Antioxidant activity of hot-water extracts and floral waters from natural plant pigments (천연색소 함유 식물추출물과 플로럴 워터의 항산화 활성)

  • The antioxidant potentials of ten kinds of medicinal plants used as natural dyeing agents were tested. Among the plant extracts and floral waters that were examined in this study, the antioxidant activities of the DPPH and ABTS radical scavengers increased with increasing amounts of the extracts. The hot-water extracts from Aphis chinensis, Caesalpinia sappan L., Rumex crispus L., and Areca catechu effectively inhibited the DPPH and ABTS radicals at concentration below 0.1 mg/mL. The floral water obtained through steam distillation from Areca catechu, Rubia akane Nakai, Coptis chinensis, and Phellodendron amurense showed relatively valid antioxidant activities. In particular, the natural colorants extracted from Rumex crispus L., Areca catechu, and Aphis chinensis effectively suppressed the photogenerated singlet oxygen induced by a photosensitizer in in-vitro assay systems. The concentrations ($IC_{50}$) of the hot-water Aphis chinensis extract required to exert a 50% reduction effect on DPPH, ABTS, and singlet oxygen were found to be 8.5, 8.0, and $210{\mu}g/mL$, respectively. The study results suggest that extracts derived from naturally occurring colorants as well as floral waters could be used as natural antioxidants in the food and health-care industries.

Antioxidant activities and inhibitory effects on oxidative DNA damage of leaf from Zelkova serrata with ethyl acetate fractions and hot water extracts (느티나무 잎 에틸아세테이트 분획물 및 열수 추출물의 항산화 및 산화적 DNA 손상 억제 활성)

  • Reactive oxygen species (ROS) has been played a critical role in damage of DNA. Recently, many effort is focusing to develop the natural antioxidants for controlling ROS. Zelkova serrata, Ulmaceae, is close as plants which are planted in front of Korea villages. Although Zelkova serrata is familiar with Koreans, those of antioxidant activities and protective effects on oxidative DNA damage haven't studied. We demonstrated antioxidant activities and inhibitory effects on oxidative DNA damage of Leaf from Zelkova serrata with ethyl acetate fractions (EA) and hot water extracts (HW). Between the extracts, EA showed higher activities in 1,1-diphenyl-2-picryl-hydrazyl, 2,2'-azino-bis[3-ethylbenzthiazoline-6-sulphonic acid radical scavenging, $Fe^{2+}$ chelating and reducing power than HW. Also, those of total phenolic content are 56.63 and 51.61 mg/g respectively. In addition, ${\phi}X$-174 RF I plasmid DNA cleavage assay for inhibitory effect by oxidative DNA damage was both EA and HW has significant protective effect on oxidative DNA damage. The results suggested that leaf from Zelkova serrata with ethyl acetate fractions and hot water extracts have surpassing potential as natural resources with antioxidant and inhibitory effect on oxidative DNA damage.

  • Analysis of general ingredients and amino acid content in dried silkworm feces which were extracted with hot water, EtOH, MeOH for developing new income source of rearing farm, seeking high additive value material and industrialization of silkworm feces. In the analysis of general ingredient content by extracting solution, raw protein and ash contents were increased with MeOH, EtOH, hot water. And raw fat showed higher contents by the order of EtOH > MeOH > hot water. The inorganic matter content showed a little difference among collection times of silkworm feces. Hot water was most effective extracting solution for total amino acid, and no significant difference between EtOH and MeOH. According to the time, there were no significant difference in amino acid contents. When extracted with hot water, Glu content was highest content and order of others is Asp > Gly > Ala > Leu > Thr > Ser among 18 amino acids.

The Antioxidant Ability and Nitrite Scavenging Ability of Poria cocos (복령(Poria cocos) 균사체의 항산화성 및 아질산염 소거작용)

  • This study was conducted to investigate the efficacy of antioxidant ability and nitrite scavenging ability of Poria cocos Poria cocos mycelium and Poria cocos fermented with rice were extracted with water and ethanol for measurement of degree of fat acidification against fish oil. Lower TBARS values of Poria cocos mycelium and Poria cocos fermented with rice were shown compared with control. Iron chelating ability of Poria cocos mycelium against Fe$^{2+}$, Cu$^{2+}$ was higher than Poria cocos fermented with rice. Electron donating abilitv of water extract and ethanol extract of Poria cocos mycelium were 76.8% and 79.9%, respectively. Higher nitrite scavenging ability was shown at ethanol extract than water extract of Poria cocos mycelium.ium.

