Taxonomic Studies on Cercospora and Allied Genera in Korea (I) (한국산 Cercospora 및 관련 속의 분류학적 연구 (I))

  • The mycoflora of Korea, especially the cercosporoid taxa, have been poorly studied in the past. For this reason, a systematic revision of these taxa has been initiated, with the eventual aim of producing a monograph of the Korean cercosporoid taxa. The present study circumscribes ten species; viz., Cercospora althaeina, C. arachidicola, C. armoraciae, C. carotae, C. ipomoeae, Mycovellosiella ferruginea, Passalora miurae, Pseudocercospora araliae, P. cannabina and P. vitis. Distinguishing morphological characteristics are described and illustrated for each species.

Effects of Job's Tear(Yul-Moo) Extracts on Mouse Immune Cell Activation (율무 추출물이 마우스 면역세포 활성에 미치는 영향)

  • Natural products are increasingly appreciated as a lead for drug discovery and development. A number of investigators have studied various activities of natural products and have found that they have not only nutritional effects but also beneficial properties to cure various diseases and to maintain good health. Job's Tear(Yul-Moo) is a grass crop that have long been used in traditional medicine and a nourishing food. Job's Tear has been reported to exhibit anti-inflammatory, stomachic, antiallergic activity, and antispastic effects and has been used in China for the treatment of warts, rheumatism, and neuralgia although its mechanism remains unclear. Previous results in our laboratory demonstrated that the ethanol extract and water extract of Job's Tear exerted an immune regulatory function on mice cells in vitro. The present study was performed to investigate the ex vivo effect of Job's Tear on immune function. Seven to eight weeks old mices(Balb/c) were fed ad libitum on chow diet and water extract of Job's Tear were orally administrated every other day for two or four weeks at two different concentrations (50 and 500mg/kg B.W.). Proliferation of mice spenocytes and antibody production to sheep red blood cells(SRBC) using hemolytic plague forming cell assay were used to indicate the immune activity. Splenocytes proliferation of Job's Tear with mitogen stimulation such as Con A and LPS was enhanced at 50 mg/kg B.W. concentrations compared to those of control group. In case of antibody production to sheep red blood cells, the number of antibody- secreting cells was increased by administration of 50mg/kg B.W. concentration in mice immunized as a T-dependent antigen. From the present study, Job's Tear water extracts may be suggested to stimulate the mice immune response by enhancing the splenocytes proliferation and the number of plague forming cells.

Development and Quality Properties of Cereal Bars (씨리얼바 제조 및 품질특성)

  • The purpose of this study was to develop new functional cereal bars for breakfast with 9 types of cereal(corn, whole wheat, rice, brown rice, black rice, indian millet, sprouting brown rice, black soybean, job's tear) and their chemical and sensory properties were evaluated. Process of cereal bars are cereal mixing ${\to}$ formation ${\to}$ baking ${\to}$ cooling ${\to}$ topping. Moisture content of sunsik-type cereal bars were $9.4\%$, and puffed-type cereal bars were $10.1\%$. L-values of sunsik cereal bars were lower than that of puffed type cereal bars, and a-value was the highest in sunsik-type cereal bars with fructooligosaccaride. Texture measurement showed that hardness of sunsik-type cereal bars was higher than that of puffed-type cereal bars. Sensory evaluation resulted that sunsik-type cereal bars showed the high quality Score.

Studies on the Sensory Characteristics of Korean Tea and Related Products (국산차(國産茶)의 관능적 품질특성에 관한 연구)

  • The sensory quality characteristics of 7 different types of Korean traditional tea products were analyzed. For the standardization of sensory testing condition, the optimum drinking temperature were measured with 50 students, and all the samples tested were found to fall in the range of $60-70^{\circ}C$. The optimum concentrations of tea for drinking were generally met with the amount recommended by the producer. A total of 45 sensory describing terms expressing the taste, odor, and mouthfeel were collected. Using the sensory describing terms as the character notes, flavor profile analysis was made for each tea product with 8 members of trained panel. The differences in quality characteristics of 29 test samples were evaluated and shown in the chart constructed by the quantitative descriptive analysis method.

