Purufication and Characterization of Extracellular Collagenase from Vibrio mimicus (Vibrio mimicus 가 생산하는 collagenase의 정제 및 특성)

  • A collagenase was isolated from the culture filtrate of Vibrio mimicus (ATCC 33658). The enzyme was purified to homogeneity by ammonium sulfate precipitation and DEAE-Sephadex A-50 chromatography, which an activity recovery of 22%. The molecular weight of the purified enzyme was estimated to be 42 kDa by SDS-polyacrylamide gel electrophoresis and gel filtration, indication a monomer structure. The optimum pH and temperature od the enzyme for insoluble collagen (Type I) were around 7.75 and 28$\circ$C, respectively. Some chelating agents and serine protease inhibitor inactivated the enzyme, but L-cysteine and histidine did not affect the activity. The amino acid composition indicated that the collagenase contained high amounts of amino acid residues of glycine and alanine. The K$_{m}$ and R$_{cat}$/K$_{m}$ values for the collagenase, using insoluble collagen (type I) as substrate, were 2.86 mg/ml and 972.28 U/mg-protein, respectively.

  • (A hybrid-type wireless infrared data communication module was fabricated by using the light emitting andabsorption diodes with the one-chip of integrated digital circuits. The light emitting diode with the peak spectrum of 850 nm was made from compound semiconductor material of AIGaAs and shows high speed signal transmission with the delay time of 60 nsec for the light direction angle of 30".   • 본 연구는 목재로서 활용가치가 적은 수피(bark)를 활용하여 수질오염물질을 제거할 수 있는 여과 시스템에 대한 기초연구로서 소나무의 일종인 loblolly pine(Pinus taeda L.) 수피의 인 ($PO_4-P$) 흡착특성을 연구하였다. 실험에 사용된 수피는 입상형태로서 polyallylamine hydrochloride로 전처리하여 회분식 등온 흡착실험을 수행하였다. 회분식 등온흡착실험은 수용액 pH 3~pH 8범위에서, 인의 농도별(10, 20, 30, 40 mg/L)로 수행하였다. pH 5에서 가장 높은 인 제거 효율을 나타냈으며, 수용액의 pH는 실험 후 pH 3으로 감소하였다. 이러한 감소는 수피에 의한 phosphate의 흡착이 Lewis acid-base 반응으로서 이 과정에서 $H^+$의 방출로 인하여 나타난 현상인 것으로 여겨지며, 주된 반응 메카니즘은 더 연구할 필요성이 있다. 인 흡착은 초기에 빠른 속도로 진행되었으며 대략 200분 이후에 평형에 도달하였고, 시간이 지날수록 흡착양이 증가하는 경향을 보였다. 인의 흡착특성 결과는 Langmuir 등온흡착식과 Freundlich 등온흡착식으로 설명될 수 있으며, 등온흡착식 적용결과, 흡착제에 대한 최대 흡착능은 7.14 mg/g 이며 다른 흡착제와 비교하여 더 높았다. 실험결과와 모델에 의한 흡착능을 비교하고자 pseudo second-order model을 적용하여 흡착 동역학 상수를 구하였다. 또한 EDXA분석으로 회분식 흡착실험 후 수피와 인이 결합되어있는 것을 확인할 수 있었다.

A Study on the Minimum Fluidization Velocity and Expansion of Various Media (여과재(濾過材)의 종류(種類)에 따른 최소유동상(最小流動狀) 속도(速度) 및 팽창(膨脹)에 관한 연구(硏究))

  • Backwashing is one of the most important processes in water treatment. Several models have been utilized to predict minimum fluidization velocity and expansion of media. However, it is not unusual that the actual bahavior of media dose not agree well with the prediction. This study has investigated the applicability of models in predicting the minimum fluidization velocity of sand media. However even the better model has predicted minimum fluidization velocity 1.1 to 1.8 times higher than actual fluidization velocity. The expansion rate of sand media was inspected. It is found that the actual expansion rate was greater than the predicted. In this study condition, use of $d_{10}$ instead of $d_{60}$ was better to predict the expansion of media. On the contrary to the sand media, the actual expansion of anthracite media was less than that predicted. Sometimes it is reported that the dual media has been overflown during backwashing and mixed severely at the interface. It is because the grain size distribution of anthracite has not been selected properly. The numerical values for media expansion found in this study could be referred as the useful data in operating and/or designing filter media.

Analysis of Melatonin Content from Domestic Edible Plants (국내산 식용식물체의 멜라토닌 함량 분석)

  • Melatonin, which is a hormone secreted from pineal gland of brain and known to prevent oxidative damages of various tissues, was analyzed in 26 domestic edible plants. For the preparation of melatonin fraction, 50% ethanol extract prepared from lyophilized plant powder was filtered and applied on TLC plate. Melatonin position on TLC developed with acetone was identified by fluorescence light and extracted with methanol. This methanolic fraction was injected into HPLC comprising ODS-A column, fluorescence detector, and mobile phase consisting of a mixture (30 : 70, v/v) of 70% ammonium acetate and methanol at a flow rate of 1.0 mL/min. Melatonin was identified at the retention time of 17 min. Results revealed that celery, leek, broccoli, and cauliflower had higher melatonin contents than others.

A Filtering Method of Trajectory Query for Efficient Process of Combined Query (복합질의의 효율적 수행을 위한 궤적질의 필터링 기법)

  • The combined query which consists of the region and trajectory query finds trajectories of moving objects which locate in a certain region. The trajectory query is very informant factor to determine query performance because it processes a point query continuously to find predecessors. This results in bad performance due to revisiting nodes in an index. This paper suggests an efficient method for the combined query based on the 3-dimensional R-tree which has good performance of the region query. The basic idea is that we define the least common search line which enables to search single path and a filtering method based on prediction without revisiting nodes.

