Antimicrobial Activity of the Aerial Part of Artemisia capillaris Extracts on the Food-Borne Pathogens (인진호 추출물의 식중독 세균들에 대한 생육억제 효과)

  • The solvent extracts of the aerial part of Artemisia capillaris extracted by using several solvents with different polarities were prepared and their antimicrobial activities were examined. The antimicrobial activities and cell growth inhibitions were investigated to each strain with the different concentrations of the aerial part of Artemisia capillaris Acetone extract showed the highest antimicrobial activity. Minimum inhibitory concentrations (MIC) for each strain were appeared to 20 mg/mL at Staphylococcus aureus and Bacillus subtilis, 40 mg/mL at Vibrio parahaemolyticus, and 80 mg/mL at Salmonella tyhimurium. The cell growth inhibitions were shown on Staphylococcus aureus, Bacillus subtilis, Vibrio parahaemolyticus, and Salmonella typhimurium for 48 hours. The acetone extract was further fractionated sequentially with hexane, chloroform, ethyl acetate, and butanol for purifying crude acetone extract. The solvent fraction of hexane, chloroform, ethyl acetate, and butanol showed the different antimicrobial activity, respectively.

Solvent Fractionation of Fig Leaves and its Antimicrobial Activity (무화과 잎의 용매분획 및 항미생물 활성)

  • Fig leaves were extracted with methanol and then fractionated with ethyl acetate and various buffers to get active fractions, and their antimicrobial activities in each fraction were determined. Acidic fraction and phenolic fraction of fig leaves showed strong antimicrobial activities, but the basic fraction and neutral fraction did not show any activities. The degree of antimicrobial activities in phenolic fraction against tested bacteria were higher than those in acidic fraction, but these against yeasts and mold were almost equivalent to those in acidic fraction. Especially, phenolic fraction showed the strongest activities against Staphylococcuns aureus and Pseudomonas aeruginosa. Although there were some differences among microorganisms, minimum inhibition concentrations(MIC) in acidic and phenolic fractions against tested microorganisms were 0.05 to 0.1 g eq./ml and 0.01 to 0.25 g eq./ml, on the basis of fresh sample weight, respectively. Minimum lethal concentrations(MLC) in acidic and phenolic fractions were 0.25 to 2.00 g eq./ml and 0.05 to 1.50 g eq./ml, respectively. These supposed the antimicrobial activities in phenolic fraction were generally higher than those in acidic fraction and also phenolic fraction had lower MIC and MLC values than acidic fraction.

Antioxidant and antimicrobial activity of solvent fractions from black bamboo leaves (오죽 잎 용매분획물의 항산화 및 항균 활성)

  • To investigate the natural antioxidant and antimicrobial phytochemicals from black bamboo (Phyllostachys nigra MUNRO) leaves, the solvent fractions from crude methanol extract were made with n-hexane, ethyl acetate, and butanol, and their antioxidant and antimicrobial activities were determined. The antioxidant activities were examined by 1,1-diphenyl-1-picrylhydrazyl (DPPH) method and ferric ion reducing antioxidant power (FRAP) method, and the antimicrobial activities against Staphylococcus aureus were tested by paper disc agar diffusion method. Total phenolic contents and total flavonoid contents of the solvent fractions were also determined. The ethyl acetate fraction with the highest total phenolic contents among all fractions showed the strong antioxidant activities by DPPH method and FRAP method, and antimicrobial activities against S. aureus at all test concentrations. Caffeic acid, ferulic acid, quercetin, and kaempferol were analyzed by HPLC in the ethyl acetate fraction from black bamboo leaves by the comparison with the standard chemicals. It is supposed that the ethyl acetate fraction from black bamboo leaves could be used as natural preservatives in the food industry.

The Effects on Antimicrobial and Cytotoxicity of Solanum Iyratum Fractions (배풍등 분획물의 항균 및 암세포 증식 억제효과)

  • In this study, we investigated antimicrobial and cytotoxicity effects to each fraction extracted from Solanum lyratum (SL), which were extracted methanol (SLM) and then the extract was fractionated into five different types : hexane (SLMH), ethyl ether (SLMEE), ethylacetate (SLMEA), butanol (SLMB) and aqueous (SLMA). The antimicrobial activity was analyzed by the paper disc method. Among the various solvent fractions, SLMEA showed the strongest antimicrobial activies. The cytotoxicity of SL fractions on HeLa, MCF-7, HT-29 and HepG2 cells was evaluated by MTT assay. Among various partition layers, SLMEE showed the strongest cytotoxic effects to all cancer cell lines. We also observed that quinone reductase (QR) was induced by all fraction layers of SL to HepG2 cells. Since the QR-induced effects of SLMEE on HepG2 cells at $160{\mu}g/ml$ concentration showed 2.1 when compared with a control value of 1.0, inducer of QR for cancer protection may be contained in this fraction.

