Anti-Inflammatory Activity of Ethanol Extract of Sargassum miyabei Yendo via Inhibition of NF-κB and MAPK Activation (NF-κB와 MAPKs 활성 저해를 통한 미야베 모자반(Sargassum miyabei Yendo) 에탄올 추출물의 항염증 활성)

  • The aim of this study was to investigate the anti-inflammatory effect of Sargassum miyabei Yendo ethanol extract (SMYEE) using RAW 264.7 cells and croton oil-induced Balb/c mice. SMYEE inhibited the production of pro-inflammatory cytokines [interleukin (IL)-6, tumor necrosis factor $(TNF)-{\alpha}$, and $IL-1{\beta}$] and nitric oxide in lipopolysaccharide (LPS)-induced inflammatory response. In addition, SMYEE suppressed the expression of inducible nitric oxide, cyclooxygenase-2, and nuclear factor-kappa B. Further, SMYEE inhibited the expression of mitogen-activated protein kinases (MAPKs), such as extra cellular signal-regulated kinase 1/2, p38, and c-Jun N-terminal kinase. In ear edema test, edema formation in the SMYEE treatment was lower than that in the positive control and was similar to that in the prednisolone treatment group. Photomicrographs of mice ear tissue showed a reduction in dermal thickness and number of infiltrated mast cells. Therefore, our results indicate that SMYEE exerts an anti-inflammatory effect via inhibition of nuclear factor ${NF}-{\kappa}B$ and MAPK activation and can be used as a natural source of anti-inflammatory compounds.

Anti-oxidative Effect of Chungsimyeonja-um (CSYJE) via Nrf2/HO-1 Pathway Activity in Lipopolysaccharide (LPS) Induced RAW 264.7 Macrophages (대식세포에서 Nrf2/HO-1경로를 통한 청심연자음의 항산화효과)

  • Reactive oxygen species (ROS) plays an important role in maintaining homeostasis. However, excessive ROS production damages cellular components such as proteins, lipids, and nucleic acids and promotes skin aging. In this study, we confirmed the antioxidant effect of CSYJE to prevent excessive oxidative stress. First, DPPH and ABTS assays were performed to confirm the antioxidant effect of CSYJE and the radical scavenging activity was confirmed depending on the concentration. As a result of performing the MTT assay to confirm the cell viability, it was confirmed that there was no cytotoxicity at a concentration of 1,000 ㎍/mL. As a result of western blotting to confirm the expression levels of the antioxidant-related proteins nuclear-E2-related factor 2 (Nrf2) and Heme oxygenase-1 (HO-1), it was confirmed that the expression was increased in a concentration-dependent manner. After inducing ROS with lipopolysaccharide (LPS), an intracellular ROS-causing substance, DCF-DA was performed to confirm the inhibitory effect of ROS production, and the inhibition of ROS production was confirmed to concentration-dependent. Real-time RT-PCR was performed to confirm the mRNA expression level of inflammatory cytokines and inflammatory mediator caused by ROS generation, mRNA expression was reduced in a dose dependent manner. Therefore, this study confirmed the antioxidant effect of CSYJE through the Nrf2/HO-1 signaling pathway, which suggests that CSYJE can be used as an antioxidant cosmetic material by inhibiting free radicals.

  • Sasa borealis is a major source of bamboo leaves used for traditional medicine in Korea. Obesity is a serious health problem in industrialized countries that has been implicated in various diseases, including type 2 diabetes, hypertension, cancer, and coronary heart disease. Recent reports have proposed mechanisms to reduce obesity by decreasing preadipocyte differentiation, and proliferation in 3T3-L1 preadipocyte. The preadipocytes play a key role by differentiation into mature adipocytes and increasing fat mass. In this study, we investigated whether ethanol-soluble extracts and ethyl acetate-soluble fractions from Sasa borealis inhibits intracellular accumulation of lipid droplets in a dose-dependent manner in 3T3-L1 cells (an important model system for studying adipogenesis). The down-regulation of PPAR${\gamma}$ and C/EBP${\alpha}$ (key adipogenic transcription factors) were confirmed by the reverse transcription polymerase chain reaction (RT-PCR). Ethyl acetate-soluble fractions from Sasa borealis attenuated the expression of PPAR${\gamma}$ and C/EBP${\alpha}$. These results suggest that Sasa borealis inhibits adipogenic differentiation by regulating adipogenic transcription factors in 3T3-L1 cells. Therefore, Sasa borealis extracts may be a good candidate for the management of obesity.

