Association Analysis between Chromogranin B Genetic Variations and Smooth Pursuit Eye Movement Abnormality in Korean Patients with Schizophrenia (한국인 조현병 환자에서 Chromogranin B 유전자와 안구운동 이상의 연합에 대한 연구)

  • Objectives According to previous studies, the Chromogranin B (CHGB) gene could be an important candidate gene for schizophrenia which is located on chromosome 20p12.3. Some studies have linked the polymorphism in CHGB gene with the risk of schizophrenia. Meanwhile, smooth pursuit eye movement (SPEM) abnormality has been regarded as one of the most consistent endophenotype of schizophrenia. In this study, we investigated the association between the polymorphisms in CHGB gene and SPEM abnormality in Korean patients with schizophrenia. Methods We measured SPEM function in 24 Korean patients with schizophrenia (16 male, 8 female) and they were divided according to SPEM function into two groups, good and poor SPEM function groups. We also investigated genotypes of polymorphisms in CHGB gene in each group. A logistic regression analysis was performed to find the association between SPEM abnormality and the number of polymorphism. Results The natural logarithm value of signal/noise ratio (Ln S/N ratio) of good SPEM function group was $4.19{\pm}0.19$ and that of poor SPEM function group was $3.17{\pm}0.65$. In total, 15 single nucleotide polymorphisms of CHGB were identified and the genotypes were divided into C/C, C/R, and R/R. Statistical analysis revealed that two genetic variants (rs16991480, rs76791154) were associated with SPEM abnormality in schizophrenia (p = 0.004). Conclusions Despite the limitations including a small number of samples and lack of functional study, our results suggest that genetic variants of CHGB may be associated with SPEM abnormality and provide useful preliminary information for further study.nwhile, smooth pursuit eye movement (SPEM) abnormality has been regarded as one of the most consistent endophenotype of schizophrenia. Production of Poly(3-hydroxybutyrate) Using Waste Frying Oil (Waste frying oil를 사용한 Poly(3-Hydroxybutyrate) 생합성)

  • In this study, the optimal growth and poly(3-hydroxybutyrate) (PHB) biosynthesis of Pseudomonas sp. EML2 were established using waste frying oil (WFO) as a cheap carbon source. The fatty acid composition of WFO and fresh frying oil (FFO) were analyzed by gas chromatography. The unsaturated and saturated fatty acid contents of the FFO were 82.6% and 14.9%, respectively. These contents changed in the WFO. The compositional change in the unsaturated fatty acid content in the WFO was due to a change in its chemical and physical properties resulting from heating, an oxidation reaction, and hydrolysis. The maximum dry cell weight (DCW) and PHB yield (g/l) of the isolated strain Pseudomonas sp. EML2 were confirmed under the following culture conditions: 30 g/l of WFO, 0.5 gl of $NH_4Cl$, pH 7, and $20^{\circ}C$. Based on this, the growth and PHB yield of Pseudomonas sp. EML2 were confirmed by 3 l jar fermentation. After the cells were cultured in 30 g/l of WFO for 96 h, the DCW, PHB content, and PHB yield of Pseudomonas sp. EML2 were 3.6 g/l, 73 wt%, and 2.6 g/l, respectively. Similar results were obtained using 30 g/l of FFO as a carbon source control. Using the FFO, the DCW, PHB content, and PHB yield were 3.4 g/l, 70 wt%, and 2.4 g/l, respectively. Pseudomonas sp. EML2 and WFO may be a new candidate and substrate, respectively, for industrial production of PHB.

