Effect of Follicular Fluid on Sperm Swim-up Separation with Sucrose Layer (난포액이 Sucrose 층을 이용한 정자의 Swim-up 분리에 미치는 효과)

  • To establish a system for sperm swim-up separation through sucrose layer, indiscreet sperm migration should sufficiently to block but movement of sperm shouldn't inhibit. Thus, the effects of sucrose levels in sucrose layer, incubation times and types of sucrose layer on sperm separation were examined. And the results obtained were as follows; 1. Layer of 10mM sucrose inhibited sperm swim-up migration through sucrose layer. 2. Incubation for 25 minutes without sucrose layer significantly increased sperm swim-up migration. However, incubation for 10 minutes to induce swim-up through sucrose layer significantly stimulated sperm migration and maintained sperm movement. 3. There was no significant difference between Type I and Type II in barrier effect of sucrose layer. However, sucrose layer of Type II with shorter distance of barrier was efficient for sampling. To elucidate a function of follicular fluid on sperm chemotaxis using in vitro system of sucrose layer of Type II and incubation for 10 minutes, the effects of dilution, heat treatment, and protein and lipid extracts of follicular fluid on sperm swim-up separation were examined. And the results obtained were as follows; 4. Follicular fluid stimulated sperm migration and movement, and significantly-attracted capacitated-sperm at 10% level. 5. Follicular fluid heated at 55$^{\circ}C$ for 30 minutes maintained the effect of follicular fluid stimulating sperm migration and movement. 6. Follicular protein stimulated sperm movement that was reduced by filtration of the protein. 7. Follicular lipid didn't significantly stimulate sperm migration and movement. 8. Both of follicular protein and lipid reduced the effect of follicular fluid stimulating sperm migration and movement. In conclusion, sucrose layer could be used for a barrier against indiscreet sperm migration by swim-up. And follicular fluid stimulated migration and movement of sperm and attracted capacitated-sperm through sucrose layer. Especially, heat-resistant protein of follicular fluid stimulated sperm migration.

Assessment of Sperm Activity of Black Porgy(Acanthopagrus schlegeli) Acclimated in Freshwater on Cryopreservation Condition (담수순화 감성돔(Acanthopagrus schlegeli) 정자의 냉동보존 조건별 활성평가)

  • Various cryoprotective agents (CPA) were tested to establish the best conditions for the cryopreservation of sperm from black porgy Acanthopagrus schlegeli acclimated and raised in freshwater (BFW). Survival rates of frozen/thawed sperm from BFW were higher in the order of dimethy sulfoxide (DMSO), glycerol, ethylene glycol (EG) and methanol. Sperm motility was higher in the order of glycerol, DMSO, EG and methanol. These effects were the same in thawed sperm from black porgy raised in seawater (BSW). Thus, optimum CPA for sperm cryopreservation of BFW and BSW were DMSO and glycerol where the highest survival rates and sperm motility were found at the concentration of 10%. In particular, the survival rates and motility of thawed sperm from BFW and BSW after cryopreservation using 10% DMSO were better than when cryopreserved using 10% glycerol. On the other hand, for the thawed sperm from both BFW and BSW, the longer the preservation period was, the lower the survival rates and sperm motility were. Notably, the higher the concentration of CPA was, the lower the survival rates and sperm motility were.

Effect of Thawing Rate on the Function of Cryopreserved Canine Sperm (융해 속도가 동결.융해된 개 정자의 기능에 미치는 영향)

  • Sperm cryopreservation methods have been improved over the last few decades. However, an optimized thawing rate has not yet been established. Therefore, we investigated the effect of thawing rate on sperm function after cryopreservation. The ejaculates collected from beagle dogs were cryopreserved and then thawed at two different thawing rates ($37^{\circ}C$ for 1 min or $70^{\circ}C$ for 15 sec). The thawed sperm were evaluated for motility, viability, morphology, plasma membrane integrity, phosphatidylserine (PS) translocation, and intracellular $H_2O_2$ level. The sperm thawed rapidly at $70^{\circ}C$ showed improved motility, viability, normal morphology, plasma-membrane integrity and non-PS translocation compared to the sperm thawed slowly at $37^{\circ}C$ (P < 0.05). However, the intracellular $H_2O_2$ levels were not significantly different between the rapid- and slow-thawed sperm (P > 0.05). In conclusion, sperm rapid thawing at $70^{\circ}C$ could improve the function of cryopreserved canine sperm, and the appropriate thawing rate would enhance the quality of the cryopreserved sperm.

