Studies on the Photosynthesis of Korean Ginseng 1. Seasonal Changes in Photosynthetic Ability of Youngaged Korean Ginseng (Panax ginseng C. A. Meyer) (고려인삼엽의 광합성능력에 관한 연구 -제 1 보 저년생 고려인삼 광합성능력의 계절적 변이-)

  • This study was carried out to investigate the effect of seasonal changes on some of the morphological and physiological characteristics, including the photosynthetic abilities and dark respiration, of young ginseng plants due to planting location under shading. The results obtained are as follows: 1. Seedlings and 2-year old plants planted in the back rows appeared to have broader leaf area, and their leaf weight greatly increased in September. Chlorophyll content was significantly reduced in September rather than in June and the plants in the back rows had more chlorophyll content than those in the front rows. 2. There was no difference in the light compensation point between the front and back rows in June, while in September the light compensation point of 2-year old ginseng leaves was much lower for plants in the back rows compared with those in the front rows. A difference in the light saturation point was not noticeable between plants in the front and back rows in June and September. But the light saturation point of 2-year old ginseng leaves at $15^{\circ}C$ was high in June, while it was high at $20^{\circ}C$ high in September. 3. Maximum photosynthetic ability was attained at $15^{\circ}C$ in June and at $20^{\circ}C$ in September. During June no significant difference in photosynthetic ability was found between plants in the front and back rows, but in September the amount of photosynthesis was significantly increased at the leaves of seedlings as well as 2-year old plants planted in the back rows. 4. The optimum temperature for maximum photosynthesis in 2-year old plants ranged from $14.0^{\circ}C$ to $14.5^{\circ}C$ in June and from $19.5^{\circ}C$ to $20.5^{\circ}C$ in September. However, the optimum temperature for maximum photosynthesis in the seedlings was from $21.2^{\circ}C$ to $21.6^{\circ}C$ in September, but a significant difference in the optimum temperature for the maximum photosynthesis in seedlings and 2-year old plants was not noticeable between the front and back rows. 5. The respiration rate was rather high in seedlings compared with 2-year old plants. During September the respiration rate in seedlings was much lower in the back rows than in the front rows. The rate of increase in the respiration of 2-year old plants was higher at September than June. The increase in respiration rate due to temperature was more significant in seedlings than 2-year old plants. 6. In September, the level of $Q_{10}$ in 2-year old plants was much lower than that found in seedlings. During June, 2-year old plants showed lower $Q_{10}$ levels at a temperature difference between $15^{\circ}C$ and $25^{\circ}C$; but in September this occurred at a temperature difference between $20^{\circ}C$ and $30^{\circ}C$.

Temporal and Spatial Variation Analysis of Suspended Solids, Ionic Contents, and Habitat Quality in the Woopo Wetland Watershed (우포늪 수계에서 부유물, 이온농도 및 서식지 특성에 대한 시 ${\cdot}$ 공간적 변이 분석)

  • The main objective of present study was to evaluate how seasonal rainfall influenced natural habitat conditions of 10 metric habitat variables along with ionic conditions and suspended solids in the Woopo Wetland during August 2002-July 2003. Largest spatial variabilities in total suspended solids (TSS) occurred during the summer monsoon and the inorganic suspended solids (ISS), expressed as a inorganic proportion of total solids, showed linearly increasing trend from the upstream to downstream. This phenomenon was mainly attributed to counter flow of turbid water from the main Nakdong-River. During the flooding, ISS : TSS ratio showed large increases (92%) in the downstream than the upstream (43%). For this reason, transparency declined (mean=0.13 m, range=0.08-0.21 m) largely in the downstream reach and thus, chlorophyll-a concentration showed low values (range: $4.2-8.6\;{\mu}g\;L^{-1}$), indicating a direct influence on primary productivity or algal growth by inorganic turbidity. In the 2nd survey, ISS averaged 4.0 mg $L^{-1}$ (3.3-4.8 mg $L^{-1}$), thus the ISS decreased by 14 fold, compared to the ISS in the 1st survey during the flooding, while organic suspended solids (OSS) values were greater than those of ISS, indicating a dominance of organic solids. This condition was similar to solid contents in the 3rd survey, but showed a large difference compared to the 4th survey during the growing season. Habitat health assessments, based on 10 metric habitat variables, showed that QHEI values were greatest in the growing season (May) than any other seasons and largest spatial variations occurred in the 2nd survey. Overall, dataset suggest that seasonal episodic flooding during the monsoon may largely contribute nutrient cycling and sediment contents in the Woopo Wetland and Topyung Stream.

