Stabilities of Plasmid Vectors in Zymomonas mobilis (Zymomonas mobilis Plasmid Vector의 숙주세포 내에서의 안정성에 관한 연구)

  • The stabilities of plasmid vectors in Zymomonas mobilis were tested in batch and continuous cultures. It was found that the growth of the host Zymomonas strain was greatly affected by the size of plasmids as well as the composition of nutrient media: the host cells grew taster when harboring plasmids of smaller sizes and in n non-selective medium. All the Zymomonas plasmid vectors containing antibiotics selective markers and Zymomonas replication origins could be maintained in a stable manner over 30 generations without being integrated into host chromosomes.

  • A series of tests was performed to determine a threshold concentration of water-soluble organic permeant(ethanol) for permeability to be increased in clay and to estimate long-term permeability behaviors, effects of overburden pressure and compaction conditions on permeability in clay. Results of study are as follows ; 1. A threshold concentration of water-soluble organic permeant(ethanol) in clay was about 7O~8O% and its dielectric constant was 40, and dielectric constant seems to be closely related with absolute permeability. 2. Permeability of long-term tests was more or less larger than that of short-term tests. 3. Overburden pressures applied for a long time elapsed have little effects on the restriction of permeability increase. 4. Since water content has no effects of compaction capability when it is compacted with OMC or wet side of OMC, its permeability is to be estimated as same level of dry side of OMC. 5. Clays matured in the humid chamber are increased in permeability 2 times larger than unmatured ones. Thixotropy, therefore, should be considered in the design procedures because field conditions of construction would be quite similar to this..

Larval migration inhibition activity of ivermectin, doramectin and ethanol against Anisakis simplex in vitro (시험관내에서 이버멕틴, 도라멕틴, 에타놀에 대한 아나사키스 유충의 운동성 억제효과)

  • This experiment has been investigated in order to examine larval migration inhibition activity of ivermectin, doramectin and ethanol against Anisakis simplex (A. simplex) in vitro. A. simplex larvae were obtained from the mackerel acquired from the fish market of Cheongju. They were divided into many groups and placed in culture dishes (40 larvae each) containing RPMI-1640, in the absence or presence of different concentrations of ivermectin, doramectin and ethanol. Ivermectin had a complete inhibition of larval migration at 72 h in all groups ($10-300{\mu}g/ml$). Ethanol reduced the migration of the larvae, its maximum activity being an high doses (7.5%, 10% ethanol) when it was 100% efficacy at 4 h. Doramectin had no efficacy in vitro. Being needed that further studies with ivermectin and doramectin, it is recommended that in vivo test with laboratory animals should be carried.

  • A simple and modified method is developed to determine a lipase activity. A linear relationship exists between the color intensity and the free fatty acid liberated by enzyme action. The range of determination is from 0.05 to $1.5\;{\mu}moles$ of long chain fatty acid (oleic acid) and 0.2 to $2.0\;{\mu}moles$ of short chain fatty acid (caproic acid). The cumbersome procedure of the removal of the upper aqueous phase which was required in the previous copper soap extraction method was eliminated by the movement of solvent phase to upper phase in the respective biphasic system with a mixture solvent (chloroform: n-hexane:ethanol = 49:49:2) and copper reagent saturated with sodium chloride.

Effects of Saponin and Ethanol Extract of Panax Ginseng against Thioacetamide - Induced Hepatotoxicity in Rats (Thioacetamide 유발 흰 쥐 간독성에 대한 인삼 사포닌 및 에타놀 추출물의 효과)

  • Panax ginseng has been used for various diseases including hepatic disorders. The aim of the present study was to investigate the hepatoprotective effects of ethanol extract and saponin of Panax ginseng in thioacetamide-intoxicated rats and to compare with silymarin, a known hepatoprotective agent. Male Sprague-Dawley rats were given single intragastric administration of thioacetamide. Aqueous solutions of ethanol extract and saponin of Panax ginseng with or without silymarin were administered intragastrically daily for six days from four days before until one day after thioacetamide administration. At the end of the treatment, the rats were fasted overnight and sacrificed. As a result, thioacetamide caused significant increase in serum levels of AST, ALT, 5'-nucleotidase and bilirubin. Thioacetamide increased $Ca^++$ content but decreased protein content in liver tissue. These thioacetamide-induced biochemical changes were prevented both by ethanol extract of ginseng and silymarin, but not by ginseng saponin. Silymarin did not potentlate the effect of either ethanol extract or saponin of ginseng on these parameters. Thioacetamide-induced confluent necrosis was not protected by the test drugs. In conclusion, ethanol extract of ginseng protects the liver possibly by stabilizing the cell membrane and by inhibiting thioacetamide-induced $Ca^++$ increase in the hepatocytes, which was comparable to that of silymarin.

  • In our continuing effort to investigate compounds having anti-inflammatory activity from natural products, Ailanthus altissima was examined. Among six compounds isolated from Ailanthus altissima, Luteolin-7-O-glucoside (L7G) along with ethanol extract of Ailnathus altissima (EAa) were chosen to determine their inhibitory activity on secretory recombinant phospholipase $A_2s$ enzyme activity in vitro. As a results, EAa inhibited human recombinant $sPLA_2-V$ ($IC_{50}$ of about 100 ${\mu}g/ml$) and $cPLA_2$, ($IC_{50}$ of about 59 ${\mu}g/ml$), while L7G showed strong inhibitory effect on $sPLA_2-A$, V and $cPLA_2$ with an $IC_{50}$ value of approximately 40 ${\mu}M$, respectively.