Physicochemical Treatment for the Reduction of Fusarium spp. Infested in Adlay (Coix lacryma-jobi L.) Seeds (율무 종자 오염 Fusarium 속 진균 저감화를 위한 이화학적 처리)

  • Background : The aim of the present study was to identify an effective physicochemical control method to reduce Fusarium species infestation in adlay (Coix lacryma-jobi L.) before and after harvesting. Methods and Results : We observed that prochloraz emusifiable concentrate and hexaconazol prochloraz emusifiable concentrate strongly inhibited the mycelial growth of 10 Fusarium species. Strong growth inhibitions and cell lysis were observed following treatment with 4% NaOCl solution. The total number of fungi detected were lower follwing treatment with thiophanatemethyl triflumizole wettable powder ($1.1{\times}10^4CFU/g$), hexaconazol prochloraz emulsifiable concentrate ($1.2{\times}10^4CFU/g$), carboxin thiram dustable powder ($1.6{\times}10^4CFU/g$) and prochloraz emulsifiable concentrate ($1.7{\times}10^4CFU/g$) than in the non-treated control ($7.7{\times}10^4CFU/g$). The reduction of Fusarium fungi varies with the concentration and soaking time of NaOCl solution. Fungal detection was not observed after soaking in NaOCl solution for 24 h and harmful effects were not observed for plant growth by NaOCl after soacking for 6 - 12 h. Conclusion : Soaking seed for 6 - 12 h in 4% NaOCl could be an effective method of disinfectant treatment for the control of Fusarium fungi in adlay seeds.

Seed Transmission of Bipolaris coicis, B. cynodontis, B. maydis and Curvularia lunata causing Leaf Blight of Job's tears (율무에 잎마름 증상을 일으키는 Bipolaris coicis, B. cynodontis, B. maydis 및 Curvularis lunata의 종자전염)

  • Bipolaris coicis, B. cynodontis, B. maydis and Curvularia lunata were leaf blight fungi detected from 45 seed samples of Job's tears and B. coicis was the predominant species in seed samples followed by C. lunata, B. cynodontis and B. maydis. When the seed components were plated on test tube agar, B. coicis and C. lunata were highly detected from invelucre, glume, endosperm and stamen, but not detected from plumule. Seed infection with B. coicis casued seed rot, coleoptile blight and seedling blight of Job's tears. Conidial characteristics of leaf blight fungi were as follows; B. cynodontis was fusiform, brown, slightly curved, 0~5 distoseptate, and 16.8-48$\times$7.2-16.8${\mu}{\textrm}{m}$ in size; B. cynodontis was fusiform, brown, slightly curved, 0~8 distoseptate, and 16.8-72$\times$9.6-19.2 ${\mu}{\textrm}{m}$; B. maydis was fusiform, brown, distintly curved, 0~10 distoseptate, and 28.8-110.4$\times$12-21.6 ${\mu}{\textrm}{m}$. C. Lunata was fusiform, brown, typically curved, 0~3 distoseptate, and 7.2-24$\times$4.8-12 ${\mu}{\textrm}{m}$. B. coicis was highly pathogenic to Job's tears and corn, weakly pathogenic to rice, but not pathogenic to wheat and barley. c. lunata was highly pathogenic to Job's tears (No. 2), corn, wheat and barley, weakly pathogenic to Job's tears (No. 1), but not pathogenic to rice. All treatments were effective to inhibition of leaf blight fungi when carboram, benoram, fludioxonil, prochloraz, thioram, and tap water treated to infected seeds.

Identification of Seed-borne Penicillium spp. on Gramineae Crops Based on Morphological Characteristics (형태적 특성에 의한 벼과작물 종자전염 Penicillium spp.의 동정)

  • A total of 81 isolates of Penicillium were isolated from postharvest seeds of barely, Job's-tears, maize, sorghum and rice from 1997 to 2003. Based on the morphological characteristics, they were identified as P. chrysogenum, P. citrinum, P. cyclopium, P. oxalicum, P. polonicum, P. purpurogenum and P. viridicatum. P. chrysogenum was detected from Job's-tears, rice and sorghum seeds, P. citrinum from maize seeds, P. cyclopium from sorghum seeds, P. oxalicum from barely, maize, sorghum and rice seeds, P. purpurgenum from maize, rice, sorghum seeds, P. viridicatum from Job's-tears, maize and rice seeds, P. polonicum from Job's-tears, maize, rice and sorghum seeds. Among these species, P. cyclopium, P. polonicum and P. purpurogenum were first reported in Korea. Especially, about 50% of the Penicillium isolates detected from the seeds were P. polonicum. Identification of the Penicillium species using morphological characteristics was difficult especially for the species belonging to the subgenus Penicillium such as P. polonicum.

