Phytoestrogen Extraction for Relaxation of Female Menopause Symptoms from Natural Products (천연물로부터 여성갱년기 증상완화를 위한 식물성 에스트로겐의 초음파 추출특성)

  • Phytoestrogens are one of quasi-estrogens which are not generated within the endocrine system, but consumed by intaking phytoestrogenic plants. Phytoestrogens, also called as "dietary estrogens", are various botanic chemical compound groups naturally occurring nonsteroidal plant compounds of spontaneous generation. Due to their structural similarities to estrogone, they may provide desirable health benefits for reducing the menopausal symptoms. In this paper, various phytoestrogens were extracted from sophora root, pomegranate and kudzu by ultrasonic extraction process. In this study, the amount of extracted phytoestrogens at various ultrasonic power strengths and extraction times were compared, which could be used as basic data in the study of biology and chemistry related to natural products.

Verification of Estrogenic Activity in Ethanol Extracts of Marine Organisms Using in vitro Test System. (In vitro 검출시스템을 이용한 해양생물 추출물로부터 에스트로겐 활성 검증)

  • In order to verify the occurrence of an estrogenic compound in natural products, the estrogenic activity was measured using an in vitro detection system. For this system, human breast cancer cell line MCF7 was transfected using an estrogen responsive CAT reporter plasmid. Estrogenic activities of photosynthetic algae spirulina and sea lettuce were evaluated using this system. Estrogenic activities of a $500\mug/ml\; and\; 50 \mug/ml$ ethanol extracts of spirulina were as much as that of $10^{-8}$M standard solution (17$\beta$-estradiol) and activity of $5\mug/ml$ ethanol extract of spirulina was as much as that of $10^{-10}$ M standard solution. However, no significant estrogenic activity was observed using sea lettuce extract. Estrogenic activities of marine animals, such as star fish and shrimp, were also evaluated using this system, however, no significant estrogenic activity was observed in these extracts. In this result, it is confirmed that spirulina extract possesses estrogenic compound.

  • 전국매립지 중 수도권, 울산, 대구 세 매립지의 침출수를 채수하여 Photobacterium을 이용한 독성 test와 Yeast Two-hybride Based Assay을 이용하여 에스트로겐성을 측정하였다. 독성도 값은 대구에서 145로 가장 높은 값을 나타냈다. 에스트로겐성은 울산이 hER 3.2ppt, mER 210ppt로 가장 높게 나타났으며, 채수지점 모두 industrial hormones에 반응성이 좋은 mER 값이 높게 나타났다. 특히 대구지점은 독성도 값은 높으나 에스트로겐성은 낮은 값을 나타내는 것으로 보아 호르몬 영향을 억제하는 antangonist chemicaldml 존재 가능성이 시사된다.

The Phytoestrogenic Effect of Daidzein in Human Dermal Fibroblasts (피부 섬유아세포에서 다이드제인의 파이토에스트로겐 효과)

  • Estrogen deficiency results in a reduction of skin quality and function in postmenopausal women. Over the past decade, many studies have supported that estrogen provides anti-aging effects as a result of the ability of estrogen to prevent skin collagen decline, restore skin elasticity, and increase skin hydration in postmenopausal women skin. Due to their structural similarity with estrogen, isoflavones have been called phytoestrogens. Photoprotective effects of isoflavones are well established while their estrogenic-like activities are not fully understood in human skin. In this study, we investigated whether daidzein, an effective isoflavone, has phytoestrogenic activity and induces transcriptional change of extracellular matrix components in dermal fibroblasts. We examined the luciferase activity of daidzein and ${\beta}$-estradiol using transiently transfected NIH3T3-ERE cells. The estrogenic receptor-dependent transcriptional activity was increased in a dose-dependent manner when treated with daidzein, with a maximum of 2.5-fold induction at $10{\mu}g/mL$ of daidzein compared with non-treated control. In addition, daidzein significantly in creased the expressions of collagen type I, collagen type IV, elastin, and fibrillin-1 in human dermal fibroblasts. By comparing with the effects of ${\beta}$-estradiol through out all the experiments, we confirmed that daidzein had estrogenic activity and function in fibroblasts. These results suggest that daidzein-based application, having both photoprotective and phytoestrogenic effects, may be a powerful approach for skin anti-aging of postmenopausal women.

  • Estrogens are known as the steroid hormones and essential regulators of developments, differentiations, and fertility in animals including humans. Recently, classic focus on estrogens which are considered as female hormone is changing in the whole field of reproductive endocrinology. Especially, interest in estrogen functions in male reproduction is increasing more and more, as numerous studies about the endocrine disruptors, interrupting the endocrine system, are being carried out. To understand exactly the function of estrogen in a male reproductive system, a summary for estrogen receptors upon developmental distributions in testis will be useful. In addition to the regulatory roles of estrogen in male, unexpected exposure to exogenous estrogens causes defects of differentiation of male reproductive system and an injury of spermatogenesis. Also, this review highlights the indicator of exogenous estrogens to perturb male fertility. These approaches would give tile practical information about estrogen roles in male development and reproduction.

