  • The effects of cultural conditions in the rice straw media for cellulolytic enzymes production by Pleurotus sajor-caju were investigated. The optimum moisture content, pH and temperature for enzymes production were 60%, 7.0 and $35^{\circ}C$ in $C_1-cellulase$, and 60%, 5.0 and $25^{\circ}C$ in $C_x-cellulase$, and 60%, 7.0 and $20^{\circ}C$ in ${\beta}-glucosidase$, respectively. When light was irradiated during the cultivation period, $C_1-cellulase$ and ${\beta}-glucosidase$ production were decreased but $C_x-cellulase$ production increased at $500{\sim}1,000\;lux$. During the cultivation period, $C_1-cellulase$ production was contrary to $C_x-cellulase$ and ${\beta}-glucosidase$. Among the various materials added, rice bran was effective to $C_1-cellulase$ production, cotton seed cake and rice bran to $C_x-cellulase$ production, and defatted soybean and fish meal to ${\beta}-glucosidase$ production. The optimum concentration of rice bran for enzymes production were 20% in $C_{1-}$, $C_x-cellulase$ and 10% in ${\beta}-glucosidase$.

Effects of Dietary Proteins on Serum Insulin-like Growth Factor-I (IGF-I) and IGF-Binding Protein-3 in Korean Rockfish, Sebastes schlegeli (사료의 단백질이 조피볼락 혈액중 Insulin-like growth factor-I (IGF-I) 및 IGF-binding protein-3에 미치는 영향)

  • Insulin-like growth factor-I (IGF-I) is a mitogenic peptide with a molecular mass of 7 kDa. It is produced mainly in the liver and has important functions in the regulation of development and somatic growth. Moreover, Serum IGF-I concentration is regulated by the quantity and the nutritional quality of dietary protein. To determine the IGF-I level in Korean rockfish, Sabastes schlegeli, were fed four experiment diets that contained different protein quantities, namely $30\%,\;40\%,\;50\%\;and\;60\%$ for 70 days. Weight gain of the fish increased depending dietary protein quantity, Also, IGF-I concentrations increased according to dietary protein quantity, Feeding experiments were conducted to examine the effects of dietary protein sources on the serum IGF-I level in Korean rockfish, Fish meal (CO), soybean meal (SM), corn-gluten meal (CGM), meat meal (MM) and feather meal (FM) were used as variable protein sources of the formulated diet. IGF-I concentrations of the CO and MM groups ($277.7\pm23.2,\;291.5\pm41.2\;ng/mL$) were higher than those of the CGM and FM groups ($208.9\pm21.3,\;217.2\pm38.2\;ng/mL$). And IGFBP-3 levels by western blot analysis increased in good protein diets such as in the CO and MM groups. In conclusion, IGF-I may be a sensitive indicator the protein metabolism in fish as well as mammalian.

  • This study was conducted to determine the proper inclusion levels of squid liver powder (SLP) and dehulled soybean meal (DHSM) as a fish meal (FM) replacement in the diets for olive flounders. All the experimental diets were formulated to be isonitrogenous (50% crude protein, CP) and isocaloric (17.1 kJ energy $g^{-1}$ diet). Four diet were formulation of FM with SLP (DHSM) were tested at 0, 25, 50, 75% (SLP(DHSM)$_0$, SLP(DHSM)$_{25}$, SLP(DHSM)$_{50}$ and SLP(DHSM)$_{75}$, respectively). Triplicate groups of 12 fish with an average body weight of 5.56$\pm$0.1 g (mean$\pm$S.D.) were fed one of three experimental diets for 7 weeks. Weight gain (WG) and the specific growth rate(SGR) of fish fed SLP(DHSM)$_0$ and SLP(DHSM)$_{25}$ diets were significantly higher than those of fish fed SLP(DHSM)$_{50}$ and SLP(DHSM)$_{75}$ diets (P<0.05). Also, WG and SGR of fish fed SLP(DHSM)$_{50}$ had significantly higher WG and SGR than that of fish fed SLP(DHSM)$_{75}$ (P<0.05). The feed efficiency (FE) and protein efficiency ratio (PER) of fish fed SLP(DHSM)$_0$ and SLP(DHSM)$_{25}$ were significantly higher than those fed SLP(DHSM)$_{50}$ and SLP(DHSM)$_{75}$ (P<0.05). The whole-body crude lipid content of fish SLP(DHSM)$_{75}$ was significantly higher than that of fish fed SLP(DHSM)$_{50}$ (P<0.05). However, there were no significant differences in whole-body moisture, crude protein and crude ash of fish fed all diets. Based on these growth performance results, the SLP(DHSM) combination replace up to 25% FM protein by the ANOVA test in juvenile olive flounder diets.

Based on the fish growth performance results obtained from this study, the SLP (SPC) premix can substitute up to 25% of the FM in the diets of juvenile olive flounder.