Antioxidant Activity of Extracts from the Lespedeza bicolor (싸리(Lespedeza bicolor) 추출물의 항산화성에 관한 연구)

  • This study was designed to investigate the antioxidant activity of Lespedeza bicolor extract, by measuring electron-donating ability (EDA) using 1,1-diphenyl-2-piciyl hydroxyl (DPPH), superoxide dismutase-like activity (SODA) by pyrogallol, and nitrite-scavenging ability. The EDA of water and ethanol extracts in nitrite-scavenging ability of ethand extract showed the highest values of $82.02\%$ and $75.56\%$, respectively. The SODA increased with the concentations of extract, which was $17.21\~44.08\%$ in ethanol extract $19.89\~29.86\%$ in pressure-water extract and $16.53\~20.0\%$ in water extract respectively. The nitrite-scavenging ability of ethanol extract (pH 1.2, 1000ppm) was $99.53\%$ that was higher than both $85.84\%$ in water extract and $74.72\%$ in pressure-water extract at pH 3.0 was also the highest as $98.20\%$.

Effects of Fermented Water Extracts from Ligularia fischeri on Hepatotoxicity in Ethanol-Induced Rats (에탄올 투여 랫드에서 곰취 열수 추출 발효물이 간 독성에 미치는 영향)

  • This study was conducted to determine the effects of fermented water extracts from Ligularia fischeri (LAF) on reduction of hepatotoxicity induced by ethanol in rats. Ethanol-treated Sprague-Dawley rats were divided into the following eight groups: ethanol-treated group (control), ethanol and ursodeoxycholic acid-treated group (positive control), ethanol and non-fermented water extracts from Ligularia fischeri (LA)-treated groups [100, 200, and 400 mg/kg BW (body weight)], ethanol and LAF-treated groups (100, 200, and 400 mg/kg BW). ${\gamma}$-Glutamyl transferase activities of the ethanol+LA-treated (100, 200, and 400 mg/kg BW) groups and ethanol+LAF-treated (400 mg/kg BW) group decreased significantly compared to those in the control group (P<0.05). Aspartate aminotransferase activities of the ethanol+LAF-treated (100, 200, and 400 mg/kg BW) groups and ethanol+LA-treated (200 and 400 mg/kg BW) groups decreased significantly compared to those in the control group (P<0.05). Alanine aminotransferase and lactate dehydrogenase activities of all groups significantly decreased compared to those in the control group (P<0.05). The total cholesterol, low density lipoprotein-cholesterol, and triglyceride levels of all groups tended to decrease compared to those in the control group, but the differences were not significant. Superoxide dismutase activity of liver tissues was enhanced in the ethanol+LAF-treated (400 mg/kg BW) group (P<0.05). The contents of malondialdehyde in liver tissues decreased in the ethanol+LAF-treated groups (P<0.05). All treated groups showed well preserved lobular architectures with no evidence of steatosis or liver damage compared to the control group. As the results of this study, LAF may improve the plasma lipid profile and alleviate hepatic damage by ethanol.

Comparison of physiological activity of medicinal mushrooms produced in Korea and China using different extraction solvents (한국 및 중국산 약용버섯류의 추출용매에 따른 생리활성 성분 비교)

  • In the present study, physiological activities of hot water extracts and 70% ethanol extracts of three types of medicinal mushrooms (Hericium erinaceus, Cordyceps militaris, and Ganoderma lingzhi) produced in Korea and China were investigated. Both the hot-water and 70% ethanol extracts of H. erinaceus and C. militaris from Korea displayed the highest 2,2-diphenyl-1-picrylhydrazy radical scavenging activities. Nitrite scavenging activities of hotwater extracts of G. lingzhi from Korea and China (41% and 39%, respectively) were higher than the activities of 70% ethanol extracts. Total polyphenol contents of hot water extracts of H. erinaceus and C. militaris from Korea were higher than those of 70% ethanol extracts. The ethanol extracts of G. lingzhi from Korea displayed the highest total polyphenol content. C. militaris from Korea displayed the highest ${\beta}-glucan$ level (45.11%). ${\beta}-glucan$ content of H. erinaceus from China (30.87%) was higher than H. erinaceus from Korea (16.94%). The findings indicate that healthy ingredients can be maximally extracted using the optimal solvents for each mushroom. These results will be useful in understanding the difference in physiological activities between the solvents used for the extraction of medicinal mushrooms from Korea and China.