Limiting Nutrients for Phytoplankton Growth in the Seomjin River Estuary as Determined by Algal Bioassay Experiment (생물검정실험에 의한 섬진강 하구역 식물플랑크톤 성장의 제한영양염 평가)

  • limiting nutrients for phytoplankton were determined by dissored inorganic nitrogen/phosphorous (DIN/DIP) in situ and algal bioassay experiment in the Seomjin River estuary during a study period from March 1999 to October 2001. DIN/DIP ranged from 14.7 to 681.1 during the study period. DIN/DIP was over 16 at the upper and middle estuarine region where salinity was lower than 25 psu and chlorophyll a concentration was high, probably indicating P-limitation in this region while the ratio was less than 16 at the high saline (> 25 psu) region, reflecting the supply of DIP from Gwangyang Bay and thereby indicating N-limitation at the lower estuarine region. These results suggested that the spatial distribution of DIN/DIP in the study region was controlled by the high supply of phosphate from Gwangyang Bay, the low input from Seomjin River and the active uptake by phytoplankton within the estuarine system. The bioassay experiments using Skeletonema costatum, Thalaasiosira rotula and in situ phytoplankton assemblage displayed relatively higher growth of phytoplankton in the P-added culture media, indicating P-limitation. This result was well consistent with the spatial distribution of inorganic nutrients. S. costatum showed a rapid adaptation to the low salinity compared to other phytoplankton species. This phenomenon seemed to account for the strong (> $90\%$ in total cell number) S. costatum bloom in autumn in this estuary. Moreover, although phytoplankton growth rate was higher in the P-added culture media at the end of culture experiment of in situ phytoplankton, the fast growth in the trace metal-added media at the beginning of the experiment suggested a possibility of limitation by other micro-nutrients such as trace metal and vitamin etc.

Possibility of establishment of a tree nursery at Saemangeum Reclaimed Land and Classification of 36 Landscape Trees Based on Salt Tolerance (새만금 간척지에서 36종 조경수의 양묘 가능성 검증과 내염성 분류)

  • The objectives of this study were to investigate the possibility of establishing a tree nursery at Saemangeum reclaimed land and to classify landscape trees based on the salt tolerance. A tree nursery (2.0 ha) was made in Gunsan Okgu area in 2012 with underground drain lines on the reclaimed land established in 2010. Salt content of the nursery soil within the 60 cm depth in 2013 was 5.13 dS/m and 8.20 dS/m for the pre-desalinated and non-desalinated lands, respectively. Thirty-six woody plant species (22 tree species and 14 shrub species at ages of 1 to 4) with a total of 3,943 individuals were planted in early April, 2013 and their growth performance was monitored until September of the same year. The average survival rate of the transplanted plants was 71.4% in late September, suggesting the high possibility of establishing a tree nursery at the reclaimed land. Based on the survival rate and tree vigor (amount of healthy leaves and crown development), the following 17 species with some salt tolerance were classified into three groups: "salt tolerant group" (3 species, Tamarix chinensis, Cudrania tricuspidata, Ilex serrata), "recommended group" (5 species, Pinus thunbergii, Albizia julibrissin, Ligustrum obtusifolium, Rosa rugosa, Pleioblatus pygmaeus), "plantable group" (9 species, Zelkova serrata, Hibiscus syriacus, Elaeagnus umbellata, Sorbus alnifolia, Sophora japonica, Metasequoia glyptostroboides, Quercus acutissima, Ulmus parvifolia, Robinia pseudoacacia). Seven tree species that had been adapted to the reclaimed land for three to four years before being transplanted to new reclaimed land in Gunsan Okgu area showed average survival rate of 98%, suggesting that pre-conditioned trees would survive well in the reclaimed land.

