Flow Properties of Sulfated Polysaccharides in the Tunic of the Ascidin Halocynthia roretzi (우렁쉥이 껍질 유래 황산다당의 유동 특성)

  • The flow properties of sulfated polysaccharides purified from the ascidian Halocynthia roretzi tunic were investigated. The sulfated polysaccfarides produced aqueous solutions of low apparent viscosity with pseudoplastic flow behavior. The respective activation energies of TCA-treated and TCA-nontreated solutions were $2.2248{\times}10^4\;and\;1.442{\times}10^4J/kg{\cdot}mol$ at a 150L/s shear rate. The viscosity of the sulfated polysaccharides solution was increased by the addition of sugar, while it was not changed by the addition of NaCl.

  • Functional properties such as anti-blood coagulation, angiotensin converting enzyme (ACE) inhibitory activity, fat binding capacity, foaming properties, emulsifying properties and chemical components of sulfated polysaccharides isolated from ascidian tunics were investigated. The sulfated polysaccharide mainly consisted of sulfate, uronic acid, protein, and chondroitin sulfate, among which chondroitin sulfate showed higher concentration while sulfate and uronic acid did lower. Compositional menosaccharides were arabinose, xylose, glucose, galactose, glucuronic acid, N-acetylgalactosamine and N-acetyglucosamine. Especially, galactose content was dominant among them. And emulsifiability and foaminess of the sulfated polysaccharide was higher than the control group. Anti-blood coagulation of sulfated polysaccharide showed with respect to APTT (Activated partial thromboplastin time). ACE inhibitory activity showed about $16.7\%$.

Immune-Enhancing Effects of Polysaccharides Isolated from Ascidian (Halocynthia roretzi) Tunic (우렁쉥이(Halocynthia roretzi) 껍질로부터 분리된 다당류의 면역증강 효과)

  • In this study, the immune-enhancing effects of purified polysaccharides from ascidian (Halocynthia roretzi) tunic were investigated. Crude polysaccharides (AP) were isolated by enzyme extraction (neutrase, $60^{\circ}C$, 15h), ethanol precipitation, and lyophilization. In addition, crude polysaccharides were further fractionated into unabsorbed fractions (APF-I, fraction No. 11~17) and absorbed fractions (APF-II, fraction No. 22~37) by DEAE-sepharose CL-6B column chromatography in order to isolate immune regulating polysaccharide. The major constituents in APF-I and APF-II were total sugar (66.62% and 27.03%), uronic acid (47.53% and 15.87%), hexosamine (16.62% and 46.79%), and protein (2.43% and 4.94%), respectively. APF-I increased production of nitric oxide (NO) and cytokines, such as tumor necrosis factor-alpha (TNF-${\alpha}$) and interleukin (IL)-6 in a dose-dependent manner. The mRNA expression levels of inducible NO synthetase, cyclooxygenase-2, TNF-${\alpha}$, and IL-6 were markedly increased as determined by polymerase chain reaction analysis. The above data led us to conclude that macrophage activation of purified polysaccharides was higher than that of crude polysaccharides. The polysaccharides isolated from ascidian tunic investigated herein are useful as natural immune enhancing agents.

  • Rainbow trout, Oncorhynchus mykiss, were cultured with different levels of carotenoids in odor to investigate the feeding effect of ascidian tunic extracts on the liver fatty acid compositions of them. Dominant monoenoic fatty acids of the ascidian tunic extracts were 16:1n-7($5.9\%$), 18:1n-9($21.9\%$), and 18:1n-7($3.5\%$) and polyenoic fatty acids of them were linoleic(18:2n-6, $14.2\%$), eicosapentaenoic(20:5n-3, $3.5\%$), and docosahexaenoic acid(22:6n-3, $8.3\%$). The compositions of fatty acid in the liver lipids were affected by the tunic extract levels during the feeding. The percentage of monoenoic acids in extract diets was decreased, and that of n-3PUFA was increased during feeding 2 weeks. But the n-3PUFA contents were decreased in 4 weeks. The 20:4n-6 content in rainbow trout fed extract diet was higher than that in the control and pink diet groups. The rainbow trout fed with ascidian tunic extracts showed an increase of essential fatty acids in the fish tissue, compared to the control or pink diet feeding groups.

  • The attraction activity of the natural baits for sea eels(Astroconger myriaster) was surveyed by using fishing traps in the 3 different fishing ground of southern coastal waters of Korea. Twenty four different baits were experimented and weight for each bait was 50g, the optimal minimum weight owing to our preliminary test. Among the above 24 baits, anchovy(Engraulis japonica), sardine(Sardinops melanosticta), or marine worms(Neanthes japonica)were most effective. In case of sardine bait, the body showed higher attraction activity than the head or viscera and the builded juice of anchovy or sardine showed higher attraction activity than spoiled one.

