Cultivable Bacterial Community Analysis of Saeu-jeotgal, a Korean High-Salt-fermented Seafood, during Ripening (배양법을 이용한 새우젓갈 숙성과정 중 박테리아상 분석)

  • To determine the dominant bacterial species during the Saeu-jeotgal ripening process, the cultivable bacterial population was examined over a 135-day period using six different growth media. The greatest numbers of bacteria were identified when marine agar was used for culture, with maximum cell density identified at day 65 (2.51 × 107 colony forming units/g). Over the course of 135 days, the bacterial diversity was analyzed eight times. A total of 467 isolates, comprising 87 species from 42 genera, as well as 16 isolates belonging to previously unknown species, were identified. The number of species detected decreased from 39 at day 1 to 13 at day 135. The order of dominance at the genus level was as follows: Staphylococcus, Salimicrobium, Kocuria, and Psychrobacter. Staphylococcus and Salimicrobium accounted for 2% of the diversity at day 1, and then increased to 39% and 36%, respectively, at day 135. The dominant species Staphylococcus equorum, Salimicrobium salexigens, and Kocuria palustris accounted for 23.6%, 16.1%, and 10.9% of all isolates, respectively. Importantly, both St. equorum and Sm. salexigens remained viable at a NaCl concentration of 21% (w/v), which indicates their strong involvement in the ripening of Saeu-jeotgal.

Long Head of the Biceps Tendon Lesion Associated with Rotator Cuff Tear (회전근 개 파열과 동반된 상완 이두 건 장두의 병변)

  • Purpose: To evaluate pathologic patterns and outcomes of treatment of a biceps tendon lesion associated with a rotator cuff tear. Materials and Methods: We reviewed 92 patients (i) who underwent surgery for a cuff tear, (ii) for whom the biceps lesion could be observed retrospectively, and (iii) had a minimum follow-up of 2 years. The pathology of biceps tendon was classified into 4 types: tenosynovitis, fraying or hypertrophy, tear, and instability. All but the 4 with massive cuff tears were repaired. The biceps lesions were treated with debridement in 30, tenotomy in 10, tenodesis in 8, and recentering in 4. UCLA scoring was used for clinical results. Results: Seventy patients had a biceps lesion, 19 tenosynovitis, 22 fraying or hypertrophy, 21 a tear, and 8 instability. A biceps lesion was observed in 63% of cases of cuff tears below the medium size, and in 88% of cases with cuff tears above the large size. UCLA scores according to the pathology of the biceps lesion were 29.6 in the absence of a biceps lesion, and 28.3 in its presence. UCLA scores in patients with tenotomy or tenodesis for associated biceps tendon lesions were 28.2. Conclusion: There is a greater incidence and severity of a biceps lesion with a larger cuff tear. Therefore, the cause of a biceps lesion might be related to the cause of the cuff tear. Among the several options of treatment for biceps lesion, tenotomy or tenodesis may be particularly effective in providing pain relief.

Comparative Effectiveness of Adjunctive Aripiprazole versus Bupropion Uses to Selective Serotonin Reuptake Inhibitor on the Specific Symptom of Depression : A post-hoc, Multi-Center, Open-Label, Randomized Study (세로토닌 재흡수 억제제에 대한 아리피프라졸 및 부프로피온 부가요법의 우울증 세부증상에 대한 효과 비교 : 다기관, 개방표지, 무작위 연구)

  • Objective : The purpose of this study was to examine the effects of adjunctive aripiprazole versus bupropion on specific symptoms of depression. Methods : Data were from 6-week, randomized, prospective, open-label multi-center study in 103 patients with major depressive disorders. Participants were randomized to receive aripiprazole (2.5-10 mg/day) or bupropion (150-300 mg/day) for 6 weeks. Change in four subscales of the 17-item Hamilton Depression Rating Scale (HAM-D17) that capture core depression symptoms was determined, and change in individual HAM-D17 items was also assessed. Changes in three composite subscales-anxiety, insomnia, and drive were also examined. Results : Within-group change in the four core subscales was large [effect size (ES)=1.30-1.47] and it was similar to that in the HAM-D17 total score. Differences between aripiprazole and bupropion were significant for each of the four core subscales and the HAM-D17 total score favored aripiprazole (p<0.001). On three composite scales, both treatments caused substantial changes in anxiety (within-group ES=1.10 (aripiprazole) vs. 1.00 (bupropion)], insomnia (ES=0.75 vs 0.50), and drive (ES=1.17 vs 1.15). Conclusion : This results suggested that both aripiprazole and bupropion adjunctive therapies with selective serotonin reuptake inhibitors resulted in significant and clinically meaningful changes in core symptom subscales for depression.