An Effective Method of Isolating Immunoglobulins from Bovine Plasma Proteins (도축혈액으로부터 면역단백질의 효과적인 분리법)

  • Imunoglobulins from bovine plasma proteins were isolated by IMAC which $Cu^{2+}$ was chelated on a chelating sepharose fast flow gel. Most plasma proteins were eluted by 1st (0.01 M $Na_2HPO_4$, 0.5 M NaCl, pH 4.0) and 2nd elution buffers (0.01 M imidazol). According to the reverse phase HPLC analysis, it was found that proteins which were eluted by 1st elution buffer were mainly composed of serum albumin, while most IgG and transferrin were eluted by 2nd elution buffer. When protein fractions obtained by 2nd elution buffer was applied to ultra filtration system (molecular weight cut off: 100 kD), IgG was further purified. These results indicate that IMAC is an excellent tool for isolating imunoglobulins from plasma proteins.

Anti-Thrombogenic and Anti-Inflammatory Effects of Solvent Fractions from Leaves of Zanthoxylum Schinifolium (Sancho Namu) in Rats Fed High Fat Diet (고지방식이 흰쥐에서 산초나무 Butanol 및 Methylene Chloride 분획의 항혈전 및 항염증 작용)

  • This study was performed to investigate anti-thrombogenic, anti-inflammatory effects of n-BuOH (B) and $CH_2Cl_2$ (MC) fractions extracted from Sancho (Zanthoxylum. schinifolium) leaves in rats fed high fat diets. The experimental animal groups were consisted of eight including one 5% fat (N) and one 20% fat (H) without the test materials in diets and six H groups of feeding three levels (50, 100 and 150 mg/day) of the B and the MC fractions from Z. schinifolium, respectively. Plasma activated partial thromboplastin times and thrombin times of H group were decreased compared to the N group, but they were increased by feeding the MC fraction of 50 mg and over. Polymorphonuclear leukocyte 5#-lipo-xygenase activities and leukotriene $B_4$ contents of the H group were significantly increased compared to the N group, but they were decreased in the 100 mg and 150 mg of B fraction or the 150 mg of MC fraction fed groups. Liver cytochrome $P_{450}$, $O_2^-$, $H_2O_2$ and GSSG contents were increased by the high fat diet but decreased by feeding the B fraction or the MC fraction, while GSH content and glutathione S-transferase activity lowered by high fat diet were increased by feeding the two solvent fractions. The effects of the solvent fractions were evident at the level of 100 mg/day and over. The present results confirmed that two solvent fractions from the leaves of Z, schinifolium have enhancing effects on anti-thrombosis and anti-inflammation partly by antioxidant action and partly by direct modulation of the respective processeds. In conclusion, the n-BuOH and $CH_2Cl_2$ fractions from leaves of Z, schinifolium can be utilized as the proper ingredients of functional foods for preventing chronic degenerative disease.

Antioxidant Activities of Ulva lactuca Extracts with Different Solvents (갈파래(Ulva lactuca) 용매별 분획의 항산화활성)

  • The fractions of Ulva lactuca were studied to verify the antioxidant activities. The fractions from the ethanol extract of U. lactuca were prepared by the systematic extraction procedure with the solvents such as hexane, ethyl ether, ethyl acetate, butanol and $H_2O$. Furthermore, ethyl ether, ethyl acetate and aqueous fractions of U. lactuca were purified using HPLC. The antioxidant activities of purified samples from ethyl ether, ethyl acetate, and aqueous fractions were investigated using 1,1-diphenyl-2-picryl-hydrazil (DPPH). L-ascorbic acid, a positive control showed the highest DPPH radical scavenging activity. In addition, purified sample from aqueous fraction also showed relatively high activity. Purified sample from ethyl acetate fraction showed moderate activity, but purified sample from ethyl ether fraction showed the lowest activity. Dose dependent patterns were observed on all three samples tested. The lipid peroxidation inhibition activities of these three purified samples were also investigated. Purified sample from ethyl ether fraction of U. lactuca showed the highest activity and as strong activity as that of $\alpha-tocopherol$, a positive control. These results suggest that U. lactuca may be a useful candidate for a natural antioxidant agent.

