Studies on the Pathogenicity of Aphelenchoides sp. and Rhabditis sp. attacking Cultivated Mushroom, Agaricus bisporus(Lange) Sing (Aphelenchoides sp.와 Rhabditis sp.의 양송이에 대한 병원성에 관한 연구)

  • Four species of nematodes attacking mushroom beds were found in samples taken from 35 mushroom farms throughout Korea. These were Rhabditis sp., Aphelenchoides sp., Ditylenchus sp. and Aphelenchus sp. ,Rhabditis sp. was found from compost and casing from all mushroom farms and the frequencies of Aphelenchoides sp. was 31.4% in the both compost and casing. Both Ditylenchus sp. and Aphelenchus sp. showed 2.7% of frequencies in the compost, none in casing. Temperature and moisture content of compost affected pathogenicity of Aphelenchoides sp. on mushroom mycelia grown in compost. The higher temperature and moisture content the sooner the damage became apparent, and the more rapid was subsequent destruction of mycelia. There was no mycelial destruction at the lowest temperature of $10^{\circ}C$. Rhabditis sp. completely disintegrated mycelia grown in the compost, in the early stage, the numbers of Rhabditis sp. rose gradually and then increased suddenly to reach a peak but soon declined. At first, the pH of Rhabditis-infested spawned compost declined but then rose gradually as mycelia was disintegrated by nematodes. The trend in pH of infested unspawned compost was similar to those of uninfested, unspawned compost. Cultures inoculated with surface-disinfected dead Rhabditis sp. and with tap water used in the nematode extraction procedures showed no mycelial injury associated microorganisms containing within or outside the nematodes even though added by artificial wounding of the mycelia. Cultures artificially wounded showed no injury away from the wounds without the presence of living Rhabditis sp., such wounded mycelia slowly regenerated. On the other hand, artificial wounding accelerated the breakdown of mycelia in the presence of living Rhabditis sp.

Critical Temperature for Early Marginal Transplanting of Japonica Rice in Korea (우리나라 자포니카 벼 품종의 조기이앙 한계온도 분석)

  • We investigated critical temperature for early marginal transplanting (CT-EMT) of the contemporary japonica rice varieties in Korea through the field, pot seedling tray, and the phytotron experiments during 2020 to 2023. The lowest mean temperature for 10 days from transplanting (MT-10DFT) that resulted in earlier heading date was 12.4℃ and the highest MT-10DFT that did not show the earlier heading date was 12.0℃ in the field study when the MT-10DFT varied by changing transplanting date. The lowest MT-10DFT that induced the increased biomass but not the earlier heading date was 11.6℃ and the highest MT-10DFT that showed neither the increased biomass nor the earlier heading date was 11.4℃. Compared to the 10-day later transplanting, the dates of the first root development, initiation of the chlorophyll recovery, and the first tiller development were earlier when the MT-10DFT was 9.1℃ or higher, 10.5℃ or higher, and 11.6℃ or higher, respectively, in the pot seedling tray and field experiments. The earliness of the first tiller development was a practical index for the estimation of CT-EMT during the early growth stage of rice. The response of transplanted rice to temperature treatments with the diurnal change of 10℃ in the phytotron study was similar to that shown in the field study. The data shown for constant temperature without a diurnal change revealed that the extent of positive effects of high temperature at day-time was greater than the extent of negative effects of low temperature at night-time on the early growth of transplanted rice. It was concluded that the critical MT-10DFT for early marginal transplanting of japonica rice in the temperate environments was between 11.4 to 11.6℃ based on the plant growth and between 12.0 to 12.4℃ based on the plant development.