A Study on Biological Activities of Opuntia humifusa Cladode Extracts (손바닥 선인장 (Opuntia humifusa) 줄기 추출물의 생리활성)

  • Biological activities of the hot water and ethanol extracts from Opuntia humifusa cladodes were investigated. 1,1-diphenyl-2-picryl hadrazyl (DPPH) electron donating ability of hot water and ethanol extracts was 79.07 and 82.54%, respectively. Hot water extract generally showed better cytotoxic activity than ethanol extract against each cell line. HeLa and AGS cell lines treated with hot water extract had more than 50% cytotoxic activities. Based on the antimicrobial activities against four microbial strains, both extracts inhibited growth of Staphylococcus aureus KCCM 11593, whereas affected cell growth of three other microorganisms, Escherichia coli (KCCM 11234), Pseudomonas aeruginosa (ATCC 27853), and Salmonella typhimurium (ATCC 11862), in proportion to the concentration of extracts. The inflammatory activities against hot water extract (34.31%) showed higher than that of ethanol extract (25.59%). The effect of extracts on 3T3-L1 preadipocytes differentiation showed that differentiation of treated group with 80 and 100 ${\mu}g/mL$ of hot and ethanol extracts were increased more than treated group with isobutyl methyl xanthine (IBMX) + dexamethasone. These results indicate that the O. humifusa cladodes extracts can be used as a functional material due to their effective biological activities.

Hepatoprotective Activity of Dandelion (Taraxacum officinale) Water Extract against D-Galactosamine-Induced Hepatitis in Rats (D-Galactosamine으로 유발된 간손상에 대한 민들레 열수추출물의 예방효과)

  • This study aimed to investigate the protective effect of dandelion water extract (DWE) on liver injury induced by D-galactosamine (GalN) in Sprague-Dawley rats. Fifty rats were divided into 5 groups; normal control (C), DWE-control (DWE-C: saline injection after feeding 3% DWE diet), GalN-control (GalN-C: GalN injection after normal diet), DWE I (GalN injection after feeding 1.5% DWE diet), and DWE II (GalN injection after feeding 3% DWE diet). After 2 weeks, the acute hepatitis was induced by GalN (650 mg/kg, i.p.) and 24 hrs later, all rats were sacrificed. The DWE supplement ameliorated the serum alanine and aspartate aminotransferase (AST, ALT) as well as alkaline phosphatase (ALP) and tumor necrosis $factor-{\alpha}\;(TNF-{\alpha})$. Hepatic antioxidative enzyme activities, such as catalase, GSH peroxidase, GSH reductase, and Mn-superoxide dismutase (SOD) were slightly or significantly elevated by the treatment of DWE. Moreover, the histological examination corresponded with these biochemical observations. According to these findings, dandelion could be used as a potential therapeutic material for treating chemically induced acute hepatitis.

Antioxidant and Anti-inflammatory Effect of Extracts from Flammulina velutipes (Curtis) Singer (팽이버섯 추출물의 항산화 및 항염증 활성)

  • The potential antioxidant and anti-inflammatory effect of water and ethanol extracts from Flammulina velutipes (Curtis) Singer (FVS) on hydrogen peroxide ($H_2O_2$) and LPS-induced oxidative damage in PC-12 and RAW264.7 cells were investigated. The DPPH radical scavenging activities of the water extract from FVS was the highest, and the 50% inhibitory concentration value was 0.388 mg/mL. Also, the antioxidant activities of water and ethanol extracts were determined by ferric reducing antioxidant power, 2,2'-azino-bis-(3-ethybenzothiazoline-6-sulphonic acid) radical scavenging activity. In addition, water extract from FVS showed a strongly inhibitory effect on lipid peroxidation by measuring ferric thiocyanate values. The water extract decreased cell apoptosis in PC-12 cells against $H_2O_2$-induced oxidative damage. In addition, FVS extracts exhibited the strongest nitric oxide (NO) inhibition activity. It was also found that FVS extract inhibited LPS-induced iNOS and COX-2 expression in RAW264.7 cells. The findings of the present study suggest that extracts of FVS exhibit anti-oxidative and anti-inflammatory activity against oxidative stress and/or stimulated cells.