  • Six phenolic acids were isolated from Jindalrae flowers (Rhododendron mucronulatum Turcz.), an edible plant in Korea. These compounds were identified as chlorogenic acid, 3,5-O-dicaffeoylquinic acid, 4,5-O-dicaffeoylquinic acid, caffeic acid, ferulic acid, and p-coumaric acid on the basis of IR, UV, $^{1}H$ and $^{13}C$ NMR, FAB-MS, ES-MS and/or El-MS data. Chlorogenic acid (0.2 g) present in both ethyl acetate and ethyl ether fractions comprised up to 38.5% of the total phenolic acid amount (0.52 g) finally recovered by means of polyamide C-200 column chromatography, preparative TLC, recrystallization, and Sephadex LH-20 column chromatography The antioxidant activities were measured in an ethanol solution of linoleic acid in the presence of ferric thiocyanate. The antioxidant efficiency increased in the order of p-coumaric acid<${\alpha}-tocopherol$

Effects of Ethanolic Extracts of Scutellaria Radix on the alveolar bone formation in the extract socket of rat (황금(Scutellaria Radix)의 에타놀 추출물이 백서 치조골 형성에 미치는 영향)

  • The purpose of this study was to evaluate the effects of ethanolic extracts of Scutellaria Radix on the alveolar bone formation in the extract socket of rat. Thirty-six Sprague-dawley rats were used in this study. Mean body weight of rat was $130{\pm}5g$. Experimental animal were administered 0.4% ,${\beta}-aminopropionitril$(Sigma, USA) with the solid commercial food for 5 days. All the maxillary 1st molar of the rats were extracted by using of the tissue forcep under the general anesthesia with Pentobarbital sodium(Tokyo Chemical Co, Japan) injection into intraperitoneal space. All the extracted rats were divided into two group, experimental group which were feeded the solid food mixed ethanolic extracts of Scutellaria Radix, and control group which were feeded same food without reagent. At 1, 3, 5, 7, 9 and 14th days after tooth extraction, rats in both groups were serially sacrificed respectively. All the specimen were treated as usual method and prepared Hematoxylin-eosin stain for the light microscopic observation. The results were as follows : 1. Bone formation of extracted socket starts from the area on remained periodontal ligament than other area. 2. In the case of administration of the extracted Scutellaria Radix showed rapid healing process of connective tissue than non-administrated group. 3. In the case of administration of the extracted Scutellaria Radixshowed rapid osteogenesis than non-administrated With above results, it was concluded that ethanolic extracts of Scutellaria Radix may play a favorable role on the healing process of exatraction socket after extraction in rats. It was suggested that further study to evaluate the different concentration and administration method of ethanolic extracts of the Scutellaria Radix into same experimental model.

The Experimental Model on Morphological Changes of the Liver Cell Treated with Ethanol (에타놀 처치 간세포(肝細胞)의 형태학적 변화에 대한 실험모델)

  • In order to prepare an experimental model on the basis of morphological changes of liver, 90 rabbits were allocated into four groups and 5, 15, 30 and 50% of ethanol (5ml/day) were dosed to each group for 5, 10, 20, 30, 60 and 90 days, and observed the livers with light and electron microscope. The results obtained are summarized as follows: 1. Light microscopically, the hydropic changes were found in the liver from 10th day after 5% and 15% ethanol ingestion, and these findings were remarkable as time goes by. Necrosis of hepatocytes, however, was most remarkable on 60th and 90th day after ingestion. The proliferation of interlobular connective tissue was found from 30th day in this group. 2. The hydropic necrosis of liver cells was found from 5th day in 30% and 50% ethanol dosing groups. The proliferation of interlobular connective tissues and infiltration of the lymphocytes were observed from 30th day in 30% ethanol ingestion group, however, it was found from 20th day in 50% ethanol ingestion group. 3. Electron microscopically, the appearance of various lipid droplets and dilated rER, slightly increased sER and glycogen field, dense mitochondria with scanty cristae and hydropic change were observed on 5th day after 5% ethanol ingestion. These findings were more remarkable on 30th and 90th day. Especially, disorganized outer and inner membrane of mitochondria and autophagic vacuoles were appeared on 90th day. The similar findings were also found in 15%, 30% and 50% ethanol ingestion groups, however, the appearance of fibrillar and myelin-like structures were observed from 30th day after 15% ethanol ingestion. The severe hydropic change and increased sER were observed on 90th day after 15% ethanol ingestion. From 5th day after 30% and 50% ethanol ingestion these findings were more remarkable.

Chemical Properties of Fucoidans from Hizikia fusiformis and Sargassum fulvellum (톳과 모자반 fucoidan의 화학적 특성)

  • Fucoidans were isolated from Hizikia fusiformis and Sargassum fulvellum and characterized on their chemical properties. Crude fucoidans were extracted at $65^{\circ}C$ for 1hr with acid solution of pH 2.0, and cetylpyridinum chloride was used for partial purification. The yields of partially purified fucoidans were $2.51\%$ for H. fusiformis, and $65\%$ for S. fulvellum, respectively. The partially purified fucoidans were separated into 3 fractions by DEAE-Sephadex A-25 ion exchange column chromatography and the major fractions were refractionated with fractional preripitation with ethanol. $60-70\%$ ethanol precipitated fraction(P-70) of H. fusiformis and $60-65\%$ ethanol precipitated fraction(P-65) of S. fulvellum turned out to be homogeneous by cellulose acetate electrophoresis and gel filtration chromatography. The molar ratios of fucose, galactose, and sulfate In the purified fucoidans were 1 : 0.66 2.74 for H. fusiformis and 1 : 0.24. 1.46 for 5. fulvellum. The averaged molecular weights of the purified fucoidans from H. fusiformis and S. fulvellum were 26,000 and 105,000, respectively.
