  • In a continuation of the 1st series of these studies, a separation of the components in organic phases obtained from Szilard Chalmer reactions of aromatic chloro derivatives was attempted to clarify the conditions of isolating the main component. Though it was unable to isolate all constituents of organic phases, the main component was well isolated by means of a simple distillation or a thin layer chromatography. It was identified that the main component was a recombined product of the recoiled $^{38}$ Cl atom with the original sample. In cases of solid samples the main component amounted about 80-60% of the radio activity of the organic phase, whereas the main component from liquid samples gave less than 70% of the radio activity of the organic phase. A prolonged irradiation could increase the yield of a main component, but it was, in case of a radiation senintive compound, decreased as the irradiation is extended. The number of aide produets separated from an organic phase of a liquid sample was greater than that of a solid sample, and hence the main component of a solid sample was isolated easier. The overall fields of the labelled main products of 22 kinds of compounds were listed and a procedure of the separation of the main component by a thin layer chromatograply was proposed and evaluated. The feasibility of the present procedure for labelling purpose was also reviewed with respects to the chemical structure of a compound.

  • New visible and fast assay system have been developed for tobacco transformant introduced with adenosine deaminase (ADA) marker gene, which converts cytotoxic adenosine analogues to non-toxic inosine analogues and ammonia. Ammonia was changed to blue color in the solution of phenol-nitoprusside and alkaline-hypochlorite. It was possible to detect activity of ADA visibly on the holes of 96 well plate using tiny explant of transgenic tobacco leaves within 1 hour incubation time. As substrates of ADA enzyme from transgenic plant on the plate, a number of adenosine analogues such as 9-D-arabinofuranosyl adenine, cordycepin, 2'-deoxyadenosine, adenosine and xylofuranosyl adenine were possible for detection of ADA activity. Optimal condition of substrate for ADA enzyme was each 10 mM and pH 7.5 in adenosine solution. Especially, transgenic plant did not convert adenosine to inosine and ammonia in the presence of ADA inhibitor deoxycoformycin, which means that ammonia produced from transgenic plant is due to expression of ADA gene. Now, we show that this detection system can be easily, sensitively, fast and cheaply as well as visibly assayed in vitro as GUS gene system with very small size of transformant explant.

  • This study was conducted to assess the effect of heptapeptide, composed of seven amino acids, on the activation of human hair cells isolated from human hair follicles. We have confirmed that the heptapeptide could bind to Lgr5 from the results of surface plasmon resonance (SPR) analysis. Heptapeptide enhanced the proliferation of human hair follicle dermal papilla cells (HHFDPCs) in a dose dependent manner. It induced the protein level of nuclear β-catenin, and the expressions of β-catenin downstream target genes, including LEF1, Cyc-D1 and c-Myc, in HHFDPCs. Heptapeptide significantly induced the phosphorylation of Akt and ERK, and the mRNA expressions of growth factors, including hepatocyte growth factor (HGF), keratinocyte growth factor (KGF) and vascular endothelial growth factor (VEGF), in HHFDPCs. In addition, heptapeptide significantly increased mRNA expression levels of differentiation-related transcription factors of human hair germinal matrix cells (HHGMCs) and differentiation markers of human hair outer root sheath cells (HHORSCs). Additionally, we investigated the effect of heptapeptide on human hair follicle stem cells (HHFSCs) differentiation and found that the heptapeptide reduced the mRNA and protein levels of stem cell markers, while it increased those levels of differentiation markers. These results have indicated that the heptapeptide promotes proliferation or differentiation of various types of hair follicle constituent cells through the induction of Wnt/β-catenin signaling. From the results, we have suggested that the heptapeptide in this study could be applied as a new functional material for the improvement of hair growth and alopecia.

  • Spontaneously infected and non-infected dairy cows were assessed in a cross-sectional study aimed at determining whether bovine leukocyte markers may diagnose intra-mammary infections (bovine mastitis). Animals located in herds where bovine mastitis was highly prevalent were investigated (n = 31 animals). The expression of three cell-surface markers (CD11b, CD4 and CD8) was assessed, and the somatic cell count (SCC) and bacteriological analyses (both cultures and PCR tests) were also conducted. Cows identified as infected revealed statistically significant higher milk leukocyte CD11b, CD4 percentage and milk CD4/CD8 ratios than non-infected cows. Immunological markers may diagnose spontaneous bovine mastitis.

  • The objective of this study is to derive the appropriate perception location for changes in driving behavior of autonomous vehicles in urban road environments based on traffic safety signs. For this purpose, 32 types of signs that induce changes in driving behavior were selected from currently used traffic safety signs and classified as three types according to changes in driving behavior. Based on this, three scenarios were designed: stop, speed change, and lane change scenarios. These were used to confirm the impact on traffic flow. As a result of the analysis, it was found that each scenario needs to receive information on traffic safety signs in advance to ensure changes in traffic flow and safety. Consequently, the appropriate perception location can be used as a basis for establishing standards for delivering message sets to autonomous vehicles or revising traffic safety signs for them. In addition, this study is expected to contribute to the establishment of safe and efficient driving strategies on urban roads as autonomous vehicles are introduced in the future.

  • Purpose: We investigated whether a relationship exists between tumor control dose 50 ($TCD_{50}$) or tumor growth delay (TGD) and radiation induced apoptosis (RIA) in syngeneic murine tumors. Also we investigated the biological markers that can predict radiosensitivity in murine tumor system through analysis of relationship between $TCD_{50}$, TGD, RIA and constitutive expression levels of the genetic products regulating RIA. Materials and Methods: Syngeneic murine tumors such as ovarian adenocarcinoma, mammary carcinoma, squamous cell carcinoma, fibrosarcoma, hepatocarcinoma were used In this study. C3H/HeJ mice were bred and maintained in our specific pathogen free mouse colony and were $8{\sim}12$ weeks old when used for the experiments. The tumors, growing in the right hind legs of mice, were analyzed for $TCD_{50}$, TGD, and RIA at 8 mm in diameter. The tumors were also analyzed for the constitutive expression levels of $p53,\;p21^{WAF1/CIP1},\;BAX,\;Bcl-2,\;Bcl-X_L,\;Bcl-X_S$, and p34. Correlation analysis was peformed whether the level of RIA were correlated with $TCD_{50}$ or TGD, and the constitutive expression levels of genetic products regulating RIA were correlated with $TCD_{50}$, TGD, RIA. Results: The level of RIA showed a significant positive correlation (R=0.922, p=0.026) with TGD, and showed a trend to correlation (R=-0.848), marginally significant correlation with $TCD_{50}$ (p=0.070). It indicates that tumors that respond to radiation with high percentage of apoptosis were more radiosensitive. The constitutive expression levels of $p21^{WAF1/CIP1}$ and 34 showed a significant correlation either with $TCD_{50}$ (R=0.893, p=0.041 and R=0.904, p=0.035) or with TGD (R=-0.922, p=0.026 and R=-0.890 p=0.043). The tumors with high constitutive expression levels of $p21^{WAF1/CIP1}$ or p34 were less radiosensitive than those with low expression. Conclusion: Radiosensitivity may be predicted with the level of RIA in murine tumors. The constitutive expression levels of $p21^{WAF1/CIP1}$ or p34 can be used as biological markers which predict the radiosensitivity.
