Development of Dissolution Testing Method for Piracetam Tablets and Fenoterol Hydrobromide Tablets in Korean Pharmaceutical Codex (고시 수재 의약품 중 피라세탐 정 및 브롬화수소산페노테롤 정의 용출시험법 개발)

  • Although the dissolution test can serve as an effective tool for quality control and predictor of in vivo performance, there are a number of drugs with no established dissolution specifications in Korean Pharmaceutical Codex (KPC). Among those commercially available, Piracetam Tablets and Fenoterol hydrobromide Tablets were selected to develop the dissolution testing method. The dissolution condition was determined based on the "Guidelines on Specifications of Dissolution tests for Oral dosage forms" of Korea Food & Drug Administration (KFDA). The dissolution test for Piracetam Tablets was carried out under sink condition with distilled water as dissolution medium, paddle rotation speed at 50 rpm and medium volume of 900 ml. More than 80% of its label claim was released within 30 min. In case of Fenoterol hydrobromide Tablets, distilled water was also found to be suitable to ensure sink condition. The rotation speed of 50 rpm and 900 ml of dissolution medium were used to evaluate the dissolution profile. The dissolution rate of fenoterol hydrobromide was over 90% in 15 min. The HPLC analysis methods were validated in terms of accuracy, precision, specificity, linearity, quantitation limit and range. The results suggested that the analytical methods used are simple and suitable to measure the dissolution rate of piracetam and fenoterol hydrobromide. Therefore, the analysis methods could be utilized in setting dissolution specifications of Piracetam Tablets and Fenoterol hydrobromide Tablets in the revised version of KPC.

Toxicity Reduction and Improvement of Anticancer Activities from Rhodiola sachalinensis A. Bor by Ultra High Pressure Extracts Process (초고압 공정에 의한 홍경천의 독성 감소 및 항암활성 증진)

  • This study was performed to investigate the reduction of toxicity and improvement of anticancer activities from R. sachalinensis by ultra high pressure extracts process. The cytotoxicity on human kidney cell (HEK293) and human lung cell (HEL299) was showed below 20.4% and 21.6% as compare to normal extracts in adding 1.0 $mg/m{\ell}$ concentration. This showed that toxic materials through ultra high pressure processing is broken or degraded. Because bond such as hydrogen bond, electrostatic bond, Van der waals bond, the hydrophobic bond, can be broken by high pressure. The anticancer activity was also increased in over 7% by high pressure processing in A549, AGS, MCF-7 and Hep3B cells. The result showed that extraction by high pressure have low cytotoxicity and high anticancer activity. So, the high pressure extraction technology can play an important role in eruption of new material with high biological activity.

Antimicrobial Activities of Scutellariae Radix Extract against Vibrio parahaemolyticus (Vibrio parahaemolyticus에 대한 황금추출물의 항균활성)

  • To investigate the antimicrobial effects of Scutellariae Radix extract against Vibrio parahaemolyticus from food samples, Vibrio parahaemolyticus strains isolated from Tapes philippinarum were examined for their sensitivity to Scutellariae Radix extract. Total 66 Vibrio parahaemolyticus strains were isolated from Tapes philippinarm 72 samples (91.7%). The serotypes of isolated Vibrio parahemolytics were K-I group 7 strains (10.6%), K-IV group 5 strains (7.6%), K-II group 2 strains (3.0%), K-V group 2 strains (3.0%), K-VII group 2 strains (3.0%), K-VI group 1 strains (1.5%), K-VIII group 1 strains (1.5%) and antisera UT K-group 46 strains (69.7%) on antisera agglutination test, but K-III group and K-IX group strains were not found. The growth curves of isolates showed lag phase, logarithmic phase, stationary phase and death phase as typical sigmoid curve on the shellfish samples. After 6 hours, the group containing Scutellariae Radix extract differs from the control on shellfish samples in the growth inhibition curves, and Vibrio parahaemolyticus were inhibited in more than 1000 ppm Scutellariae Radix extract. The morphological changes were observed by transmission electron microscope and the microbial cells membrane was destroyed by Scutellariae Radix extract.

