Ultrasound-assisted Extraction of Total Flavonoids from Wheat Sprout: Optimization Using Central Composite Design Method (밀싹으로부터 플라보노이드성분의 초음파 추출 : 중심합성계획모델을 이용한 최적화)

  • The process of extracting active ingredients from wheat sprout using ultrasound assisted method was optimized with a central composite design model. The response value of the central composite design model established the extraction yield and the total flavonoids content, main effects and interactive effects were analyzed depending on independent variables such as the extraction time, volume ratio of ethanol to ultrapure water, and ultrasonic irradiation power. The volume ratio of ethanol to ultrapure water and ultrasonic irradiation power were relatively large for the extraction yield and the extraction time was most significantly affected the total flavonoids, Considering both the extraction yield and total flavonoids content, the optimal extraction conditions were as follows: the extraction time of 17.00 min, volume ratio of ethanol to ultrapure water of 50.25 vol%, ultrasonic irradiation power of 551.70 W. In this case, the extraction yield and total flavonoids content were 28.43 wt% and $29.99{\mu}g\;QE/mL\;dw$, respectively. The actual experimental extraction yield and total flavonoids content under this condition were 8.73 wt% and $29.65{\mu}g\;QE/mL\;dw$, respectively with respective error rates of 1.05 and 1.13%.

  • 동양의학에서 강정 강장제로서의 귀히 여겨지는녹용의 유효성분과 또한 녹각과 그 유효성의 차이를 찾기 위해 무기질, 아미노산과 지방산 조성 등을 비교 검토하였다. 무기질 조성은 녹용과 녹각에서 Ca의 함량이 제일 높았고, Na과 K 함량도 상당히 높았으며 Hg 등 중금속은 전혀 검출되지 않았으며, 녹각에서는 Mn과 Fe이 미량 검출되었다. 녹용의 지방산은 포화지방산보다 불포화지방산 함량이 월등히 높았으며 그 중에서도 동맥경화증을 경감시킨다고 하는 docosahexaenoic acid가 43.73%로 제일 높았고, 녹각에서는 녹용에 비해 전반적으로 함량이 낮았다. 아미노산은 녹용에서 총 17종이 검출되었는데, glycine 함량이 제일 높았고, alanine과 proline 순이었다. 이는 collagen이 녹용의 상당부분을 차지한다는 것을 시사하며, 녹각에서는 비록 함량은 적었지만 10종이 검출되었으며, glutamic acid, proline, glycine과 alanine 등도 상당히 함유되어 있었다.

녹용 물추출액이 흰쥐혈액중의 급성기 반응 단백질에 미치는 영향

  • 녹용의 알려진 약리작용 중에서 녹용의 상용량 투여에 의해 나타나는 것은 강장작용과 창상치유작용이므로 이러한 작용이 진정한 녹용의 효능으로 간주될 수 있으나 이러한 작용을 생물활성 검색법으로 활용하여 녹용의 유효성분을 검색하기에는 재현성과 간편성이라는 면에서 어려운 점이 없지 않다. 녹용의 효능이 생체의 질소평형이 음의 방향으로 진행되었을 때 이를 정상 수준으로 복귀시키는 생물활성에 기인할 것이라는 가정하에서, 실험적으로 생체내 질소편형을 음의 방향으로 전환시킬 수 있는 급성기 반응 (acute-phase responce)을 야기시킨 다음 녹용의 효과를 연구하였다. 흰쥐의 급성기 반응물질 (acute-phase reactants, APR) 중에 서 positive ARP인 ceruloplasmin, $\alpha$-cysteine pretense inhibitor(= T-kininogen)는 비색정량법으로 간편하게 측정될수 있음에 착안하여 이 단백질을 지표로 삼아 turpentine oil로 급성 염증을 유발시킨 전후에 녹용 물추출액의 효과를 측정하였다. 그 결과 녹용의 상용량에서 괴소 4일 이상 경구투여한 흰쥐에서 급성 염증유발 후 4일에서 ceruloplasmin 의 혈중농도가 현저히 감소함을 알수 있었다. 급성 염증 유발후 녹용의 경구투여한 실험에서는 적어도 녹용 상용량의 3배 이상을 투여하여야 만 ceruloplasmin 혈중 농도를 감소시킬 수 있었다. 본 연구에서 수립된 생물활성을 이용하여 녹용의 유효 성분을 단리할 계획이다.

  • 우리나라 사람들에게는 유달리 높은 간염 발병률이 나타나고 있고 더우기 바이러스성 간염의 경우 풍토병 내지 전염병이라고 할 정도의 양상을 띠고 있다. 현재까지는 이러한 간질환에 적절한 치료약이 없는 점을 감안하여 저자들은 생약으로 부터 간질환에 유효한 성분을 분리하여 치료약으로 사용할 수 있도록 하기위하여 집중적인 연구를 지난 5년간 계속하여 왔다. 연구의 내용은 동서양의 고전및 연구보문을 조사하여 간보호 및 치료학으로 사용한 생약에 관한 문헌적 조사, 이들 생약중 채집 및 구입이 가능한 식물을 구하여 생약엑기스 제조, 이들 엑기스를 간염 동물 모델의 하나인 $CCl_{4}$로 유발시킨 간독성에 대한 보호작용의 검색, 보호작용을 나타내는 생약중에서 자원적 측면을 고려하여 국내에서 많이 생산되는 차전자(Plantago asiatica seeds)를 선택하였고, 차전자로 부터 간보호 작용을 나타내는 유효성분으로 iridoid 계열물질인 aucubin을 분리하였다. 그러나 aucubin이 차전자에 소량 밖에 없으므로 aucubin이 다량 함유된 식나무(Aucuba japonica)로 aucubin 추출자료생약을 바꾼후 aucubin의 간보호 작용을 간염 동물모델인, $CCl_{4}$ D-galactosamine 및 $\alpha$-amanitin등으로 유발시킨 간독성에 대한 보호작용을 보여 주었기 때문에, 이러한 간보호작용의 기전을 규명하는 연구를 진행하였고, 다음은 aucubin 자체의 급성 독성 및 기타 독성 유발여부를 밝히는 연구 등으로 요약 될 수 있다.

