Analysis of Trifluoroacetyl group in PVA-polyvinyltrifluoroacetate Copolymer by the Pyrolysis Gas Chromatography (열분해 기체크로마토그래피를 이용한 PVA-polyvinyltrifluoroacetate 공중합체중의 trifluoroacetyl 기의 정량)

  • The composite mole fraction of monomers in PVA-PVFAc copolymer could be calculated from the analysis of trifluoroacetyl group by the pyrolysis gas chromatography without breaking of C-F bonds in polymer. A linearity between trifluoroacetyl peak areas in pyrogram and sample weights was obtained within the range below 3mg. The data of trifluoroacetyl contents derived from gas chromatogram of copolymers with various D.S. were in good agreement with results by the Specific Ionmeter.

  • The endosperm protein, vicilin, of ginseng (Panax ginseng C.A. Meyer) was purified by ammonium sulfate precipitaion, gel permeation and ion exchange column chromatography. Vicilin is a glycoprotein composed of 2 subunits with molecular masses of 55,000 (large subunit) and 44,000 (small subunit). The anti-vicilin antibody was raised in rabbit, and purified by DEAE Affi-Gel Blue affinity chromatography. The endosperm cells of the seed were reacted with this anti-vicilin antibody and colloidal gold conjugated secondary antibody. Gold particles were labelled on the elaborating granules of Golgi complex, electron-dense granules and protein bodies in the endosperm cells. These results indicated that the vicilin, which was synthesized in rough endoplasmic reticulum and transported to Golgi, was elaborated in saccules of the Golgi and then transported into protein bodies by electron-dense granules.anules.

  • Transesterification reactions were carried out with myo-inositol and five fatty acid methyl esters such as methyl laurate, methyl myristate, methyl palmitate, methyl stearate and methyl oleate in the dimethylsulfoxide solvent. Their products were separated by both thin layer chromatography and column chromatography, and myo-inositol monoesters were quantitatively separated by counter current distribution. We measured their surface tension, foaming power and emulsifying power, determined critical micelle concentrations by the color method, and evaluated their hydrophilic-lipophilic balance. The results show that myo-inositol monoesters exhibit surface activites.

  • We have isolated an anti-complementary polysaccharide from the hot water extracts of Phellinus linteus mycelia. Anti-complementary polysaccharide, PL-5-IIIa, was purified by ultrafiltration, gel permeation chromatography using Sepharose CL-4B. GPC (Sepharose CL-4B) and its homogenicity was demonstrated by HPLC. Using gel permeation chromatography with standard dextrans, its molecular weight was determined as about 800,000 dalton. The purified PL-5-IIIa was identified as a protein bound polysaccharide comprising of 29.6% protein and 64.2% carbohydrate which was composed of fucose(15.8%), galactose(43.1%) and mannose(40.6%).

  • Apolipophorin-III (apoLp-III) was isolated and purified from the last instar larval hemolymph of Galleria mellonella by gel chromatography (Sephadex G-100) and ion exchange chromatography (CM-52). In the present study, I wanted to show that apoLp-III is taken up into the adult ovary in Galleria mellonella. Adult ovary tissues were incubated at room temperature for 30 min with fluorescein isothiocyanate (FITC)-labeled apoLp-III. Fluorescence microscopy and sodium dodecylsulfate (SDS)-polyacrylamide gel electrophoresis (PAGE) revealed that adult ovary tissues internalize fluorescence-labeled apoLp-III. The results suggest that apoLp-III is taken up by the adult ovary.

  • 악취물질은 산업의 발전과 경제성장에 따른 생활수준의 향상으로 인하여 급격히 늘어나는 대기오염 물질 중의 하나로, 악취문제는 인간의 오감을 통해서 느끼는 감각공해의 대표적인 물질이다. 악취물질의 대표적인 성분인 암모니아는 무색의 자극성이 매우 크고 부식성이 있는 알칼리 기체로 1 ppm 정도에서 감지되어 다른 악취물질보다 감지되는 농도가 높다. 또한 그 배출원이 화학공장 등의 산업시설 뿐만 아니라 양돈, 양계장 등에서도 배출되어 우리 주위에서 쉽게 감지할 수 있는 물질이다. (중략)

  • immunotoxin으로 쓰일 수 있는 리보솜 불활성화 단백질 (RIP, ribosome-inactivating protein)을 보리 씨앗에서 분리하였다 이 단백질은 분자량이 약 30,000 kDa 정도되고, pl가 9.0 보다 높다. 이러한 성질을 이용하여 Na-phosphate 완충용액으로 추출하고, 황산암모늄 60-80% 포화로 분획화하였다. 다음 CM-cellulose를 이용한 이온 교환 크로마토그래피, Sephacryl S-200 HR 컬럼을 이용한 gel filtration을 하여 순수히 분리하였고, 이를 전기영동하여 확인하였다.

Purification and Characterization of Polyphenol Oxidase from Flammulina velutipes (팽나무버섯 polyphenol oxidase의 정제 및 특성)

  • Polyphenol oxidase from Flammulina velutipes was purified and characterized. Purification of polyphenol oxidase was achieved by ammonium sulfate precipitation, Superdex G-200 gel filtration chromatography, Phenyl superose affinity chromatography, Mono-Q anion exchange chromatography and Superdex S-200 gel filtration chromatography on FPLC. After these purification steps specific activity of purified polyphenol oxidase increased to 199.1 units/mg. Polyphenol oxidase from F. velutipes was composed of a single polypeptide with molecular weight of about 40 kDa. Optimum pH and temperature for the enzyme reaction were found to be 6.0 and $25^{\circ}C$, respectively. The activity of the enzyme gradually decreased at acidic pH between 3 and 5, and the enzyme lost its activity at alkaline pH between 8 and 10. This enzyme exhibited high substrate specificity to o-diphenols. Km-values for L-DOPA and caffeic acid were found to be 3.97 mM and 1.78 mM, respectively. 2-mercaptoethanol, L-ascorbic acid, sodium bisulfite, EDTA and $Mg^{2+}$ inhibited the activity of pholyphenol oxidase and $Cu^{2+}$, $Fe^{2+}$, $Zn^{2+}$ and $Ni^{2+}$ increased enzyme activity. The activity of enzyme was well maintained at $-70^{\circ}C$ for over 4 months, and at $-20^{\circ}C$ for 1 months.