  • In this study, hexavalent chromium (Cr(VI)) plating wastewater in strong acidic condition was treated by reduction and alkalization. Ferrous sulfate ($FeSO_4$), known to reduce Cr(VI) to Cr(III) rapidly at acidic pH, was used as a reductant of Cr(VI). The optimum reduction condition of Cr(VI) was observed at iron to chromium dose ratio of 3:1 by mole concentration. The precipitation of Cr(III) as $Cr(OH)_3$, was achieved by the pH adjustment in the limestone aeration bed. The precipitates were removed less than the upper limit of chromium for effluent at pH over 5.0. The continuous removal of Cr(VI) was performed using the process consisting of reduction vessel, limestone aeration bed, and sedimentation tank coupled with metal screen membrane. As pH was maintained around 5.0 in the limestone aeration bed, insoluble chromic hydroxide flocs was formed continuously. Most chromic hydroxide flocs were filtered by the metal screen membrane with 1450 mesh size, and the treated water to meet the upper limits of chromium for effluent (Cr Conc. 0.25~0.90 mg/l) was obtained in 30 minutes. Periodic backwashing decreased the fouling on the membrane rapidly.

  • In this study, the biochemical characterization of strong proteolytic bacteria and their extracellular pretense isolated from fermented fish paste were experimented for the purpose of industrial large scale-production by accelerated fermentation. The results obtained were as follows: Among 4 strains isolated from fermented fish paste, B. subtilis p-4 and B. licheniformis p-5, which grow well at $40^{\circ}C$, pH 7.0 and $1\%$ of salt contents, were the best proteolytic bacteria and were shown $0.48hr^{-1}$, $0.49hr^{-1}$ of specific growth rate in TPY medium, respectively. Maximum enzyme activity of B. subtilis p-4 was 335n mole-Tyr/min.ml after 30 hrs and that of B. licheniformis p-5 was 300n mole-Tyr/min.ml after 28 hrs of shaking culture. Purified pretense produced by B. subtilis p-4 and B. licheniformis p-5 showed maximum activity at $50^{\circ}C$, pH 7.0 and molecular weight were estimated to be 18,000, 30,000 by sephadex G-100 gel filtration, respectively. These were supposed to be a kind of metal chelator sensitive neutral pretense from the result of high sensitivity against EDTA, o-phe-nanthroline and metal ions such as $Cu^{2+},\;Ni^{2+},\;Zn{2+}$.

Instant Tea Preparation from Cassia tora Seeds (결명자로부터 인스턴트차 제조)

  • In order to develop a type of instant tea cassia tora seed, changes of constiuent amounts before and after roasting $(3{\pm}0.5min\;at\;170{\pm}5^{\circ}C)$ effective extracting solvent and it's optimum concentration, dehydration method and sensory scores were investigated. Moisture, lipid, reducing sugar, emodin and rhein were decreased by roasting, whereas protein, and total sugar of cassia tora increased. Color intensity and extracting efficacy were also increased. Of three kinds of solvent (water, methanol and ethanol), ethanol was highest in redness and yellowness of exeracts, and filtrate yield. Effective extracting concentration of ethanol for cassia tora was 50%(V/V). Freeze dry product(FDP) and spary dry product(SDP) showed coffee and aloe-like smell, coffee like brown color, rapid soluble in cold and hot water, and fast caking in air during storage. Free sugar contents were high in order of raffinose>fructose >glucose>maltose>sucrose Metallic contents were high in order of sodium>calcium>potassium>magnesium>iron. In sensory score for color, taste and aroma, FDP and SDP were both above 8.0 point, however, of them FDP more or less higher than SDP In addion, score in developing value as an instant tea was above 8.0 point (valuable).

Use of Hydrogen Peroxide with Ozone to Simultaneously Reduce MIB and Quench Ozone Residual in Existing Water Treatment Plants Sourcing Water from the Han River (한강을 원수로 하는 오존/과산화수소 고도정수처리공정에서의 MIB제거 및 잔류오존 농도에 관한 연구)

  • This paper provides a detailed account of pilot testing conducted at South Lake Tahoe (California), the Ddukdo (Seoul) water treatment plant (WTP) and the Bokjung (Seongnam) WTP between February, 2010, and February, 2012. The objectives were first, to characterize the reactions of ozone with hydrogen peroxide (Peroxone) for Han River water following sand filtration, second to determine empirical ozone and hydrogen peroxide doses to remove a taste-and-odor surrogate 2-methylisoborneol (MIB) using an advanced oxidation process (AOP) configuration and third, to determine the optimum dosing configuration to reduce residual ozone to a safe level at the exit of the process. The testing was performed in a real-time plant environment at both low- and high seasonal water temperatures. Experimental results including ozone decomposition rates were dependent on temperature and pH, consistent with data reported by other researchers. MIB in post-sand-filtration water was spiked to 40-50 ng/L, and in all cases, it was reduced to below the specified target level (7 ng/liter) and typically non-detect (ND). It was demonstrated that Peroxone could achieve both MIB removal and low effluent ozone residual at ozone+hydrogen peroxide doses less than those for ozone alone. An empirical predictive model, suitable for use by design engineers and operating personnel and for incorporation in plant control systems was developed. Due to a significant reduction in the ozone reaction/decomposition at low winter temperatures, results demonstrate the hydrogen peroxide can be "pre-conditioned" in order to increase initial reaction rates and achieve lower ozone residuals. Results also indicate the method, location and composition of hydrogen peroxide injection is critical to successful implementation of Peroxone without using excessive chemicals or degrading performance.
