Tricholoma gigantium, an edible mushroom, belongs to Tricholomataceae of Tricholoma and is distributed at Jeju-Do in Korea. It is also well-known as the medicinal mushroom in Taiwan. The cultivation method using the compost was developed in Korea in 1995. To develop a mass cultivation method and a superior strain, four strains were collected and tested. To establish the optimal cultivation conditions, various examinations were accomplished. Bag cultivation was more effective than box cultivation and the optimal relative humidity was more than 80%. Although the mycelial growth was tested in the substrate supplemented with different additives, such as rice bran and wheat bran, there's no significant difference between them. It suggested that the environmental conditions were more important than the substrate additives for cultivation.
For the artificial removal of air pollutants such as pesticides, environmental toxicants, and pathogenic microorganisms in the greenhouse or the living environment, the accelerated photodegradation and the biocidal effects of some photosensitizers (PS)/photocatalysts (PC) were tested under the sunlight and/or artificial light. The selected photosensitizers/photocatalysts included the semiconductors (PC-1 and PC-2), the oxidizers (PC-3, PC-4, PC-5 and PC-6), the aromatic ketone (PS-7) and the aromatic amine (PS-8). In the case of dichlorvos, all the photocatalysts selected showed more accelerated photodegradation than the control without photocatalysts under both the sunlight and artificial light. Whereas, only the photocatalyst PC-1 accelerated the degradation of methyl tert-butyl ether about 17 times more than the control under both the sunlight and artificial light. Procymidone was much more degraded by the photosensitizer PS-8 and the two photocatalysts (PC-1, PC-6) than by PS-7. In the preliminary experiments to diminish the population of the microorganisms in the air, the photocatalyst PC-1 added to the suspensions of Pseudomonas putida, Phytophthora capsici, and Salmonella typhimurium obviously inhibited the microbial growth under the artificial light. The photocatalyst PC-1 showed a bactericidal activity against Salmonella typhimurium spread on the nutrient broth agar medium. These results suggest that the photosensitizers/photocatalysis under the light can remove some air pollutants and hence they can be used to reduce the exposure of the workers in the horticultural facilities and/or the public in the environment to the harmful pollutants.
This study was conducted to investigated the distribution, ecological character and life cycle of Black Soldier Fly(BSF), Hermetia illucens, to determine effects of BSF on composing ability to the livestock feces. The distribution of BSF was defined in all parts of the country in Korea. Its main habitat was found to be areas near cattle sheds, feces sheds, living waste dump grounds, and food waste dump grounds. Excellent species of the bio-conversion using the livestock feces were selected out of two kinds of solider flies(Hermetia illucens and Ptecticus tenebrifer). And effects of BSF on composing ability to the livestock feces were pig feces 0.5 g, cattle feces 0.43 g and chicken feces 0.3 g per a larvae of BSF. In the examination of the ability of BSF to decompose food waste, volume of the livestock feces decreased by weight pig feces 77, cattle feces 83.62 and chicken feces 80%. Observed characteristics of BSF larvae and pupae stage were by developmental stage may be summarized as follows: size and length of larvae and pupae of BSF were higher than in pig feces and mixed sawdust treatments. In order to develop artificial indoor rearing techniques, The pupation and emergence rate were not visible the difference but important factor of the mating and egg-laying was a sun light. And it was visible the difference in mating ratio. Mating rate of H. illucens by seasonal change was the highest from May to July. Mating mostly occurred between 10 : 00 and 12 : 00 during which light intensity is high. As the egg-laying medium for artificial egg collection, calf feed and food waste were most effective. The ratio of egg-laying medium is waste food : sawdust(1 : 1) in volume. For egg-laying materials, flower foam and wood with holes were used; holes sized 3 ~ 5 mm in diameter, 7 ~ 10 mm in depth were most preferred for egg-laying.
This study was conducted to determine the effects of mycorrhizal inoculation and ortet ages on the rooting of Quercus acutissima cuttings. The cuttings taken from 12-week-, 2-year-, 4-year-, and 20-year-old plants were rooted in mid summer in the rooting medium (vermiculite 2 : peatmoss 1 by volume) with or without Pisolithus tinctorius (Pt) and Glomus sp. inocula and IBA under intermittent misting system in an open shed. The average percentages of rooting were about 82%, 49%, 29%, and 13% for cuttings taken from 12-week-, 2-year-. 4-year-, and 20-year-old seedlings, respectively. Pt inoculation, enhanced rooting of cuttings at all age classes, except 12-week-old seedlings, with the highest enhancement(22%) observed in cuttings taken from 20-year-old trees. The highest percentage of rooting in each age group eras 88.9% in 12-week-old seedlings treated with Pt plus 3.000ppm IBA, 75% in 2-Year-old plants with 1,000ppm IBA, 58.3% in 4-year-old plants with 3.000ppm IBA and 22% in 20-year-old plants. The addition of Glomus sp. fungus inoculum failed to enhance rooting. Pt mycorrhizal inoculation enhanced root dry weight, length, and diamter of adventitious roots at cuttings taken from 12-week- and 20-year-old trees, except the cuttings taken from 4-year-old seedlings. Rooted cuttings had more total nitrogen content in the leaves than unrooted cuttings, and the greater rooting response was associated with the higher phosphorus content in the leaves.
