Biodegradation of Kerosene by Pseudomonas aeruginosa K14 (Pseudomonas aeruginosa Kl4를 이용한 등유(Kerosene)의 생물학적 분해)

  • In this study, we isolated 32 strains of kerosene degrading bacteria from oil contaminated soil by enrichment culture. Isolates were screened for kerosene degradation efficiencies and K14 were selected which had the highest removal efficiency for 1,000 mg/L of kerosene. K14 were identified as Pseudomonas aeruginosa by morphological, biochemical test and 16S rDNA analysis. The optimal culture condition were determined as initial inoculated cell concentration, 1.0 g/L; substrate concentration, 1,000 mg/L; temperature $30^{\circ}C$; pH 7. When we enforced batch test in this condition, K14 degraded 72% of kerosene with 1,000 mg/L during 72 hr. And, at low concentration (200 mg/L), K14 degraded 95.8% of kerosene during 48 hr. As a result, kerosene biodegradation by Pseudomonas aeruginosa K14 could be useful for clean up of groundwater and soil contaminated with crude oil.

Sampling and Analysis of Soil Pore Water for Predicting the Diffusion and Behavior of Soil Pollutant Using Soil Lysimeter (토양라이시미터를 이용한 토양오염확산.거동 예측을 위한 토양공극수 채취와 분석)

  • This case study is about the sampling and interpretation of soil pore water in order to understand and to predict the diffusion and behavior of soil pollution. For the measurement of polycyclic aromatic hydrocarbons(PAHs) in two representative hydrocarbon-contaminated sites, the extraction system of the soil pore water was set up with respect to soil depths and the behavior of contaminants was interpreted. The soil solution extraction system consisted of peristaltic pump, and extraction and sampling compartment, and can measure simultaneously the soil water pressure. The concentration of PAHs with respect to extraction pressure and time decreased due to dilution through soil pore water. Particularly, the concentration of PAHs was more reduced under the unsaturated oxic condition than saturated anoxic condition. Therefore, the soil solution extraction with respect to soil water pressure can interpret the extent of equilibrium between porewater and soil surface.

Analysis of Total Petroleum Hydrocarbon in Domestic Distribution Petroleum (국내 유통 중인 석유제품 내 석유계 총 탄화수소화합물(TPH) 분석)

  • Over 60~70% of the domestic soil contamination have occurred by petroleum products. B T E X including benzene, toluene, ethylbenzene, xylene and total petroleum hydrocarbon (TPH) have to be inspected for the contaminated soil by petroleum products. An accurate contamination analysis is necessary to estimate the are of contaminated soil and also establish an appropriate purification scheme. In this study, we analyzed a sectional TPH pattern for current domestic distributed petroleum products. Also, the TPH content was analyzed by compensating the defect of current Korea standard analytic methods for soil where the analytic range is just for $C_8{\sim}C_{40}$. The light distillate petroleum products such as gasoline and solvent 1 showed the maximum of 85% difference in the TPH content between the standard analytic and improved methods.

  • Ultrasonic waves have several mechanical, chemical, and biological effects on a saturated soil medium. Their mechanical effects, popularly known as cavitation. Cavitation is the rapid and repeated formation, and resulting implosion, of imcrobubbles in a liquid, resulting in the propagation of microscopic shock waves. In a soil-liquid system, their mechanical effects generate high differntial fluid-particle velocities and microscopic shock waves. The velocity perturbations are capable of dislodging oil in the system by overcoming the forces binding oil to sand particles. In this study, a series of laboratory experiments involving the simple flushing and ultrasonic flushing were carried out. An increase in permeability and oil removal rate were observed in ultrasonic flushing tests. Some practical implications of these results are discussed in terms of technical feasibility of in situ implementation of ultrasonics.

Soil Health Assessment of Soil Washing and Landfarming Treated Soils (토양세척 및 토양경작 정화 토양의 건강성 평가)

  • To restore the ecological function of contaminated soil and maximize the ecological services provided by the soil, besides the toxicity orrisk caused by pollutants, the functional aspects of the soil ecosystem should be considered. In this study, a method for evaluating the health of cleaned soil was presented, and the applicability of the proposed evaluation method was examined by applying it to soil treated with washing and landfarming. Productivity, habitat, water retention capacity, nutrient cycling, carbon retention capacity, and buffering capacity were used as soil health evaluation indicators. The results showed that the soil health was not completely recovered after remediation, and even in the case of the washed soil, the health was lower than before remediation. On the other hand, there was no significant change in soil quality due to oil pollution, but soil health deteriorated. Unlike the slightly improved soil quality after landfarming treatment, soil health was not completely restored. Therefore, the results of this study indicate that it is desirable to consider both soil quality and health when evaluating the remediation effect. The soil health evaluation method proposed in this study can be usefully utilized for the sustainable use of cleaned soil and to promote ecosystem services.

Culture-Independent Methods of Microbial Community Structure Analysis and Microbial Diversity in Contaminated Groundwater with Major Pollutants (주요 오염물질로 오염된 지하수에서 미생물의 무배양식 군집분석방법과 미생물상에 대한 조사방법 연구)

  • This review inquired the recently applied molecular biological and biochemical methods analyzing the microbial community structure of groundwater and, as a result, summarized the functional or taxonomic groups of active microorganisms with major contaminants in groundwater. The development of gene amplification through PCR has been possible to figure out microbial population and identification. Active microbial community structures have been analyzed using a variety of fingerprinting techniques such as DGGE, SSCP, RISA, and microarray and fatty acid analyses such as PLFA and FAME, and the activity of a specific strain has been examined using FISH. Also, this review included the dominant microflora in groundwater contaminated with fuel components such as n-alkanes, BTEX, MTBE, and ethanol and chlorinated compounds such as TCE, PCE, PCB, CE, carbon tetrachloride, and chlorobenzene.

