돼지 췌장 유래 엘라스타제의 변이원성 시험

  • 돼지 췌장 유래 엘라스타제의 돌연변이원성을 알아보기 위하여 포유동물 세포를 이용한 돌연변이 시험과 마우스를 이용한 소핵시험을 실시하였다. 엘라스타아제에 의하여 CHO-K1-BH4 cell이 6-thioguanine에 대해 내성을 가지게 되는 변이의 빈도를 조사하였다. 시험결과 엘라스타아제의 농도 증가 (0-30mg/ml)에 따른 변이빈도의 증가가 관찰되지 않았으며, 변이빈도도 음성대조군 변이빈도 치의 2배 이상을 보이지 않았다. 마우스를 이용한 소핵시험에서는 엘라스타아제를 1250, 2500 그리고 5000mg/kg의 용량으로 마우스에 1회 경구 투여 하였으며 양성대조군과 음성대조군에는 각각 mitomycin C 2mg/kg, 10skim milk 20ml/kg 을 각각 복강내와 경구로 투여하였다. 시험결과 엘라스타제 투여군의 소핵 출현빈도는 음성대조군과 비교하여 증가하지 않았다. 위의 결과로 보아 엘라스탄제는 이 시험계들에서 돌연변이원성이 없는 것으로 사려된다.

Application as a Cosmeceutical Ingredient of Euryale ferox Seed Extract (가시연꽃 종자 추출물의 화장품 원료로서의 특성)

  • In our search for the natural cosmetic ingredients, we found that Euryale ferox seed extract exhibited the strong antioxidative activity. Five active compounds were isolated from the ethyl acetate extract through various chromatographic methods and their structures were determined by NMR and MS spectral analysis. These compounds were identified as fucosterol (1), 3-(4-hydroxy-3-methoxybenzyl)-4-[(7'R),5'-dihydroxy-3'-methoxybenzyl]tetrahydrofuran (2), resorcinol (3), pyrogallol (4) and 4-O-methylgallic acid (5).We evaluated the antioxidative, antielastase activities and melanogenesis inhibitory effects of these compounds. The $SC_{50}$ values of compounds 2 ~ 5 for free radical scavenging activity were $17.0\;{\sim}\;100.2\;{\mu}M$ and especially compounds 4 and 5 were 6-fold more effective than ferulic acid as a positive control. And compounds 2 ~ 4 inhibited human neutrophil elastase with $IC_{50}$ values of $18.8\;{\sim}\;78.2\;{\mu}M$ and compound 3 also inhibited melanin synthesis in B16F10 melanoma cells with an $IC_{50}$ value of $492.8\;{\mu}M$. These results suggest that Euryale ferox extract having a lot of various active ingredients may be useful as a natural multi-functioning agent.

A Study on the Latest Research Trends in Natural products with Anti-Aging Effects (항노화 효능을 가진 천연물에 대한 최신 연구 동향에 관한 연구)

  • In this study, four types of retinol, retinyl palmitate, adenocin, and polytoxylate-dretinamide, which are the ingredients of the Ministry of Food and Drug Safety Notice, included in the study. also we looked at trends in research on Sciadopitys verticillata, Prunella vulgaris, Celosia cristata L., Brazilin, Persicaria hydropiper, Astragalus membranaceus Bunge, Forsythiae Fructus, Lithospermum root, Rheum undulatum L. and Cistanche deserticola Y. C. Ma a natural material that has the efficacy of antioxidant aging. The anti-aging study so far has been found to be centered mainly on collagen production and elastase synthesis inhibition mechanisms. However, given that the aging process of the skin is caused by various ageing processes, it is believed that anti-aging studies using safe and effective natural materials that can help the skin age with various mechanisms should be conducted.

A Study on the Effectiveness of a Mixed Fermented Extract of Vitamin Tree Fruit, Acai Palm Fruit, Mango, Lemon, Apricot and Blueberry as an Active Ingredient (비타민나무열매, 아사이팜열매, 망고, 레몬, 살구 및 블루베리의 혼합 발효추출물의 효능 연구)

  • In this study, the cell viability, radical scavenging ability, elastase activity, moisturizing activity, and antimicrobial activity of mixed fermented extract of vitamin tree fruit, acai palm fruit, mango, lemon, apricot and blueberry as an active ingredient were measured. The experiment was performed at three concentrations(0.5 / 1.5 / 2%), since cytotoxicity was not shown at these three concentrations. Based on this, a radical scavenging experiment was conducted to compare with L-ascorbic acid, a representative raw material of an antioxidant material. The results showed that the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging ability was 95.1 ± 0.6%, 94.3 ± 0.7%, 95.3 ± 0.6%, 95.1 ± 0.7%, 95.1 ± 0.3%, 95.5 ± 0.3%, and 95.4 ± 0.4% respectively, when the concentration of the mixed fermentation extract was 2%. As the concentration increased at the three concentrations (0.5/1.5/2%) and the content of blueberry extracts increased, sample 7 showed excellent elastase activity at a concentration of 2% to 0.9 cm, such as retinol crystal, and moisturizing activity was also found to be 71.5%, 75.6%, and 81.6%, respectively. Particularly, at the 2% concentration of the mixed fermented extract, the antibacterial activity effect on Propionibacterium was very excellent in sample 7. Therefore, it is considered that it is worth developing the mixed fermented extracts of 2% concentration as a cosmetic component with antioxidant, moisturizing, and antibacterial properties.