In order to elucidate the systematic taxonomy and genetic characters of Canoderma lucidum, cultural characteristics of the fungus were investigated. Mycelial growth of Ganoderma lucidum were favorable on oat meal agar medium, and optimum temperature and pH of the medium for mycelial growth were $30^{\circ}C$ and 5.5-6.0 respectively. Irradiation of white fluorescent lamp inhibited mycelial growth and critical time for inhibition of mycelial growth was 4-8 hours. Concentric zones and mycelial strands of Ganoderma lucidum was induced by irradiation of white fluorescent lamp and formation of mycelial sectors was influenced by nutrient source of media and irradiation of white fluorescent lamp. These characters were different among the isolates, but no relationship was observed between these characters and the fruiting body type of the fungus. Basidiospores were formed directly from the mycelium cultured on artificial media without producing fruit body.
The marine microalga Isochrysis galbana Parke was studied to optimize its growth conditions in flask culture. Important medium components studied include nitrogen source, buffer, trace elements and vitamins. Environmental conditions include pH, temperature, light intensity, mixing extent and working volume. The medium prepared from natural sea-waters gave a higher final cell density than the medium prepared from synthetic sea-water Nitrate was a better source than ammonium. In the range of 0.4∼2mM, the final cell density was proportional to the initial nitrate concentration and the cell yield was estimated to be 8.5g dry cell wt/g N. For phosphate, optimal growth was observed in 0.1∼1.0mM but a considerable variation in pH was resulted. The addition of Tris at 5mM or 7mM could stabilize the medium pH, but this significantly reduced both growth rate and final cell density, The effect of trace elements and vitamins was negligible. Optimal temperature and initial pH were $20^{\circ}C$ and 8. When the intensity of incident light was varied in the range of 400∼2100 lux, the growth rate increased from 10mL to 70mL, the final cell density decreased although the initial growth rate did not change. Optimal agitation speed was 100rpm when working volume was 30mL. With optimal conditions, the maximum specific growth rate obtained was 0.021hr-1 and the final cell density was 1.1g/L.

항만건설에 있어서 항만의 내부시설 중에서 무엇보다도 가장 중요한 시설물은 선박을 안전하게 접안시켜 하역할 수 있는 안벽시설물이다. 안벽구조 형식의 결정은 항만의 이용목적 등에 따라서 달라지지만 항만의 건설입지조건 등에 의해서도 달라진다. 안벽구조형식 중에서 잔교 식 안벽은 무엇보다도 단기간에 건설이 용이하여 지금까지 각국에서 널리 사용되어 왔고 장래에도 이용도가 증가되리라 생각한다. 최근에는 해안선을 이용한 위락시설이 건설되면서 잔교 식 안벽구조물을 설치하여 보조시설물로 이용하는 경우가 많다. 과거에 설계되어 잔교를 설계할 경우는 일반적으로 항내의 정온이 잘 유지되는 경우에 대해서 설치하는 경우가 많기 때문에 파랑에 의한 반사율과 잔교 상부에 작용하는 양압력을 고려해야할 필요성이 거의 없었다. 그러나 최근에는 태풍이 내습할 경우 기존의 항내로 높은 파랑이 침입하는 경우가 발생하고 있어 항내에서도 잔교의 파괴로 인한 자연재해가 대형화되는 경우가 발생하고 있다. 또, 처음부터 안벽을 설계할 때에 대형화의 잔교 식 안벽구조물을 설치하는 경우도 있다. 이런 잔교 식 안벽 구조물을 잔교의 상부 판에 작용하는 양압력 분포와 잔교 전면의 반사율 등이 구조물의 유지관리 등에 미치는 영향이 매우 크기 때문에 반사율 검토와 양압력을 고려한 설계가 필요하다. 본연구의 대상은 일정 해역에 잔교 식 안벽을 설계하고자 할때 최적의 안벽 설계가 될 수 있도록 수리모형실험을 실시하여 구조물의 안전과 항내정온에 기초가 되는 자료를 도출하고자 하였다. 따라서 본 수리모형실험으로 인한 연구는 잔교 식 안벽에 대한 반사율과 상부에 작용하는 양압력, 잔교말뚝(pile)에 작용하는 수평압력을 검토하여 잔교 식 안벽 설계에 기초자료를 제공하고자 한다. 혈청을 이용한 동결보존을 대체할 수 있을 것으로 생각된다.지에 더해주면 세포의 증식이 개선될 것이다. 그래서 몇 가지 첨가물을 이용해 세포의 증식력에 변화가 나타나는지 알아보았다. 첨가물을 이용한 실험에서 IGF-I의 경우 장기간 배양에서 세포의 수를 안정적으로 유지하고 계대 횟수를 증가시키는 효과를 보였다. 이는 IGF-I이 어느정도 세포의 증식을 유지시켜주는 역할을 하기 때문인 것으로 생각된다. 무혈청 배지에서 비적응 CHO 세포의 계대 배양에 한계가 있는 것은 세포주기가 멈추기 때문인 것으로 생각된다. 세포주기가 멈추는 growth factor와 같이 세포의 증식을 지속적으로 유도할 수 있는 물질이 무혈청 배지에서는 부족하기 때문인 것으로 생각되고, IGF-I과 같은 첨가물을 통해 극복할 수 있는 문제라고 여겨진다.관점과 주거교육가치관 요소와의 관계를 알아본 결과, 전통적 관점은 주거교육가치관 요소 중 오직 주거관리적 요소와 관계가 있었으나 그 정도는 낮으며 실천적 관점과 구조적 관점은 주거가치관의 각 요소에 따라 약간 다르기는 했으나 주로 보통의 관계를 보였다.