Comparison of the Total Phenolic and Flavonoid Contents and Antioxidant Activities of Four Kinds of Sand Dune Plants Living in Taean, Korea (태안에서 서식하는 사구식물 4종의 폴리페놀, 플라보노이드 함량과 항산화 활성 비교)

  • This study attempted to investigate the antioxidant properties of four different species of sand dune plants (Calystegia soldanella, Messerschmidia sibirica, Vitex rotundifolia and Rosa rugosa). In order to validate the antioxidant activity of these plants, we first determined the total amount of flavonoid versus phenolic contents (TFC/TPC) and extracted crude flavonoids for measuring antioxidant activities that were determined by DPPH, ABTS and FRAP assays with radical scavenging effects. We found that highest amounts of TPC were detected in R. rugose with values of 110.20 mg/g (leaves) and 65.71 mg/g (stems), while the highest amounts of TFC in V. rotundifolia with values of 38.07 mg/g (leaves) and 6.55 mg/g (stems). We further examined how closely related the amounts of TFC/TPC in antioxidant activities and found that R. rugose has the highest activities of radical scavenging with values of $63.4{\mu}g/ml$ and $51.2{\mu}g/ml$ determined by DPPH and ABTS assays compared with the value of $21.2{\mu}g/ml$ by FRAP assay. It is of note that there is a statistically significant correlation between the resulting antioxidant activities and the total ratio of TFC and TPC, suggesting that the different amounts of TFC/TPC may directly contribute to the various antioxidant activities.

Isolation of Bioactive Compounds from the Ethylacetate Fraction of Fermented Garlic Complex and their Tyrosinase Inhibition Activities (대산(大蒜)을 포함하는 복합발효물의 에틸아세테이트 분획으로부터 Tyrosinase 저해활성 성분의 분리 및 동정)

  • Fermented complex from garlic and nine medicinal plants were developed as a natural whitening material. Tyrosinase inhibition activity was determined and four active compounds were isolated. The nutritional components of fermented garlic complex (FGC) were analyzed to confirm the applicability as a functional food material. Tyrosinase inhibitory effect of FGC was 88.6%. Methanol extract was partitioned with EtOAc, n-BuOH and H2O. From the EtOAc fraction (47 g), which showed the highest yield, active fractions were separated by repeated TLC, silica gel and ODS column chromatography to isolate active compounds. The chemical structures of the isolated compounds were analyzed by NMR and MS spectra. Phenylpropanoid compounds of 2,4,5-trihydroxy-benzenepropanoic acid (1) (1.9 mg) and 2,3,5-trihydroxy-benzenepropanoic acid (2) were confirmed. In addition, 2,4-dihydroxy-hydrocinnamic acid (3) (3.3 mg) and (+)sesamin (4) (6.1 mg) were isolated. These compounds will be useful as index compounds or functional compounds in FGC.

Component Analysis and Antioxidant Activity of Adenophora triphylla (잔대의 함유성분 분석과 항산화 활성)

  • The levels of minerals, amino acids, fatty acids and sugars and antioxidant capacity of Adenophora triphylla (AT) root were investigated. The moisture, crude protein, crude lipid, crude ash and carbohydrate contents of the AT root were $3.4{\pm}0.3,\;9.4{\pm}0.1,\;1.6{\pm}0.1,\;3.7{\pm}0.2$, and $81.9{\pm}0.5$, respectively. Among the minerals present in root, potassium content was the highest. Total amino acid contents were $103.5{\pm}9.1$ mg/g. Palmitic acid was the most abundant fatty acid in AT root. The vitamin E and C levels in AT root were $1.1{\pm}0.1$ and $5.2{\pm}0.5$ mg/100 g, respectively. The antioxidant activity of 70% ethanol extract and fractions of the AT root has been determined by the scavenging of the stable radical DPPH; the result showed that the chloroform fraction was the most active, as the amount required for 50% reduction of DPPH after 30 min ($RC_{50}$) was $103.7{\mu}g$, followed by 70% ethanol extract ($265.4{\mu}g$), hexane fraction ($104.4{\mu}g$), ethyl acetate fraction ($104.4{\mu}g$), butanol fraction ($108.9{\mu}g$) and aqueous fraction ($484.1{\mu}g$).