PCR Detection of Terephthalic Acid Degrading Comamonas testosteroni in Soil (PCR을 이용한 토양 중 Terephthalic Acid 분해 Comamonas testosteroni의 검출)

  • Eleven bacterial strains which are able to utilize terephthalic acid as a carbon and an energy source for growth were isolated from the soil of 7 water quality evaluation points in Kyonggi area of Korea. Phthalic acid isomer degrading activity of the isolates from the 4 contaminated points was higher than those from the 3 clean points. Among 11 isolates, 4 isolates which have high terephthalic acid degrading activity and degrade two phthalic acid isomers were identified by partal 16S rDNA sequence determination. One of them was identified as Pseudomonas putida, and the others as Comamonas testosteroni. Thus a large number of phthalic acid isomer degrading bacteria in domestic soil were inferred as C. testosteroni. On the basis of these results, the PCR detection of C. testosteroni in soil was applied to monitor soil contamination by phthalic acid isomers. The DNA of C. test-osteroni extracted from 4 g soil was directly detected by PCR with C. testosteroni specific primer pair. The amount of PCR products was different according to sampling sites and more PCR products were obtained from contaminated sites than those from clean sites (Gulpo-chun>Anyang-chun>Hwangguji-chun>Shin-chun>Huk-chun>Pukhan-river>Kapyeong-chun). This result was coincided with that of the viable cell counts for terephthalic acid degrading bacteria.

Evaluation of the Colonization of Lactobacillus plantarum in Mouse Gut by Terminal Restriction Fragment Length Polymorphism Analysis (Terminal Restriction Fragment Length Polymorphism 분석을 이용한 Lactobacillus plantarum의 생쥐 장관 정착 평가)

  • T-RFLP (terminal restriction fragment length polymorphism) analysis, one of the most highly adopted culture-independent microbial community analysis methods, was applied to evaluate the colonization of probiotics in experimental animal gut. Lactic acid bacteria that exhibited cinnamoyl esterase activity were isolated from Korean fermented vegetables and identified by 16S ribosomal RNA sequence analysis. Lactobacillus plantarum KK3, which demonstrated high chlorogenic acid hydrolysis by cinnamoyl esterase activity, and acid/bile salt resistances, was cultured, freeze-dried, and fed to mice and the microbiota in their feces were monitored by T-RFLP analysis. The T-RF of L. plantarum was detected in the feces of mice after the start of administration and lasted at least 31 days after the initial 7 day feeding. T-RFLP analysis was considered a useful tool to evaluate the gut colonization of probiotic L. plantarum. In order to prove that L. plantarum was from viable cells, we reisolated L. plantarum in the feces using cinnamoyl esterase activity media as the screening step. The colonization of L. plantarum KK3 in the mouse gut was confirmed by this research.

Stereo Vision-Based Obstacle Detection and Vehicle Verification Methods Using U-Disparity Map and Bird's-Eye View Mapping (U-시차맵과 조감도를 이용한 스테레오 비전 기반의 장애물체 검출 및 차량 검증 방법)

  • In this paper, we propose stereo vision-based obstacle detection and vehicle verification methods using U-disparity map and bird's-eye view mapping. First, we extract a road feature using maximum frequent values in each row and column. And we extract obstacle areas on the road using the extracted road feature. To extract obstacle areas exactly we utilize U-disparity map. We can extract obstacle areas exactly on the U-disparity map using threshold value which consists of disparity value and camera parameter. But there are still multiple obstacles in the extracted obstacle areas. Thus, we perform another processing, namely segmentation. We convert the extracted obstacle areas into a bird's-eye view using camera modeling and parameters. We can segment obstacle areas on the bird's-eye view robustly because obstacles are represented on it according to ranges. Finally, we verify the obstacles whether those are vehicles or not using various vehicle features, namely road contacting, constant horizontal length, aspect ratio and texture information. We conduct experiments to prove the performance of our proposed algorithms in real traffic situations.