Dose-Response Characteristics of Uterotrophic Activity in Immature Female Rats Treated with Estrogen Agonist and Antagonist

  • 자궁내 반응에 대한 용량 의존적인 효과를 검색하기 위해 에스트로겐 agonist 및 antagonist를 사용하여 미성숙 랫드에서 uterotrophic assay를 실시하였다. 김(2000) 등은 이미 난소절제랫드를 이용한 3-day uterotrophic assay에서 에스트로겐성 물질에 대한 검색 시험을 실시한 바 있다. 양성대조물질인 17-ethinyl estradiol (EE)을 미성숙랫드에 매일 3일간 피하 투여하였으며, EE의 에스트로겐 활성을 차단하기 위한 항에스트로겐성 물질인 ZM189.154은 EE와 병행 투여하였다. 본 시험 결과 EE 1.0$\mu\textrm{g}$/kg 이상 투여군에서는 자공 비대와 fluid 저류등이 대조군에 비해 육안적으로 뚜렷하게 나타났다. 자궁무게(wet 및 blotted)는 용량 의존적으로 증가하였으며, 특히, EE 1.0$\mu\textrm{g}$/kg 이상에서는 유의성있는 증가를 나타내었다. 또한 질무게 증가도 자궁무게 증가와 일치 하였다. EE 0.3$\mu\textrm{g}$/kg을 투여한 후 ZM189, 154 0.1 mg/kg 및 1.0 mg/kg 투여와 비교할 때 EE에 의해 유도된 에스트로겐 활성이 용량 의존적으로 감소되었으나 유의성 있는 결과는 ZM189, 154 1.0 mg/kg군에서만 나타났다. 본 연구결과 EE는 uterotrophic assay에서 자궁 무게 (wet와 blotted)를 용량 의존적으로 증가시켰으며, ZM189, 154 1.0mg/kg은 EE의 에스트로겐 활성을 유의하게 감소시켰다. 따라서 미성숙 랫드를 이용한 3-day uterotrophic assay는 내분비계 장애물질 검색 시험법으로 우수하다는 것을 알 수 있었다.

Roles of Estrogen in the Reproductive Organs of Male Rodents : Focus on ERKO Mice Model (설치류 수컷 생식기관에서의 에스트로겐 기능 : 에스트로겐 수용체 녹아웃(ERKO) 생쥐 모델을 중심으로)

  • It has been known for many years that administration of estrogen, a so-called female hormone, has harmful effects on male fertility. However, studies employing transgenic mice deficient in estrogen receptors reveal substantial roles for estrogen in male fertility. The aim of this article is to review and summarize the current knowledge on the estrogen receptor localization in male reproductive organs including male germ cells of rodents. Also, informations about the mice models with disrupted estrogen signaling and associated phenotypes will be provided.

Effects of Estrogen on Nitric Oxide and Aortic Atherosclerosis in Cholesterol-Fed Rabbits (고지방식 가토에서 nitric oxide와 대동맥 동맥경화증에 대한 에스트로겐의 영향)

