Plant regeneration from protoplasts-derived from embryogenic callus of Citrus (감귤 embryogenic callus 원형질체 배양에 의한 식물체 재분화)

  • This study describes conditions for plant regeneration from protoplasts-derived from embryogenic callus of satsuma mandarin. Plants were generated via somatic embryogenesis. Protoplasts isolated directly from nucellar callus induced from immature ovule of satsuma mandarin cv. Okitsu (Citrus unshiu Marc.) were cultured in 0.6M $BH_3$ medium. Cell division and plating efficiency were affected by protoplast culture method. The liquid over solid method was the most effective for formation of microcalli. Most of microcalli grew rapidly and transferred onto embryoid formation medium. Optimum embryoid formation medium was MT medium containing 1.5 g/L malt extract, 0.146 M sucrose and the medium for plantlet regeneration was MS medium containing 0.09M sucrose, 1.0 mg/L $GA_3$. No differences were noticed in growth habits and leaf characters such as shape, thickness, and colour between protoplast-derived plants and nucellar seedlings. This plant regeneration system from protoplasts-derived from embryogenic callus provides an alternative way for producing new scion and rootstock cultivar from citrus species which can not be crossed.

Spatial Distribution and Sampling Plan for Pink Citrus Rust Mite, Aculops pelekassi (Acari: Eriophyidae) in Citrus Orchard (감귤원에서 귤녹응애 공간분포 분석과 표본조사법 개발)

  • The dispersion indices, spatial pattern and sampling plan for pink citrus rust mite (PCRM), Aculops pelekassi, monitoring was investigated. Dispersion indices of PCRM indicated the aggregated spatial pattern. Taylor's power law provided better description of variance-mean relationship than Iwao's patchiness regression. Fixed-precision levels (D) of a sequential sampling plan were developed using by Taylor's power law parameters generated from PCRM on fruit sample (cumulated number of PCRM in $cm^2$ of fruit). Based on Kono-Sugino's empirical binomial the mean density per $cm^2$ could be estimated from fruit ratio with more than 12 rust mites per $cm^2$: $ln(m)=4.61+1.23ln[-ln(1-p_{12})]$. To determine the optimal tally threshold, the variance (var(lnm)) for mean (lnm) in Kono-Sugino equation was estimated. The lower and narrow ranged change of variance for esimated mean showed at a tally threshold of 12. To estimate PCRM mean density per $cm^2$ at fixed precision level 0.25, the required sample number was 13 trees, 5 fruits per tree and 2 points per fruit (total 130 samples).

Spatial Distribution Pattern of Ascotis selenaria (Lepidoptera: Geometridae) larvae in a Small-Scale of Citrus Orchard (소규모 감귤원에서 네눈쑥가지나방 유충의 공간분포 특성에 대한 이해)

  • This study was conducted to understand the settlement process of Ascotis selenaria larvae into citrus orchards with respect to oviposition site and analysis of the spatial distribution pattern of the larvae. A. selenaria eggs were not found on citrus trees in field and green house, but not on citrus trees in the field. A. selenaria larvae showed a significant clump distribution in the greenhouse. In the open citrus field, the index of dispersion was around 1.0 in most cases, with a weak clumping degree. However, the d-statistic was between -1.96 and 1.96, indicating a statistically significant random distribution. In addition, the Green's index (a clumping index) was very low in all cases, even though the clump distribution was accepted. for most samples, the probability distribution of larval frequency in the field satisfied the probability distribution functions of Poisson (random pattern) and the negative binomial (clump pattern) distribution. In addition, the temporal distribution of the larvae in the open field showed a pattern which was formed by colonizers from outside oviposition sites. Further, the difference in larval spatial distribution between field and greenhouse orchards was discussed.

