Measurement of 150 kDa protein of Taenia solium metacestodes by antibody-sandwich ELISA In cerebrospinal fluld of neurocystlcercosls patients (뇌 유구낭미충증 각자 혈청 및 뇌척수액에서의 유구낭미충 낭액항원의 측정)

  • An antigenic protein in cystic quid of Taenia solium metacestodes (CF) of 150 kDa was measured by antibody-sandwich ELISA in serum and cerebrospinal quid (CSF) of neurocysticercosis patients. Capture antibodies were rabbit antisera against CF (RACF) and a monoclonal antibody (MAb) against 150 kDa protein in CF. Lower limit of antibody-sandwich ELISA was 8 ng/ml of the protein. Except CF, no tested helminthes extracts reacted. Levels of the protein in 351 sera from 255 patients (55 surgery confirmed and 202 antibody and CT/MRl confirmed) were below sensitivity of the assay. Of 276 CSF from 212 patients, 31 samples (11.2%) showed positive findings. This assay, therefore, was not sensitive enough to be a diagnostic. Instead, the 150 kDa protein appeared in CSF in such situations as in 2 days after prasiquantel treatment, or as in a patient infected with a racemose cysticercus with degenerated cyst wall. Of cases whose follow-up CSF were assayed, 2 cases showed that the protein appeared intermittently, These results suggest strongly that appearance of free 150 kDa protein is associated with cyst wall rupture. In CSF which contained the 150 kDa protein over 61 ng/ml, the protein was recognized in 505-PAGE before and after immunoprecipitation.

The changes of histopathology and serum anti-sparganum IgG in experimental sparganosis of mice (마우스 피하 스파르가눔증에 있어서 감염 경과에 따른 조직병리학적 병변 및 혈청 항체가의 변화 양상)

  • The present study is intended to observe the chronologic changes of experimental sparganosis by histopathological observation and detection of circulating anti-sparganum IgG antibody using ELISA. Each of 25 mice was infected with aye spargana, and they were examined after 1, 2, 4, 10 weeks or 6 months from infection. The followings are summarized results. - 1. The plerocercoids were detected in the subcutaneous tissue of the trunk, neck or axilla, but a few often extended into the skeletal muscle. The recovery rates were 72% at the first week, 80% at the second week, 95% at the fourth week, 92% at the tenth week and 100% at .the sixth month. The larvae grew slowly in both length and weight until 6 months. 2. Histopathologically, most of the larvae were observed alive in the soft tissue or skeletal muscle. Numerous eosinophils, neutrophils, Iymphocytes and plasma cells were infiltrated focally around the worms by the second week, but they surrounded the worms to form a layer of inflammatory reaction after 4 weeks of infection. Also histiocytes and fibroblasts began to appear around the inflammatory cells at 4 weeks. After 10 weeks, the worms encircled by a thin fibrous layer were found. After 6 months, the worms were surrounded by either fibrous tissue or active inflammatory cells. The inflammation looked more severe in the tracks left by the worms, rather than around the worms. 3, The level of anti-sparganum IgG antibody in the serum showed an increase by the fourth week, and a rapid and continuous increase was observed thereafter by the tenth week after infection. The high level of the IgG antibody was maintained up to 6 months forming a plateau curve. The present results suggest that the tissue reaction and antibody production in subcutaneous sparganosis become distinctive by the fourth week after infection.

