Ultrastructure of the Integument of Adult Paragonimus westermani (폐흡충 표피의 미세구조)

  • The present study was performed to observe the ultrastructure of the integument of adult Paragonimus westermani. Dogs experimentally infected with 60 metacercariae of F. westermani were autopsied 4 months after the infection. Adult p. westermani were extracted from the dogs and the fine structure was studied by means of scanning and transmission electron microscope. The findings are as follows: 1. Scanning electron microscopic findings showed that the spines and the papillae are distributed at whole body surface but the well developed spines or papaillae are shown around the oral sucker and ventral sucker. 2. At the end of the body, excretory pore was found, the shape was irregular. 3. Transmission electron microscopic findings showed that plasma membrane, tegument, basal lamina, connective tissue, circular muscle layer, longitudinal muscle layer. nerve axon and tegumental cell were observed. 4. In higher magnification, plasma membrance and bar-shaped granules were found at the outer surface of the tegument.

Acoustic Emission Characteristics and Fracture Behaviors of GFRP-Aluminum Honeycomb Hybrid Laminates under Compressive and Bending Loads (GFRP-알루미늄 하니컴 하이브리드 적층판의 압축 및 굽힘 파괴거동과 음향방출해석)

  • This paper investigated acoustic emission (AE) characteristics in association with various fracture processes of glass fiber reinforced plastic skin/ aluminum honeycomb core (GF-AH) hybrid composites under compressive and bending loads. Various failure modes such as skin layer fracture, skin/core interfacial fracture, and local plastic yield buckling and cell wall adhesive fracture occurring in the honeycomb cell wall were classified through the fracture identification in association with the AE frequency and amplitude analysis. The distribution of the event-rate in which it has a high amplitude showed a procedure of cell wall adhesive fracture, skin/core interfacial debonding and fiber breakage, whereas distribution of different peak frequencies indicated the plastic deformation of aluminum cell wall and the friction between honeycomb walls. Consequently, the fracture behaviors of GF-AH hybrid composites could be characterized through a nondestructive evaluation employing the AE technique.

The Dietary Effect of Royal Jelly Supplementation on Epidermal Levels of Filaggrin and Free Amino Acids during Menopause in Rats (폐경기 노화 유도 전후의 로얄제리 식이공급이 백서 표피의 필라그린과 유리아미노산 함량 및 관련 대사 효소의 단백질 발현 변화에 미치는 영향)

  • Epidermal hydration is mainly maintained by natural moisturizing factors (NMFs). Of these various NMFs, free amino acids (AAs) are major constituents generated by filaggrin degradation. The reduction of these AAs has been reported in aging skin induced during menopause. In this study, we examined whether the dietary supplementation of royal jelly (RJ) during the pre- and post-menopausal period alters epidermal levels of filaggrins, free AAs, and peptidylarginine deiminase-3 (PAD-3) (an enzyme involved in filaggrin degradation processes). Sprague Dawley rats were divided into five groups: groups fed a control diet for 12 weeks, in which an ovariectomy (OVX) or sham operation (SHAM) were underwent at week 4; groups fed a diet with 1% RJ harvested in different area of Korea (RJ1 and RJ2); and a group fed a diet with isoflavone (IF), the typical functional food for menopause prevention, for 4 weeks before and 8 weeks after an ovariectomy operation. In the epidermis of group OVX, total filaggrins (including profilaggrin and filaggrin) were reduced; these levels in groups RJ1 and IF were similar or less than in group OVX. However, total AAs, which showed no apparent difference between groups SHAM and OVX, were highly increased in groups RJ1 and IF. Specifically, aspartate (Asp) and proline (Pro), the major AAs in functioning NMF, were highly increased in group RJ1. Although total filaggrins, profilaggrin, filaggrin and PAD3 increased, total AAs (including Asp and Pro) in group RJ2 were modest or less than in group RJ1. The PAD3 alteration was not apparent among the four other groups. Taken together, we demonstrate that the diet supplementation of RJ1 enhanced filaggrin degradation (but not through the increased protein expression of PAD3), and increased total AAs, Asp and Pro. RJ1 could be a dietary supplementation for preventing the skin aging induced during menopause.