Effects of Food Material Water Extracts on Content of Testosterone and Dihydrotestosteron in Serum and Skin of Rat (식품소재 물 추출물이 쥐 혈청과 피부의 Testosterone 및 Dihydrotestosteron 함량에 미치는 영향)

  • Effect of mixture of food material water extracts(modouri) composed of Job's tears, maize, buckwheat Japanese mushroom, lovage, licorice and jujube(13 : 50 : 15 : 2 : 5 : 5 : 5, v/v)] on the content of testosterone(T) and dihydrotestosterone(DHT), biochemical and histological changes of rat were investigated. Animal experiments(30 rats) were divided into 3 experimental groups(control, modouri and propecia). The summarized results were as follows: Activities of GOT, GPT, ALP, ${\gamma}$-GTP and content of total cholesterol and total lipid are normal in modouri group. Therefore modouri does not give rise to any damage in the liver. Also in the histological view, modouri does not have any hepatotoxic effect and increase the number of hair folicle. Total(T+DHT) and DHT content in rat serum and skin are significantly decrease in modouri group compare to the control but there is not any significant difference with propecia.

Antigenotoxic Effect of Dominant Bacterial Isolates from Kimchi in vitro (김치로부터 분리된 우세 균주들의 in vitro 항유전 독성효과)

  • This study was carried out to isolate the colonies of dominant fermented bacteria from Kimchi (Korean native fermented foodstuffs) and investigate their inhibitory potentials on mutagenesis and carcinogenesis induced by N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) as direct carcinogen. For this purpose, single cell gel electrophoresis technique (SCGE assay, or comet assay) which is a sensitive and rapid technique for detecting the presence of DNA strand breaks in individual cells was used. DNA damages of Kimchi isolates were compared with that of the positive control, MNNG. Among 3 isolates from Tongbaechu Kimchi, two isolates B-1 and B-2 showed antigenotoxicities (p<0.01). All 4 isolates from Yulmu Kimchi had antigenotoxicities (p<0.05). Also, 3 of 5 isolates from Chonggak Kimchi and 2 of 9 isolates from Kaktugi were antigenotoxic (p<0.05 and p<0.01, respectively).

Extracts of Adlay, Barley and Rice Bran have Antioxidant Activity and Modulate Fatty Acid Metabolism in Adipocytes (율무, 보리, 미강 유기용매 추출물의 항산화능과 포도당 및 지방산 대사에 미치는 영향)

  • Adlay, barley and rice bran were extracted using various concentrations of methanol(10% and 80%) and chloroform : methanol(2 : 1) to examine the biological activities of these raw grains. Extraction with 80% methanol resulted in high Vitamin C Equivalent Antioxidant Capacity(VCEAC), in the order of barley > rice bran > adlay, as determined by DPPH and ABTS assays. In addition, the extracts of adlay and rice bran showed high cellular antioxidant activity in HepG2 cells possibly due to the presence of polyphenol glycosides in these grains. We examined the expression of glucose/fatty acid metabolizing genes in differentiated 3T3-L1 adipocyte cells. Glut1 was downregulated after treatment with rice bran and no changes in the expression of Glut4 was observed. In contrast, genes involved in fatty acid metabolism, CD36 and aP2, were upregulated. Since these physiological changes were matched with peroxisome proliferator activating receptor $\gamma$(PPAR $\gamma$) agonism, we suggest that the extracts from adlay, barley and rice bran may play preventive roles against aging and diabetes via antioxidant activity and increased uptake of fatty acids by adipocytes.