  • The sensitivity and specificity of crude and affinity-purified antigens of Clcnorchis sinensis obtained from the infected rabbits were studied. Stage-specific antigenic proteins from the eggs, metacercariae and adult worms were characterized by SDS-polyacrylamide gel electrophoresis (SDS-PAGE) and enzyme-linked immunosorbent astray (ELISA). The results were as follows: 1. The antibody.binding antigen (ABA) purified from whole worm crude antigen (IVWA) by CNBr-activated Sepharose 4B affinity chromatography made :l specific bands against rabbit antisera on Ouchterlony gel diffusion plate, while WWA made 7 bands. Major WWA protein bands by SDS-PAGE were found at 16, 300~18, 500 and 28, 000~29, 000 daltons, while major ABA protein bands were at 18, 000~21, 000 and 29, 000~31, 000 daltons. The reactivity of ABA with rabbit anti-sera in ELISA was remarkably less sensitive than that of WWA. 2. Molecular weights of egg antigen (EGA), metacercarial antigen (MEA) and adult worm antigen (WWA) of C. sinensis ranged from 15, 000-200, 000 daltons, 15, 000-100, 000 daltons and 11, 000~80, 000 daltons, respectively. Major WWA proteins consisted mainly of polypeptide bands of low molecular weight, less than 31, 000 daltons, while those of EGA and MEA consisted of higher molecular T.eights than 30, 000 daltons. 3. The ELISA reactivities of WWA to rabbit anti.sera were remarkably greater than those of MEA. EGA showed negative reaction throughout the experiments. WWA showed higher optical density (O.D.) than 1.0, when reacted with rabbit anti-sera obtained at 4~6 weeks after the infection. In the rabbit anti-sera later than 12 weeks after the infection, the O.D. reacting witll WWA showed a plateau without variation. MEA shoT.ed relatively low O.D. values (<0.6), when reacted with anti-sera from lightly in(ected groups throughout the experiments, althougll there were some wealth positive cases (O.D.>0.6) ill heavily infected groups. MEA reacted with rabbit anti-sera showed negative results on Ouchterlony gel diffusion plates. Summarizing the above results, it is suggested that the whole worm antigen prepared from the adult worms of C. sinensis is most highly antigenic. However, this antigen might reveal cross reactions with other trematodes such as Paragonimus westermani, therefore, purification of antigenic proteins from the crude antigen is essential 18 increase the sensitivity and specificity for the immuncdiagnosis of clonorchiasis.

  • Biological control against mushroom fly, Lycoriella mali, was performed by using Bacillus thuringiensis subsp. israelensis Bti-D and Bti-U, isolated from dead mushroom fly in oyster mushroom houses. Control values of the bacterial strains Bti-D and Bti-U against L. mali in bottle culture of oyster mushroom were 74.4% and 64.2%, respectively, and the value in small tray culture were 75.8% and 56.8%, respectively. In the experiment to develop the mass, cheap media for Bti-D and Bti-U isolates, the Biji broth (bean curd residue, called Biji in Korean language) was selected as a culture medium for an inexpensive and mass cultivation by the measurement of optical density of the two bacteria grown in the different media tested. Insecticidal effect of the formulation contained different ingredients that were prepared by using the Bti-D strain cultured in the Biji broth was tested in tray and bottle culture of oyster mushroom. The WCS formulation that contained corn starch as bio-gel (86.4%) was more effective to control the mushroom fly than living cells (69.1%) in bottle culture of oyster mushroom. Moreover, insecticidal effect of the WCS formulation was improved when water of pH 8 was used for dilution of the formulation. Effect of the WCS formulation using water of pH 8 and chemicals, Zuron (dimillin) W.P. on the control of mushroom fly and the productivity of oyster mushroom was investigated in tray culture of oyster mushroom. The Zuron W.P. was more effective to control the mushroom fly than the WCS formulation. However, compared with no treatment, the productivity of the mushroom treated with the WCS formulation was improved than that of the mushroom with Zuron W.P.

Effects of the Molting-Hormone Mimetid Insecticide Tebufenozide: on Chironomus riparius Larvae (탈피교란물질인 Tebufenozide가 Chironomus riparius (Diptera: Chironomidae)에 끼친 영향)