The Relationship of the Helicobacter pylori Virulence Factor Gene Subtype in Gastric Adenocarcinoma (위선암에서 Helicobacter pylori 독성인자와 유전자 아형의 관련성)

  • Purpose: The H. pylori cagA gene, vacA gene and iceA gene are considered to be important virurence factors that have been implicated in the development of gastric adenocarcinoma. It was reported that the presence of IS605 elements may be responsible for rearrangements and lead to partial or total deletions of the cag pathogenicity island (PAI) and the virulence of cag PAI may be changed. However, different results regarding the association between these virulence factors and clinical disease have been reported from different geographic regions. This study evaluated the relationship between H. pylori virulence factors such as cagA, vacA, iceA, IS605 and gastric adenocarcinoma. Materials and Methods: H. pylori isolates were obtained from 54 infected patients (24 cases of gastric adenocarcinoma, 30 cases of control). H. pylori isolates were identified by PCR with ureC gene and 16S rRNA. PCR was performed to examine cagA, vacA, iceA and IS605 genotypes. Results: Significant difference was found in the negative rates of cagA between gastric adenocarcinoma group and control ($62.5\%\;vs.\;33.3\%$ P=0.033). No significant difference was found in the prevalence of iceA, vacA between gastric adenocar cinoma and control. The genotype of cagA+ vacA s1-m1 iceA1 was predominant in H. pylori isolates irrespective of the clinical outcome. IS605 in PAI was not found in gastric adenocarcinoma gruop and control. The positive rates of IS605 in genome were $33.3\%$ in gastric adenocarcinoma group and $36.7\%$ in control (P>0.05). In gastric carcinoma, the positive rate of $cagA^{+}/IS605$ was lower than in control ($12.5\%\;vs\;40.0\%$, P=0.025) and the positive rate of cagA-/IS605 was higher than in control ($54.2\%\;vs\;23.3\%$, P=0.02). Conclusion: H. pylori virulence factors had not related significantly with gastric adenocarcinoma. Further study is needed to examine the specificity of H. pylori strains.

  • Soft carbons are prepared by heat-treatment of cokes with different amounts of phosphoric acid (2, 4.5, and 10 wt% vs. cokes) at $900^{\circ}C$ to be used as anode materials for lithium ion batteries. From electrochemical measurements combined with structural analyses, we confirm that abundant nano-pores are existed in the microstructure of soft carbons prepared with the phosphoric acid, which are responsible for further lithium ion storage. Significant increase in reversible capacity of soft carbon is attained in proportion to added amount of the phosphoric acid. We also demonstrate the effect of structural modification with phosphoric acid on electrochemical performance of soft carbon to elucidate the origin of additional capacity.

  • Installation of digital tachograph, black box, and ADAS have been enforced to commercial vehicles for preventing violent driving and accidents by the Traffic Safety Act in Korea. Nevertheless, the damage caused by road hazards has increased 1.5 times in 2016 compared to 2013. So, developing new technologies that can identify road hazard using the sensors installed in commercial vehicles are conducting by the Ministry of Land, Infrastructure and Transport. As a part of the technologies, this research analyze the error range of GPS installed in commercial vehicles that vary according to the driving speed. As a result, the average error was 9.72m at the driving speed of 100km/h, and the error was 2.1 times larger than the average error of 4.69m at the driving speed of 40km/h. The event point proper integration/separation range(m) was analyzed to be 20m with a recognition rate of 90% or more at the same point regardless of driving speed. The results of this research can be used as basic data for improving the accuracy of location-based data would be collected using commercial vehicles.

  • In preparation of self-repairing polymer-modified waterproofing asphalt-montmorillonite (MMT) composite, silylation-modification characteristics of cation ($Na^+$) exchanged K-10 (Na-MMT-K) using 3-aminopropyltriethoxysilane (APS) were studied and the optimal conditions of its silylation-modification process were proposed by use of the results of XRD analysis on silylation-modified Na-MMT-K (S-Na-MMT-K) under various conditions. According to XRD results, it was confirmed that peaks of Na-MMT-K were simultaneously consistent with those of K-10 and natural or Ca-MMT modified Na-MMT. Similarly, S-Na-MMT-K was observed to have two basal spacings ($d_{001}$), among which the area-ratio of a secondary (001) peak ($2{\theta}=3.9{\sim}4.2^{\circ}$) to a primary (001) peak ($2{\theta}{\sim}8.838^{\circ}$) was suggested to be a criterion to represent a degree of APS silylation-modification. Then, the optimal conditions on APS-stirring period prior to APS-MMT reaction, APS-MMT reaction period, APS concentration and reaction temperature at the highest area-ratio were turned out to be 20 min, 2~3 hr, 7.5 w/v% and $50^{\circ}C$, respectively.
