In this study, the physicochemical properties of irradiated (gamma-ray and electron-beam) soybeans with different processing (dry and powder) and origins (Korea, China, and USA) were investigated and compared. The results of photostimulated luminescence (PSL) screening indicated that all non-irradiated soybeans showed photon counts (PCs) ${\leq}700$, while all irradiated soybeans showed positive values-gamma-ray 5,815-39,591 count/min; electron beam 5,791-60,055 count/min. The results of thermoluminescence (TL) analysis of all irradiated soybeans indicated that the $TL_1$ glow curves exhibited maximum peaks at 150-250. TL ratio of irradiated samples was ${\geq}0.1$; therefore, the clear identification of irradiated samples was guaranteed by analysis of the $TL_1$ curve shape and TL ratios. The results of electron spin resonance (ESR) signal of 3 irradiated and dried soybeans showed two side peaks mutually spaced at 6.0 mT (cellulose radical). Non-specific signal was detected for all irradiated soybean powders; hence, ESR analysis could not be performed.
The discrimination of the agricultural origin, especially locally produced of imported products such as black rices was investigated by using electronic nose. Volatile components from these products were discriminated by six metal oxide sensors without pretreatment. Pattern recognition was carried out. Principal component analysis showed the differences between imported and locally produced ones. The number of 57 from 69 species of black rices were recognized as locally produced one (83.33%) and 11 from 13 species one (imported black rices) was correctly discriminated. Unknown habitat of black rice could be identified by artificial neural network system whether the imported or not. Also commercial electronic nose (E-nose 5000) that was combined with metal oxide sensor and conducting polymer sensor showed 92.75% (locally produced black rices) and 92.31% (imported one) of discrimination.
Discrimination of geographic origin of agricultural products is a important issue in Korea because the price difference between Korean domestic and imported cereals is a key among some reasons. NIRS (Near Infrared Reflectance Spectroscopy) has been applied to classify the geographical origin of soybeans. Total 135 samples (Korean domestic 92 and foreign 43) were used to obtain calibration equation through 400~2,500 nm wavelength. The math treatment with 1st derivative and 4 nm gap and the modified partial least squares(MPLS)regression was outstanding for calibration equation. The standard error of calibration and determination coefficient in calibration set(n=115) was 6.65 and 0.98, respectively. And it showed that the extra 20 samples for validation equation were identified their authentication correctly. This study describes that the application of NIRS would be possible for discrimination of geographical origin between Korean domestic and imported soybeans.
Consumers are increasingly concerned about the origin of foods, so the geographical origin of foods has been a major topic of debate and extensive research. Various instrumental methods (e.g. high performance liquid chromatography (HPLC), gas chromatography (GC), capillary electrophoresis (CE), electronic nose, near-infrared spectroscopy (NIRS), nuclear magnetic resonance spectroscopy (NMR), DNA analysis, multi-isotope analysis) in conjunction with statistical analysis, were developed and applied in attempt to provide reliable answers to their geographical origin. This study reviews current developments in the application of various methods for a clear geographical origin of foods. The limitation of discrimination analysis for geographical origin was also discussed.
녹용은 사슴의 뿔을 통칭해 일컫는 말로서 우리나라는 전 세계 녹용 소비량의 80%를 점유하고 있다. 하지만 국내에서 선호되는 것은 러시아산 붉은 사슴 즉, 원용으로 칭하는 것으로 가격 면에서 가장 고가이다. 따라서 수입되는 녹용의 일부가 러시아 산으로 둔갑 판매되는 경우가 발생되고 있다. 본 연구의 목적은 현재 주로 수입되는 녹용으로부터 미토콘드리아 DNA(mtDNA)의 D-loop 유전자의 일부를 PCR로 증폭하고 이들의 염기서열 분석을 통해서 각 수입지역 녹용에 특이적인 RFLP 마커를 탐색하고 이를 녹용의 유전자 감별에 적용코자 하는 것이다. 결과적으로 TaaI과 HaaI 두 종류의 제한효소가 녹용의 원산지를 구별할 수 있는 RFLP 마커로 이용 가능성이 확인되었다.

Sesame seed (Sesamum indicum L.) is an important seasoning in Korea and most korean consumer tend to eat the korean sesame seed as the best than other ones produced in oriental countries such as China and Japan. Near infrared reflectance spectroscopy (NIRS) was applied for discrimination according to geographical origin (Korea, China and so on) of sesame seeds. Near-infrared spectroscopy among the many kinds of techniques could provide a rapid screening, low cost solution to discriminate geographical origin of sesame seed. The objective of this study is to determine if NIR technique could be used to discriminate between the korean sesame seed and non-korean sesame seed by using the new method. Rapid, precise and nondestructive analysis method for determination of the geographic origin of sesame seeds were discriminated relative accurately according to geographical origin using PLS regression method.