  • We investigated the effects of molting-hormone insecticide tebufenozide on D7 (the day of hatching from egg) larvae of the midge Chironomus riparius in growth developments. D7 instar larvae were exposed test concentrations were chosen control, 10${\mu}g \;L^{-1}$, 30${\mu}g \;L^{-1}$, 60${\mu}g \;L^{-1}$ and 100${\mu}g \;L^{-1}$ of tebufenozide. In general, dead larvae showed 16% on the next day after insecticide treatments (D12), and observed 44% from D12 to D16 in this exposed days. Dead larvae of C. riparius was abruptly increased on D12 and also continuously increased along the days in 10${\mu}g \;L^{-1}$ treatments. The converged day was from D12 to D16 at move 30${\mu}g \;L^{-1}$ treatments in this study. Therefore, dead larvae obviously increased along these concentrations of tebufenozide. In control condition,78% of the test individuals have grown the pupae. But the larvae have developed the pupa stage from 5% to 17% of the test organism in 10${\mu}g \;L^{-1}$ and 30${\mu}g \;L^{-1}$ treatments. And 75% of the test individuals was arrived the adult through the molting process in control condition. While the other condition was rarely observed the adult. Usually, the emerged period of the test individuals was gathered the D26-D29 in control. The dead pupa showed from D19 to D20 in 30${\mu}g \;L^{-1}$ treatments, D32 in control and D33 in 10${\mu}g \;L^{-1}$ treatments. The observed periods of dead pupa were D32-D34 in control and D33-D37 in 10${\mu}g \;L^{-1}$ treatments. Consequently, due to molting hormone disruption, development of midge was postponed relatively low concentration such as 10 treatments of tebufenozide.

Studies on INtestinal TRematodes in Korea XVII. Development and Egg Laying Capacity of Echinostoma hortense in Albino Rats ans Human Experimental Infection (한국의 장흡충에 관한 연구 XVII. 호르텐스극구흡충의 흰쥐내 발육, 충란산출양상 및 인체실험감염)

  • The worm development and egg laying pattern of Echinostoma hortense (Trematoda; Echinosto-matidae) were studied in albino rats and the brief clinical course was observed in human volunteers. A total of 21 rats were infected with 20~69 metacercariae each and two humans were with 7 and 27 metacercariae, which were collected from the loaches. For recovery of worms, the rats were sacrificed at irregular intervals from the 6th to 150th day after infection and the human volunteers were treated with praziquantel and purged with magnesium salt on the 26~27th day. The stools of the rats and humans were examined for the eggs. The results were as follows: 1. The worm recovery rate from the rats was not affected by the increase of infection time but varied individually; 9.1~50.0% (31.1 % in average). From humans, 14.3% and 37.0% (32.4% in average) of challenged were recovered. 2. In the rats, it was revealed that the worms rapidly grew for the first 14 days to become 7.59mm in average length and 1.17mm in average width but the growth became much slower thereafter until the 150th day; 7.95mm in length on the 21 th day, 9.04mm on the 28th day, 10.21mm on the 49th day and 12. 62mm on the 150th day. During the early stage of infection, the growth of genital organs (male or female) was expressed as sigmoid curves whereas non-genital organs (such as suckers) was simply as straight lines. 3. The prepatent period of this fluke was 10~12 days in the rats and 16~17 days in men. After the start of oviposition, the egg production by the worms remarkably increased, reached maximum on the 32~33th day, followed by decrease thereafter. The maximum value of E.P.G./worm was 390. 4. The major subjective symptoms in human volunteers were abdominal pain and diarrhea during the early stage of infection. The results show that human is as susceptible as the rats to E. hortense infection and the amount of egg production in the rats is greatly affected by the age of worms.