Biological Activities of Magnolia denudata Desr. Flower Extracts (목련(Magnolia denudata Desr.) 꽃 추출물의 생리활성)

  • The antioxidant, antiproliferation, and nitrate synthesis inhibitory effects of Magnolia denudata extracts (ME) were evaluated. The ME was extracted with 70% (v/v) ethanol and fractionated with solvents of hexane, chloroform, ethyl acetate, n-buthanol and aqueous. The ethyl acetate fraction contained the highest phenolic and flavonoid contents of 427.10 mg garlic acid eq/g and 356.05 mg catechin eq/g, respectively. The ethyl acetate fraction showed strong 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity with a 50% inhibition concentration ($IC_{50}$) of 0.20 mg/mL and total antioxidant activity was 0.90 mg AA eq/100 mg. From the results of cytotoxic effects of HCT116, NCL-H460, and HepG2 human cancer cells by MTT assay on the ME and its solvent fraction, chloroform fraction showed the highest cytotoxic effect ($IC_{50}$ value: 0.14, 0.37, and 0.41 mg/mL, respectively). Nitrate synthesis inhibitory effect of ME and its solvent fractions on nitric oxide synthase activity in LPS stimulated RAW 264.7 cells were decreased in dose-dependent manners, and $IC_{50}$ value of hexane and chloroform fractions were 0.39 and 0.49 mg/mL, respectively.

  • An entomopathogenic bacterium, Photorhabdus temperata ssp. temperata (Ptt), suppresses insect immune responses and facilitates its symbiotic nematode development in target insects. The immunosuppressive activity of Ptt enhances pathogenicity of various microbial pesticides including Bacillus thuringiensis (Bt). This study was performed to select a cheap and efficient bacterial culture medium for large scale culturing of the bacteria. Relatively cheap industrial bacterial culture media (MY and M2) were compared to two research media, Luria-Bertani (LB) and tryptic soy broth (TSB). In all tested media, a constant initial population of Ptt multiplied and reached a stationary phase at 48 h. However, more bacterial colony densities were detected in LB and TSB at the stationary phase compared to two industrial media. All bacterial culture broth gave significant synergism to Bt pathogenicity against third instars of the diamondback moth, Plutella xylostella. Production of bacterial metabolites extracted by either hexane or ethyl acetate did not show any significant difference in total mass among four culture media. Reverse phase HPLC separated the four bacterial metabolites, which were not much different in quantities among four bacterial culture broths. This study suggests that two industrial bacterial culture media can be used to economically culture Ptt in a large scale.

Isolation and Purification of Tyrosinase Inhibitors from the Seeds of Thuja orientalis L. (백자인(Thuja orientallis L.)으로부터 tyrosinase 저해제의 분리 및 정제)

  • Previously, the methanolic extracts of thirty Korean medicinal plant seeds were screened for tyrosinase inhibitors using a rapid and simple TLC method, which was superior to a conventional spectrophotometrical in vitro assay. As a result, the methanolic extracts of Thuja orientalis seeds was found to have strong tyrosinase inhibitory activity. To isolate active tyrosinase inhibitors, the seeds were defatted with n-hexane under reflux, and then extracted twice with methanol under reflux at 90$^{\circ}C$. The methanolic extract was evaporated to a small volumn in vacuo, and then successively fractionated with ether, ethyl acetate and n-butanol. The ether extract showing significant tyrosinase inhibitory activity was solubilized with 5% NaHCO$_3$and then acidified with 6N HCI. The ether souble acidic fraction was successively ohromatographed on silica gel, Sephadex LH-20 and preparative TLC. Among four compounds isolated, two of them showed stronger tyrosinase inhibitory activity, comparable to that of L-ascorbic acid (IC$\sub$50/=28$\mu\textrm{g}$/$m\ell$). These results suggest that Thuja orientalis seeds may be useful as potential sources of antibrowning agents in fruits and vegetables, and anti-melanoma agents in cosmetics and phamaceuticals.