  • One dimensional unsteady heat conduction was studied on soybean curd as a water-protein-fat food system. This heterogeneous soybean curd could be treated as homogeneous material as for the unsteady heat conduction by 'effectiver' thermal diffusivity. Measurements were made with a apparatus designed and constructed by the author and also made with ice to establish the reliability, the reproducibility and the accuracy of the apparatus. It was found that the effective thermal diffusivity of soybean curd was substantially dependent on the fraction of protein and fat as well as water.

  • This experiment was conducted for the production of alismatis rhizoma (Alisma plan­tago-aquatica var. orientale Samuelsson) with high root tuber yield and alisol-B monoacetate content by treatment of different fertilizers levels. The results obtained were as follows: Shoot growth tended to in­crease as the fertilizers levels increased. Root tuber yield increased by 25% and 20% with 20-10-20 and 30-15-30 treatment, respectively. Alisol-B monoacetate contents increased as fertilizers levels increased, being the highest in 30-15-30 treatment.

Changes of Flavor Compounds in Persimmon Leaves(Diospyros kaki folium) during Growth (감잎의 성장시기별 향기성분의 변화)

  • This study was conducted to investigate the change in volatile flavor components of persimmon leaves during growth. The flavor components of persimmon leaves were analyzed by gas chromatography (GC) and combined gas chromatography-mass spectrometry(GC/MS). The flavor compounds were collected by simultaneous steam distillation and extraction(SDE) method, and were separated and identified resulting in 126 components, including 23 alcohols, 18 aldehydes, 4 esters. 15 acids, 37 hydrocarbons, 14 ketones, 6 phenols. and 9 others in persimmon leaves. The most abundant components of persimmon leaf were alcohols including iinalool. cis-3-hexanal. 1-$\alpha$ -terpineol. 3.7.11.15-tetramethyl-2-hexadecen-1-ol and aldehydes including trans-2-hexanal. nonanal, 2-decenal and hydrocarbons including 1.1-dimethylethyl cyclohexane, 1-methyl-4-(2-methylpropyl) -benzene. During growth, many other components were formed and dissipated after the 20th of June.

  • This paper presents consideration on validity of instantaneous correlation power theory. The proposed power theory is defined and analyzed by time domain approach, thus it is easy to understand and instrument. The power is decomposed into active, fundamental reactive and harmonics components based on the autocorrelation and crosscorrelation signal techniques between voltage and current waveforms. On the compensation property, active power filter deal with three components only. Also, for real time control of active power filter, the power models with difficult concept are not cost effective. To verify the validity of the instantaneous correlation power theory, experimental work for voltage type DSP based active power filter is achieved. The power of thyristor controlled motor drives is decomposed into three orthogonal components by proposed power theory. From compensation results, validity of proposed theory is confirmed. feedback controller needs the information on some motor parameters. New recursive adaptation algorithms for rotor resistance and mutual inductance which can be applied to our nonlinear feedback controller are also presented in this paper. The recursive adaptation algorithms make the estimated values of rotor resistance and mutual inductance track their real values. Some simulation and experimental results show that the adaptation algorithms are robust against the variation of stator resistance and stator inductance.

  • Harmonics and fundamental reactive current of nonlinear loads in serious unbalanced power condition, are compensated by current synchronous detection(CSD) theory which is also acceptable for single phase power system. But, the CSD theory is not suitable any more, in case of controlled independently harmonics and reactive component. Therefore, a new algorithm, the expanded current synchronous detection(ECSD) theory for a three phase active power filter based on decomposition of fundamental reactive, distorted components, is proposed in this paper. The proposed ECSD theory is experimented and tested comparison with a few power theories under asymmetrical condition in power system.

Growth and Mineral Composition of Young Radish in Soils Amended with Sewage Sludge and Lime (하수오니 및 석회시용이 열무의 생육과 화학성분에 미치는 영향)

  • Growth and mineral composition of young radish in soil amended with sewage sludge (ISS) with or without liming were investigated. Levels of EC, T-N, and micronutrients were increased in the ISS treatment (50 Mg/ha) compared to the NPK treatment but those of available P and exchangeable cations were decreased. Chlorosis was appear on the young radish leaf at the ISS treatment, but this phytotoxic symptom was corrected with lime amendment (3 Mg/ha). Yields of young radish leaf and root in the combined treatment of ISS and lime were increased about 29 and 48%, respectively, compared with those in the ISS treatment. Contents of P, K, Ca, and Mg in young radish were higher in the combined treatment to ISS and lime, whereas those contents in the ISS treatment were lower than those in the NPK treatment. Contents of Cu, Zn, and Ni in soil and young radish leaf were positively correlated with total N content in young radish leaf, while contents of P, K, Ca, and Mg were negatively correlated. Contents of Cu, Zn, and Ni in leaf were negatively correlated with yield of young radish. Contents of Cu and Zn in leaf corresponding with a 5% yield reduction of young radish were 22.4 and 349 mg/kg, respectively.