Effect of COY (Cooking Oil and Yolk mixture) and ACF (Air-circulation Fan) on Control of Powdery Mildew and Production of Organic Lettuce (난황유와 공기순환팬의 상추 흰가루병 방제효과 및 생산에 미치는 영향)

  • Powdery mildew of lettuce that is a newly reported disease became a threat to organic cultivation of lettuce in Korea since the disease caused by Podosphaera fusca resulted in a half of yield loss in heavily infected fields. To improve micro-environmental conditions around lettuce, ACF (air-circulation fan) was installed on inside roof of plastic house at 6 m intervals. The ACF increased 57% of lettuce yield and reduced 71.4% of lettuce seedling death. COY (cooking oil and yolk mixture) consisted of cooking oil 0.3% and egg yolk 0.08% reduced lettuce seedling death from 89.3% to 92.9% under the greenhouse. Seven-day interval spray of COY resulted in high control values of powdery mildew of lettuce ranging from 89.6% to 96.3%, which was comparable to a fungicide, Azoxystrobin. Lettuce yield was increased about two times compared to a non-treated conventional cultivation. Qualities of lettuce such as hardness and chlorophyll content were also improved by COY and ACF combination. Effect of COY on control of the disease was improved when $CaCO_3$ or $SiO_2$ 1,000 ppm was supplemented. Results indicated that the COY made of cooking oil such as canola emulsified with yolk was highly effective on control of powdery mildew of lettuce and suitable for organic agriculture, especially when combined with ACF.

Photomixotrophic Growth of Solanum tuberosum L. in vitro with Addition and Omission of Organic Materials at Thee Initial Sucrose Levels in the Medium (세 수준의 자당이 첨가된 배지에서 유기물의 첨가 유무에 따른 Solanum tuberosum L.의 기내 광혼합영양생장)

  • The most commonly used inorganic nutrient compositions such as Murashige & Skoog medium have been optimized for heterotrophic growth. Therefore, they may not be optimal for photomixotrophic and photoautotrophic growth of plantlets. In photomixotrophic micropropagation, emdium sugar level is often lowered, while light and $CO_2$ levels in vessel are raised, and chlorophyllous explants are used to facilitate photosynthetic carbon acquisition. In a factorial experiment effect of addition (+) and omission(_) of organic materials (OM, 0.5 g ${\cdot}$ $m^{-3}$ each of thiamine, nicotinic acid and pyridoxine and 100 ${\cdot}$ $m^{-3}$ myo-inositiol) combined with three sucrose levels (0, 15, and 30 kg ${\cdot}$ $m^{-3}$) was tested on the growth of potato plantlets. Each of nodal cuttings with a leaf was cultured on 0.1${\times}$$10^{-4}m^{-3}$) MS agar ( 8 kg ${\cdot}$ $m^{-3}$) medium (pH 5.80 before autoclave) in glass test tubes (100 mm${\times}$25mm) capped with a sheet of transparent film with a 6 mm diameter gas permeable filter (5.1 air exchanges ${\cdot}$$h^{-1}$). Cultures were maintained in a room for 27 days at $23^{\circ}C$, 50% RH, 350-450${\mu}mol\;{\codt}\;mol^{-1}CO_2$, 16 h ${\cdot}$ $d^{-1}$ photoperiod at 13${\mu}mol\;{\codt}\;m\;{\codt}\;s^{-1}$ PPFD provided by white cool fluorescent lamps. Growth of potato plantlet in the +OM and -OM treatments were similar, while medium pH was 0.2 scale lower in the latter. Dry weight, % dry matter, and stem diameter enhanced, while shoot to root dry weight ratio, leaf area, chlorophyll concentration per gram dry weight, and medium pH decreased with increasing initial sucrose level. Interaction between OM and sucrose levels was observed in shoot length and medium pH. Results indicate that OM can be omitted from the medium without detrimental effect while addition of sucrose was beneficial for the photomixotrophic growth of potato plantlets under raised light and $CO_2$.