  • To detect naturally occurring bioactive compounds from unused marine resources, the screening for the 5-lipoxygenase(potato lipoxygenase-II) inhibitors in Asterina pectinifera, Halocynthia roretzi skin, Nototodarus sloani ink, Anthocidaris crassispina skin, Sargassum horneri, Agarum cribrosum, Odonthalia corymbifera and Desmarestia ligulata was carried out. Water, ether, acetone and methanol fractions extracted from Sargassum horneri had strong inhibitory effect on enzymatic lipid oxidation by potato lipoxygenase-II, and their $IC_{50}$ were 320, 18, 9.5 and $100\;{\mu}g/mL$, respectively. The $IC_{50}$ of ether fraction extracted from Asterina pectinifera and acetone fraction extracted from Nototodarus sloani ink were 29.5 and $34.3\;{\mu}g/mL$, and these extracts showed relatively excellent inhibitory activity. Nonpolar solvent (ether, acetone) extracts of tested marine organisms had more inhibitory effect against 5-lipoxygenase than the polar solvent(water) extracts.

Screening for Antimicrobial Compounds in Unused Marine Resources by the Paper Disk Method (수산 미이용자원 중에 존재하는 항균성 물질의 검색)

  • To detect naturally occurring bioactive compounds in unused marine resources such as marine by-products and algae, the screening test for the antimicrobial compounds containing in Asterina pectinifera, Halocynthia rotetzi skin, Nototodarus sloani ink. Anthocidaris crassispina skin, and four algae(Sargassum horneri, Agarum cribrosum, Odonthalia corymbifera and Desmarestia ligulata) was carried out. The minimum inhibitory concentrations(MIC, ${\mu}g/disk$) of ether and methanol fractions extracted from Sargassum horneri against Bacillus subtilis were 80 and 70 ${\mu}g/disk$, respectively. The MICs of the ether, acetone and methanol fractions extracted from Sargassum horneri against Staphylococcus aureus were 160, 180 and 70 ${\mu}g/disk$, respectively. In addition, the MICs of ether and acetone fractions extracted from Odonthalia corymbifera against Staphylococcus aureus were 50 and 170 ${\mu}g/disk$, respectively. These nonpolar fractions showed the strongest antimicrobial effect among several fractions extracted from tested marine resources.

  • Dried raw Ascidians(Halocynthia roretzi) shells harvested from fish farms in southern coast area in Korea were used to extract ${\beta}$-carotene using supercritical carbon dioxide($SCO_2$) and with ethanol as a co-solvent at the range of temperatures and pressures, from 25 to $65^{\circ}C$ and 100 to 350 bar respectively. The size of the dried Ascidians shells was around $850{\mu}m$. The system used this study was a semi-batch flow type high pressure unit. The efficiency of ${\beta}$-carotene extraction using $SCO_2$ with and without co-solvent, ethanol, influenced to pressure and temperature changes. The highest solubility of ${\beta}$-carotene in $SCO_2$ was 1.35 mg/g for ${\beta}$-carotene at $35^{\circ}C$ and 350 bar. With addition of 2(v/v%) ethanol the recovery of ${\beta}$-carotene was 93%. As a result of using n-hexane and methanol for rinse, at $35^{\circ}C$ and 350 bar the amount of ${\beta}$-carotene by methanol rinse was 5 times higher than that of n-hexane rinse.

  • To detect bioactive compounds present in unused marine resources, the screening for the 5-lipoxygenase inhitors in Asterina pectinifera, Halocynthia roretzi skin, Nototodarus sloani ink, Anthocidaris crassispina skin, SArgassum horneri, Agarum cribrosum, Odonthalia corymbifera and Desmarestia ligulata was carried out. THe ether and acetone extracts of Sargassum horneri had the strongest antioxygnic activityon lipid oxidation by potato lipoxygenase-1 (one of 5-lipoygenase) among the tested marine samples and their $IC_{50}$ were 0.3 and 1.1g/ml, respectively. The ether and acetone extracts of Asterina pectinifera, the acetone extracts of Halocynthia roretizi, and the acetone extracts of Nototodarus sloani ink had strong inhibitory activity and their $IC_{50}$ were 72.5, 65, 13.3 and $13.3\mu\textrm{g}/ml$, respectively. In addition, the $IC_{50}$ of the acetone extracts of Agarum cribrosum and Desmarestia ligulata, and the ether extracts of Desmarestia ligulata were 15.5, 35 and $30.5\mu\textrm{g}/ml$, respectively. The nonpolar solvent (ether, acetone) extracts of tested marine organism had more antioxigenic effect against 5-lipoxygenase than the polar solvent(water) extracts.
