Rye(Secale cereale cv. Chunchu), barley(Hordeum vulgare cv. Dusan 29#), and italian ryegrass(Lolium multiflorum cv. Barmultra) were treated at 0, 0.1, 0.3, and 0.6 % of NaCl solution for 30 days. Plant height, leaf length, and leaf area were decreased with increasing NaCl level, and leaf area was decreased mainly due to decrease of leaf length. Root length was decreased at higher NaCl levels, but root number was increased. Shoot dry weight was decreased by increasing NaCl levels in rye and italian ryegrass, but barley did not show significant change. Dry weight of root was more decreased than that of shoot at higher NaCl levels. Chlorophyll content was decreased, but electric contuctivty of shoots and roots was increased at higher NaCl levels. Na+ content in shoot and root was increased with increasing NaCl level but the degree was not same. Reducing power of roots was that three crops as affected by NaCl was not equal. Barley was the most tolerant to NaCl stress.
In order to optimize the conditions of callus induction and plant regeneration in seed culture of Italian ryegrass, the effects of basic medium and carbon sources on seed culturability and genotypic difference of plant regenerability were investigated. MS medium was better than N6 and B5 medium in enhancing callus growth and plant regeneration. Sucrose was superior to maltose in plant regeneration as carbon source in the medium. The genotypic difference in plant regenerability was obvious among four cultivars of Italian ryegrass tested. 'Rio' and 'Jeanne' showed to have higher regenerability with the frequency of 38% and 56%, respectively.
A system for the production of transgenic plants has been developed for Italian ryegrass(Lolium mult리orum Lam.) via Agrobacterium-mediated transformation of embryogenic callus. Mature seed-derived calli were infected and co-cultured with Agrobacterium EHA101 carrying standard binary vector pIG121Hm encoding the hygromycin phosphotransferase(HPT), neomycin phosphotransferase II (NPTII) and intron-oontaining $\beta$g1ucuronidase( intron-GUS) genes in the T-DNA region. The effects of several factors on transformation and the expression of the GUS gene were investigated. Inclusion of 200${\mu}M$ acetosyringone(AS) in inoculation and co-cultivation media lead to a significant increase in stable transformation efficiency. Increasing Agrobacterium cell density up to 1.0 in $OD_{600}$ during infection increased transfonnation efficiency of embryogenic calli. The highest transfonnation efficiency was obtained when embryogenic calli were incoulated with Agrobacterium in the presence of 0.1% Tween20 and 200${\mu}M$ AS. Hygromycin resistant calli were developed into complete plants via somatic embryogenesis. GUS histochemical assay and PCR analysis of transgenic plants demonstrated that transgenes were integrated into the genome of Italian ryegrass.

In order to study the effect of barley, Italian ryegrass (IRG), and legume mixture on nitrogen fixation and transfer to grasses on spring paddy field, an experiment was carried out from Oct. 2006 to June 2007 in Naju, Korea. A split plot design with three replications was used for the experiment. One reference plot was assigned for each treatment to determine nitrogen fixation. Main plots consisted of Chinese milk vetch, crimson clover, forage pea, and hairy vetch with barley, respectively. Subplot treatment were barley or IRG with four seeding ratio of legumes (50:50, 60:40, 70:30, and 80:20). To estimate N fixation by legumes, $^{15}N$ isotope dilution technique was used. $^{15}N$ fertilizer [$(^{15}NH_4)_2SO_4$ solution at 99.8 atom N] was uniformly applied to $600cm^2$ in the middle of each plot on April 15, 2007. Plots were harvest by hand on June 8, 2007. Dried sample were ground to a fine power and analyzed for total N isotope N. $^{15}N$ was determined using elemental analyzer-isotope ratio mass spectrometry. The calculation of N transfer was determined with the isotope dilution method. The content of N was higher in legumes than that in barley or Italian ryegrass. Nitrogen level in forage pea was significantly higher than that of other legumes. There were significantly differences in N content between legumes in IRG mixture. Atom % $^{15}N$ excess was significantly different in legumes with barley. The 60:40 sub plot had higher (p<0.05) atom % $^{15}N$ than other seeding ratio treatments. The enrichment ranged from 0 to 0.58. Compared to barley, the enrichment of IRG with its accompanied legumes was higher, ranging from 0.38 to 1.0. The N derived from the atmosphere (Ndfa) ranged from 0% to 49.5% with barley-legume mixture. It ranged from 0 to 60.5% in IRG-legume plots. N transfer from legumes to neighboring grasses was 12.3 to 90.9 kg/ha for barley-legume mixture and 31.7 to 107.8 kg/ha for IRG plots. IRG plots showed higher N transfer for IRG-legume mixture in general based on difference method. Based on $^{15}N$ dilution method, the N transfer was 0 to 36.1 kg/ha for barley-legume mixture and 0 to 50.6 kg/ha for IRG plots. There was a tendency toward higher N transfer on the difference method than that of the $^{15}N$ dilution method.