Dietary effect of green tea extract on hydration improvement and metabolism of free amino acid generation in epidermis of UV-irradiated hairless mice (자외선 조사와 병행된 녹차추출물 식이공급이 무모생쥐의 표피보습 개선 및 유리아미노산 생성 관련 대사에 미치는 영향)

  • Purpose: Ultraviolet (UV) irradiation decreases epidermal hydration, which is maintained by reduction of natural moisturizing factors (NMFs). Among various NMFs, free amino acids (AA) are major constituents generated by filaggrin degradation. This experiment was conducted to determine whether or not dietary supplementation of green tea extract (GTE) in UV-irradiated mice can improve epidermal levels of hydration, filaggrin, free AAs, and peptidylarginine deiminase-3 (PAD3) expression (an enzyme involved in filaggrin degradation). Methods: Hairless mice were fed a diet of 1% GTE for 10 weeks in parallel with UV irradiation (group UV+1%GTE). As controls, hairless mice were fed a control diet in parallel with (group UV+) or without (group UV-) UV irradiation. Results: In group UV+, epidermal levels of hydration and filaggrin were lower than those in group UV-; these levels increased in group UV+1% GTE to levels similar to group UV-. Epidermal levels of PAD3 and major AAs of NMF, alanine, glycine and serine were similar in groups UV- and UV+, whereas these levels highly increased in group UV+1% GTE. Conclusion: Dietary GTE improves epidermal hydration by filaggrin generation and degradation into AAs.

  • Lipid prooxidants in sardine skin was characterized. Prooxidants in the sardine skin extract with 0.05M phosphate buffer was purified by successive chromatography on Sephadex G-200, DEAE-Sephadex A-50 and CM-Sephadex A-50. Prooxidants of sardine skin exist mainly in the intermediate molecular weight fractions. Observations of the thermounstability and optimum pH(pH 7.0) suggest that the major prooxidants are enzymes and hemoproteins. They can oxidize well both free and esterified linoleic acid and form conjugated hydroperoxides. From these results, the major prooxidants in sardine skin are assumed to be lipoxygenase-like enzymes.

소 비장유래 Macrophage의 체외배양시 배양액 중 호르몬적 변화

  • 소에 있어서 수정란이식이 활성화되고, 수정란이식 기술이 첨단기술도입의 근간이 되면서, 수태율의 향상은 첨단기술의 활성화에 필수적인 요소가 되었다. 또한 수태율 향상을 위해 임신유지를 도와주는 progesterone의 정기적인 투여나 수정란이식 시기에 hCG 등을 투여하여 수태율을 향상시키고자 여러가지 방법이 시도되고 있으나, 수태율 향상에 대한 명확한 기술이 정립되지 않은 상태이다. 따라서 본 연구는 착상시 황체의 Progesterone의 분비를 촉진시키는 것으로 보고되고 있는 비장유래의 macrophage의 체외배양을 통하여 배양액 중 호르몬적 변화를 관찰하고 착상을 유도하는 기전을 밝히고자 시도하였다. 임신 및 비임신 도축암소의 비장을 채취하여 알루미늄박으로 포장하여 얼음에 채운 뒤 실험실로 운반하였고, 비장을 70% alcohol로 외부를 잘 세척한후 표피를 제거하였다. 표피를 제거한 비장조직을 가위로 잘게 세절하고 buffer A 용액으로 조직속의 세포를 분리하였다. 분리된 세포는 1,700 rpm으로 5분간 3회 세정하였으며, 세정된 세포는 유리 petri dish에 넣어 39$^{\circ}C$, 5% $CO_2$, 95% air인 배양기에서 2시간 동안 배양을 실시하였다.   • The albinic phenomenon of flat-fish (Paralichthys olivaceus) was investigated by measuring protein content, tyrosinase activity, amino acid composition, and contents of vitamin A and C. These materials in the flat-fish feed-stuff were also tested. The amount of skin protein was higher than that of muscle in normal flat-fish. Catechol and L-dopa oxidase activity did not differ between normal and albinic flat-fish. The free amino acid of skin in normal flat-fish was 7.5 times that in albinic one. Sulfur-containing amino acid in normal flat-fish was also 6.3 times that in albinic ones. Vitamin A was not detected in both of flat-fish. The content of vitamin C in normal flat-fish was 7.8 times that in albinic one. The contents of protein, sulfur-containing amino acid and vitamin C in micro-encapsulated feed (one commercial feed in Japan) were the highest among the feed-stuff used in this experiment. The melanin formation of flat-fish skin was affected by substrates such as aromatic amino acid and cofactor such as sulfur amino acid.