  • This study evaluated fermented soybean meal (FSM) as a fish meal (FM) replacement and determined the appropriate amount of FSM in juvenile olive flounder diet. Twenty-four aquaria with a flowing-water system were stocked with fish averaging 20.9 g at a density of 25 fish/tank. Five experimental diets were prepared replacing FM with 0, 10, 20, 30, or 40% FSM based on FM protein (designated $FSM_{0}$, $FSM_{10}$, $FSM_{20}$ $FSM_{30}$, and $FSM_{40}$, respectively). Two additional diets were prepared that replaced 30 or 40% of the FM with FSM with added amino acids (methionine and lysine) (designated $FSM_{30+AA}$, and $FSM_{40+AA}$, respectively). Fish (triplicates) were fed one of the eight experimental diets (50% crude protein and 16.7 kJ available energy $g^{-1}$ diet) for 8 weeks. Survival did not differ among the treatments during the feeding experiment. There were no significant differences in weight gain (WG) or specific growth rate (SGR) among the fish fed diets with up to 30% of the FM replaced. However, fish fed $FSM_{40}$ or $FSM_{40+AA}$ had a reduced WG and SGR, as compared to $FSM_0$ (control) (P < 0.05). The feed efficiency and apparent digestibility showed a similar trend (P < 0.05). The proximate composition in the whole body of fish differed only between the control and $FSM_{40}$ for the crude protein level and between the control and $FSM_{30+AA}$ for the crude lipid level. The whole-body amino acid composition did not differ among treatments. No significant differences were found between the diet groups with and without amino acid supplementation, indicating that amino acid supplementation had no effect. The major finding of this study is that fermented soybean meal may replace up to 30% of fish meal without amino acid supplementation for normal growth of juvenile olive flounder.

The Effect of Partial Replacement of Fish Meal by Squid Sepia esculenta Liver Powder on the Growth and Body Composition of Juvenile Black Rockfish Sebastes schlegeli (조피볼락(Sebastes schlegeli) 치어 사료에 오징어(Sepia esculenta) 간 분말에 대한 어분대체 효과)

  • An 8-week feeding experiment was conducted to evaluate three types of squid Sepia esculenta liver powder (SLP) as a dietary protein source for replacement of fish meal (FM) in the juvenile black rockfish, Sebastes schlegeli. For replacement of FM, six experimental diets were formulated with 5 or 10% of either of three types (A, B, C) of SLP: SLP-$A_5$, SLP-$A_{10}$, SLP-$B_5$, SLP-$B_{10}$, SLP-$C_5$, and SLP-$C_{10}$. One of the control diets contained 100% FM as the protein source, and the other was a commercial diet (Com). Fish with an average body weight of $6.50{\pm}0.03g$ ($mean{\pm}SD$) were allocated randomly in triplicate groups of 50 to aquaria and fed the experimental diets until satiation. The weight gain (WG) and specific growth rate (SGR) of fish fed the FM, SLP-$A_5$, SLP-$A_{10}$, and SLP-$B_5$ diets were higher than those of fish fed the Com, SLP-$B_{10}$, SLP-$C_5$ and SLP-$C_{10}$ diets. No significant differences were observed in WG and SGR among the diet groups, with the exception of the SLP-$C_{10}$ diet group. The feed efficiency (FE) and protein efficiency ratio (PER) of fish fed the SLP-$A_5$, SLP-$A_{10}$, and SLP-$B_5$ diets did not differ from those fed the control diet. However, fish fed SLP-$B_{10}$ and SLP-$C_{10}$ had lower FE and PER than the fish fed the FM, commercial, SLP-$A_5$, SLP-$A_{10}$, and SLP-$B_5$ diets. Both the SLP-A and SLP-B diets, but not the SLP-C diets, replaced up to 10% and 5% of FM for juvenile black rockfish, respectively. The results of this experiment provide information that will assist in formulating an inexpensive and practical diet containing SLP for juvenile black rockfish.

Effects of Dietary Soybean Meal in Rainbow Trout (Oncorhynchus mykiss) II. On the Apparent Digestibility, Excretion and Body Composition (무지개송어(Oncorhynchus mykiss) 사료원으로써 대두박의 이용 효과 II. 소화율, 배설 및 체조성 변화)