Effect of Application Rate of a Controlled Release Fertilizer on the Changes in Medium EC and Growth of Subirrigated Vinca and Salvia (저면관수 재배에서 완효성 비료의 양이 배지의 EC 및 일일초와 살비아의 생장에 미치는 영향)

  • Plug seedlings of vinca (Catharanthus roseus L. 'Pacifica Punch') and salvia (Salvia splendens F. Sellow ex Roem & Schult 'Maestro') were transplanted into square plastic pots (145 mL volume) filled with a soilless growing medium. To determine the effect of application rate on the growing medium EC and growth of plants, 0, 0.5, 1.0, 1.5, 2.0 and 4.0 g per pot of a controlled release fertilizer (14-14-14 Osmocote, 14N-6.2P-11.6K) were mixed with the growing medium. Plants were subirrigated daily with tap water. In both vinca and salvia, growing medium EC increased as application rate was elevated. Growing medium EC was relatively constant over a whole crop period when the application rate was less than 1.5 g per pot, while it decreased throughout the experiment at higher application rates such as 2.0 to 4.0g per pot in both species. The greatest leaf area, plant height, and shoot dry weight of vinca were obtained when plants were fertilized with 2.0 to 4.0 g per pot of the fertilizer, resulting in a growing medium EC of $1.0{\sim}1.7dS{\cdot}m^{-1}$ throughout the experiment. Leaf area, shoot dry weight, and chlorophyll content of salvia increased with elevated application rates. Leaf area, shoot dry weight, and chlorophyll content of salvia were the greatest when plants were fertilized with 4.0 g per pot, resulting in growing medium EC of $1.0{\sim}4.0dS{\cdot}m^{-1}$ throughout the experiment. Plant height of salvia was the greatest when plants were fertilized with 2.0 to 4.0g per pot. Concentrations of nitrogen (N), phosphorus (P), potassium (K), magnesium (Mg), and sulfur (S) in the shoots of vinca increased, while concentration of calcium (Ca) decreased with elevated application rates. Concentrations of boron (B) and manganese (Mn) in the shoots of vinca increased as the application rate decreased.

Trophic State Index (TSI) and Empirical Models, Based on Water Quality Parameters, in Korean Reservoirs (우리나라 대형 인공호에서 영양상태 평가 및 수질 변수를 이용한 경험적 모델 구축)

  • The purpose of this study was to evaluate trophic conditions of various Korean reservoirs using Trophic State Index (TSI) and predict the reservoir conditions by empirical models. The water quality dataset (2000, 2001) used here were obtained from the Ministry of Environment, Korea. The water quality, based on multi-parameters of dissolved oxygen (DO), biological oxygen demand (BOD), chemical oxygen demand (COD), total phosphorus (TP), total nitrogen (TN), suspended solid (SS), Secchi depth (SD), chlorophyll-${\alpha}$ (CHL), and conductivity largely varied depending on the sampling watersheds and seasons. In general, trophic conditions declined along the longitudinal axis of headwater-to-the dam and the largest seasonal variations occurred during the summer monsoon of July-August. Major inputs of TP occurred during the monsoon (r=0.656, p=0.002) and this pattern was similar to solid dynamics of SS (r=0.678, p<0.001). Trophic parameters including CHL, TP, SD, and TN were employed to evaluate how the water systems varies with season. Trophic State Index (TSI, Carlson, 1977), based on TSI (CHL), TSI (TP), and TSI (SD), ranged from mesotrophic to eutrophic. However, the trophic state, based on TSI (TN), indicated eutrophic-hypereutrophic conditions in the entire reservoirs, regardless of the seasons, indicating a N-rich system. Overall, nutrient data showed that phosphorus was a primary factor regulating the trophic state. The relationships between CHL (eutrophication index) vs. trophic parameters (TN, TP, and SD) were analysed to develop empirical models which can predict the trophic status. Regression analyses of log-transformed seasonal CHL against TP showed that the value of $R^2$ was 0.31 (p=0.017) in the premonsoon but was 0.69 (p<0.001) during the postmonsoon, indicating a greater algal response to the phosphorus during the postmonsoon. In contrast, SD had reverse relation with TP, CHL during all season. TN had weak relations with CHL during all seasons. Overall, data suggest that TP seems to be a good predictor for algal biomass, estimated by CHL, as shown in the empirical models.