Improvement of Control Efficacy of Mancozeb Wettable Powder against Citrus Melanose by Mixing with Paraffin Oil (파라핀유와 혼용 살포에 의한 만코제브 수화제의 감귤 검은점무늬병 방제 효과 증진)

  • This study was carried out to determine the effect of mixing with paraffin oil on rainfastness of mancozeb on citrus fruits and assay the improvement of control effect of mancozeb against citrus melanose by mixing with paraffin oil. In artificial rainfall condition (7.2 mm/hr), the attached contents of mancozeb on detached fruits were the most in treatment of mancozeb 0.2% + paraffin oil 0.1% as the contents was $7.43{\mu}g/cm^2$ after treatment of rainfall for 10 hr. In open field condition, the contents of attached mancozeb on fruits were significantly more in treatment of mancozeb 0.2% + paraffin oil 0.1% or 0.25% than the other treatments 1, 15 and 25 days after treatment 2009 and 2010 seasons. The disease incidence was significantly lower in treatment of mancozeb 0.2% + paraffin oil 0.1% or 0.25% than treatment of mancozeb 0.2% only 2009, 2010 and 2011 seasons. Based on this study, it was suggested that the control effect of mancozeb against citrus melanose can be improved by mixing with paraffin oil.

Physicochemical Properties and Sensory Evaluation of Citrus Unshiu Produced in Cheju (제주산 온주밀감의 특성과 관능평가)

  • Soluble solids $(^{\circ}Brix)$, acid content and vitamin C content of Citrus miyakawa unshiu and C. okitsu unshiu juice harvested middle of November 1993 in Seuho-Dong and Topyung-Dong, Seogwipo-si, south area of Cheju were $10.7{\sim}11.0$, 1.04% and $41.19{\sim}44.94\;mg/100\;g$ juice, respectively. Carbohydrate was consisted of about 1/2 sucrose, 1/4 glucose, and 1/4 fructose. Fruit weight, peel thickness, soluble solids, pH, acid content and Brix/Acid ratio had a good correlation in linear function with increasing fruit size, respectively. However, hardness and edible part ratio were not showed correlation with increasing fruit size. Middle size of citrus fruit was favorable to panelists, and the citrus peel color of red yellow was more favorable than pale yellow color in sensory evaluation. Sensory evaluation score showed a linear correlation among citrus fruits of C. okitsu unshiu produced in Namwon-ri, Seuho-Dong, Seogwipo-si, south area of Cheju, and that of Chochun-ri, Ara-Dong, Cheju-si, north area of Cheju, and the score was higher according to citrus fruit produced in optimum cultivation area. With increasing Brix/Acid ratio of citrus juice, high sensory evaluation score was gained in panelists. These data obtained are supposed to be applied to the quality evaluation of Citrus unshiu produced in Cheju.

Anti-Inflammatory Effect of Peel Extracts from Citrus Fruits (감귤류 과피 추출물의 항염증 효과)

  • The following study was presented to investigate the anti-inflammatory effect of peel extracts (PE) from three citrus fruits: Citrus unshiu, Citrus limonia Osbeck and Citrus hallabong. According to this study, cytotoxicity, NO-production and protein levels of iNOS (inducible nitric oxide synthase) in macrophage cell were analyzed, which had been incubated in murine macrophage cell line RAW 264.7 cell of PE from those three citrus fruits. According to Citrus unshiu peel extracts (CUP), Citrus limonia Osbeck peel extracts (CHP) and Citrus hallabong peel extracts (CLP) treatment, the result showed that there was no cell growth inhibited below 2 mg/mL. Comparing the NO-production of the cell with LPS (100 ng/mL) and the treatment without LPS, significant increase of NO-production was detected. However NO-production also showed decrease trend, as the concentration increased. For each treatment, at the concentration of 1 mg/mL, NO-ihibitory activity showed significant result with following order: CUP > CHP > CLP. According to the result from Western blot, the inhibitory activities of iNOS protein from CUP and CHP showed fairly similar performances. Also inhibitory activity of COX-2 showed the following order: CUP > CHP> CLP. There was no doubt that all the treatments of CUP, CHP and CLP have anti-inflammatory effect and also that the inhibitory activity of the CUP treatment was the strongest among those three.

