  • This study was conducted to evaluate the effects of saponin contained plant extracts on in vitro rumen fermentation characteristics and methane production. Ruminal fluid was collected from rumen cannulated Hanwoo steers fed rice straw and concentrate (5:5). Collected rumen fluids, corn starch and buffer including saponin contained plant extracts (ginseng, Ogapi, soapwort, tea plant and yucca; 0.5%/15 ml) were incubated at $39^{\circ}C$ for 24 h. All incubations were repeated five times. Rumen pH in all treatments was lower (p<0.05) compared with that of the control (no addition) during incubation time. The concentration of total VFA in all treatments was higher (p<0.05) than that of the control after 12h incubation. Compared with the control, the concentration of acetate and propionate in all treatments was lower and higher after 6h incubation, respectively. The concentration of $NH_3$-N in all treatments was lower (p<0.05) than that of the control except for Ogapi or yucca extracts supplementation. The number of protozoa in all treatments was significantly (p<0.05) lower than that of the control except for soapwort extract supplementation. The total gas production and methane production in all treatments was higher (p<0.05) and lower (p<0.05) compared with the control, except for ogapi or soapwort extracts supplementation after 12h incubation, respectively. Therefore, reduction in methane production by saponins may could be results from decreased protozoal population without any negative in vitro fermentation.

  • This study was conducted to elucidate the characteristics of equine meat traits and to evaluate the utilization of dried-citrus pulp abundantly produced in Jeju Island as feed ingredient in the diet of finished meat horses. Treatment groups consisted of wheat bran (A) and the dried-pellet mixture of citrus pulp (20%) and wheat bran (80%) (B). Feeding of agricultural by-products were 2 times in a day at the level of 1.5% of body weights. Water and dried-grass forage was not restricted. The average daily gain was similar in both groups. Feed conversion of treatment B showed lower level than that of A. Carcass rate of B was relatively lower than that of A, but meat yield grade of B was higher than that of A. For meat color traits, $L^*$ of B was lighter than that of A, and $b^*$ of group B was lower than that of group A. From the analysis of physical characteristics of equine meats, shearing force had decreased in the treatment B but the levels of drip loss and water holding cavity had increased in the treatment B compared to A. The level of crude fat is lower in the treatment B than the treatment A. For the mineral contents, the treatment B showed higher levels of Ca, K, Mg, Cu and Mn than that of A. For the fatty acid contents, the treatment B showed increased levels of stearic acid, oleic acid, eicosenoic acid, and arachidonic acid compared to A. Consequently, these results suggested that dried-citrus pulp might be utilized as replacement material for equine forage. • BACKGROUND: A lasting release of low levels of persistence chemicals including pesticides and pharmaceuticals into river has a bad influence on aquatic ecosystems and humans. The present study monitored pesticide residues in the Yeongsan and Seomjin river basins and their tributaries as a fundamental study for water quality standard of pesticides.METHODS AND RESULTS: Nine pesticides(aldicarb, carbaryl, carbofuran, chlorpyrifos, 2,4-D, MCPA, methomyl, metolachlor, and molinate) were determined from water samples using SPE-Oasis HLB(pH 2) and LC/MS/MS. Validation of the method was conducted through matrix-matched internal calibration curve, method detection limit(MDL), limit of quantification(LOQ), accuracy, precision, and recovery. MDLs of all pesticides satisfied the GV/10 values. Linearity(r2) was 0.9965- 0.9999, and a percentage of accuracy, precision, and recovery was 89.4-113.6%, 3.1-14.0%, and 90.8-106.2%, respectively. All pesticides exclusive of aldicarb were determined in the river samples, and there was a connection between the positive monitoring results and agricultural use of the pesticides.CONCLUSION: Monitoring outcomes of the present study implied that pesticides were a possible non-point pollutant source in the Yeongsan and Seomjin river basins and tributaries. Therefore, it is required to produce and accumulate more monitoring results on pesticides in river waters to set water quality standards, finally to preserve aquatic ecosystems.

  • This study was carried out to investigate the effect of selenium treatment on the growth of vegetables sprout. Four vegetables, such as cabbage, lettuce, pak-choi and leaf mustard were examined under various selenium treatments (0, 1, 5, 10, 25, 50, 100 mg $L^{-1}$). Seed germinations in cabbage, pak-choi and leaf mustard were significantly inhibited at high concentration of selenium treatment. However, seed germination in lettuce was not much inhibited. Growth characteristics, such as soot length, root length, fresh weight and chlorophyll contents, were not much decreased at 1 mg $L^{-1}$ of selenium and then significantly inhibited with the increase of selenium concentration at above 5 mg $L^{-1}$ in all four vegetables. The selenium content increased linearly with the increase of selenium concentration. At the range of 1 to 25 mg $L^{-1}$ of selenium treatment, selenium contents in vegetables were 0.11 to 1.15 of cabbage, 0.16 to 0.61 of lettuce, 0.13 to 1.31 of pak-choi and 0.14 to 1.13 mg $g^{-1}$dw of leaf mustard, respectively. These results showed that treatment of selenium with the range of 1 to 5 mg $L^{-1}$ could be used to produce the selenium enriched vegetable sprouts.

