Extractive Nitrogenous Constituents and Their Monthly Variation of Fresh Laver Porphyra dentata (잇바디돌김의 함질소 엑스성분조성 및 월별변동)

  • In order to investigate the composition and monthly variation of extractive nitrogenous components in the laver Porphyra dentata cultured at the south coast of Korea, the fresh laver was analyzed separately for the amounts of free amino acids, combined amino acids, ATP and its related compounds, and quaternary ammonium basis in fresh laver were measured. The extractive nitrogen contents of fresh laver extracts were 760~870 mg/100g (dry basis). Twenty-seven to thirty-one kinds of free amino acids were detected in the laver extracts and their total amounts were 2,404~3,966 mg/100g (on dry basis). The laver extracts showed rich in free amino acids such as alanine, taurine, glutamic acid, glutamine and aspartic acid. Sixteen to twenty-three kinds of combined amino acids were detected in the extracts and their total amounts were 1,429~2,692 mg/100g (on dry basis). Proline, glutamic acid, glycine, phosphoserine, serine were the amin combined amino acids in the extracts. The amounts of ATP and related compounds were 73.3~94.4 mg/100 g (2.04~4.43 $\mu$mol/g, on dry basis). Homarine and trigonelline were detected in all specimens but $\beta$-alaninebetaine, ${\gamma}$-butyrobetaine were found in some. Small amounts of trimethylamine were detected in all samples. Free and combined amino acids were occupying almost 90% of extractive nitrogen. Most of free and combined amino acids showed a marked monthly variation with a maximum in January and March, and a minimum in February and April. The fresh laver P. dentata did not differ much from the fresh laver P. yezoensis in qualitative com-position of extractive components, but their contents were generally low level.

The Components of the Fermented Soy Sauce from Gorosoe and Bamboos SaP (고로쇠 및 대나무 수액간장의 성분조성)

  • Soy sauce was prepared with the addition of Gorosoe and bamboos sap instead of tap water to make ordinary soy sauce, respectively. The changes of such compounds during fermentation as minerals, free sugars, organic acids and free amino acids were analyzed. Total mineral contents in the 90 days fermented soy sauce from Gorosoe and bamboos sap were increased by 2.0 and 4.4 times as compared with those of control samples, respectively. Especially the increase of calcium, potassium and magnesium in soy sauce of sapes were derived from those of saps. The sugars were found to be only glucose and galactose in control samples, but they were detected not only above sugar but also fructose and sucrose in soy sauce of saps. Butyric acid in detected organic acid were dominant in all stray sauce samples. In the free amino acid composition of control samples, leucine, phenylalanine, isoleucine, lysine and glutamic acid were abundant amino acids. Amino acid such as isoleucine, leucine and phenylalanine were decreased, the rest 11 kinds of amino acids containing phosphoserine were increased during the fermentation of control samples. Increasing and decreasing patterns of free amino acids during fermentation of sap soy sauce were similar to control samples. Total nitrogen, amino acid nitrogen and ammonia nitrogen were increased during their fermentation.

Comparison of Free Amino Acids and Nucleotides Content in the Olive Flounder Paralichthys olivaceus Fed with Extruded Pellet (건조 배합사료로 사육한 넙치 어육의 유리아미노산 및 핵산 관련 성분의 비교)

  • This study analyzed the taste components of dorsal muscle and fin muscle collected from olive flounder cultured with extruded pellet (EP) compared with those of olive flounder cultured with raw fish moist pellet (MP) as a control. The olive flounder in this study were cultured for 10 months with either the formulated extrude pellets (FEP), commercial extruded pellets (CEP) or the MP feed, and the average weight was 1.15 kg. The major nucleotides and related compounds in the dorsal muscles of the fish fed with MP, CEP and FEP diets were AMP and IMP. The fin muscle of the olive flounder contained the largest amounts of IMP($5.91{\mu}mol/g$). The K value of the dorsal muscle in olive flounder fed with MP, CEP and FEP showed 9.63%, 9.83% and 5.84%, respectively. The free amino acids in the dorsal muscle of olive flounder showed significantly differences in the composition depending on the feed, but the distribution of free amino acids showed similar patterns. Among free amino acids, taurine showed the highest content in all the experimental groups, and the asparagine and citrulline contents were significantly higher in the groups fed with CEP and FEP than in the group fed with MP. The significant differences were observed in the contents of specific free amino acids in dorsal muscle and fin flesh depending on the feed for cultured olive flounder.