  • Although sludge production has been increased due to the number of the wastewater treatment plants expanded, needs of the techniques for the sludge reduction and disposal has been issued importantly because the sludge dumping to ocean is prohibited from 2012 by the London Dumping Convention. Therefore, the sludge solubilization using electrolysis as an alternative techniques for the sludge disposal was carried out in this study. Iridium coated titanium based insoluble electrodes were used and 20 volt was applied to the electrolysis reactor using DC power supply. Supernatants of the treated sludge was monitored: The soluble COD, TN, TP of it was increased to 151%, 22% and 6% respectively. And the sludge floc size distribution was changed, that is, the flocs ranged from 0.1 to 1.0 ${\mu}m$ were increased. All of these results indicate that the cells were lysed and the internal matters bursted out of the cell after electrolysis. As well as the reduction of the sludge production, the soluble organic matters from the cells could be used as an external carbon sources in the advanced wastewater treatment plants.

Correlation of Heavy Metal Concentrations between Total Digestion and Aqua Regia Digestion for Sediments from Yeongsan and Seomjin Watersheds (완전분해와 전분해 방법에 의한 영산강.섬진강수계 퇴적물의 중금속농도 상관관계)

  • In this study, the correlations of concentrations in sediment heavy metals between two pretreatment methods, total digestion and aqua regia digestion, have been investigated. Total 43 samples had been collected from streams and lakes in Yeongsan and Seomjin watersheds. They were decomposed in two pretreatment methods and five metal components (Cd, Cr, Ni, Pb, Zn) were analyzed. Overall average concentrations of heavy metals were Zn>Cr>Pb>Ni>Cd in the order. The concentrations of aqua regia digestion were 35.0% (Cd), 53.8% (Cr), 66.2% (Ni), 64.4% (Pb) and 76.4% (Zn) of the concentrations of total digestion and heavy metal concentrations of total digestion were higher. Heavy metal concentrations in sediments from lakes were higher than those from streams. The correlation equations between the concentrations of total digestion and aqua regia digestion were obtained. The concentrations of Zn and Ni, which showed strong linear correlations, increased in aqua regia digestion as the concentration in total digestion increased. The linear correlation coefficients between two digestion methods for most analyzed metals were above the average correlation. However, these results can be applied with limitations due to heterogeneity of sediments.

Biological Leaching of Cu, Al, Zn, Ni, Co, Sn and Pb from Waste Electronic Scrap using Thiobacillus Ferrooxidans (廢電子스크랩에서 Thiobacillus ferrooxidans를 이용한 Cu, Al, Zn, Ni, Co, Sn 및 Pb의 浸出)

  • In order to recover valuable metals from the waste electronic scrap, bioleaching of Cu, Zn, Al, Co, Ni, Sn and Pb was carried out using Thiobacillus ferrooxidans as a leaching microorganism in a shaking flask. In a preliminary study, to obtain the data on the leaching of Cu, Zn, Al, Co and Ni, the metal leaching was accomplished using metal powers instead of electronic scrap. The leaching percentaga of Cu, Zn, Co, Al and Ni powers was reduced with the increase of metal power concentration in solution. Below the metal concentration of 0.5 g/L, more than 85% of Cu, Co and Zn powers was leached out. At the electronic scrap concentration of 100 g/L, Thiobacillus ferrooxidans were able to leach more than 90% of the available Cu and Co while Al, Zn and Ni were able to leach less than 40%. Pb and Sn were not detected in the leachate. Pb was precipitated as PbSO$_4$, whereas Sn precipitated probably as SnO.

Antimicrobial Effects of Scutellariae Radix Extract against Listeria monocytogenes (Listeria monocytogenes에 대한 황금추출물의 항균효과)

  • To investigate the antimicrobial effects of Scutellariae Radix extract against L.monocytogenes from foods, L. monocytogenes strains isolated from livestock, processed food from meat and milk, and frozen foods, were examined for their sensitivity to Scutellariae Radix extract. 30 L. monocytogenes strains were isolated from total 178 samples(16.9%); 13(14.0%) strains from beef 6(20.7%) strains from pork, 9(39.2%) strains from chicken and 2 (16.7%) strains from frozen foods but was not found from processed products, The serotypes of isolated L.monocytogenes were serotype O-1 strains (23, 76.7%) and serotype O-4 strains(7, 23.3%) on antisera agglutination test. The growth curves of isolates were shown lag phase, logarithmic phase, stationary phase and death phase as typical sigmoid curve on the preservative-free hams. After 6 hours. Scutellariae Radix extract contain group differ from control group on preservative-free ham samples, and the isolates were inhibited in more than 1000 ppm Scutellariae Radix extract on the inhibitory growth curve of L.monocytogenes. The mor-phological changes were observed by transmission electron microscope and the microbial cells membrane was destroyed by Scutellariae Radix extract.