Blood Pressure Modulating Effects of Black Raspberry Extracts in vitro and in vivo (복분자 추출물의 항고혈압 활성)

  • This study aimed to investigate the effects of 50% ethanol extract of ripe black raspberry (Rubus occidentalis, RBR) on hypertension in human umbilical vein endothelial cells (HUVECs) and in spontaneously hypertensive rats (SHR). Angiotensin converting enzyme (ACE) inhibition and activation of nitric oxide production by endothelial nitric oxide synthase were significantly regulated by RBR in HUVEC cells. Moreover, the SHR showed significantly higher levels of blood pressure, ACE, renin, endothelin-1, and interleukin-6 than Wistar Kyoto rats (WKY). However, treatment with captopril and RBR decreased the levels of these hypertension-related events in the SHR. The renal arteriole showed greater media thickness/lumen diameter (%) in the SHR than in the WKY. However, media thickness/lumen diameter (%) was reduced in SHR by treatment with captopril and RBR. In addition, the number of eosinophilic cardiac muscle cells was decreased in the heart muscles after treatment with captopril and RBR. Therefore, this study suggests that 50% ethanol extract of RBR may be useful for the prevention and treatment of high blood pressure.

The Improving Effect of Paeoniae Radix on Dextran Sulfate Sodium-induced Colitis in Mice (Dextran Sulfate Sodium으로 유도된 궤양성 대장염에 대한 작약의 개선 효과)

  • Paeoniae Radix has been used as a traditional medicine for various diseases including hepatic disease. However, the inhibitory effect of Paeoniae Radix on intestinal inflammation has not been fully understood yet. The aim of this study was to evaluate the effect of Paeoniae Radix on colitis induced by dextran sulfate sodium in mice. To investigate the protective effects of Paeoniae Radix, the colitis mice were induced by drinking water containing 5% dextran sulfate sodium for 7 days. Mice were randomized into groups receiving Paeoniae Radix (100 mg/kg), sulfasalazine (150 mg/kg) as a positive control, or water as a negative control. We evaluated the effects of Paeoniae Radix on clinical signs induced by dextran sulfate sodium, measuring weight loss, colon length, and disease activity index. Additionally, to find a possible explanation for the anti-inflammatory effects of Paeoniae Radix, we evaluated the effects of Paeoniae Radix on the interleukin-6 and cyclooxygenase-2 levels in colitis tissue. The results indicated that mice treated with dextran sulfate sodium showed measurable clinical signs, including weight loss and reduced colon length. However, Paeoniae Radix treatment significantly improved the weight loss and disease activity index as clinical symptoms. Moreover, Paeoniae Radix inhibited the interleukin-6 and cyclooxygenase-2 expression levels in colon tissues treated with dextran sulfate sodium. Taken together, the findings of this study suggest that Paeoniae Radix may be useful for treating intestinal inflammation, including ulcerative colitis.

  • Cognitive impairment and Alzheimer's disease are serious social problems associated with the rising elderly population in Korea. 1,2,3,4,6-Penta-O-galloyl-β-ᴅ-glucopyranose (PGG) is a gallotannin isolated from medicinal plants such as Rhus chinensis. This study was performed to evaluate the effect of PGG on biomarkers related to cognitive function in human neuroblastoma SK-N-SH cells. Inhibition of acetylcholinesterase (AChE) activity is considered to be one of the main therapeutic strategies. PGG inhibited AChE activity in the test tube as well as in SK-N-SH cells. In addition, PGG induced protein and mRNA expression of brain-derived neurotrophic factor (BDNF), which is a mammalian neurotrophin that plays major roles in the development, maintenance, repair, and survival of neuronal populations. As one of the underlying molecular mechanisms that induce BDNF expression, PGG induced the activation of Ca2+/calmodulin (CaM)-dependent protein kinase II (CaMKII)-cAMP response element binding protein (CREB) pathway. In conclusion, PGG may be an useful material for improving cognitive function.

Inhibitory Effects of Illicium verum Hooker fil. Dichloromethane Fractions on Adipocyte Differentiation (팔각회향 dichloromethane 분획물에 의한 지방세포 분화 억제 효과)

  • Fat accumulation in adipocytes occurs through the process of adipogenesis in which preadipocytes differentiate into adipocytes. Obesity is a metabolic disorder caused by excessive accumulation of fat in the body, which increases the incidence of cardiovascular diseases, hypertension, type 2 diabetes, hyperlipidemia, and various cancers. Recently, inhibition of adipocyte differentiation was shown to be a potential antiobesity strategy. In this study, the inhibitory effect of dichloromethane fractions from Illicium verum Hooker fil. water extract on the differentiation of 3T3-L1 preadipocytes to adipocytes was investigated. Dichloromethane fractions from I. verum Hooker fil. significantly inhibited adipocyte differentiation when applied during the adipocyte differentiation process, as assessed by measuring fat accumulation using Oil-red O staining. In addition, dichloromethane fractions from I. verum Hooker fil. reduced important adipogenic transcription factors, such as CCAAT/enhancer binding protein ${\alpha}$ ($C/EBP{\alpha}$), $C/EBP{\beta}$, and peroxisome proliferator activated receptor ${\gamma}$ ($PPAR{\gamma}$). The expression of FAS and LPL, which are terminal differentiation markers of mature adipocytes, was also reduced in the 3T3-L1 adipocytes treated with dichloromethane fractions from I. verum Hooker fil. In addition, the treatment significantly inhibited mitotic clonal expansion, which is essential for adipocyte differentiation, by arresting the G1 phase of the cell cycle. Taken together, these results suggest that dichloromethane fractions from I. verum Hooker fil. may be a natural material with antiobesity effects.