Anti-melanogenesis Activities for the Terpenes from the Acantophanax koreanum Stems (섬오갈피나무 줄기 유래 테르펜 화합물의 멜라닌 생성 저해 활성)

  • A. koreanum was investigated to identify the constituents possessing whitening effects. As anti-melanogenesis activities were screened for the ethanol extract and solvent fractions, n-hexane (Hex) and ethyl acetate (EtOAc) fractions showed the most potent activities. Three constituents were isolated from the n-Hex fraction of A. koreanum; kaurenoic acid (1), $16{\alpha}$-hydro-17-isovaleroyloxy-ent-kauran-19-oicacid (2), $16{\alpha}$-hydroxy-17-isovaleroyl-oxy-ent-kauran-19-oic acid (3). The chemical structures of the isolated compounds were elucidated based on the spectroscopic data including $^1H$ and $^{13}C$ NMR spectra, as well as comparison of the data to the literature values. Whitening effects were studied for the isolated compounds. Upon the anti-melanogenesis test using ${\alpha}-MSH$ stimulated B16F10 melanoma cells, the compounds 1, 2 and 3 inhibited the cellular melanogenesis and intracellular tyrosinase activities effectively. Based on these results, A. koreanum stems extract could be potentially applicable as whitening ingredients in cosmetic formulations.

Investigation of Antimicrobial Activity and Stability of Ethanol Extracts of Licorice Root (Glycyrrhiza glabra) (감초의 에탄올 추출물의 항균활성 및 안정성 조사)

  • Conditions for extraction of antimicrobial materials from licorice root, Glycyrrhiza glabra, were optimized. Among solvents tested, 95% ethanol gave highest antimicrobial activity, and was chosen as optimal extracting solvent. Extraction temperature and time were optimal at room temperature and for 12 hr, respectively. Minimal inhibitory concentration (MIC) of 95% ethanol extracts was determined against 14 microorganisms. Reference microorganisms included 6 Gram(-) bacteria, 4 Gram(+) bacteria, and 4 yeast strains. Ethanol extract exerted very strong growth inhibition on Gram(+) bacteria, while was moderately effective for Gram(-) bacteria and yeasts. Treatment at $180^{\circ}C$ for 30 min or extreme pHs merely destroyed antimicrobial activity of ethanol extract. These findings suggest ethanol extract of G glabra may be useful as natural preservative.

Antioxidative Activity and Chemical Characteristics from Different Organs of Small Black Soybean (Yak-Kong) Grown in the Area of Jungsun (정선산 쥐눈이콩의 부위별 이화학적 특성 및 항산화 효과)

  • Nutritional composition, contents of phenolic compounds, and antioxidative activities of small black soybean were investigated. Proximate composition of the soybean was crude fat, crude protein, carbohydrate, and crude ash at 15.9, 34.7, 38.5, and 3.7%, respectively. Potassium was the predominant mineral, followed by phosphorus, calcium, and magnesium. Glucose and galactose were the major sugars. Twelve different kinds of fatty acids were identified in whole soybean, dehulled soybean, and soybean hull. The 75% methanol and ethyl acetate extract fractions from the soybean hull contained total phenolic compounds at 4.64 and 13.53 g/100 g, respectively. The methanol extract of soybean hull showed strongest antioxidative activity among samples tested. Ethyl acetate fraction of the soybean hull exhibited DPPH radical scavenging activity at level similar to those of ${\alpha}-tocopherol$ and BHT. Phenolic compounds were the major biological components in the soybean hull. These results suggest that small black soybean can be used as a new material for functional food.

Antibacterial Activity of Oriental Medicinal Herb Extracts against Skin Pathogens (한약재 추출물의 피부 염증 유발 세균에 대한 항균활성)

  • The antibacterial activity of methanol extracts from 70 kinds of oriental medicinal herbs on four strains of Staphylococcus aureus and S. epidermidis was investigated. The results showed that C. japonica, C. sappan, R. javanica, R. tanguticum, and S. miltiorrhiza had an antibacterial activity on all the strains used. Among these, C. japonica and R. javanica, which showed excellent antibacterial activity, were extracted with water, ethanol, methanol, and ethyl acetate for further study of antibacterial activities. The results showed that the boiled water extract of C. japonica had the best antibacterial activity. Assuming that the antibacterial activity of C. japonica originated from berberine, which has been reported many times, the berberine content of the boiled water extract of C. japonica was analyzed and the result was 13.88%. To compare the antibacterial activity of berberine with that of other antibiotics, berberine chloride and three other kinds of antibiotics were investigated, which showed that berberine chloride had an antibacterial activity on KCCM 35494 S. epidermidis only when the concentration was higher than 600 mg/l, and it did not show antibacterial activity in the other strains. Based on these results, it was concluded that the antibacterial activity of the boiled water extract of C. japonica on the strains used in this study originated from high concentration of berberine or substances other than berberine. Therefore, identification of the substance will be necessary.
