자화된 $SF_6$ 유도결합형 플라즈마를 이용한 SiC 식각 특성에 관한 연구

  • Silicon carbide (SiC)는 높은 power 영역과 높은 온도영역에서도 작동 가능한 우수한 반도체 물질이다. 또한 우수한 열적 화학적, 안정성을 가지고 있어 가흑한 조건에서의 소자로써도 사용 가능하다. 현재 SiC 적용분야로는 우수한 전기적, 기계적 성질을 이용한 미세소자(MEMS)와 GaN 와 거의 유사한 격자상수를 가지는 것을 이용한 GaN epitaxial 성장의 기판으로도 사용되어진다. 그러나 SiC 는 기존의 습식식각 용매에 대해 화학적 안정성을 가지고 있기 때문에 전자소자의 제작에 있어서 플라즈마를 이용한 건식식각의 중요성이 대두되어지고 있다. 소자제작에 있어 이러한 건식식각시 식각 단면의 제어, 이온에 의한 낮은 손상 정도, 매끄러운 식각 표면, 그리고 고속의 식각 속도둥이 요구되어진다. 본 실험에서는 식각 속도의 증가와 수직한 식각 단면둥을 획득하기 위하여 SF6 플라즈마에서 Source power, dc bias voltage, 그리고 외부에서 인가되는 자속의 세기를 변화시쳐가며 식각 속도, 식각 마스크와의 식각 션택비, 식각 단면둥과 같은 SiC 의 식각 특성을 관찰하였다. 식각 후 식각 단면은 주사전자 현미경(SEM)을 통해 관찰하였다. 본 실험에서의 가장 높은 식각 속도는 분당 1850n 로써 이때의 공정조건은 1400W 의 inductive power, -600V 의 dc bias voltage, 20G 의 외부자속 세기이었다. 또한, 높은 inductive power 조건과 낮은 dc bias voltage 조건에서 Cu는 $SF_6$ 플라즈마 내에서 식각부산물의 증착으로 인해 SiC 와 무한대의 식각선택비를 보였다. 이러한 Cu 마스크를 사용한 SiC 의 식각에서는 식각 후 수직한 식각 단변을 관찰할 수 있었다. 것올 알 수 있다. 따라서, 기존의 pve 보다 세라믹 기판의 경우가 수분 흡수율이 높아 더 오랫동안 전류를 흐르게 하여 방식성이 개선된 것으로 판단된다.을 통해 경도가 증가한 시편의 경우 석출상의 크기가 5nm 이하로 매우 작고 대체로 기지와 연속적인 계면을 형성하나, 열처리가 진행될수록 석 출상의 크기가 커지고 임계크기 이상에 이르면 연속적인 계면은 거의 발견되지 않고, 대부 분 불연속적이고 확연한 계면을 형성함을 관찰 할 수 있었다. 알루미나(${\alpha}-Al_2O_3$) 기판 위에 증착한 $(Ti_{1-x}AI_{x})N$ 피막은 마찬가지로 (200) 우선 방위를 나타내었으나, 그 입자의 크기가 수십 nm로 고속도강위에 증착한 피막에 비해 상당히 크게 형성되었다. 또한 열처리 후에 AIN의 석출이 진행됨에도 불구하고 경도 증가는 나타나지 않고, 열처리가 진행됨에 따라 경도가 감소하는 양상만을 나타내었다. 결국 $(Ti_{1-x}AI_{x})N$ 피막이 열처리 전후에 보아는 기계적 특성의 변화 양상은 열역학적으로 안정한 Wurzite-AlN의 석출에 따른 것으로 AlN 석출상의 크기에 의존하며, 또한 이러한 영향은 $(Ti_{1-x}AI_{x})N$ 피막에 존재하는 AI의 함량이 높고, 초기에 증착된 막의 업자 크기가 작을 수록 클 것으로 여겨진다. 그리고 환경의 의미의 차이에 따라 경관의 미학적 평가가 달라진 것으로 나타났다.corner$적 의도에 의한 경관구성의 일면을 확인할수 있지만 엄밀히 생각하여 보면 이러한 예의 경우도 최락의 총체적인 외형은 마찬가지로 $\ulcorner$순응$\lrcorner$의 범위를 벗어나지 않는다. 그렇기 때문에