A Comparative Study of Sulfate and Chloride Intrusion in Mortar Sections: An Approach Using Laser Induced Breakdown Spectroscopy and Ion Exchange Membrane (LIBS와 이온교환막을 활용한 모르타르 단면 침투 황산염과 염화물 분석)

  • This research aimed to conduct an empirical assessment of the penetration of chloride and sulfate ions into mortar sections using an anion exchange membrane(AEM) and laser-induced breakdown spectroscopy(LIBS). The study involved a simultaneous ion chromatography(IC) analysis and LIBS analysis performed on mortars immersed in varying concentrations of chloride and sulfate. The findings revealed that at the wavelengths specific to Chloride(837.59nm) and Sulfur(921.30nm), the LIBS intensity achieved using AEM surpassed that obtained with a paper substrate at equivalent penetration concentrations. A robust correlation was confirmed between LIBS intensity and chloride ion concentration. Furthermore, when juxtaposed with IC analysis concentration outcomes at identical depths, the AEM displayed a higher intensity. The research noted an enhancement in LIBS intensity and a diminution in errors within the low-concentration section when deploying AEM. However, for the Sulfur wavelength of 921.3nm, there remains a need to augment the sensitivity of the LIBS signal within the low-concentration section in future studies. The findings underscore the potential of employing AEM and LIBS for precise analysis of chloride and sulfate ion penetration into mortar sections. This strategy can aid in bolstering assessment precision and mitigating errors, particularly in regions with low concentrations. It is recommended to further research and develop methods to amplify the sensitivity of the LIBS signal for sulfur detection in low-concentration sections. In sum, the study accentuates the significance of employing advanced techniques like AEM and LIBS for efficacious and precise analysis in the domain of mortar section assessment.

Recycling Organic Sludge into Modification Block by Hydrothermal Carbonization (HTC) (열수가압탄화공법을 적용한 유기성 슬러지의 응용 블록 제조에 관한 연구)

  • This study applied a hydrothermal carbonization (HTC) method to carbonize sewage sludge in order to satisfy the criteria of the Waste Management Act for recycled products and to explore the possibility of recycling sludge into modification blocks. Cement was mixed with carbonized sludge generated at the optimal temperature and reaction time during HTC. After that, the compressive strengths of the modification blocks were measured by conducting both a performance and leaching test. The results of the leaching test showed that heavy metals were not detected, and the specific gravity and absorption rates were less than 1.7 and 10%, respectively, indicating that all species satisfied the criteria. The results of the compressive strength test showed that a mixing ratio of 5% and 7% with cement cured for 28 days satisfied the criteria of A, B, and C type blocks but a mixing ratio of 3% with cement did not satisfy the criteria of A type blocks after 28 days. However, after additional curing for 42 days, the mixing ratio of 3% also satisfied the A type block criteria. Therefore, the optimal mixing ratio of carbonized sludge and cement was considered to be between 3% and 5% and confirmed that the modified blocks could be utilized as aggregates.

Hydrochemical and Microbial Community Characteristics of Spring, Surface Water and Groundwater at Samtong in Cheorwon, South Korea (강원도 철원 샘통과 주변 지표수 및 지하수의 수리화학 및 미생물 군집 특성 연구)

  • Hydrochemical characteristics and microbial communities of spring (Samtong), surface water, and groundwater in Cheorwon, Korea, were analyzed. Field surveys and water quality analyses were undertaken at 10 sampling points for five spring, two surface, and three groundwater samples on 15 December 2022. Hydrochemical analysis revealed that most water samples were Ca-HCO3 type and that water-rock interactions were the predominant mineral source. Radon concentrations were <1 kBq m-3 for surface water, 1~10 kBq m-3 for spring water, and 1~1,000 kq m-3 for groundwater. Microbial cluster analysis showed that the main phyla were Proteobacteria, Planctomyceta, Verrucomicrobia, Acidobacteria, and Actinomycetota.Non-metric multidimensional scaling (NMDS) analysis indicated that water temperature, pH, and Si content were closely related to microorganism content. NMDS and canonical correspondence analysis results revealed that environmental factors affecting spring water were temperature, and Mg and Si concentrations, particularly for Acidobacteria and Proteobacteria, and Pseudomonas brenneri. Both hydrochemical and microbial community analyses yielded similar results at some spring and groundwater sampling points, likely due to the effects of a basalt aquifer.
