Antiproliferative Effect of Extracts from Corydalis heterocarpa on Human Cancer Cells (염주괴불주머니 (Corydalis heterocarpa) 추출물의 암세포 성장 억제 효과)

  • Whole plants of Corydalis heterocarpa were extracted twice with $CH_2Cl_2$ and MeOH in turn. The combined crude extracts were concentrated in vacuo and then partitioned between $CH_2Cl_2$ and $H_2O$. The organic layer was fractionated with n-hexane and 85% aq. MeOH, and the aqueous fraction was also further fractionated with n-BuOH and $H_2O$, successively. Growth inhibition effects of crude extracts and their solvent fractions were evaluated in AGS, HT1080, U-937, MCF-7 and HT-29 human cancer cells using MTT assay. The inhibitory effects of solvent fractions were increased in a dose-dependent manner. Among these tested samples, 85% aq. MeOH fraction showed the most potent inhibitory effect on the growth of human cancer cells. These results suggest that active compounds having much stronger anticancer effect can be isolated from Corydalis heterocarpa.

  • A 42-year-old female was referred to our department after find out submucosal tumor type lesion on cardia at local clinics. She experienced no specific symptom. On gastroscopy, two distinct neoplasms were detected. One of which was located in the cardia anterior wall of the stomach with the size of 2.0×1.1 cm, and the other one was localized in the cardia posterior wall of the stomach and its size was 1.5×1.2 cm. We performed endoscopic submucosal dissection. Pathological evaluation revealed the diagnosis of gastrointestinal stromal tumor (GIST) at the cardia anterior wall and malignant lymphoma from the mass localized cardia posterior wall of the stomach. We would like to report these rare synchronous tumors which were successfully treated by endoscopic resection in the same patient

Antioxidant and Cytotoxic Activity of Compounds from the Stem of Juncus effusus (등심초로부터 분리된 화합물의 항산화 활성 및 암세포 성장 억제 효과)

  • The stems of Juncus effusus were extracted with 70% aqueous ethanol and the concentrated extract was partitioned with ethyl acetate, n-butanol and $H_2O$, successively. Two compounds were isolated from the ethyl acetate fraction through the repeated silica gel and Sephadex LH-20 column chromatographies. According to the results of physico-chemical and spectroscopic data including NMR and IR, the chemical structures of the compounds were determined as dehydroeffusol (1) and effusol (2). Dehydroeffusol and effusol exhibited potent scavenging activity for DPPH and ABTS radicals with the $IC_{50}$ values as $130{\pm}3.21$ and $79{\pm}1.53{\mu}M$ in DPPH assay, and as $39{\pm}3.51$ and $24{\pm}2.73{\mu}M$ in ABTS assay, respectively. The compounds also significantly inhibited the proliferation of human cancer cell lines, AGS and A549.

  • A 75-year-old man presented with worsening dyspnea and intermittent dysphagia of one month's duration. A plain chest X-ray showed severe tracheal indentation by the right superior mediastinal mass. A chest CT established the diagnosis of a saccular aneurysm arising from the right proximal subclavian artery. Resection of the aneurysm and arterial revascularization was done through a median sternotomy with supraclavicular extension. Aneurysm wall and thrombus culture results were negative and pathology showed an atherosclerotic aneurysm. After the operation, dyspnea and dysphagia were reduced, but he died of advanced stomach cancer 8 months later.

Studies on the Biological Activity of Pleurotus ferulea (아위버섯(Pleurotus ferules) 추출물의 생리활성 탐색)

  • This study was peformed to screen the biological activities of Pleurotus ferulea (K5 and K8 strains). The cap of K5 strain is well developed than stalk, and vice versa in K8. The ethanol extract of Pleurotus ferulea exhibited significant free radical scavenging activity (35∼36%), suggesting possible effect on many degenerative diseases originated from the reaction of oxygen species. Acetylcholinesterase inhibitor is proven to be the most effective factor for Alzheimer disease induction, and ethanol extract of Pleurotus ferulea significantly inhibited acetylcholinesterase activity (25∼35%) in uitro. Moreover, ethanol extract of Pleurotus ferulea suppressed liver fibrosis by 3∼12% in uitro. However, Pleurotus ferulea feruled to inhibit glucose uptake in human intestinal cell line. Viability of gastric and colon cancer cells was also not affected by Pleurotus ferulea extract. In conclusion Pleurotus ferulea exhibited significant effect on free radical scavenging, acetylcholinesterase inhibiton and brain cell protection. However, Pleurotus ferulea failed to affect glucose uptake, and cytotoxicity of gastric and cancer cells. In general, K8 revealed more significant effects than K5.

  • In this study isolation and identification of squalene from amaranth and antineoplastic activity of its residue extract except squalene were determined to examine the utilization of grain amaranth in Korea. The content of squalene in amaranth grain was about 0.43%. The isolated squalene showed 99% pureness containing the identical molecular weight and structure provisionally in comparison with that of animal squalene from Sigma Co. by means of GC /Mass spectrum. Antineoplastic activity against human gastric and colon carcinoma cell line was measured in extract (except squalene) from Amaranth using MTT method. Extract from remaining plant good showed significant cytotoxic effect at the concentration of less than 230$\mu\textrm{g}$/ml.