Manure recycle is an emerging issue in agriculture in Korea these days. Farmers are keeping eye on legume mixture with grasses for nitrogen fixation and transfer to companion crops by legumes. We had a trial to investigate the effect of different legume mixtures on nitrogen fixation and transfer in spring soil. The treatment was arranged in a randomized complete block design with three replications. Three different mixtures were used(rye+hairy vetch, Italian ryegrass+crimson clover, oat+pea) and sowed in pots with paddy soil from western part of Korea(Seo Chon County). Pots with rye+vetch were transplanted from field of RDA(rural development administration) in Suwon on 2 February 2007 and other mixture treatments were sowed on early March with different sowing rate(7:3=Italian:crimson, 6:4=oat:pea). $(^{15}NH_4)_2SO_4$ solution at. $99.8\;atom%^{15}N$ was applied to the each pot at the rate of $2kg\;N\;ha^{-1}$. Application was done on 6 April at rye+vetch pots and remainder were applied on April 16. Forage were harvested from each pot at ground level in heading stage and separated into legume and grass. Total N content and $^{15}N$ value were determined using a continuous flow stable isotope ratio mass spectrometry. DM yield of rye+vetch, Italian+crimson and oat+pea were 6,607, 3,213 and 4,312 kg/ha, respectively. Proportion of N from fixation was 0.73(rye+vetch), 0.42(Italian+crimson) and 0.93(oat+pea). The percentages of N transfer from legume to grass were from 61% to 24% in different method by treatment and -35% to 21% in isotope dilution method.
As an initial step for future genetic manipulations to improve forage characteristics of Italian ryegrass (Lolium multiflorum Lam.), an efficient tissue culture system was established and the factors affecting plant regeneration were evaluated. MS medium containing 5mg/L 2,4-D was optimal for embryogenic callus induction from mature seed and had a strong effect on successive plant regeneration. The plant regeneration frequency was observed at above 70% when embryogenic calli induced were transferred to N6 medium supplemented with 1mg/L 2,4-D and 5mg/L BA. Among several basic media tested, MS and N6 medium were optimal for callus induction and plant regeneration, respectively. Genotype was an important factor in plant regenerability. 'Jeanne' showed the highest regeneration frequency of 73%. A short tissue culture period and high-frequency regeneration system established in this study will be useful for molecular breeding of Italian ryegrass through genetic transformation.
In an effort to optimize tissue culture responses of Italian ryegrass(Lolium multiflorum Lam.) for future genetic manipulations to improve forage characteristics, the effects of culture medium supplements on tissue culture responses were investigated with mature seeds of three cultivars, 'Jeanne', 'Florida-80' and 'Metro', as explant tissues. For all explants, MS medium containing 5mg/L 2,4-D was optimal for embryogenic callus induction from mature seed and had a strong effect on successive plant regeneration. The optimal concentration of dicamba for the induction of embryogenic callus from mature seeds was 7mg/L. The highest plant regeneration frequency was observed when embryogenic callus was transferred to N6 medium supplemented with 1mg/L 2,4-D and 5mg/L BA. Plant regeneration frequency of callus cultured in the dark was higher than that of cultured in the light. Casein hydrolysate and L-proline improved both in embryogenic callus induction from mature seeds and plant regeneration. High-frequency regeneration system established in this study will be useful for molecular breeding of Italian ryegrass through genetic transformation.
Gray leaf spot (GLS) is a serious fungal disease caused by Pyricularia oryzae Cavara, recently reported on the important turf and forage species, perennial ryegrass (Lolium perenneL.). This fungus also causes rice blast, which is usually controlled by host resistance, but durability of resistance is a problem. Few instances of GLS resistance have been reported in perennial ryegrass. However, two major QTL for GLS resistance have been detected on linkage groups 3 and 6 in an Italian x perennial ryegrass mapping population. To confirm that those QTL are still detectable in the next generation and can function in a different genetic background, a resistant segregant from this population has been crossed with an unrelated susceptible perennial clone, to form a new mapping population segregating for GLS resistance. QTL analysis has been performed in the new population, using two different ryegrass field isolates and RAPD, RFLP, and SSR marker-based linkage maps for each parent. Results indicate the previously identified QTL on linkage group 3 is still significant in the new population, with LOD and percent of phenotypic variance explained ranging from 2.0 to 3.5 and 5% to 10%, respectively. Also two QTL were detected in the susceptible parent, with similar LOD and phenotypic variance explained. Although the linkage group 6 QTL was not detected, the major QTL on linkage group 3 appears to beconfirmed. These results will add to our understanding of the genetic architecture of GLS resistance in ryegrass, which will facilitate its use in perennial ryegrass breeding programs.