Evaluation of Water Quality after Rehabilitation of Cheonggye Stream using AGP Test (조류성장잠재력 조사를 이용한 청계천 복원 이후 수질 평가)

  • Algal growth potential (AGP) test was performed to evaluate the water quality and changes in phytoplankton communities before and after a heavy rain event at six sampling sites in Cheonggye Stream (St. 1 and 2), Jungnang Stream (St. 3 and 4), and Lower part of Han River System (St. 5 and 6) after rehabilitation of Cheonggye Stream, October 2005. To test AGP on each sampling site, cyanobacterium Microcystis aeruginosa was applied as a standard alga. Total nitrogen (TN) showed high values at Jungnang Stream, while St. 4 recorded highest values in this study. However, TN values of Cheonggye Stream and Lower Part of Han River showed similar levels. Total phosphate (TP) also showed high values at Jungnang Stream, while St. 4 recorded highest. However, TP in Cheonggye Stream were extremely low levels. Although chlorophyll-${\alpha}$ (chi-${\alpha}$) contents before the rain event were similar through the sampling sites, chl-${\alpha}$ after the rain increased dramatically at Jungnang Stream and Lower part of Ban River. In particular, after the rain, TP was the highest at St. 4, where Cyclotella sp. dominated the phytoplankton community. When compared with control, AGP values before the rain were comparatively low in all sites, while those after the rain highly increased with the dose-dependently of field water added, due perhaps to the increased nutrients by rainfall. Similar results were observed in Cheonggye Stream. Therefore, for the aesthetic fostering for the citizens, although Cheonggye Stream was Presently being sustained by treated water supply, they have a potential of outbreak of phytoplankton by the increased nutrients supply when a heavy rain comes.

Effect of Sowing Dates on Turf Vegetation of Creeping Bentgrass (파종기에 따른 Creeping Bentgrass 잔디초지의 식생변화)

  • This study was conducted from March 16 to July 6 in 2004 at Jeju Island to investigate the influences of sowing dates(on March 16, March 26, April 5, April 15 and April 25) on creeping bentgrass vegetation. The result obtained were summarized as follows; Plant height was 22.7 cm at March 16 planting. It was longest but after that planting, plant height gradually shorted. Then it was shortest at April 25 planting(16.6 cm). Root length and Minolta SPAD-502 chlorophyll reading value were directly proportional plant height response. Leave and root weight were greatest at March 16 planting. It were 1,373 kg /10a and 2,374 kg /10a, respectively. These weight decreased gradually as planting was delayed from March 16 to April 25. Degree land cover and density of creeping bentgrass were $98.0\%$ and $99.3\%$, respectively, at March 16. After that planting they were decreased ($97.5\%$, $98.7\%$). But degree land cover and density of weed tended to increased gradually as the planting was delayed. The number of weed species were increased from March 16 to April 25. It showed increase that Poa annua, Stellaria media and Chenopodium album var. centrorubrum(at March 16 planting), Poa annua, Digitaria adscendens and Chenopodium album var. centrorubrum(at March 26 planting), Digitaria adscendens, Chenepodium album var. centrorubrum and Stellaria media(at April 5 planting), Digitaria adscendens, Stellaria media and Chenopodium album var. centrorubrum(at April 15 planting), Digitaria adscendens, Polygonum hydropiper, Chenopodium album var. centrorubrum(at April 25 planting). Based on the these findings, optimum sowing date for growth of creeping bentgrass seems to be about early seeding in atmospheric phenomena and volcanic ash soils of Jeju island.
