의 병리조직학적 특성)

  • Aeromonas veronii was isolated from the haemorrhagic ulcer of the snakehead that had been infected in natural condition, This bacterium was injected hypodermically into the healthy snakeheads and the effect was compared to the naturally infected fish. Both groups showed severe necrosis, falling off of epidermal tissue and hypodermal muscle. In both groups, severe histophathological changes were observed in gill, digestive tract and kidney just before death. Artificially injected fish showed necrosis of tissue in skin, gill and digestive tract from 2 days after injection. Then it showed necrosis or cell atrophy of tissue in kidney from 5 days after injection, and in liver and spleen just before death. Snakehead infected with haemorrhagic ulcer died within 9 days after infection, showing the symptom of skin damage and metabolic inhibition in respiration" digestion, excretion, etc. It was concluded that Aeromonas veronii (CA26) that was isolated from the naturally infected fish is the main bacterium causing haemorragic ulcer in the snakehead.

Effects of Provitamin D on Skin Pigmentation and Immunity (프로 비타민 D 의 피부색 조절 및 면역 효능)

  • Vitamin D is a fat-soluble vitamin that is mainly produced in the skin by UV rays. Along with melatonin, it is a representative chronobiotic substance, and the skin plays an important role in distinguishing between day and night. However, vitamin D cannot be used directly in cosmetics because it is a vitamin that acts as a coenzyme and plays a hormonal role in regulating the expression of various types of genes. Therefore, it was to investigate the skin efficacy of provitamin D (7-dehydrocholesterol), a vitamin D precursor that can be used in cosmetics. Our findings reveal that pro vitamin D can effectively inhibit the expression of tyrosinase, the melanin-producing enzyme, thereby attenuating melanin synthesis. This skin tone regulatory effect has been corroborated in vitro using artificial skin models. Additionally, pro vitamin D demonstrated anti-inflammatory properties by suppressing the expression of TNFa and, upon conversion to vitamin D through UV exposure, it was observed to induce the production of the antimicrobial peptide CAMP (LL-37). The inhibitory effect of pro vitamin D on melanin production appears to be a result of it reducing the UV absorption capacity of melanin, thereby inducing the conversion of pro D to vitamin D. Utilizing pro vitamin D in cosmetic formulations could not only modulate skin tone and enhance skin immunity but also expect to contribute to other cutaneous benefits as anti-aging and barrier function improvement with everyday UV exposure. This multifaceted efficacy positions pro vitamin D as a promising ingredient in advancing the formulation of skin care products.