  • This study was performed to observe the chronological changes in the tegumental structure of M. yokogawai using scanning electron microscope. The subjected worms were encysted metacercariae obtained from the sweetish, and 2-day, 1-week and 4-week old worms experimentally reared in albino rats. The results are as follows: 1. The tegument of encysted metacercariae showed many transverse shallow rugae, which were more remarkable in posterior half body, i.e., posterior to ventral sucker. The whole surface was armed with many scale-like spines; 7~8 pointed ones on anterior body and 2~3 pointed on posterior body. The ciliated knob-like papillae (Type I) were abundant around oral and ventral suckers, which grouped 2, 3 or 4 in number in most cases. A few round swellings of tegument(Type lI were observed only on oral sucker. 2. The tegumental surface of 2-day old worms showed deeper rugae, and the anterior half covered with knob-like processes of distal cytoplasm and the posterior half with cobblestone-like ones. Interspinous space became more wide and 9 pointed spines appeared on anterior dorsal surface. The sensory papillae enlarged but not changed in their distribution. 3. The tegument of 1-week old worms revealed knob-like cytoplasmic processes in posterior half body and velvety ones around oral sucker. The scale-like spines of anterior half body changed remarkably to the slender ones of posterior body at the level of ventral sucker. In dorsal surface, the arrangement of the Type I papillae were bilaterally symmetrical. 4. The tegument of 4-week old worms were finely differentiated and the posterior tegument covered with velvety cytoplasmic processes. The spines had remarkably grown in length and width but the density remained nearly unchanged. The papillae also became larger but their morphology and distribution were not different from younger worms. However, the round elevation of cytoplasmic ridges (Type III papilla) appeared bilaterally on inner wall of oral sucker, approximately 8 in number. From the above results, it is considered that the tegument of juvenile iH. yokogawai continued to differentiate until 4 weeks after infection.

Effect of anthelmintics on the early stage of Enterobius vermicularis (요충 초기 발육단계에 대한 구충제의 효과)

  • In order to determine the susceptible age of Enterobius vermicular is to anthelmintics and to observe the chronologie growth of female E. vermicularis in man, experimental infections were done. About 500 eggs were challenged to 19 volunteers. After 4, 8, 16, 20, 24, 28, 32 and 35 days of infection, each case was treated by either mebendazole or pyrantel pamoate. On the 40th day of infection all cases including control were treated again to terminate the experimental infection and to evaluate the effect of previous treatment. Each case collected 3-day stools to harvest the expelled worms. The results could be summarized as follows: 1. The infection rates of females were in range of 0.6~13.1 % in control cases. Because the collected worms showed comparable growth and development by day, the worms were concluded to be derived from experimental infection. 2. Cases that were treated with mebendazole on 4, 8 and 16 days after infection expelled 37.5%, 2.5% and 67.5% of the number expelled by a control case on the 40th day. Cases treated thereafter expelled no worms on the 40th day. 3. Cases that were treated with pyrantcl pamoates on 4, 8, 16, 24, 28, 32 and 35 days, expelled 90.7%, 25%, 45.3%, 8%, 2.7%, 5% and 29.3% of the number collected from control cases in respect. 4. All the worms collected were females. The total body length increased consistently and comparably from the 20th day of infection. Those collected on the 20th day were 2.5~3.0 mm long with vagina, sac-like structure and strands of ovaries; 24 day-old worms may have short uterus, 28 day-old worms had long uterus without eggs, 32 day-old worms began to produce eggs, 35 day-old worms showed wide variations in egg deposit in uterus, and 40 day-old worms had uterus filled with eggs from vulva to anal levels. From the above results, it was inferred that the life span of female Enterobius vermicularis was longer than 40 days, and the developmental stages of worms younger than 16 days resisted considerably to both mebendazole and pyrantel pamoate.

Studies on the mechanism in the induced to unfertilized eggs(male sterility of Silkworm) by protected environment during pupae period (용기의 보호환경에 따른 불수정란(웅성불임잠)의 유발기구에 관한 조사연구)