  • Male breast cancer is rare, accounting for approximately 1% of breast cancers. Metastasis from extra-mammary malignancy to the breast in men is extremely rare. The most common primary tumors that metastasize to the breast in male are prostate, lung, stomach, colorectal cancer, melanoma, and sarcoma. To our knowledge, only a few cases of bladder cancer presenting with metastasis to the male breast have been reported, and metastasis with infiltration rather than mass is extremely rare. We report imaging findings on mammography and ultrasonography in a 59-year-old male with bladder cancer metastatic to the breast.

Growth Inhibitory Effects of $Doenjang$, Prepared with Various Solar Salts, on Cancer Cells (다양한 천일염으로 제조한 된장의 암세포 성장 억제 효과)

  • The growth inhibition effect of $Doenjang$ that was prepared with various kinds of solar salt was investigated. $Doenjang$ was prepared using the bacterial koji and five kind of salt with 12% salt concentration (w/w): purified salt $Doenjang$, one-year aged solar salt $Doenjang$, four-year aged solar salt $Doenjang$, topan solar salt $Doenjang$, and boiled solar salt $Doenjang$. The $Doenjangs$ were fermented and aged for 18 months. The growth inhibition effects of the water extracts and the methanol extracts of the $Doenjangs$ were measured on AGS human gastric adenocarcinoma cells, HT-29 colon carcinoma cells, and BJ human foreskin normal cells using MTT assay. The water and methanol extracts of the $Doenjang$ samples showed growth inhibition effects on the cancer cells, in the following order of the samples with the strongest to the weakest effect: the four-year aged solar salt $Doenjang$, the topan solar salt $Doenjang$, the boiled solar salt $Doenjang$, the one-year aged solar salt $Doenjang$, and the purified salt $Doenjang$. The methanol extracts of the four-year aged solar salt Doenjang (AGS: 55% and HT-29: 48%) showed the strongest growth inhibition effect. In addition, decreased cancer cell numbers and morphological changes in the cancer cells (AGS and HT-29) were observed when the methanol extract of the four-year aged solar salt $Doenjang$ was treated. None of the $Doenjang$ extracts showed a growth inhibition effect on the BJ normal cells, though.

  • Background : Cell adhesion molecules knave been Implicated In the various stages of tumor progression and metastasis. ICAM-1 plays a important roles in cell-cell interactions in inflammatory and immune response of several diseases. Recently, elevated levels of sICAM-1 in circulation was reported as association with liver metastasis in gastric, Colonic, gall bladder and pancreatic cancer, with reduced survival in malignant melanoma. This study was performed to measure the sICAM-1 in patients with lung cancer and to evaluate the relations between staging of lung cancer and level of sICAM-1. Methods : Serum sICAM-1 was measured in 36 patients with lung cancer according to the pathologic types and clinical staging before therapy and in 8 controls with ICAM-1 ELISA kit. Results : Serum sICAM-1 levels were elevated in patients with lung cancer except small cell type. Also progression and metastasis of lung cancer associated with elevation of sICAM-1 levels. Conclusion : These results suggest that higher levels of serum ICAM-1 reflect the progression and metastasis of lung cancer and it may be used as a marker with diagnostic and prognostic significance.

Clinical Analysis of MALT Lymphoma in the Stomach (위장의 점막 연관성 림프조직 림프종(MALT Lymphoma)의 임상적 고찰)

  • Purpose: The aim of this study was to analyze the clinical and the histopathological characteristics of mucosa associated lymphoid tissue (MALT) lymphomas in the stomach. Materials and Methods: We retrospectively reviewed the medical records of 22 patients who had been treated at Samsung Medical Center from Jan. 1995 to Sep. 2000 and who had been pathologically proven to have a MALT lymphoma. The factors we analyzed were operative procedure, tumor stage, and histopathological characteristics. Results: Of 3658 patients with a gastric malignancy, 22 patients proved to have a MALT lymphoma ($0.6\%$). There were 7 men and 15 women whose ages ranged from 25 years to 70 years (mean, 48.8 years). Forteen cases were located in the antrum, 4 ($18\%$) in the body and 4 ($18\%$) in the fundus or the high body. Nineteen of these patients were managed with total gastrectomy and splenectomy and 3 with radical subtotal gastrectomy. Histopathologically the tumor was limited to the mucosa in 3 patients ($1.36\%$), to the submucosa in 13 ($59.1\%$) and extended to the muscularis propria in 6 ($27.3\%$). Lymph node involvement was seen in 12 patients ($54.6\%$). There was no splenic or hepatic involvement. Bone marrow involvement was not seen in any patients. H. pylori was identified in 11 patients (50$\%$). During the mean follow-up period of 32.7 months, there were no reports of tumor recurrence or death. Conclusion: MALT lymphomas rarely disseminate by the time of diagnosis and rarely involve the bone marrow. Lymph node involvement is relatively high and a total gastrectomy is effective in managing patients with a MALT lymphoma.