Study on Cosmeceutical Activities from Fermented Ginseng Berry Extracts (진생베리 발효추출물의 화장품 약리활성)

  • Ginseng berry contains a large amount of Ginsenoside Re and has anti-inflammatory, anticancer, hypoglycemic and whitening effects. In this study, Rhizopus Oligosporus strain was used to establish ginseng berry fermentation process and cosmetic pharmacological activity of ginseng berry fermented product was analyzed.. The electron donating ability of ginseng berry extract by fermentation shown 81% at 1,000 ㎍/mL concentration. The ABTS+ radical scavenging ability of shown 100.2% at 1,000 ㎍/mL concentration. The tyrosinase inhibitory effect which is related to skin-whitening, was 57% at the concentration of 1,000 ㎍/mL. The elastase inhibitory effect which is related to skin-wrinkle, was 47% at 1,000 ㎍/mL concentration. Also, the collagenase inhibition effect was 33% at 1,000 ㎍/mL concentration. From these results, ginseng berry extracts by fermentation is considered to have anti-inflammatory, anti-wrinkle effect and whitening effect. Therefor, ginseng berry fermented product is expected to be very useful as an anti-inflammatory and anti-aging cosmetic raw material.

The study of oligopepetide synthesis and biological activity (올리고 펩타이드 유도체의 합성과 생리활성에 관한 연구)

  • In this study, we synthesized Oligo Peptide (Lys-Val-Ala-Arg-Pro: KVARP) and peptide derivatives using Solid Phase Peptide Synthesis(SPPS). KVARP was commonly known to improve whitening of skin. We measured bio-activity of the synthesized compounds. The whitening effect was measured in tyrosinase inhibition and the result showed to be highly effective with 93% inhibition rate at $5000{\mu}g/m{\ell}$ of Geranic-KVARP, on the other hand the IC50 value was $68{\mu}g/m{\ell}$. The wrinkle-reducing effect was measured by elastase inhibition at a concentration of 63% at $400{\mu}g/m{\ell}$ of Salicylic-KVARP, and the IC50 value was $253{\mu}g/m{\ell}$. In the DPPH assay, Caffeic-KVARP showed more than 95% antioxidant activity at $400{\mu}g/m{\ell}$ with high concentration and IC50 value was $31{\mu}g/m{\ell}$. The anti-inflammatory effect of Nitric Oxide inhibition was 67% at $400{\mu}g/m{\ell}$ of Lipoic-KVARP. Therefore, the four types of KVARP derivative that were synthesized from various experiments has shown that it could have potential to be used to develop new medicines, cosmetics as well as in various industries.

Inhibition effect against elastase, collagenase, hyaluronidase and anti-oxidant activity of thinning Green ball apple (그린볼 사과(Green ball apple; Malus pumila Mill.) 적과의 항산화 및 elastase, collagenase, hyaluronidase 저해 효과)

  • In this study, the anti-oxidative, health functional, and beauty food activities of water and ethanol extracts from newly bred Ruby S apple (Malus pumila Mill.). The results of measuring the 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity by treating the phenolic compound with thinning green ball apple at a concentration of 50-200 ㎍/mL showed that the water and ethanol extracts at a concentration of 200 ㎍/mL showed 94.69 and 92.24%, respectively. 2,2'-Azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) radical scavenging activities showed 100.30 and 99.16%, respectively, in 200 ㎍/mL of water and ethanol extracts. The water and ethanol extracts of Green ball showed antioxidant protection factor of 1.76 PF 1.76, respectively. The water and ethanol extracts showed 101.46 and 99.64% anti-oxidative effect on thiobarbituric acid reactive substances at phenolic concentration of 200 ㎍/mL. The water and ethanol extracts showed 33.28 and 32.14% hyaluronidase inhibition, respectively, at phenolic concentration of 150 ㎍/mL. The water and ethanol extracts showed 47.33 and 40.92% elastase inhibition and 46.19 and 65.58% collagenase inhibition at phenolic concentration of 200 ㎍/mL, respectively. About these experiments, thinning Green ball apple was found to exhibit anti-oxidation activity as well as hyaluronidase, elastase and collagenase inhibitory activities. Therefore, thinning Green ball apple can be considered a potential sources for new functional materials.