Redifferentiation of the Cutaneous Pigment System during the Wound Healing Process in the Goldfish, Carassius auratus (금붕어 (Carassius auratus L.) 상처치유과정중 피부색소체계의 재분화에 관한 연구)

  • The regeneration and differentiation of the cutaneous pigment system in the goldfish, Carassius auratus during the wound healing process were studied with high magnification electron microscope. The cutaneous pigment cells of the normal tissues were composed of three kinds of dermal chromatophores-xanthophores, leucoiphores and melanophores. While xanthophores contain two kinds of pigment granules-pterinosomes and carotenoid vesicles, leucophores and melanophores contain amorphous pigment granules (leucosomes) and oval shaped electron dense melanin pigment granules (melanosomes) respectively. After injury, primary wound healing responses being carried out by migration of epidermal cells and hemocytes spreading over the wound surface at the day of wounding. And at the time of primary wound closure, 5 to 7 days after wounding, rER rich cells-presumably common precursors of dermal chromatophores-immigrated into the wound area. First redifferentiated chromatophores appeared 3 weeks after wounding. Pigment granules of the chromatophores were emerged from the cytoplasmic Golgi complex via rough endoplasmic reticulum. Pinocytotic vesicles which associated with accumulation of pigment material, appeared only at the inner surface of the chromatophores adhering to the rER rich cells, characteristically. The differentiation of each chromatophore in addition to integumental wound repair were accomplished within 4 weeks after wounding at most cases, however the total numbers and densities of these repaired chromatophores still primitive state. Moreover, It has been revealed that complete repair of chromatophores at wounded tissues from burns requirs more than 3 months in normal environment.

Protective Effect of Cosmetics Containing Red Beet against Cigarette Smoke-induced Oxidative Damage in Human Skin (레드비트를 함유하는 화장품의 담배 연기에 의한 피부 지질 산화 방지 효과)

  • In cosmetics market, anti-pollution products recently come up with new solution for skin health. Environmental oxidation mechanisms are realized as bio-marker of atmospheric pollution upon skin by environmental pollutant such as ozone, UV rays, particulate matter (PM) as well as cigarette smoke. The exposure of cigarette smoke directly or indirectly causes the oxidation of the stratum corneum skin lipids, resulting in the conversion of squalene to squalene monohydroperoxide and/or generation of malondialdehyde (MDA) as a product of lipid peroxidation. The aim of this study is to see whether new cosmetics product containing red beet has anti-oxidation effect on skin exposed by cigarette smoke. So as to determine oxidative damage to human skin at biochemical level, each unit area of volar forearms was exposed to cigarette smoke through device (3.3 cm, diameter) for fifteen minutes, then measured MDA using standardized TBARS assay kit. Compared to negative control (untreated and unexposed area), the level of MDA was significantly increased at positive control (untreated and exposed area) more than 3.7 times, indicating the pollutant induced-oxidative damage on the skin barrier. Whereas, the pre-applied area with the cosmetics products containing red beet revealed a decrease of 25% compared with positive control. As reports, these data demonstrated that cigarette smoke induce peroxidation of stratum corneum skin lipids. Conclusively, we suggest that anti-pollution effect of the cosmetics product containing red beet is beneficial to prevent the oxidation of skin lipids by atmospheric pollution.

  • Thermal power generation accounts for the highest percentage of domestic power generation, among which coal-fired boiler generation accounts for the highest percentage. Coal boilers generate harmful substances and fine dust during coal combustion and have a serious effect on air pollution. So, fluidized-bed boilers have been introduced as eco-friendly coal boilers. It uses a fluid medium which affect the combustion temperature of coal. Because of it fluidized-bed boilers emit less pollutants than original one. Water-wall tubes play an important role in this fluidized bed boiler. Due to the fluid medium, the wall damage is more severe than the existing boiler. However, there is no quantitative maintenance technique in Korea yet. Remote field eddy current testing is a non-destructive evaluation technique that is often used for inspection of inner and outer wall of tube. it can inspect with non-contact and high speed. However, it is an inspection that proceeds from inside the pipe, and the water-wall tube is not able to enter the interior. In this study, we designed and simulated an external remote field eddy current sensor suitable for water-wall tube of a fluidized - bed boiler using simulations. By obtaining a signal similar to the existing remote field eddy current test, the criteria for the external remote field eddy current sensor design can be presented.