  • A feeding experiment for 160 days was conducted to evaluate effects of dietary soybean meal (SBM) on the apparent digestibility, ammonia excretion and body composition of rainbow trout (Oncorhynchus mykiss). Seven diets containing $0\~70\%$ SBM were formulated based on iso-nitrogenous and iso-caloric basis of $44\%$ crude protein and approximately 4,200 Kcal/kg of gross energy. To evaluate the nutritional utilization of the SBM diets at the end of 60 and 120-day rearing with test diets, digestion rates of protein and lipid of SBM diets were measured. Fish fed the diets containing above $46\%$ SBM showed higher apparent digestibility for protein, but lower for lipid than did fish fed the control diet in both trials. Protein and fat contents in the carcass were similar for all experimental fish, except for fish fed $58\%$ and $70\%$ SBM which showed lower fat content than the others, but composition of fatty acid and amino acids were not affected by dietary SBM levels. Gill and urinary post-prandial ammonia ($NH_3-N$) excretions were measured at 12 and 24 hours after single feeding of the experimental diets. Excretions of $NH_3-N$ measured after 24 hours were almost 1 times higher than those measured after 12 hours. Total excretion fer the 24 hours by fish fed $70\%$ SBM was $15\%$ higher than that of fish fed the control diet. Results of present study may suggest that the rainbow trout which were fed above $34%\;or\;46\%$ of SBM diet showed a decrease gradually in lipid bioavailability compared to the control group.

  • Preliminary creel-holding test of live anchovies which are to be used as bait for the skipjact fishery, was conducted on the fish caught by set net and lift net. They were held in the creels constructed with bamboo frame and minnow netting, during the period of Oct. 24-Nov. 28 in 1972. If was found that the survival rate of the creel-held anchovies varied by the size of fish, the towed distance and speed of fish-carrying creel in which the fish were accomodated after catchi. 1. The survival rate of the medium size anchovies, 7.8-9.6cm in length and 4.6-4.8 g in weight, was $70-92\%$ during the holding period of 20-35 days. Whereas, only $16\%$ of juvenile anchovies, 5.3cm in length and 1.0 g in weight, were survived for 19 hours after catch. 2. During the time of transport, the farther and faster the creel containing fish were towed, the less fish were survived in the subsequent holding period. 3. Though both the set net and lift net could be fairly used to catch live anchovy, no decision between these two gears could be made in this experiment to determine the superiorness for the subsequent survival rate.

Antioxidant and Antibacterial Activities of Lactobacillus-fermented Artemisia annua L. as a Potential Fish Feed Additive (양어 사료첨가제로서의 유산균 발효 개똥쑥의 항산화 및 항균활성)

  • Fermented medical herbs using Lactobacilli have attracted significant attention due to their enhanced biological activities. A traditional medicinal plant, Artemisia annua L., was fermented using a probiotic strain, L. plantarum SK3494. The strain was isolated from Artemisia princeps var. orientalis and molecularly identified through sequence similarities and phylogenetic tree analysis. The antioxidant activity of L. plantarum-fermented A. annua L. (LFA) was determined using the DPPH free radical scavenging assay. Cellular antioxidant activity of LFA was examined using the superoxide radical reduction assay in MAT-C cells. Total polyphenol contents (TPC) and flavonoid contents (TFC) of LFA were determined. The antibacterial activity of LFA against fish pathogens was also determined in this study. The viable cell number (9.38 log10 CFU/ml) and pH (4.1) results showed good adaptive ability of the selected strain during fermentation. LFA was found to have enhanced antioxidant activity compared to non-fermented A. annua L. (NFA) based on the DPPH assay. Cellular antioxidant activity was present in both LFA and NFA. After 24 hr and 48 hr of fermentation, the LFA also showed antibacterial activities against fish pathogens Photobacterium damselae subsp. damselae and Vibrio ichthyoenteri. These results suggest that L. plantarum-fermented A. annua L. may have potential as a feed additive in aquaculture.

Effects of Dietary Soybean Meal in Rainbow Trout (Oncorhynchus mykiss) III. Bioavailability of Phosphors (무지개송어(Oncorhynchus mykiss) 사료원으로써 대두박의 이용 효과 III. 인(P)의 이용성)

  • Feeding experiment was conducted to evaluate effects of dietary soybean meal (SBM) on the phosphorus bioavailability of rainbow trout (Oncorhynchus mykiss). Seven diets, containing 0~70% SBM, were made based on iso-nitrogenous and iso-caloric basis of 44% crude protein. Ajpparent digestibility of phosphorus was measured 60 and 120 days after feeding. The digestibility coefficient increased gradually as the level of SBM increased up to 46~58%, but decreased at 58~70% SBM level. Phosphorus concentrations in feces decreased with an increased in dietary SBM level. Patterns of post-prandial phosphorus excretion were not similar to those of ammonia excretion, but the amount decreased with the increased SBM level in the diets. Total excretion of phosphorus for 24 hours was 19.68mg.kg-1 body wt..day-1 in the control group, but excreted phosphorus by the 58% and 70% SBM groups was 63.4% and 56.1% of the control group, respectively. Phosphorus concentration of whole body decreased with an increase in dietary SBM level for 120 days of feeding. However, there were no significant differences in phosphorus concentrations of liver among the experimental groups, except for 70% SBM diet group.