Identification of DNA Markers Related to Resistance to Herbicide Containing Mesotrione in Tongil Type Rice (통일형 벼에서 메소트리온계 제초제 저항성 연관 DNA marker 탐색)

  • This study was conducted to identify DNA markers related to resistance to herbicide containing mesotrione in Tongil type rice. Two Tongil type elite lines; Milyang154 and Suweon382, showed resistance to mesotrione, whereas the others were susceptible at 20 days after mesotrione application, and severe growth inhibition was observed in the remaining 13 lines. As a result of analysis of mesotrione resistance using 190 $F_2$ populations derived from a cross of Hanareum2 (susceptible) and Milyang154 (resistant), the mesotrione resistance locus was shown to be a single dominant gene with a 3:1 segregation ratio ($X^2=1.19$, P=0.31). To identify a DNA marker closely linked to the mesotrione resistance gene, bulked segregant analysis (BSA) was adopted. The DNA marker RM3501 was identified on chromosome 2 with a recombinant value of 0.53 to the mesotrione resistance gene. Mst1(t) was located between SSR (simple sequence repeat) markers RM3501 and RM324 with a physical map distance of 10.2 Mb-11.4 Mb on chromosome 2. The band pattern of agarose gel electrophoresis of the SSR marker RM3501 showed the same segregation pattern with respect to mesotrione treatment in 20 Tongil type varieties and a $BC_2F_2$ segregation population derived from a cross between Unkwang (resistant) and Hanareum2 (susceptible). Thus, the RM3501 DNA marker could be used in breeding programs for Marker Assisted Selection in mesotrione resistant rice breeding.

  • Understanding the phytotoxic mechanism of methyl bromide (MB), an essential fumigant during the quarantine and pre-shipment process, is urgently needed to ensure its proper use and reduce international economic losses. In a previous study, two main MB-induced toxic mechanisms such as reactive oxygen species (ROS) and auxin distribution were selected by analyzing transcriptomic analysis. In the study, a 3-week-old A. thaliana was supplied with 1 mM ROS scavengers [N-acetyl-L-cysteine (NAC) or L-glutathione (GSH)] and 1µM indole-3-acetic acid(IAA) three times every 12 h, and visual and gene expression assessments were performed to evaluate the reduction in phytotoxicity by supplements. Phytotoxic effects on the MB-4h exposed group were decreased with GSH application compared to the other single supplements and a combination of supplements at 7 days post fumigation. Among these supplements, GSH at a concentration of 1, 2, and 5mM was suppled to A. thaliana with MB-fumigation. During a long-term observation of 2 weeks after the fumigation, 5 mM GSH application was the most effective in minimizing MB-induced phytotoxic effects with up-regulation of HSP70 expression and increase in main stem length. These results indicated that ROS was a main key factor of MB-induced phytotoxicity and that GSH can be used as a supplement to reduce the phytotoxicity of MB.