The Effects on Melanogenesis in B16F10 Melanoma Cells and the Anti-inflammatory Activities of an Ethyl Acetate Fraction from Glechoma hederacea var. longituba (긴병꽃풀(Glechoma hederacea var. longituba) ethyl acetate 분획물의 항염증 활성 및 B16F10 세포의 멜라닌 생성에 미치는 영향)

  • This study aimed to confirm the possibility of being used as a cosmetic material material through the verification of the whitening and anti-inflammatory activities of an ethyl acetate fraction from Glechoma hederacea var. longituba (GHEA). The observed electron donating and ABTS+ radical scavenging abilities of GHEA were 89.6% and 88.7%, respectively, at 1,000 ㎍/ml concentration, with a tyrosinase inhibitory effect of 22.3% at the same concentration. For cell viability, a rate of 80% or more was observed in all concentrations that treated GHEA on melanoma and macrophage cells. Protein and mRNA expression inhibition was measured by Western blot and RT-PCR for 25, 50, and 100 ㎍/ ml concentrations, and it was confirmed that expression decreases in a concentration-dependent manner as GHEA concentration increases. The inhibition of the whitening-related factors MITF and TRP-2 were superior following GHEA treatment than those of the control group treated with kojic acid of 100 ㎍/ml concentration. For tyrosinase, the lowest mRNA expression rate was 29.1% at 100 ㎍/ml which confirmed excellent inhibition. In analyzing the effects of pro-inflammatory cytokines IL-1β, IL-6, and TNF-α on protein and mRNA expression, IL-6 and TNF-α showed high protein and mRNA inhibition compared to a vitamin C control group. Based on these experimental results, GHEA could be applied as a natural cosmetic material.

Anti-obesity effects of Glycyrrhiza uralensis ethanol extract on the inhibition of 3T3-L1 adipocyte differentiation in high-fat diet-induced C57BL/6J mice (감초 주정추출물의 3T3-L1 지방세포 분화 억제 및 고지방 식이로 유도된 C57BL/6J 마우스에 대한 항비만 효과)

  • The anti-adipogenic activity of Glycyrrhiza uralensis was investigated by examining the effects of its ethanol extract (GUE) on a mouse model with a high-fat diet (HFD) and 3T3-L1 preadipocytes during adipocyte differentiation. GUE administration for eight weeks significantly reduced weight gain in mice fed an HFD. GUE effectively inhibited 3T3-L1 preadipocyte differentiation and lipid droplet accumulation. This inhibitory effect is associated with the downregulation of key adipogenic regulators, including PPARγ and C/EBPα, and the modulation of adipose metabolism regulators, such as Fasn and Fabp4. LC-Q-TOF-MS analysis identified twelve phenolic and flavonoid compounds, including liquiritigenin and licorice saponin, in the GUE. These findings demonstrate that the anti-obesity effect of the GUE is attributed to the biological activity of its phenolic and flavonoid compounds. Therefore, the GUE has potential anti-obesity activity. Moreover, further studies on the isolation of bioactive components from the GUE and the investigation of the underlying molecular mechanisms of the GUE are required to establish its efficacy in metabolic disorders, including obesity.

Study on the Properties of Lagerstroemia indica Extract as an Anti-acne Cosmetic Material (배롱나무 꽃 추출물의 항여드름 화장품 소재로서의 특성 연구)

  • This study is to propose Lagerstroemia indica flower as an anti-acne cosmetic material, and confirmed the sebum control, anti-inflammatory, and antioxidant effects using Lagerstroemia indica flower extract (LIFE) . As a result of evaluating the sebum control effectiveness, it was confirmed that LIFE inhibited the production of sebum excessively induced with palmitic acid up to 65%. Subsequently, it was confirmed that LIFE has an antibacterial effect and the ability to inhibit lipase activity against Cutibacterium acnes (C. acnes), which mainly appears in acne lesions, and can also reduce the inflammatory response caused by virulence factors secreted by C. acnes. It was also confirmed that LIFE inhibited the secretion of nitrogen monoxide (NO) and prostaglandin E2 (PGE2), which are inflammatory mediators induced by LPS in macrophages, by 75% and 54%, respectively, and that it also had a high DPPH radical scavenging ability similar to that of ascorbic acid. These results suggest that LIFE, a natural extract, can be used as an anti-acne material to relieve and treat acne, a complex disease, by controlling sebum production and having antibacterial and lipase activity inhibiting against C. acnes, and anti-inflammatory, antioxidant properties.