Regulatory Mechanism of Lipopolysaccharide(LPS)-Induced Interleukin-8 Gene Expression in Mononuclear Phagocytic Cells (단핵식세포에서 내독소에 의한 인터루킨-8 유전자 발현 조절기전에 관한 연구)

  • Background : In acute lung injury, activated neutrophils play an important role in tissue damage. For neutrophils to participate in lung inflammation, chemotactic factors released from mononuclear phagocytes are needed to bring these cells to the local site of inflammation, with interleukin-8 (IL-8) being one of the most specific and important chemotactic factors for neutrophils. IL-8 also induces the expression of adhesion molecules and activates neutrophils to release various inflammatory mediators. Lipopolysaccharide(LPS) is one of the most important causes of adult respiratory distress syndrome and can cause release of many inflammatory cytokines including IL-8 leading to acute lung injury. But little is known about the regulatory mechanism of LPS-induced IL-8 gene expression in mononuclear phagocytes. Method : Human alveolar macrophages(HAM) and peripleral blood monocytes(PBMC) were isolated from healthy volunteers. Time and dose relationship of LPS-induced IL-8 mRNA expression was observed by Northern blot analysis. To evaluate the regulatory mechanism of LPS-induced IL-8 gene expression, pretreatment of actinomycin D(AD, $5{\mu}g/ml$) and cycloheximide(CHX, $5{\mu}g/ml$) was done and Northern blot analysis for IL-8 mRNA and ELISA for immunoreactive IL-8 protein in culture supernatant were performed. Results : 1) In HAM, dose and time dependent LPS-induced IL-8 mRNA expression was observed with peak mRNA level at 8 hours post-stimulation. 2) In PBMC, dose and time dependent LPS-induced IL-8 mRNA expression was also observed with peak mRNA level at 4 hours post-stimulation. 3) AD decreased expression of LPS-induced IL-8 gene expression at both mRNAand protein levels in both types of cells. 4) CHX decreased expression of LPS-induced IL-8 gene expression at protein level in both cell types but in HAM, superinduction of IL-8 mRNA was observed while decreased expression of IL-8 mRNA was observed in PBMC. Conclusion : Time and dose dependent LPS-induced IL-8 gene expression was observed in mononuclear phagocytes which is at least partly regulated pretranslationally. LPS-induced IL-8 mRNA expression in HAM needs no de novo protein synthesis and may be under the control of a labile repressor protein while de novo protein synthesis may be needed in PBMC.