  • 순천만은 순천시의 남쪽에 위치하여 여수반도와 고흥반도의 사이에 있는 만 지형 형태로 만 전체를 순천만이라고도 하나, 보통 고흥군반도와 여수반도 사이의 만을 여자만이라 하고 만내부에 위치한 여자도라는 섬의 위쪽으로 순천시의 해안하구에 형성된 지역을 순천만이라 일컫는다. 순천만은 순천시에서 유입되는 동천과 이사천 및 해룡천의 하류에 형성되어 있어서 육지부에서 유입되는 퇴적물과 유기물로 인하여 갯벌과 갈대등이 잘 형성된 습지로서 2003년 12월 31일 해양수산부 갯벌 습지보존지역으로 지정되어 관리되고 있으며, 2004년에는 동북아 두루미 보호 국제네트워크에 가입하였고, 2006년 1월 국내 연안습지로는 최초로 람사르협약에 "Suncheon Bay"로 등록되었으며, 2016년 6월에는 람사르습지로 지정된 국내외적으로 중요한 습지이다. 순천만습지에 형성되어 있는 갯벌($28.0km^2$)은 세계 5대 연안습지 중 하나로서, 넓은 사니질 갯벌과 갈대군락이 잘 발달되어 있는 생태계의 보고(寶庫)이자 소중한 생태자원으로서, 순천시에서는 순천만을 자연생태공원으로 지정하여 보호 관리하고 있으며, 각종 자연학습자료들과 영상물들을 갖춘 생태전시관과 갈대숲 탐방로, 용산전망대, 야생화 정원, 담수습지, 갈대정자, 갯벌관찰대 등 사계절 생태체험을 위한 각종 시설들을 잘 갖춰놓고 많은 관광객의 이용할 수 있도록 하고 있다. 갯벌은 바닷가의 넓은 벌판이란 뜻으로서 삼면이 바다인 우리나라는 갯벌의 전체 면적이 약 $2,500km^2$에 달하는데 여기에는 많은 종류의 다양한 생물들이 살아가고 있으며 어민들의 생계에 지대한 영향을 주고있다. 이러한 갯벌은 퇴적된 입자의 구성에 따라 펄갯벌, 모래갯벌, 혼합갯벌 등으로 구별되는데 이에 따라 갯벌 생태계를 구성하고 있는 생물들의 종류도 바뀌게 된다. 순천만갯벌과 여기에 조성되어 있는 습지환경에 따라 확인되고 있는 수산자원으로는 새꼬막, 꼬막, 눈알고둥, 갯고둥, 비툴이고둥, 돌조개, 접시조개, 새알조개, 가무락조개, 바지락, 우럭, 가재붙이, 방게, 칠게, 농게 등의 저서 생물들과 짱뚱어, 문절망둑 등의 어류가 있으며, 해조류로 우뭇가사리 등이 있고, 인근의 어민들의 어업형태는 꼬막 등 패류의 채취나 종패를 뿌려 일정기간 양성하여 수확하는 양식업, 육상부에서 폐염전 등을 활용한 전어나 새우 등을 양식하는 양식업, 수산물을 직접 손이나 간단한 도구를 이용하여 잡는 맨손 어업 형태가 주를 이루고 있는 것으로 나타나고 있다. 이와 같이 국내외적으로 중요할 뿐만 아니라 인근의 어민들의 생계에도 지대한 영향을 미치고 있는 순천만의 습지는 뻘층이 깊고, 분해성 미생물이 다양하게 서식하여 유기물 분해능력이 뛰어나며, 유기영양분이 풍부하여 우리나라에서 가장 질이 좋은 습지로 평가되고 있으나, 순천시 등 순천만 인근에 거주하고 있는 인간의 활동에 따른 간섭에 많은 영향을 받고 있으며, 이에 따라 끊임없는 생태환경이 변화하고 있어서 순천만의 효율적인 보전 및 지속가능한 이용을 위해서는 생태계에서 가장 기본적인 요소인 수 저질 환경의 지속적이고 체계적인 조사 및 관리가 필요하다. 한편, 오염물질의 70% 이상은 하천이나 강을 통해서 해역으로 유입된다고 알려져 있기 때문에 생태계의 보고(寶庫)라고 알려진 순천만의 지속적인 보존 및 관리를 위해서는 유입수계 하천의 수질현황 및 오염물질의 주요 배출원을 파악하고, 이에 대한 저감대책을 수립할 필요가 있다. 따라서 본 연구는 순천만의 수저질 특성과 여기에 유입되는 하천의 수질환경 현황 및 오염원을 파악함으로써 순천만의 보전을 위한 효율적 관리방안을 제시하는 것을 목적으로 수행되었다. 연구의 결과에 따르면, 순천만의 수질평가지수에 의한 등급(WQI)은 III등급으로 나타나고 있으며, 득량만, 광양만 등에 비해 비교적 높은 유기물 및 T-N, T-P의 농도 분포를 보이고 있는 것으로 조사되었다. 이는 순천만에 유입되는 하수종말처리장의 방류수와 도시하수가 유입되어 그대로 방류되고 있는 해룡천 및 연안에 위치한 어촌으로부터 직접 방류되고 있는 일부 정화조 유출수 등, 다양한 원인에 의한 것으로 판단되며 이들의 관리가 부실할 경우 순천만의 갯벌과 습지의 지속가능한 생태환경유지는 쉽지 않다. 따라서 이를 효율적으로 관리하기 위해서는 순천만 연안의 오염물질 방류를 총량관리로 전환하여 철저히 관리하는 것이 필요할 것으로 판단된다.