Manufacturing Method and Characteristics of the Dongrok(copper chloride) pigments (동록(염화동) 안료의 제조방법 및 특성에 관한 연구)

  • Hayeob pigment is known as one of the traditional dark green pigments, but the color, raw material, and manufacturing method have not been clearly identified. However, comparing the analysis results of the particle shape and constituent minerals of Hayeob pigments revealed through pigment analysis studies of colored cultural properties such as Dancheong, Gwaebul, and paintings, Hayeob pigments appear to be the same as Dongrok pigments produced by salt corrosion. Therefore, in order to restore Hayeob pigment, the manufacturing method of Dongrok pigment was studied based on the records of old literature. The Dongrok pigment manufacturing method confirmed in the old literature records is a natural corrosion method in which copper powder and a caustic are mixed and then left in a humid condition to corrode. Based on this, artificial corrosion using a corrosion tester was adopted to corrode the copper powder more efficiently, and an appropriate mixing ratio was selected by analyzing the state of corrosion products according to the mixing ratio of the caustic agent. In addition, the manufacturing method of Dongrok pigment was established by adding a salt removal process to remove residual caustic agents and a purification process to increase chroma during pigment coloring. The prepared Dongrok pigments have a bluish green or green color, show an elliptical particle shape and a form in which small particles are aggregated, and a porous surface is observed. The main constituent elements are copper(Cu) and chlorine(Cl), and the main constituent mineral is identified as atacamite [Cu2Cl(OH)3]. As a result of an accelerated weathering test to evaluate the stability of the prepared Dongrok pigments, it was found that the greenness partially decreased and the yellowness significantly increased as deterioration progressed. Before deterioration, the Dongrok pigments had lower yellowness compared to the Hayeob pigments of the old Dancheong, but after deterioration, yellowness increased significantly, and it was found to have a similar chromaticity range as Dancheong's Hayeob pigments. As a result, the prepared Dongrok pigments were confirmed to be similar to Dancheong's Hayeob pigments in terms of color as well as particle shape and constituent minerals.

Studies on the Artificial Cultivation of Morchella esculenta in Ascomycetes (자낭균 곰보(Morchella esculenta)버섯의 인공재배에 관한 연구)

  • This study was executed in an attempt to investigate a artificial requisites of fruitbody occurrence. Environmental requirements on habitat for fruitbody occurrence of collected cultures resulted in leading to $13-16^{\circ}C$ and 75% relative humidity, and requiring silt loam of soil texture which had more nutritional substances than a dry field. Optimal temperature was $25^{\circ}C$, medium PDB, and pH 5.0 in cultural conditions. Mannose required of 5% in ASI 59002, 59003, 59004, but 3% in ASI 59001 was selected as optimum carbon source. The substrates stimulating sclerotium formation were cotton waste, or cotton waste + oak sawdust (mixture ratio of 8:2), which had 20% additive of wheat barn respectively. Sclerotium was formed well in the substrate adjusted chemical properties by applying 2% of calcium sulfate. Sclerotium formation was the most effective in the treatment of peat moss + oak sawdust (mixture ratio of 5:5) + 30% of wheat barn.

Effect of Unexpected Foaming Incident on Nitrogen Removal in a Vertical Subsurface Wetland (수직지하 흐름형 습지에서 거품발생이 질소제거에 미치는 영향)

  • A lab-scale vertical flow subsurface (VFS) wetland composed of three parallel columns with polypropylene synthetic fiber as main substrate was operated. Piggery stormwater diluted from swine excreta was fed to the wetland on the basis of three different hydraulic regimes or hydraulic retention time (HRT) of 2, 4, and 8 days with daily internal recirculation. Then, monitoring of common water quality parameters was carried out. Unexpectedly, an increase of effluent COD concentration accompanying the appearance of foams was observed during a distinct period in the wetland with HRT 2, 4, and 8 days, successively. Subsequently, a series of experiments was conducted to investigate the origin of the foams. Foams and the increase of COD concentration were found to be induced by the release of organic matter from the synthetic polypropylene fiber which was fed with piggery stormwater. Meanwhile, nitrogen removal was found to be enhanced during a period which overlapped the distinct foaming period signifying that foaming played two important functions in biological nitrogen removal. Foams which form rapidly and then burst easily could hold up and then release oxygen for nitrification. Foams which contain organic surfactants could serve as carbon sources for denitrification as well. Hence, nitrogen removal was enhanced during the foaming stage. After that, COD concentration decreased slowly to a level prior to the foaming stage, and nitrogen removal efficiency declined as well.