군 순으로 높게 관찰되었다. 이상의 결과를 종합할 때, 임상에서 니켈-티타늄 합금 와이어에 굴곡을 부여하기 위해 열처리하는 경우 초탄성 특성은 유지될 수 있으나, 부하-변위 곡선의 상방 증가가 나타나므로, 와이어에 의한 교정력이 증가될 수 있음에 유의하여야 한다. $day^{-1}$인 인공습지), scenario 2(면적 4.2ha인 저류지)가 각각 연평균 6.9%, 4.8%, 7.1%의 감소를 보였다. The optimal condition for the production of the artificial fruiting bodies of Paecilomyces japonica in bulk was determined from the observation of the conidial, hyphal, and cultural condition and the medium. The colony was grown 32 mm diameter in 14 days on potato dextrose agar(PDA). Conidia was irregularly long oval-shaped and measured 4.07${\times}$1.56 ${\mu}$m in average. The hyaline hyphae formed transparent bundles which branched out. The fruiting body on the insect surface was measured 30 to 50 mm in length and formed up to 55 branches. Potato dextrose(PD) both was the most effective for the growth of Paecilomyces japonica among the liquid type media. About 3.1 mg mrcelia in dry weight were produced in 50 ml/PD broth. The best condition for the production of conidia under which condition 4.3${\times}$10$^8$ conidia/ml were harvested was pH 5.0 in acidity and 20$^{\circ}C$ following preculture at 24$^{\circ}C$ in temperature for 7 days.
An in vitro culture method for entomopathogenic nematode Steinernema glaseri was developed. A symbiotic bacterium was isolated from Steinernema glaseri and identified as Xenorhabdus nematophilus. Phase variation that differed in some biochemical characteristics of symbiotic bacterium was observed. Entomopathogenic nematodes carried only phase I bacterium in their guts. Phase I bacterium could be converted into phase II form in in vitro culture medium consisting of 5% yeast extract, 0.5% NaCl, 0.05% $K_2HPO_4$, $0.02% MgSO_4$.$7H_2O$. The optimum temperature for bacterial growth was $28^{\circ}C$. The pH of the culture medium increased up to 9.0-9.5 during the exponential growth period of the culture, regardless of initial pH 6-7. Various culture media such as chicken offal, dog food, bovine liver, peanut, and so on were tested for in vitro culture of the nematodes. The best medium for Steinernema glaseri production was obtained from concentrated homogenate of bovine liver and the nematode growth was highest at 80% bovine liver. In the co-culture of entomopathogenic nematode with its symbiont, the growth rate of nematodes was 2 times faster than that without its symbiont and the nematode concentration reached about $5.5\times10^4$/mL within 15 days.
Fifty four acid-resistant and bile-resistant isolates of lactic acid bacteria were initially isolated from the faces of Korea native cattle and Holstein using MRS agar and LAPT agar, and ten strains with superior activity against bile salt were finally selected LS1, LS15, and LL6 isolates showed survival of 66.5%, 82.6% and 80.7% against the simulated stomach liquid(pH 2.5), respectively. LL6 and LL7 isolates had the highest inhibitory activities against the pathogenic bacteria such as Salmonella typhimurium, Staphylococcus aureus, and Clostridium perfringens. By using API 50 CHL kit LS1, LS2 and LM1 isolates were identified as a L. fermentum. LL6 and LL7 isolates as a L. acidophilus, and LS3 isolate as a L. plantarum. The other four isolates belong to genus Lactobacillus. All the isolates tested were sensitive to some antibiotics such as ampicillin, amoxicillin and erythromycin, but resistant to colistin and ciprofloxacin. LB1, LL6 and LL7 isolates were resistant to gentamicin and neomycin. Especially, the LL6 isolate showed the highest resistance to both of the simulated stomach liquid and bile salt, in addition to the highest inhibitory activities against Sal. typhimurium, Staph. aureus and Cl. perfringens.