Meaning of Plantain(Musa basjoo) Planting and Design Use through Classic Poetry and Prose (고전 시문을 통해 본 파초(Musa basjoo)의 식재 의미와 설계용도(Design Use))

  • By studying the classic poetry and prose with a theme of Plantain, I would like to study why Plantain has been planted in our traditional garden for a long time. Also, through this study, I want to find when Plantain was introduced to in our country. We use description study method for studying the classic poetry and prose with a theme of Plantain. As a result, we found a few things like below. First, the introduction of Plantain to our country traces back to unified Silla in the poetry of Choe Chi Won. Also, Plantain was planted and loved generally as a general garden plant in the middle of Goryeo through poetry and paintings. Second, the meaning of Plantain is like this. 1) It means development and enjoyment of arts of summer because the leaves of Plantain were used for drinking alcohol and writing and poetry instead of papers. 2) In Buddhism, Plantain was thought to awaken 'gong(空)', 'mua(無我)', and 'brevity' of lesson of Buddha by the special shape and the image of falling rain to the leaves. Also, it was used widely in Buddhist temples because of the story of 'Hye ga dan bi(慧可斷臂)'. 3) In Confucianism, it is the emblem of lesson 'a wise man tries to be strong and tries not to stop following to God'(自强不息). 4) The large leaf of Plantain is called 'bongmi(鳳尾)' thanks to the image of likeness with tails of Bongwhang(鳳凰). Third, design use of Plantain is like this. 1) The large leaf of Plantain was used for giving an image of freshness and brightness in the garden in summer. 2) Our ancestor thought 'the sound of falling rain to Plantain'(芭蕉雨聲) as a picturesque sound in summer. Also, Plantain was highly utilized because our ancestor thought Plantain is the best plant for implementing 'soundscape'. Thus, the most characteristic use of several design uses is 'acoustic use'. 3) Plantain was also planted in a indoor pot for viewing. 4) Plantain was used for making food and medicine in the palace and private house as a practical use. The limit of this study is that I mainly use the text translated into Korean of database of overall Korean classics. We hope the new things related to this study would be added up to this study by translating original texts into Korean more.

Comparison of the Morphometric Changes in the Cervical Foramen: Anterior Cervical Discectomy and Fusion versus Posterior Foraminotomy (전방 경유 디스크 절제술 및 유합술과 후방 추간공 절제술에서의 경추 추간공의 형태학적 변화 비교)

  • Purpose: This study compared the change in foraminal space on magnetic resonance imaging (MRI) and the clinical outcome after anterior cervical discectomy and fusion (ACDF) versus foraminotomy in cervical foraminal stenosis. Materials and Methods: A retrospective case-control study was conducted from January 2018 to March 2019 on 186 patients who underwent ACDF and foraminotomy. One hundred and two cases were selected considering age, sex, and body mass index. MRI was performed before and on the 5th day after surgery to compare the changes in the foraminal diameter between the ACDF group (group A-51) and foraminotomy group (group B-51). Results: Between groups A and B, the average change in foraminal vertical diameter was 1.7 mm and 1.2 mm, respectively; group A was 0.5 mm larger difference (p=0.042). The average change in foraminal transverse diameter was 1.2 mm and 1.8mm, respectively; group B showed a 0.6 mm larger change (p=0.21). Both the neck disability index (NDI) and Japanese orthopaedic association (JOA) scores improved in both groups. Group A showed more improvement, but there was no significant difference (p=0.356, p=0.607, respectively). Conclusion: Foraminotomy is a useful option for patients with foraminal stenosis of the cervical spine because it showed comparable clinical and morphological results to ACDF and could minimize motion segment loss and muscle and ligament damage.