Effects of Petroleum Spray Oil on Photosynthesis Characteristics in Citrus Leaves (Petroleum Spray Oil 살포가 감귤 잎의 광합성관련 특성에 미치는 영향)

  • Recently, petroleum spray oil(PSO) has been used to control key pests in integrated pest management (IPM) of citrus and other orchards in Australia and USA. In order to clarify the influences of a newly developed PSO (D-C Tron $Plus^{(R)}$) on citrus leaves, 0.33% or 1.0% of PSO were sprayed to potted 4-year-old citrus trees under some kinds of condition, and then the changes of photosynthesis, transpiration, stomatal conductance and chlorophyll fluorescence(Fv/Fm) were determined. When sprayed with 1.0% PSO, the photosynthetic rate, transpiration and stomatal conductance of citrus leaves were decreased by 20%, and then recovered in 20 days after treatment (DAT), while there were little influences by the spray of 0.33% PSO. The value of Fv/Fm decreased more under the $34/24^{\circ}C$ temperature condition than that of under the $30/20^{\circ}C$ and $28/16^{\circ}C$ condition. The high temperature ($50^{\circ}C$ for 10 hours)-treated trees sprayed with PSO 1.0% or PSO 1.0% plus dithianon 1/2000 dilution showed not only the increase of rate in dropped leaf but also the reduced photosynthesis and Fv/Fm compared with $30/20^{\circ}C$ temperature-treated ones. From the results of this study, the spray of 1.0% PSO can inhibit the physiological activities in citrus leaf, particularly under high temperature condition after spray or the mixing-spray with a fungicide (dithianon WP, 75%).

Anti-inflammatory Effect of Citrus unshiu Peels Fermented with Aspergillus niger (Aspergillus niger로 발효한 감귤과피 추출물의 항염효과)

  • Citrus, yield of which is the highest among fruits produced in Korea, is extensively consumed for processed food items. The amount of by-products of citrus produced from the processing within a short period after the harvest is tremendous. These by-products are mostly dumped into land or neglected because of cost involved in processing them. The aim of the present study was to explore the usefulness of the by-products as a new material by examining the anti-inflammatory activity of fermented extracts of citrus peels. The peels of 'unshu' (Citrus unshiu) was fermented with Aspergillus niger and their extracts before or after fermentation were analyzed using HPLC. The analysis showed that neohesperidin level considerably increased and the two new compounds were synthesized after fermentation. The anti-inflammatory activity of the fermented extracts was examined on RAW 264.7 murine macrophage cells stimulated with lipopolysaccharide. Fermented unshu extracts significantly enhanced the decrease of nitric oxide (NO) production, iNOS and COX-2 expression, comparing with those of unfermented extracts. Also TNF-${\alpha}$ and IL-6 production, both of which are pro-inflammatory cytokine, were more inhibited in fermented extracts. These results showed that the fermentation and promotion of the function of the by-products of citrus peels will help find a new application.

Effects of Water Microspraying, Foliar Application of Calcium Carbonate and Kaolin Solution on Sunburn and Quality of Fruits in Satusma Mandarin (미세살수와 탄산칼슘 및 카올린 엽면살포가 온주밀감의 일소 발생과 과실 품질에 미치는 영향)

  • This study was carried out to predict fruit surface temperature (FST) causing sunburn symptoms and assess the correlation between solar radiation and FST in satusma mandarin (Citrus unshiu Marc. 'Iwasaki'). And to reduce sunburn when ambient temperature was at 31℃, sprinkler system was repeated spraying every at 5 minutes of 30 minutes intervals and 1% calcium carbonate and 4% kaolin solution applied on citrus tree three times to investigate quality and sunburn of fruits. It showed highly significant positive correlation between FST and solar radiation (R = 0.788, p < 0.01), The FST increased up to 16℃ than ambient temperature at daytime. To estimate FST, a linear model y = 0.099 × (air temperature) + 0.018 × (solar radiation) + 20.779 (R = 0.687, p < 0.01) was derived. The canopy temperature of citrus trees was 5.1℃ lower in sprinkler system than in control. The incidence of sunburn showed at 2.1% in sprinkler system twice lower than in control and there was different significance. Fruit quality was similar between treatments. In conclusion, sunburn can be reduced by reflecting sunlight or downing of FST through sprinkler system.

  • Diets containing 0.02 to 10.0% EM-fermented orange (EFO) were fed to Paralichthys olivaceus(8.5 g), which were reared in flow-through system for 16 weeks. Groups fed on diets containing 0.1 and 0.2% EFO grew significantly faster; their feed coefficient and daily feeding rate were also higher. The number of goblet cells present in the mid-intestine of the fish receiving dietary EFO was significantly more. Total cholesterol level in plasma of the fish fed with 0.02% EFO was lower. However, there was no significant difference in GOT and GPT among the groups receiving different levels of EFO.