  • Polydeoxyribonucleotide (PDRN) is known to accelerate wound healing process by acting on specific adenosine receptors. Differently from the PDRN substances obtained so far from piscine tissues, unique PDRN was prepared recently from Porphyra, a seaweed species. In this study, effects of the Porphyra PDRN was evaluated by inducing artificial skin wounds in Nile tilapia Oreochromis niloticus. The wound recovered almost completely in 2 weeks without any treatment, however only a partial recovery was done after one week. Facilitated healing was observed by PDRN fed for 5 days before wound induction at 10 or 20 mg/kg, against the partially recovered 1-week old wound. The healing effects were identified not only by visual observation, wound area measurement, but also by colorimetric estimations. These novel wound healing effects suggest beneficial therapeutic utility of Porphyra-derived PDRN for wound treatments in fishes occurring from various causes.

The Prediction of Durability Performance for Chloride Ingress in Fly Ash Concrete by Artificial Neural Network Algorithm (인공 신경망 알고리즘을 활용한 플라이애시 콘크리트의 염해 내구성능 예측)

  • In this study, RCPTs (Rapid Chloride Penetration Test) were performed for fly ash concrete with curing age of 4 ~ 6 years. The concrete mixtures were prepared with 3 levels of water to binder ratio (0.37, 0.42, and 0.47) and 2 levels of substitution ratio of fly ash (0 and 30%), and the improved passed charges of chloride ion behavior were quantitatively analyzed. Additionally, the results were trained through the univariate time series models consisted of GRU (Gated Recurrent Unit) algorithm and those from the models were evaluated. As the result of the RCPT, fly ash concrete showed the reduced passed charges with period and an more improved resistance to chloride penetration than OPC concrete. At the final evaluation period (6 years), fly ash concrete showed 'Very low' grade in all W/B (water to binder) ratio, however OPC concrete showed 'Moderate' grade in the condition with the highest W/B ratio (0.47). The adopted algorithm of GRU for this study can analyze time series data and has the advantage like operation efficiency. The deep learning model with 4 hidden layers was designed, and it provided a reasonable prediction results of passed charge. The deep learning model from this study has a limitation of single consideration of a univariate time series characteristic, but it is in the developing process of providing various characteristics of concrete like strength and diffusion coefficient through additional studies.

The Heading Response of Field-grown Rice Varieties of Different Heading Ecotypes in Korea (벼 품종의 출수생태 특성에 따른 포장 출수 반응 분석)

  • The heading response of rice varieties is determined by the combined effects of their basic vegetative phase (BVP), photoperiod-sensitivity (PS), and thermo-sensitivity (TS). This study was performed to analyze the effect of differences among heading ecotypes of rice on their heading response in the field. To do this, we investigated the heading response of 15 varieties grown under different day-length and temperature conditions in a phytotron. The characteristics of heading ecotypes were compared to the heading responses in the field-grown rice varieties that were used in the phytotron experiments. The ranges of these characteristics across the 15 varieties were 18 - 35 days for BVP, 12 - 61 days for PS, and 12 - 35 days for TS. In comparisons of coupled varieties that differed only in a single trait among BVP, PS, and TS, the variety with the longer BVP and that with the higher PS had a longer period from sowing to heading (DTH), while the variety with the higher TS had a shorter DTH. The comparative magnitude of DTH in the coupled varieties followed the PS trait when BVP and PS were involved, the BVP trait when BVP and TS were involved, and the PS trait when PS and TS were involved in the heading response of field-grown rice. When comparing the coupled varieties with different traits of the three examined factors, the heading response was consistent with the PS trait. The DTH in all 15 varieties was significantly correlated with the PS, and the DTH in 9 selected varieties with a relatively small PS was correlated with BVP. The reduced DTH of rice in the July 10 seeding treatment compared to those in the June 25 and May 11 seeding treatments was positively associated with PS, but not with TS. We concluded that the heading response of rice varieties under natural conditions in Korea is mainly governed by the PS trait and is also greatly affected by the BVP trait when the PS is small.