The discrimination of bracken (Korean vs. Chinese) has been attempted using capillary electrophoresis (CE). Bracken (Pteridium aquilinum) was extracted with 30% methanol and separated on a uncoated fused-silica ($50{\mu}m{\times}27cm$) capillary. Conditions for optimal analysis include: temperature, $40^{\circ}C$; voltage, 8 kV; and pressure injection time, 5 sec. The optimal separation buffer was 0.3 M borate buffer (pH 8.5) containing 40 mM CHAPS with 30% ethylene glycol. Under the optimal conditions established for CE, the ratio of specific peak area (peak PA-1) to other peak area (peak PA-2) was effective in discrimination of Korean and Chinese bracken. The mean accuracy for discrimination of Korean and Chinese brackens were 80% and 86%, respectively.
The mineral contents of domestic and Chinese Sarcodon aspratus were analyzed for comparing the differences in the mineral contents according to the production sites. The mineral contents of 10 domestic and 10 Chinese S. aspratus were analyzed by using an atomic absorption spectrophotometer and an inductively coupled plasma mass spectrometer. The sodium, magnesium, potassium, and calcium contents of domestic S. aspratus were 170.3±44.0 mg/kg, 81.3±20.8 mg/kg, 1,123.8±274.3 mg/kg, and 12.1±4.1 mg/kg, respectively, while those of Chinese S. aspratus were 112.2±40.8 mg/kg, 297.8±20.9 mg/kg, 10,938.4±2,914.1 mg/kg, and 52.5±27.7 mg/kg, respectively. There were no significant differences in the micro mineral contents and hazardous heavy metal contents between domestic and Chinese S. aspratus, while there was a significant difference in the macro mineral contents between domestic and Chinese S. aspratus. Thus, the comparative analyses of the sodium, magnesium, potassium, and calcium contents of many domestic and Chinese S. aspratus are required to determine their production sites in the future.

Gamma-irradiated peanuts, Korean and Chinese origins, were investigated on detection properties by thermoluminescence (TL), electron spin resonance (ESR), and DNA comet assay (single cell gel electrophoresis). TL measurement showed that the non-irradiated sample revealed a glow curve with low intensity at about 25$0^{\circ}C$, while the irradiated samples showed higher intensity around at 18$0^{\circ}C$. TL ratio (TL$_1$/TL$_2$) of area for TL$_1$ glow curve to TL$_2$ was below 0.05 for the non-irradiated sample and 0.2 or more for the irradiated ones, thus identifying each other. ESR spectroscopy for the irradiated peanuts using outer skin showed negligible signals induced by irradiation, indicating ESR is little applicable to the detection of irradiated peanuts. In DNA comet assay, the non-sample had no or very short tails, whereas the irradiated samples revealed the cells with long tails. Significance in the increase of their lengths depending on irradiation dose (r=0.761/Korean, r=0.768/Chinese) was also found. There was no remarkable difference in detection properties by origins of samples in all determinations, It is concluded that TL analysis or DNA comet assay is suitable for detection of irradiated peanuts and a combined method is recommendable for enhancing the reliability of detection results.

Reliable labeling of fish products can reassure consumers regarding the identity and quality of seafoods. Therefore, techniques that can identify adulteration or mislabeling are valuable. To rapidly identify two Trachurus species, Trachurus japonicus and Trachurus novaezelandiae, a highly efficient, rapid, duplex polymerase chain reaction (PCR) having two species-specific primers simultaneously was identified. This species-specific primer focused on a single nucleotide mismatch in the 3'-terminal base of a primer designed in the mitochondrial cytochrome c oxidase (COI) subunit I DNA. To optimize the duplex PCR condition, gradient PCR reactions were conducted to determine the primer annealing temperature and the primer concentration. The PCR's product was observed on the gel, suggesting that DNA molecules may be useful in differentiating the two species. The length of the amplification fragments were 103 bp for Trachurus japonicus and 214 bp for Trachurus novaezelandiae, which, along with the species-specific primer visualized by agarose gel electrophoresis, enabled accurate distinction of the species of the Trachurus genus. These results indicate that the duplex PCR, which has a species-specific primer based on single nucleotide polymorphism (SNP), can be useful for rapidly differentiating the two species of Trachurus. This duplex PCR analysis is simple, rapid, and reliable, and could be beneficial to protecting consumers' rights.