고 선량율 근접 및 온열치료 병용 삽입관의 제작과 특성

  • 악성종양을 치료하는 방법중 방사선과 온열요법은 가장 강력한 치료방법으로 연구되어왔으며 이를 병용함으로 서 상승효과를 얻을 수 있다. 인체조직에 41$^{\circ}C$ 이상의 열을 가하면 세포질의 단백질변성으로 세포에 손상을 주어 세포가 사멸하게 되며 세포의 생존율은 가열시간 즉 열량에 따라 지수적으로 감소한다. 온열은 세포주기중 방사선 저항성이 매우 큰 DNA 합성시기와 산도가 높을 때 감수성이 매우 크기 때문에 방사선과 병용요법은 상호 상승효과를 가져온다. 이와 같이 온열을 이용한 악성종양의 치료가능성은 생물학적 기초연구와 임상시험에서 경이적인 효과를 얻을 수 있었으나 아직 까지 가열방법과 온도분포측정이 큰 과제로 남아있으며 주위건강조직의 가열을 피하면서 인체 깊은 곳에 존재하는 종양에만 집중 가열하는 방법인 삽입형 온열치료방법에 대한 연구가 집중되었다. 한편 방사선 치료방법은 주위 건강조직의 피폭을 최소로 줄이고 종양에만 집중 조사가 요구되며 자궁암, 유방암, 뇌암등 부피가 작고 집중적 치료를 요하는 종양은 방사성동위원소를 이용한 근접 삽입치료 (Brachyradiotherapy)가 큰 효과를 나타내고 있다 방사선과 온열의 병행 치료를 위하여 방사선 삽입 치료에 사용한 선원 삽입관을 그대로 두고 삽입관 속에 방사성 동위원소 대신 온열 전극을 넣어 열을 가하는 방사선 온열 병용치료방법을 고안하였으며 방사선과 온열병용에 사용할 최적 삽입관의 제작과 이에 따른 온도분포의 측정과 최적삽입방법을 결정하였다. 방사선 삽입치료용 폴리에찌렌 삽입관의 외부에 금박을 입혀 라디오파 첨극을 삽입할 때 서로 연결되도록 고안 제작함으로서 방사선 삽입치료와 자입식 온열치료를 동시에 만족하게 수행할 수 있는 병용삽입관 (Flexible thermoradiotherapy probes)을 제작하였다. 전도율이 큰 금박부위가 직접 조직에 접촉됨으로 라디오파의 전달이 용이하며 금박의 길이를 2 cm 에서 5 cm 로 구분제작 함으로서 종양의 크기와 모양에 따라 선택할 수 있도록 하였다. 라디오파를 이용한 온열분포의 측정은 인체조직과 전기적 특성이 비슷한 물질인 한천 팬텀 제작하여 사용하였으며 온도분포 측정은 열전대와 서머그람으로 시행하였다. 생체조직 내에서의 온도분포와 온열효과를 관찰하기 위하여 직접 개의 뇌를 이용하여 시행하였으며 4 개의 전극을 이용하여 43$^{\circ}C$로 50분간 가열하고 일주일후 개를 회생시켜 개 뇌에 대한 조직학적 검사를 시행하였다. 한편 팬텀 표면에서 중앙부로 안테나 길이가 2 cm 인 4 개의 전극을 1 cm 간격으로 정사각형이 되도록 삽입하여 가열하였을 때 90% 등온곡선이 반경 1.25의 원형으로 균일하게 분포되었고 종단면상 삽입관의 길이에 따라 균일한 온도분포가 이루어졌다. 전극을 2 cm 간격으로 삽일 하였을 때 90% 등온곡선이 1.75 반경으로 거의 4 각형의 균일한 분포를 얻었으나 전극의 간격이 증가하면 전도율이 떨어져서 전극 중심부에 불균일한 온도분포를 형성하였다. 동물실험에서 정상 개의 뇌 실질에 자입하여 직접 정방형의 중심을 43$^{\circ}C$로 유지하며 50분간 온열 요법을 시행한 후 관찰한 조직병리학적 소견은 liquefactive necrosis, pyknosis of neuronal element 및 polymorphonuclear leukocytes들의 회백질에서 급성기에 관찰되었고 liquefactive necrosis 주위에 lipid-laden macrophage들이 관찰됨이 공통적인 특정이었으며 후기변화로 괴사조직 주위로 신경교세포의 증식이 관찰되었다.

The Effect of Cordycepin on the Production of Pro-inflammatory Cytokines in Mouse Peritoneal Macrophages (코디세핀이 마우스 복강 대식세포에서 전염증성 사이토카인의 생성에 미치는 영향)

  • The effect of cordycepin purified from Cordyceps militaris on macrophage activation was investigated in peritoneal macrophages isolated from C57BL6 mice. Lipopolysaccharide-induced mouse peritoneal cells showed that cordycepin treatment increased the expression of the inflammatory cytokines interleukin (IL)-$1{\beta}$, IL-12, and tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), leading to early inflammation-mediated reactions, the activation of immunological responses, and T lymphocyte activation. T lymphocytes, activated by a greater production of IL-6, resulted in antibody-generating immune reactions, suggesting that cordycepin was effective at inducing immunological responses. Consistent with the increase in the inflammation-mediating factors including nitric oxide (NO) and hydrogen peroxide ($H_2O_2$), the toxic response of macrophages was activated and effectively induced inflammation. These findings demonstrate that cordycepin is involved in reducing cell injury provoked by inflammatory reactions. Therefore, these results suggest that cordycepin treatment of mouse peritoneal cells induces inflammation-mediated immunological responses and immunostimulation.