Lodging-Tolerant, High Yield, Mechanized-Harvest Adaptable and Small Seed Soybean Cultivar 'Aram' for Soy-sprout (내도복 다수성 기계수확 적응 소립 나물용 콩 '아람')

  • 'Aram' is a soybean cultivar developed for soy-sprout. It was developed from the crossing of 'Bosug' (Glycine max IT213209) and 'Camp' (G. max IT267356) cultivars in 2007. F1 plants and F2 population were developed in 2009 and 2010. A promising line was selected in the F5 generation in 2011 using the pedigree method and it was evaluated for agronomic traits, yield, and soy-sprouts characteristics in a preliminary yield trial (PYT) in 2012 and an advanced yield trial (AYT) in 2013. Agronomic traits and yield were stable between 2014 and 2016 in the regional yield trial (RYT) in four regions (Suwon, Naju, Dalseong, and Jeju). Morphological characteristics of 'Aram' are as follows: determinate plant type, purple flowers, grey pubescence, yellow pods, and small, yellow, and spherical seeds (9.9 g 100-seeds-1) with a light brown hilum. The flowering date was the 5th of August and the maturity date was the 15th of October. Plant height, first pod height, number of nods, number of branches, and number of pods were 65 cm, 13 cm, 16, 4.5, and 99, respectively. In the sprout test, germination rate and sprout characteristics of 'Aram' were comparable to that of the 'Pungsannamulkong' cultivar. The yield of 'Aram' was 3.59 ton ha-1 and it was 12% higher than that of 'Pungsannamulkong' in southern area of Korea. The yield of 'Aram' in the Jeju region, which is the main region for soybean sprout production, was 20% higher than that of 'Pungsannamulkong'. The height of the first pod and the tolerance to lodging and pod shattering, which are connected to the adaptation to mechanized harvesting, were higher in 'Aram' compared to those in 'Pungsannamulkong'. Therefore, the 'Aram' cultivar is expected to be broadly cultivated because of its higher soybean sprout quality, and seed yield and better adaptation to mechanized harvesting. Protein Fractionation of Whole Crop Silages, and Effect of Borate-phosphate Buffer Extraction on In vitro Fermentation Characteristics, Gas Production and Degradation (사료작물 사일리지의 단백질 분획 및 Borate-phosphate Buffer 추출이 In vitro 발효성상, Gas 발생 그리고 분해율에 미치는 효과)

  • Protein fractionation was evaluated from whole crop silages of rye (RS), wheat (WS), triticale (TS), oat (OS), barley (BS), and rice straw silage (RSS), and in vitro trial was carried out to examine the effect of silage and extraction of soluble protein on fermentation characteristics, total gas production and degradation. Soluble protein of silages was extracted with borate-phosphate buffer, and fermentation characteristics, gas production and degradation of silages were estimated by incubating anaerobically the mixed solution of strained rumen fluid and artificial saliva (1:1, v/v) containing dried and ground silages placed in nylon bag at $39^{\circ}C$ up to 48h. Soluble protein (SP) content was lowest for RSS as 2.11% in total CP compared to those for other silages. Highest A fraction (NPN) was observed from RS (74.33% of total CP) while those from TS and RSS were relatively low (48%). B2 fraction was relatively higher for RS, RSS and WS than for TS and BS. $B_3$ fraction was lowest in WS among silages. C fraction (27.07) in RSS was higher than in other silages (1.40~9.93%). pH in incubation solution was increased (P<0.01~P<0.001) for extracted silages up to 12h but decreased (P<0.01) at 48h for non-extracted ones. Contents of ammonia-N (P<0.001) and total VFA (P<0.01~P<0.001) were higher for non-extracted silages than for extracted ones. Acetate proportion was increased (P<0.001) in buffer extracted silages while those of propionate and butyrate were decreased (P<0.001) up to 24h incubation. Increased (P<0.001) total gas production was obtained from non-extracted silages up to 12h while gas production was increased (P<0.01) in extracted ones thereafter. In vitro degradation of dry matter and CP was increased (P<0.001) in non-extracted silages but that of neutral detergent fiber was increased (P<0.001) in extracted ones without difference among silages. Difference in mean values of degradability for each silage prior to- and post extraction with borate buffer, however, was not found among silages. It may be concluded that high NPN content of silages may reduce the protein availability in silages and borate buffer soluble components in silages can stimulate the early stage of fermentation.