Antioxidant and Antiproliferative Activities of the Halophyte Angelica japonica Growing in Korean Coastal Area (한국 연안지역에 서식하는 갯강활의 항산화 및 암세포증식 억제 활성)

  • This study evaluated the antioxidizing and antiproliferative effects of Angelica japonica extract and its solvent-partitioned fractions. A dried sample of the halophyte A. japonica was extracted twice using methylene chloride (CH2Cl2) and extracted twice again using methanol (MeOH). The combined crude extracts were then fractionated by solvent polarity into distilled water (water), n-butanol (n-BuOH), 85% aqueous methanol (85% aq.MeOH), and n-hexane fractions. The antioxidant activities of the crude extracts and their solvent-partitioned fractions were assessed according to their DPPH radical and peroxynitrite scavenging abilities, formation of intracellular reactive oxygen species (ROS), DNA oxidation, NO production, and ferric reducing antioxidant power (FRAP). The crude extract showed significant antioxidant activity in the overall antioxidizing bioassay systems. Among solvent-partitioned fractions, good antioxidant activities were observed in n-BuOH and 85% aq.MeOH fractions and significantly correlated with the polyphenol and flavonoid contents of the samples. Furthermore, all samples tested, including the crude extract, not only showed cytotoxic effects against human cancer cells (AGS, HT-29, MCF-7, and HT-1080) but also prevented cell migration in a dose-dependent manner in the wound healing assay using HT 1080. Among the solvent-partitioned fractions, the 85% aq.MeOH fraction most effectively inhibited the invasion of HT-1080 cells. Therefore, these results suggest that A. japonica may be a potential antioxidizing and antiproliferative agent.

Antimicrobial and Antioxidant Activities of Various Solvent Extract from Different Parts of Loquat (Eriobotrya japonica, Lindl.) (비파 부위별 용매추출물의 항균 및 항산화 활성)

  • Antimicrobial and antioxidative activities of various solvent extracts from different parts of the loquat were investigated to process it as the functional food. The extraction yield showed higher in butanol fraction of 4.95% in peel and 4.42% in seed than others, but water fraction showed high extraction yield of 3.89% in leafs 23.6% in the seedless fruit and 21.1% in fresh, In the antimicrobial activity test, ethyl acetate fraction that leaf and fruit excluded seed inhibited higher 19, 15 mm and 16, 15 m in clear zone far Staphylococcus aureus and Escherichia coli in each. Ethyl acetate fraction that extracted from a series of polar and nonpolar solvent fractionation of methanol extracts showed better results in the hydrogen donating activity of 82% in leaf, 74% in seedless fruit, 68% in seed, 52% in peel and 30% in fresh.

Concentration of Polyunsaturated Fatty Acids from Anchovy Oil by Supercritical Carbon Dioxide (초임계 이산화탄소에 의한 멸치어유의 고도불포화지방산 농축)

  • To concentrate polyunsaturated fatty acids from anchovy oil by supercritical carbon dioxide $(SC-CO_2)$, effects of different adsorbents, cosolvents and extraction pressures on the extraction yield and fatty acid composition in the extract and the residue fractions were investigated. Anchovy oil mixed with silver nitrate-coated silica gel showed higher extraction yield and concentration of EPA and DHA in the residual fraction than that mixed with only silica gel at $60^{\circ}C/345{\;}bar$. Ethyl acetate was a promising cosolvent for concentrating polyunsaturated fatty acids in the residual fraction from anchovy oil mixed with silica gel. For the extraction pressures tested, 276 bar showed a maximum value in the extraction yield and concentration of EPA and DHA in the residue. Starting with anchovy oil containing 13.3% EPA and 16.2% DHA mixed with a silver nitrate-coated silica gel, the residue fraction containing 28.2% EPA and 38.3% DHA was obtained when ethyl acetate was used as a cosolvent with $SC-CO_2$ at $60^{\circ}C/276{\;}bar$.