  • Changes in free amino acid composition in cotyledon and axis was investigated during growth of soybean sprout. Free amino acid increased with sprout-growth, especially in axis. Arginine, proline and methionine did not appear in axis throughout. Free amino acids in total amino acids was 0.11% for soybean and 8.8% for 8 day-sprout. Free amino acid change in the course of culture was in order of Asp>His>Val>Ile>Thr>Ser>Lys>Tyr>Phe>Leu>Ala>Gly>Pro>Arg>Glu>Met. The content of free amino acid in 4 day-sprout was in order of Asp>Ser>Val>His>Ala>Ile>Lys>Thr>Arg>Leu>Glu>Phe>Tyr>Gly>Cys>Met>Pro.

Changes of Free Amino Acid Compositions and Sensory Properties in Kochjang Added Sea Tangle Powder during Fermentation (다시마를 첨가한 고추장의 숙성 중 유리아미노산 조정 및 관능적 특성 변화)

  • 우리나라의 전통 발효식품인 고추장의 관능성 및 기능성을 더하여 품질을 향상시키기 위하여 전분질원료인 찹쌀무게에 대하여 2%. 4%, 6%, 및 8%의 다시마 분말을 첨가한 후 대조구와 함께 3$0^{\circ}C$에서 12일간 숙성시키면서 유리아미노산과 지방산 조성 acl 관능적 특성을 검토하였다. 숙정 중 대조고추장과 다시마고추장 모두에서 아미노질소는 서서히 증가하는 것으로 나타났으나 아미노질소의 경우 대조고추장에 비해 다시마 고추장에서 다소 낮게 나타났다. 숙성 30일째의 아미노질소량은 각각 171.31mg%, 172.10mg%, 174.18mg%, 185.60mg 및 161.70mg%로 최고값을 보였다 유리아미노산 중에서는 glutamic acid의 함량이 가장 높은 것으로 나타났는데 숙성 이 진행될수록 증가하였다. 또한 arginine, aspartic acid, proline, serine, leucine, lysine 도 비교적 높은 함량을 가졌다. 고추장에서 분리 확인된 지방산은 lauric acid myris-tic acid, palmitic acid stearic acid , oleic acid, linoleic acid 등이었고, 이들 중 oleic acid의 조성비율이 가장 높았고 palmitic acid가 다음으로 높았으며 stearic acid, lauric acid myristic acid의 경우 숙성 후기로 갈수록 산화되어 감소하는 경향을 보였다. 숙성 60일과 120일에 실시한 관능검사의 결과 8% 다시마 첨가구를 제외하고는 대조고추장과 비교하여 다시마 첨가구를 제외하고는 대조고추장과 비교하여 다시마 첨가가 고추장의 관능적 특서엥 영향을 미치지 않았으며 다시마 첨가수준이 높아질수록 높은 값을 나타내어 고추장에 다시마를 6% 정도 첨가하는 것이 적당하였다.

Comparison of amino acid and free amino acid contents between cap and stipe in Flammulina velutipes and Pleurotus ostreatus (팽이와 느타리버섯 자실체의 갓과 대에서 아미노산 및 유리아미노산 함량 비교)

  • The composition and content of amino acid and free amino acid were separately compared in cap and stipe of fruitbody obtained from different color strains in winter mushroom and oyster mushroom strains. The result showed different kinds and amount of amino acids according to the mushroom parts, strains and kinds. Tryptophane was not detected in any mushrooms. Alanine was also not detected in all oyster mushroom strains. The content of glutamic acid was the highest and that of methionine, phenylalanine and cysteine were very low in all mushroom strains. When compared by parts, the ratio of amono acids showed similar tendency in composition. Generally the content of amono acids showed higher in caps than in stipes. The profile and content of free amino acid showed a lot of variation between cap and strip in the intra and inter mushrooms.