  • The objective of this study was to evaluate the possibility of sex preselection by gradients methods using bovine serum albumin in rabbits. Artificial insemination with separated sperm was performed, after highly motile sperm were separated by different methods using 6%, 10% and 20% bovine serum albumin. Various characteristics of separated sperm, and the conception rate and secondary sex ratio at artificial insemination with sperm separated by different methods were compared. The results obtained were as follows. 1. The conception rate of sperm separated by bovine serum albumin gradients was higher than th at of control sperm. But secondary sex ratio was not altered by this methods. 2. The sperm separated by bovine serum albumin gradients showed significantly high value in motility, percent of normal sperm and progressive motility as compared with control sperm and revealed the highest sperm recovery when separated with 6% bovine serum albumin. 3. The sperm motility, percent of normal sperm and progressive motility of the highly motile sperm frozen after being separated from raw semen with bovine serum albumin, showed significantly high value than those of control sperm.

닭 정액의 보존온도 및 희석배율이 수정률에 미치는 영향

  • This study was conducted to investigate the effects of dilution rate and stored temperature of semen at 5, 25 and 35$^{\circ}C$ on fertility in liquid rooster semen. At 5$^{\circ}C$ cold temperature, no significant difference were found in sperm mobilities on dilution rate(1:1, 1:3, 1.6) among treatments. Sperm mobility for the conservation of 3 hours at 25∼35$^{\circ}C$ were significantly higher for 1:3 and 1:6 dilution rate(semen:diluent) groups than for 1:1 dilution rate group(P<0.05). In Fertility results after artificial insemination with the conservation of 3 hours at 5∼25$^{\circ}C$ temperature, no significant difference were found in fertility on dilution rate among treatments. Fertilities after artificial insemination with the conservation of 3 hours at 35$^{\circ}C$ were significantly higher for 1.3 and 1:6 dilution rate(semen:diluent) groups than for 1:1 dilution rate group(P<0.05).

  • Objective: We tested the usefulness of swim-down technique using human follicular fluid (hFF) in sperm preparation. Methods: Semen samples were obtained from twelve male partners showing asthenozoospermia (sperm motility < 50%) at the time of routine andrologic evaluation in Seoul National University Bundang Hospital. After dividing into two aliquots, each samples were processed either by swim-down using 100% hFF or density gradient using SpermGrad. Sperm quality was assessed by computer-assisted semen analyzer (CASA). Results: Motility, Rapid motility, VCL (curvilinear velocity), ALH (amplitude of lateral head displacement), and hyperactivated sperms were significantly increased, and LIN (mean linearity) was decreased significantly after sperm preparation in both groups. Motility was significantly higher after swim-down using 100% hFF when compared with density gradient using SpermGrad ($81.2{\pm}4.7$ vs. $67.6{\pm}2.3$, p=0.02) The other parameters assessed by CASA were not different between the two methods. Conclusion: Swim-down method with 100% hFF may be a useful method in preparation of sperm from asthenozoospermia.

Effect of Pentoxifylline Concentration on Sperm Quality in Jeju Crossbred Horses (Jeju Crossbred Horses 정액 생산 시 Pentoxifylline 농도가 정자 성상에 미치는 영향)