Effect to the Copper System Pigments by the Nitrogen Dioxide(NO2) Gas (이산화질소(NO2)가 구리(Cu)계통 안료에 미치는 영향)

  • Malachite and Azurite are the typical copper system pigments which used the mural paintings since ancient times. The mural painting is at risk for damages of the painting layer by atmosphere gas because it is exposed at external environment. In this study, it did experiment about an effect to Malachite and Azurite by environmental pollution gas($NO_2$, $CO_2$, $SO_2$) then analysis and estimate about test for pieces using mural painting colored that two pigments. As a result, Malachite and Azurite were changed on $NO_2$ but not changed $CO_2$ and $SO_2$. Especially as the concentration of $NO_2$ is increased, exfoliation of the pigment layer weave remarkably formed pores on the pigment particles on SEM, the phenomenon to be pieces were observed together with smaller particles. In the case of Malachite that were exposed to $NO_2$ gas, new compounds(Rouaite : dicopper (nitrate(V) trihydroxide, $Cu_2(NO_3)(OH)_3$)) was appeared by XRD analysis. Therefore, there had been able to verify the fact that the cause exfoliation and discoloration phenomena accompanied by chemical changes for Malachite and Azurite.

The Protective Effects of an Ethyl Acetate Fraction of Raphiolepsis indica Against Oxidative Stress in HaCaT Keratinocytes (HaCaT 피부 각질세포의 산화적 스트레스에 대한 다정큼나무 에틸 아세테이트 분획물의 세포 보호 효과)

  • In a previous study, the total phenolic content in ethanol extracts of medicinal plants that naturally grow on Jeju Island were analyzed with the extracts of Raphiolepsis indica leaf found to have the highest. The current study was carried out to evaluate the total flavonoid content, radical-scavenging activity, and the protective effect of R. indica extracts and solvent fractions on oxidative stress in HaCaT keratinocytes. More specifically, total flavonoid content and 2-azino-bis 3-ethylbenzthiazoline-6-sulfonic acid (ABTS) radical-scavenging activity were measured to assess anti-oxidative activity, and protective effects against hydrogen peroxide (H2O2) were determined by 3-[4, 5-dimethylthiazol-2-yl]-2, 5-diphenyltetrazolium bromide assay on the HaCaT cells. Of the various fractions analyzed, the ethyl acetate extract of R. indica showed the highest total flavonoid content (149.13 mg/g extracts) and the lowest remaining ABTS. In addition, the ethyl acetate fraction was significantly more resistant against H2O2 than the negative control. Our results therefore suggest that an ethyl acetate fraction of R. indica protects HaCaT cells against oxidative stress and could prove useful for developing functional cosmetic materials.

Anti-Fibrotic Effects by Moringa Root Extract in Rat Kidney Fibroblast (모링가 뿌리 추출물에 대한 신장섬유화 억제 효과)

  • Fibrosis in kidney by internal and external factors causes progressive loss of renal function. Renal fibrosis is the inevitable consequence of an excessive accumulation of the extracellular matrix. TGF-${\beta}$ plays an important role in the process of renal fibrosis and stimulates the synthesis of profibrotic factors, including collagens, fibronectin, and plasminogen activator inhibitor (PAI-1). We examined the effect of Moringa oleifera Lam (moringa) extracts in a rat kidney fibrosis model. We found that moringa root extract suppresses protein expression/mRNA levels of Type I collagen, fibronectin, and PAI-1 induced by TGF-${\beta}$ in renal fibroblasts. Moringa root extract selectively inhibited phosphorylation of TGF-${\beta}$-induced $T{\beta}RII$ and the downstream signaling pathway (e.g., Smad4), and phospho-ERK, but not JNK, p38, or PI3K/AKT. These results suggest that moringa root extract can act against TGF-${\beta}$-induced renal fibrosis in rat kidney fibroblast cells by a mechanism related to its antifibrotic activity, which regulates expression of fibronectin, Type I collagen, and PAI-1 through $T{\beta}RII$-Smad2/3-Smad4 and ERK. Therefore, moringa root extract is an effective substance for fibrosis therapy and provides a new therapeutic strategy for diseases associated with elevated profibrotic factor synthesis.