Enhanced Bioslurping System for Remediation of Petroleum Contaminated Soils (Enhanced Bioslurping system을 이용한 유류오염 토양의 복원)

  • Bioslurping combines the three remedial approaches of bioventing, vacuum-enhanced free-product recovery, and soil vapor extraction. Bioslurping is less effective in tight (low-permeability) soils. The greatest limitation to air permeability is excessive soil moisture. Optimum soil moisture is very soil-specific. Too much moisture can reduce air permeability of the soil and decrease its oxygen transfer capability. Too little moisture will inhibit microbial activity. So Modified Fenton reaction as chemical treatment which can overcome the weakness of Bioslurping was experimented for simultaneous treatment. Although the diesel removal efficiency of SVE process increased in proportion to applied vacuum pressure, SVE process was difficulty to remediation quickly semi- or non-volatile compounds absorbed soil strongly. And SVE process had variation of efficiency with distance from the extraction well and depth a air flow form of hemisphere centering around the well. Below 0.1 % hydrogen peroxide shows the potential of using hydrogen peroxide as oxygen source but the co-oxidation of chemical and biological treatment was impossible because of the low efficiency of Modified Fenton reaction at 0.1 % (wt) hydrogen peroxide. NTA was more efficiency than EDTA as chelating agent and diesel removal efficiency of Modified Fenton reaction increased in proportion to hydrogen peroxide concentration. Hexadecane as typical aliphatic compound was removed less than Toluene as aromatic compound because of its structural stability in Modified Fenton reaction. What minimum 10% hydrogen peroxide concentration has good remediation efficiency of diesel contaminated groundwater may show the potential use of Modified Fenton reaction after bioslurping treatment.

Extraction of Total Petroleum Hydracabons from Petroleum Oil-Contaminated Sandy Soil by Soil Washing (토양 세척법에 의한 유류오염 사질토양의 TPH 추출 효율 평가)

  • The influences of various operating parameters on physico-chemical techniques were evaluated to remediate petroleum-contaminated sandy soil including S/L ratio, kinetic, and effect of soil particle size. The simple extraction using tap water removed only 20.6% of total petroleum hydrocarbon (TPH), and addition of NaOH enhanced the removal of TPH to approximately 30%. To meet the regulation levels, a surfactant, sodium dodecyl sulfate, was added, and the removal of TPH increased to 4 times. Probably, the carbonate minerals affected chemical aging and soprtion of petroleum, which inhibited the extraction of TPH. The soil with smaller particle size contained more TPH, and the removal of TPH was obstructed with smaller particle size. However, NaOH addition increased the removal of TPH in the smaller particles. The physico-chemical properties of soil influenced greatly the removal of petroleum even in sandy soil.

Efficient Clean-up of Oil Spilled Shorelines Using the Compressed Air Jet System and Concomitant Microbial Community Analysis (압축공기 분사시스템을 이용한 유류오염 해안의 효율적 정화 및 이에 따른 미생물군집분석)

  • The objectives of this study were to investigate effectiveness of the Compressed Air Jet (CAJ) System for cleaning up shorelines contaminated with crude oils and to examine effects of the system on total petroleum hydrocarbon (TPH) removal and microbial community changes before and after remediation of the oil-contaminated shorelines. These data will lead to better understanding of optimized remediation process. About 66% of TPH reduction was observed when the contaminated site was treated with the CAJ System 2, 3, 4, and 5 times. This treatment system was more efficient than the seawater pumping system under similar treatment conditions (by 40%). By the way, little oil degrader communities were observed despite a potential function of the air jet system to stimulate aerobic oil degraders. The apparent low population density of the oil degraders might be as a result of low concentration of TPH as a carbon source and limiting nutrients such as nitrogen and phosphorus. It was proposed that the CAJ System would contribute significantly to removal of residual oils on the shorelines in combination with addition of these limiting nutrients.

토착 미생물의 활성에 의한 유류오염 토양 정화 실험

  • Many methods have been developed for the remediation of contaminated soil and groundwater. Among those technologies, in-situ bioremediation is most likely to be cost-effective method for petroleum hydrocarbon contamination. But the in-situ bioremediation can require more time to remediate hydrocarbon-contaminated soil and groundwater than other methods. Therefore we intended to save time of in-situ bioremediation using a biological additive to activate indigenous microbes in soil. The additive, 'Inipol EAP 22' stimulates the growth of specific flora, significantly accelerating the speed at which hydrocarbons are biodegraded. And it hans been tested in accordance with protocol approved by the USEPA and is registered on the National Contingency Plan Product Schedule List. In the experiment, three soil samples contaminated with fuel oil were prepared in the same concentration. Inipol EAP 22 was not added to one sample and was added to the other two samples with 5% and 10% of hydrocarbon by weight respectively. And $CO_2$gas derived from bacterial respiration was analyzed in each samples for 15 days. As a result, 145% and 153% of $CO_2$ evolution (microbial respiration) against the sample without 'Inipol EAP 22' occurred in samples with 'Inipol EAP 22' addition of 5% and 10%, respectively