  • This study was conducted to determine the effect of pentoxifylline levels on sperm motility, survival rate, sperm membrane integrity of frozen semen and fresh-extended equine semen in Jeju cross-bred horses. As a result of sperm characteristic comparison depending on pentoxifylline levles at 30 minutes post-thaw, the progressive motilities were $53.25{\pm}2.87$ (4mM pentoxifylline) and $50.28{\pm}2.14$ (8mM pentoxifylline) and significantly higher compared to the control group($40.09{\pm}5.15$) and other treatment group (16mM pentoxifylline, $41.27{\pm}2.82$). The progressive fast motility were $22.44{\pm}1.62$ (4mM pentoxifylline,) and $22.74{\pm}3.07$ (8mM pentoxifylline) and significantly higher compared to the control group ($13.47{\pm}1.48$) and other treatment group (16mM pentoxifylline, $14.66{\pm}3.68$) (p<0.05). As a result of sperm characteristic comparison depending on pentoxifylline levles at 30 minutes post-thaw were $68.96{\pm}1.64$ (4mM pentoxifylline) and $67.90{\pm}6.72$ (8mM pentoxifylline) and significantly higher compared to the control group ($53.48{\pm}4.84$) and other treatment group (16mM pentoxifylline, $58.14{\pm}2.65$) (p<0.05). In conclusion, these results suggest that treatment groups with 4mM and 8mM pentoxifylline were higher compare to equine seperm mobility and the control group and treatment groups with more than 16mM pentoxifylline has a negative effect on sperm characteristics. After thawing, the total motility in post-thawed equine sperm has increased by 10 percent for 1 hour. these results suggest that pentoxifylline contributes to the improvement of the equine sperm motility and characteristics in post-thawed semen.

Effect of Storage Times on the Kinematics and Capacitation Status in Liquid Boar Semen (보존 기간이 돼지 액상정액의 운동역학 및 수정능 획득에 미치는 영향)

  • The objective of this study was to estimate modification of semen quality during storage. Liquid boar semen samples extended in Beltsville Thawing Solution were stored at $17^{\circ}C$ up to 5 days. While % motility and linearity significantly decreased eon day 3 in extender, the qualitative motility patterns were maintained satisfactorily. Also the storage of boar semen up to 5 days before insemination did not significantly changed the acrosome intactness. However, acrosome changed sperm significantly increased and capacitated sperm significantly decreased from day 4. No significant modifications in acrosome integrity were showed during sperm storage; these results suggest that liquid boar semen may keep the quality in extender for 3 days.

Effects of MethyIxanthine Derivatives on Induction and Enhancement of Hamster Epididymal Sperm Motility (Hamster 정소상체 정자의 운동성 유도와 증가에 영향을 미치는 Methylxanthine Derivatives 의 효과)

  • This study were performed the effect of methylxanthine derivatives on hamster epididymal sperm motility. To assess the effect of methylxanthine derivatives on motility kinematics of epididymal sperm, pentoxifylline, 2-deoxy-adenosine and hypoxanthine were added to TALP medium. 1 mM of pentoxifylline, significantly increased VCL, VAP, VSL of spermatozoa obtained from corpus and cauda epididymis. With 1 mM of 2-deoxyadenosine, VCL, VAP, VSL of spermatozoa obtained from corpus and cauda epididymis were significantly increased, but 2 mM ADE for cauda spermatozoa was effective than 1 mM. In the case of hypoxanthine, various concentrations were not significantly effective, but 2 mM HX showed higher effect than other concentrations. pentoxifylline 1 mM and 2-deoxyadenosine 1 mM significantly increased VCL, VAP of corpus and cauda epididymal spermatozoa and VSL of cauda epididymal spermatozoa. From these results it could be concluded that the addition of methylxanthine increase motility kinematics of hamster spermatozoa collected from epididymis.

  • To investigate the influences on semen parameters and fertilizing capacity of immunoglobulin (Ig) isotypes and regional distribution of antisperm antibody (ASA) on the human sperm surface. Sixty-seven ASA-positive patients were compared with 96 ASA-negative donors. ASAs in semen showed significant negative effects on both semen parameters and fertilizing capacity; in those with ASAs in the sperm head and/or tail, the reductions were significant. In the head as well as the tail, there was close correlation between fertilizing capacity and both IgG and IgA. Both semen parameters and fertilizing capacity are significantly affected by the presence of ASA in semen. In particular, antibodies IgG to sperm head and/or tail, and antibodies IgA to sperm tail appeared to have a highly detrimental effect on fertilizing capacity.