  • This study was performed to investigate the effects of harvest stage of sorghum ${\times}$ sorghum hybrid (SSH) on nutritive values and quality of round baled SSH silage manufactured with SSH grown in paddy land of Department of Animal Resources Development, National Institute of Animal Science, RDA. SSH "SS405" was harvested at two different growth stages (heading and ripen stage) and ensiled at each harvest stages. Crude protein content of round baled SSH silage increased with delayed harvest maturity, but contents of ADF (acid detergent fiber), NDF (neutral detergent fiber), TDN (total digestible nutrient) and in vitro dry matter digestibility (IVDMD) was not changed. The pH in round baled SSH silage ranged from 3.8 to 4.4 at two different harvest stages, and pH in heading stage was higher than that of ripen stage. The content of lactic acid of round baled SSH silage increased with delayed harvest maturity (P<0.05), but the content of acetic acid decreased (P<0.05). The contents of lactic acid and acetic acid in ripen stage were not influenced by manufacture method of silage. The content of acetic acid in round baled SSH silage of heading stage increased as compared to that of trench SSH silage (P<0.05). Therefore, this study suggest that round baled SSH silage manufactured in both heading and ripen stage can improve the silage fermentation.

  • We studied the accumulation and partitioning of dry weight (DW) and nitrogen (N) in different parts of field-grown 'Fuyu' persimmon to elucidate that the foliar applications of supplemental N in June or September compared favorably with the traditional soil application in securing leaf area and fruit production. We also estimated the proportion of N permanently removed from the tree at the end of a growing season. Urea was applied either to leaves in June and/or September or to the soil in June and September for three consecutive years, and the trees were excavated in November for analyses. Total DW ranged from 4.2-4.8, 8.7-9.2, and 17.1-21.5 kg in a 4-, 5-, and 6-year-old tree, respectively, without statistical difference among the four treatments. Of the total DW, 3.3-10.2% was in shoots, 5.7-10.5% in leaves, 8.3-31.4% in aerial woods, 13.0-27.0% in root, and 28.0-59.3% in fruits. As the trees became more productive, DW proportion of fruits significantly affected that of the root: in 6-year-old trees, root DW accounted for only 10.6-15.8% of the tree total when fruit DW accounted for 50-60%. N contents ranged from 24.6-28.3, 48.3-53.5, and 98.3-122.6 g in a 4-, 5-, and 6-year-old trees, respectively, without statistical difference among the treatments. Of the total N, 6.2-11.5% was in shoots, 16.7-24.3% in leaves, 17.6-23.5% in aerial woods, 17.2-37.5% in roots, and 16.9-34.4% in fruits. As in DW, the increase in the proportion of N in fruits decreased in the root most significantly. Application methods for supplemental N did not affect the proportion of DW and N removed from the tree through abscising leaves and harvested fruits. Percentage of DW removal was 41 in 4- and 5-year-old trees, but it was 61 in more productive 6-year-old trees; that of N was 39, 43, and 49%, respectively. No significant changes in the contents of DW and N in field-grown trees, as well as their percentages removed from the tree at the end of the season, demonstrated that foliar application of supplemental N was as good as soil applications with much less N.

  • This study was conducted to compare the cold tolerance of seven ground cover species, Orostachys japonica, Sedum oryzifolium, S. kamtschaticum 'SG1', S. reflexum, S. rupestre 'Blue Spruce', S. spurium 'Green Mental', and S. takesimense, which have been used for green roof and wall systems in Korea. Plants were grown in 10-cm pots and 1 g of tissues at stem-end and crown of each species were kept under either light or dark condition, respectively. For cold tolerance tests, plants were initially left at $4^{\circ}C$ and linearly cooled to 0, -4, -8, -12, -16, and $-20^{\circ}C$ at $-2^{\circ}C{\cdot}h^{-1}$ rate. Low temperature injury and regrowth rates were visually evaluated and assessed by image analysis, respectively. The lethal temperature ($LT_{50}$) of plant species was determined using electrolyte leakage measurements. S. reflexum was the most cold tolerant, showing the most survival at $-16^{\circ}C$, whereas S. oryzifolium and S. takesimense showed low temperature injury at $-8^{\circ}C$. Similar results were found with electrolyte leakage measurements at the stem end. For each species, the crown (Mean $LT_{50}:\;-12.15^{\circ}C$) was more cold tolerant than the stem end (Mean $LT_{50}:\;-10.47^{\circ}C$). In conclusion, S. reflexum and S. rupestre 'Blue Spruce' are recommended for planting in the central region of Korea during late fall and early winter, as they were more cold tolerant and showed more vigorous regrowth than the other tested plant species.