Monitoring of Concrete Deterioration Caused by Steel Corrosion using Electrochemical Impedance Spectroscopy(EIS) (EIS를 활용한 철근 부식에 따른 콘크리트 손상 모니터링)

  • The electrochemical impedance spectroscopy(EIS) method was used to evaluate the concrete deterioration process related to chloride-induced steel corrosion with various corrosion levels(initiation, rust propagation and acceleration periods). The impressed current technique, with four total current levels of 0C, 13C, 65C and 130C, was used to accelerate steel corrosion in concrete cylinder samples with w/c ratio of 0.4, 0.5, and 0.6, immersed in a 0.5M NaCl solution. A series of EIS measurements was performed to monitor concrete deterioration during the accelerated corrosion test in this study. Some critical parameters of the equivalent circuit were obtained through the EIS analysis. It was observed that the charge transfer resistance(Rc) dropped sharply as the impressed current increased from 0C to 13C, indicating a value of approximately 10kΩcm2. However, the sensitivity of Rc significantly decreased when the impressed current was further increased from 13C to 130C after corrosion of steel had been initiated. Meanwhile, the double-layer capacitance value(Cdl) linearly increased from 50×10-6μF/cm2 to 250×10-6μF/cm2 as the impressed current in creased from 0C to 130C. The results in this study showed that monitoring Cdl is an effective measurement parameter for evaluating the progress of internal concrete damages(de-bonding between steel and concrete, micro-cracks, and surface-breaking cracks) induced by steel corrosion. The findings of this study provide a fundamental basis for developing an embedded sensor and signal interpretation method for monitoring concrete deterioration due to steel corrosion at various corrosion levels.

A preliminary assessment of high-spatial-resolution satellite rainfall estimation from SAR Sentinel-1 over the central region of South Korea (한반도 중부지역에서의 SAR Sentinel-1 위성강우량 추정에 관한 예비평가)

  • Reliable terrestrial rainfall observations from satellites at finer spatial resolution are essential for urban hydrological and microscale agricultural demands. Although various traditional "top-down" approach-based satellite rainfall products were widely used, they are limited in spatial resolution. This study aims to assess the potential of a novel "bottom-up" approach for rainfall estimation, the parameterized SM2RAIN model, applied to the C-band SAR Sentinel-1 satellite data (SM2RAIN-S1), to generate high-spatial-resolution terrestrial rainfall estimates (0.01° grid/6-day) over Central South Korea. Its performance was evaluated for both spatial and temporal variability using the respective rainfall data from a conventional reanalysis product and rain gauge network for a 1-year period over two different sub-regions in Central South Korea-the mixed forest-dominated, middle sub-region and cropland-dominated, west coast sub-region. Evaluation results indicated that the SM2RAIN-S1 product can capture general rainfall patterns in Central South Korea, and hold potential for high-spatial-resolution rainfall measurement over the local scale with different land covers, while less biased rainfall estimates against rain gauge observations were provided. Moreover, the SM2RAIN-S1 rainfall product was better in mixed forests considering the Pearson's correlation coefficient (R = 0.69), implying the suitability of 6-day SM2RAIN-S1 data in capturing the temporal dynamics of soil moisture and rainfall in mixed forests. However, in terms of RMSE and Bias, better performance was obtained with the SM2RAIN-S1 rainfall product over croplands rather than mixed forests, indicating that larger errors induced by high evapotranspiration losses (especially in mixed forests) need to be included in further improvement of the SM2RAIN.

  • The contents of sulfur, sulfur-containing protein and amino acids of soybean seeds of 518 genotypes as well as their inheritance and selection efficiency in early breeding generation were measured to facilitate breeding for soybean with high sulfur-containing amino acids. Average seed sulfur content of 518 cultivars was 0.33%, and ranged from 0.20 to 0.45%, and that of 30 wild soybeans was 0.35%, and ranged form 0.19 to 0.62%. Correlation coefficients between seed sulfur content and sulfur-containing protein and amino acids were 0.924$^{**}$ and 0.974$^{**}$, respectively. Seed sulfur content was tended to be high in soybean genotypes with late maturity, seed coat bloom, or green cotyledon. Sulfur content had -0.312$^{**}$ correlation coeficient with sugar content and -0.384$^{**}$ with 100 seed weight. Seed sulfur content was inherited quantitatively, in which additive effect was greater than dominant one, and proportion of genes with positive effects was similar to those with negative ones. Estimated narrow- and broad-sense heritabilities were 0.75 and 0.88 for seed sulfur content, respectively. Heritability measured from selection in early breeding lines for high or low seed sulfur content was 60~62.5% or 50~62,5%, respectively.