Free sugar, amino acid, and beta-glucan content in Lentinula edodes strains collected from different areas (표고 수집종들의 유리당, 아미노산 및 β-glucan 함량)

  • In this study, we analyzed the nutrient composition and ${\beta}$-glucan content in Lentinula edodes fruiting bodies from different collection areas. Four types of free sugars were detected by HPLC, and the range of trehalose prevalence was 0.83% to 9%. The highest total amino acid content was observed from sawdust media cultivation of Lentinula edodes fruiting bodies, collected in China (JMI10050)The highest essential amino acid content, assessed by log cultivation of Lentinula edodes fruiting bodies, collected in Jangheung (JMI10059). Sixteen free amino acids were detected in Lentinula edodes fruiting bodies, and the major free amino acids were histidine, glutamic acid, and arginine. The highest essential amino acid including threonine, valine, methionine, isoleucine, leucine, phenylalanine, histidine, and lysine was fromsawdust media cultivation of Lentinula edodes fruiting bodies collected in China(JMI10052). The ${\beta}$-glucan content from log cultivation of Lentinula edodes fruiting bodies collected in Korea (JMI10059 and JMI10066) was higher than that from sawdust media cultivation of Lentinula edodes fruiting bodies collected in China. The highest ${\beta}$-glucan content was observed from log cultivation of Lentinula edodes fruiting bodies collected in Korea (JMI10066).

Properties of Chemical Components of Camellia japonica L. loaves According to Picking Time (동백잎의 채취시기에 따른 화학적 성분 특성)

  • The purpose of this study was to analyze the chemical component of Camellia japonica L. according to picking time. Leaves of Camellia japonica L. were picked in April and May,2003. Free sugars (fructose, glucose and sucrose) and organic acids (citric acid, tartaric acid, succinic acid, acetic acid) were present in the Camellia japonica L. leaf. The contents of total free sugars and organic acids increased as picking time was delayed. The major components of free amino acids were aspartic acid, glutamic acid and histidine, and those of total amino acids were histidine and alanine. The contents of total free amino acids and total amino acids were decreased as picking time was delayed, while the ratio of essential amino acids to the total amino acids increased. The amount of minerals (P, Ca, K, Na and Fe), chlorophyll and total polyphenol increased as picking time was delayed.

Compositions of Protein and Amino Acid in Crucian Carp and Snakehead (붕어 및 가물치의 단백질 및 아미노산 조성)

  • Compositions of protein and amino acid in the muscle of crucian carp, Carassius carassius, and snakehead, Channa argus, were determined by amino acid autoanalyzer, and the protein subunits composed of sarcoplasmic and myofibrillar proteins were also analyzed by sodium dodecylsulfate (SDS) polyacrylamide gel electrophoresis. The crucian carp muscle contained about $14.7\%$ of protein, and the protein was composed of $32.6\%$ in sarcoplasmic, $62.0\%$ in myofibrillar, $4.9%$ in alkali soluble and $0.6\%$ in stroma protein. The snakehead muscle, on the other hand, contained about $16.1\%$ of protein, and $30.7\%,\;64.1\%,\;4.7\%\;and\;0.4\%$ in the above order. The sarcoplasmic and myofibrillar proteins were composed of 10 and 19 subunits respectively, in the crucian carp, and 12 and 18 subunits in the snakehead. The total amino acid compositions in the muscle of the crucian carp and snakehead were found to be very similar except lysine and glutamic acid. The major amino acids of their muscle protein were lysine, glutamic acid, arginine and aspartic acid in order. In free amino acid content of the crucian carp, histidine occupied $52\%$ of the total free amino acid, and glutamic acid, glycine and taurine did $85\%$ in case of snakehead.

Conditions of Quantitative Analysis for free Amino Acid in Fermented Proteins (발효단백질의 유리아미노산 정량)

  • This study was performed to provide the optimal conditions of quantitative analysis for free amino acid in fermented protein foods. The water extractable free amino acid from dairy fermented foods was extracted effectively at $75^{\circ}C$ for 40 min., while it were extracted from fermented soy products at $40^{\circ}C$ for 3 hours. A close results of free amino acid content to those from amino acid analyzer were obtained using OPDA method with lysine standard after deproteinizing with 1% picric acid. 95% ethanol used as a deproteinizing reagent could give a comparable results to those from picric acid treatment in determining free amino acid content using OPDA method. Therefore, ethanol treatment was more recommendable than picric acid treatment which has some troubles in removing excess picric acid through Dowex resin column. The most desirable precipitation method for free amino acid determination using TNBS method was 95% ethanol treatment among the various deproteinizing procedure. The copper salt method was not suitable owing to its lacking reproducibility and pronounced discrepancy in determining free amino acid.