  • 배경: 에스트로겐이 동맥경화증의 진행을 억제하고 예방하는데 중요한 역할을 하는 것으로 알려졌으나 그 기전은 아직 명확하지 않다. 본연구에서는 에스트로겐이 혈관의 확장에 중요한 역할을 하고 있는 nitric oxide와 동맥경화에 어떠한 영향을 미치는지를 알아보고자 하였다. 대상 및 방법: 뉴질랜드종 웅성 가토 40마리를 실험동물로 하여 정상식이군(ND), 정상식이와 0.02ig의 estradiol-2 patch 투여군(END), 0.5% 콜레스테롤 투여군(CD), 0.5% 콜레스테롤 투여와 0.02 $\mu\textrm{g}$의 estradiol-2 patch 투여군 (ECD)의 네군으로 나누어 12주간 실험을 하여 혈중 estradiol-2, nitric oxide, cholesterol 등을 측정하고 흉부동맥과 복부동맥의 동맥경화 병변을 관찰하였다. 결과: 혈중 estradiol-2의 농도는 ED, ECD 군에서 ND, CD 군보다 의의 있게 높았다. (p<0.0001). 혈중 nitric oxide 농도는 정상 대조군 (ND)을 제외한 나머지 세군(END, CD, ECD)에서 모두 의의 있게 감소하였다.(p<0.001). 혈중 콜레스테롤은 총콜레스테롤, LDL콜레스테롤, HDL 콜레스테롤은 CD, ECD의 두 군에서 모두 증가하였으나 의의는 없었다(p<0.05). 혈중 콜레스테롤과 중성지방의 농도는 에스트로겐에 의한 영향이 관찰되지 않았다. 흉부 및 복부동맥의 동맥경화 병변은 ND군과 END군에서는 발생하지 않았으나 콜레스테롤을 투여한 CD군과 ECD군의 두 군에서 현저하게 발생하였으며 혈관별 발생정도의 차이는 없었다. 에스트로겐에 의한 병변의 발생 차이는 관찰되지 않았다. 광학현미경상 동맥경화병변은 거의 지방을 탐식한 대식구의 증식으로 이루어졌었으며, 괴사나 섬유화 병변은 관찰되지 않았다. 전자현미경상에서는 내피세포의 광범위한 손상이 관찰되었다. 병리학적으로 에스트로겐에 의한 영향은 인정하기 어려웠다. 결론: 고지방식은 혈중 nitric oxide의 생산을 크게 저하 시키며, 그 원인으로는 지방의 과산화에 의한 내피세포의 손상때문인 것으로 추정된다. 또한 estradiol-2 patch 처치에 의해서 nitric oxide가 감소된 것은 그이유를 설명하기 곤란하나 성차이 및 에스트로겐의 투여방법에 의한 어떤 영향이 있었을 것으로 추측된다. 피부를 통한 에스트로겐의 투여는 혈중 콜레스테롤이나 중성지방의 농도에 영향을 주지 못했으며, 동맥경화의 진행에도 영향이 없었다. 에스트로겐의 동맥경화에 주는 영향에 대해서는 지속적이고 광범위한 연구가 더 필요할 것으로 사료된다.

Effects of Phytoestrogen on Cell Growth and Insulin-like Growth Factor-I (IGF-I) Production in MC3T3-El Cells (식물성 에스트로겐이 MC3T3-El 골아세포의 성장과 Insulin-like Growth Factor-1(IGF-1)생성에 미치는 영향)

  • Estrogen is known to play an important role in maintaining bone mass, since the concentration of serum estrogen decrease after menopause and the estrogen deficiency results in bone loss. Phytoestrogens are plant compounds with estrogen-like biological activity, In this study, to investigate the bioactivities of phytoestrogen, which act on bone metabolism, we examined the effect of selected food-borne phytoestrogens (genistein, daidzein and resveratrol) on osteoblast proliferation and IGF-I production using MC3T3-El cells, a mouse calvaria osteoblast-like cell line. Cells were cultured in a serum free medium for 48 hr in the presence of genistein $(10^{-5}\;M)$, daidzein $(10^{-5}\;M)$ and resveratrol $(10^{-5}\;M)$. The effects of genistein, daidzein and resveratrol on the cell proliferation and growth were evaluated by total cell numbers, MTS assay and cell migration assay. Their effect was compared with the $17\beta-estradiol$. Genistein, daidzein and resveratrol exhibited stimulatory effects on the growth of MC3T3-El cells, and the most pronounced effect was shown with daidzein. In addition, these phytoestrogen increased alkaline phosphatase activity of MC3T3-El cells. These effects were similar to that of $17\beta-estradiol$ effects. Moreover, treatment with genistein, daidzein and resveratrol increased production of insulin like growth factor-I (IGF-I) in conditioned media, indicating that the growth promoting effects of these phytoestrogen were related to the changes in production of IGF-I by MC3T3-El cells. These results show that genistein, daidzein and resveratrol have a stimulatory effect on osteoblast function, and that these findings in a cell model may prove relevant to protecting against the loss of bone mass and the development of osteoporosis in human subjects.

Distributive Characterization of Estrogenic Activity in Sediments from Gwangyang Bay, Korea (광양만 퇴적물에서의 에스트로겐 활성분포 특성)

  • In this study, we tried to quantitatively study the distribution of estrogenic activity in sediment from Gwangyang Bay by E-screen assay. Besides, we compared the estrogenic activity and the concentration of chemical pollutants. The highest estrogenic activity was recorded at the stations(GY6 and GY8) close to industrial complex and the river mouth of Seomjin. These results obtained from the E-screen assay similar to those of simultaneous analytical detection of 310 chemicals. In particular, GY6 and GY8 sites are confirmed as the full agonist sites because of their RPE values were over 90% having strong estrogenic effect. Also, their EEQ(Estradiol Equivalency Quantity) values are 35.6 ng/g and 14.6 ng/g, low than that of other sites, and these results suggests that have relatively high estrogenic efficiency in Gwangyang Bay. From these results, we can estimate that the stations close to industrial complex and the river mouth of Seomjin are major sources of endocrine disrupter in Gwangyang Bay. On the other hand, when we tried to compare the endocrine disrupter activity and $COD_{Mn}$ value, that is not correlated.