  • In order to find out effects of the generative power of silk worm moth which have been brought up in the high temperature accommodation at their pupa stage. For this specific study, 9 different kinds of male silk worms have been selected as specimen. All those specimen were brought up in the normal temperature at their larvae stage and after the pupation period they have been accommodated in the condition of high temperature for a certain length in accordance with the study programme. Afterwards, those mlae specimen were copulated with Suwon jam 103${\times}$104 which were all brought up in normal conditions. This study was carried out to find the copulation function as well as the ratio of unfertilized eggs(male sterility test). Results of study have been revealed as follows: 1) Although some differences were observed, male pura which have been brought up in the condition of high temperature shown the low rate of unfertilized eggs rather than those were brought up in the normal conpition. 2) In this group the eclosion(emergency) has been found to be poor rather, than those specimen brought up in normal conditions. 3) The copulation function of Moran, Daedong, J124 and C108 specimen were found to be poor than those of Suwon jam. 4) Fertility rate of Moran, Daedong, J124 and C108 was found to be around 65%. This figure is rather lower than what we normally expect. 5) Unfertilized egg ratio of Moran, Daedong and C108 were found to be around 20% if they were brought up in the condition of high temperature for one day from the time of pupation: 40% at 2 days, and 70% at 3 days duration. More than 3 days treatment has shown no progress in the unfertilized egg ratio. 6) One day's treatment for the pupa at the later stage has shown the unfertilized egg ratio of about 10%; 20% at 2 day's treatment, 35% at 3 day's treatment, 40-60% at 4 day's treatment, more than 60% at 5 day's treatment, and the 70% of fertilized egg ratio was only observed when the treatment days come to 7 days. It was understood that the unfertilized egg ratio was high at the antepupa stage rather than that of post-pupa stage. 7) According to the result of observation the sperm in copulatory pouch and seminal receptacle out of the normal female silk worm which have been copulated with the male brought up in the condition of high temperature at their moth stage. The reproduction system found in the control zone has been found to be normal and the sperm is amountful and active in motion while the sperm found in the treatment to be limited in amount and slow in motion. The observation was made within 5 hours from the copulation. 8) According to the result of observation of sperm of seminal receptacles of the female silkworm moth, and according to that observation of sperm in the seminal receptacle in female silkworm moth, the amount of sperm and mobility in the female moth brought up in high temperature were poor comparing that were brought up in normal temperature zone. Some of them are even found to be no trace of such. 9) Appearance and mosle of the copulatory organ of the male silkworm moth was found to be no anatomical change. 10) Testis of the later pupa stage which was brought up in the high temperature was found to be almost net developed to anucleate sperm and they are degenerated at stage of between maturation division and sperm abnormal stage. Mean time at control zone, the formation of anucleate sperm was already observed.

Effect of Host Plants on the Development and Reproduction of Cotton Caterpillar, Palpita indica (Saunder) (기주식물이 목화바둑명나방의 발육과 생식에 미치는 영향)

  • The present study was aimed to examine the effect of different host plants on the biological property of cotton caterpillar, outbreak pest of cucurbit vegetables. The feeding and oviposition preferences toward cucumber and pumpkin were greater than those toward watermelon, oriental melon, and melon. The periods of egg and larva fed on oriental melon, melon, and pumpkin were longer than those on watermelon and cucumber. The longest pupal period was 10.5 days from cucumber. The hatching rate, pupation rate, emergence rate, and survival rate were observed from different host plants. The hatching rate was the highest on cucumber (87.2%) and the lowest on melon (72.8%). The pupation rate on watermelon and oriental melon (90.0% and 89.1%, respectively) was higher than that on cucumber (62.0%). The emergence rate on cucumber and pumpkin (93.5% and 92.0%, respectively) was higher than that on oriental melon (78.7%). The survival rate from hatching to emergence on watermelon (76.0%) was the highest, and the lowest on cucumber (50.0%). The adult period on melon (21.0 days) was the longest, and the shortest on cucumber (15.5 days) among 5 host plants. Average number of eggs per female on cucumber (281.8 eggs) was the highest, and the least on oriental melon (96.6 eggs). The survivorship on cucumber was the longest (30 days), and the shortest on pumpkin (17 days). The preoviposition period on oriental melon and melon (3.4 days) was longer than that on watermelon (2.1 days) and mean generation time in day (T) on melon was the longest (47.2 days) though they were not significant. The net reproductive rate per generation (R$_{o}$) on cucumber, 191.3, was the highest and the highest intrinsic rate of natural increase (r$_{m}$) was observed from cucumber as 0.127. All these results suggest that the growth ana reproduction of cotton caterpillar varies depending upon the kind of host plants. The analysis of the life table revealed that cucumber and pumpkin were the favorable host plants of cotton caterpillar.