Changes in Physicochemical Characteristics of Squid upon Acid and Heat Treatment (산 및 열처리에 따른 오징어의 이화학적 특성변화)

  • In order to acquire basic data on the development of squid processing food, we investigated changes in the composition of boiled squid upon heat treatment ($100^{\circ}C$), acid treatment (acetic acid, 0~5%), and pre-boiling ($55^{\circ}C$, $80^{\circ}C$). The proximate composition of squid was 73~78% moisture and 19~24% crude protein, treatment with acid solution had a significant effect on the proximate composition of boiled squid (p<0.05). The major free sugars were ribose and glucose in all treatment samples. The $55^{\circ}C$ pre-boiled sample had lower levels of glucose than the other samples. The total free sugar content of the non-peeled sample was the highest, followed by the $80^{\circ}C$ pre-boiled sample, whereas the sugar content in the $55^{\circ}C$ pre-boiled sample was very low. With regards to amino acid content, proline was the highest in all samples, followed by taurine and histidine. Treatment with acid solution had a significant effect on the total free amino content of boiled squid (p<0.05). The total free amino acid content of the $55^{\circ}C$ pre-boiled sample was the highest, followed by the $80^{\circ}C$ pre-boiled sample and non-peeled sample. Inosine and related compounds were not detected in any of the samples, and the adenosine triphosphate (ATP) content was low. The hypoxanthine contents of the $55^{\circ}C$ and $80^{\circ}C$ pre-boiled samples were the highest, the adenosine monophosphate (AMP) and inosine monophosphate (IMP) contents were similar, and the IMP content of the non-peeled sample was higher than those of the peeled samples. The palmitic acid content was very high and constituted 40% of total saturated fatty acids. eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) contents were also high and constituted 60% of total unsaturated fatty acids. Of these, DHA content was the highest, followed by palmitic acid and EPA, which accounted for about 85% of total fatty acids. No difference in fatty acid content was observed between acid treatment and pre-boiling. The mineral P content was the highest on average in all boiled squid samples, followed by K, Na, Mg, and Ca contents. In addition, the pre-boiling temperature and acid solution concentration had significant effects on the mineral content. Further, heavy metal, Cd, Pb, and As contents were detected only at trace amounts, and their levels were lower than standard and permissible amounts for food.

Of various NMFs, free amino acids (AAs) are major constituents generated by filaggrin degradation. In this study, we attempted to determine whether dietary supplementation of royal jelly (RJ) in UV-irradiated mice can alters epidermal levels of hydration, filaggrins, and free AAs as well as of peptidylarginine deiminase-3 (PAD3), an enzyme involved in filaggrin degradation processes. Albino hairless mice were fed either a control diet (group UV+: UV irradiated control) or diets with 1% RJ harvested from different areas in Korea (groups RJ1, RJ2, and RJ3) or imported from China (group RJ4) for six weeks in parallel with UV irradiation. A normal control group (group UV-) was fed a control diet without UV irradiation for six weeks. Reduced epidermal levels of hydration, total filaggrins, and PAD3 were observed in group UV+; in group RJ1, these levels were increased to a level similar to that of group UV-. In addition, profilaggrins, two repeat intermediates (2RI), a precursor with two filaggrin repeats, and filaggrin were increased. Although no alteration of AAs was observed in any of the groups, and glutamate and serine, major AAs of NMF in group RJ1 were higher than in group UV+. Despite the increased levels of PAD3, epidermal levels of hydration, filaggrins, glutamate, and serine in groups RJ2, RJ3, and RJ4 were similar to those in group UV+. Dietary supplementation of RJ1 improves epidermal hydration in parallel with enhanced expression and degradation of filaggrin, but not by increased protein expression of PAD3, along with increased generation of glutamate and serine.

A Study on the Expressional Characteristic of Glass outer Skin in Contemporary Space Design - Focusing on the case after 2000 - (현대 공간디자인에 있어서 유리외피의 표현특성에 관한 연구 - 2000년 이후의 사례를 중심으로 -)

  • This study is about the Characteristic of glass outer skin expressions in space designs of contemporary society, due to the rapid development of information and digital technology. These days, the glass outer skin show the various images and space expressions which symbolize the informations and the society. The contemporary space designers focus on the glass as the structure materials, and purposely use the glass to achieve the structure concept which satisfies the needs of contemporary society. Therefore, it is a valuable work to analyze the meaning of glass, which has been used for the structure. Throughout the work which has been experienced by glass outer skin, theoretical studies have been achieved and recently the 14 examples were chosen as the glass structures based on the experimental research and frequency featured in magazines after year 2000, which we have analyzed in detail. Glass outer skin reflect the phrases of time in modern society, and throughout the meaning of it, we could see that the glass outer skin serve the roles in informations, interface, imaginary modernization, and as a landmark position. Therefore, in this study, we can make a definition of glass outer skin expression based on the situations of modem times by analyzing the symbolic meaning of glass outer skin in contemporary space designs.
