Evaluation of Fertility of Artificial Induced Gynogenetic Diploid Male in Paralichthys olivaceus (유도된 자성발생성 2배체 숫컷 넙치(Parlichthys olivaceus)의 생식 능력 평가)

  • To evaluate the reproductive ability of gynogenetic diploid male. Paralichthys olivaceus. histological analysis of testis. cytological analysis of spermatozoa and fertilization test with normal aggs were studied and the results are as follow; The gonads of gynogenetic diploid male were histologically normal. and many spermatozoa were observed in their testis. Number of spermatozoa from the control and gynogenetic diploid male were $2.58\times10^9$ and $2.42\times10^9$ cells per 1 ml of milt. respectively (P> 0.05). Amount of milt per kg body weight from the gynogenetic diploid male was significantly higher (P< 0.01) than that from the control male (8.3ml). Size and morphology from the two experimental groups were not different (P>0.05). More than $80\%$ of fertilization rates and hatching rates were observed when the eggs from the control were fertilized with the gynogenetic diploid male sperms.

Water Quality and Cyanobacterial Anatoxin-a Concentration in Daechung Reservoir (대청호의 수질과 남조류 독소 Anatoxin-A 농도의 관계)

  • The current study was performed to elucidate the relationship between the anatoxin -a produced by cyanobacteria and aquatic environmental factors, Algal and water samples were collected from the Daechung Reservoir from June to November 2001. The physical factors of the water quality were measured in sifu, while the biological and chemical factors were examined in the laboratory. The concentrations of anatoxin-a in the algal and water samples were analyzed by HPLC using a fluorescence detector, and ranged from $0.61-8.68\;{\mu}g/g$ dw in the algal samples and $0.01-0.08\;{\mu}g/L$ in the water samples. The suggested maximum concentration of anatoxin-a for safe drinking water is $1\;{\mu}g/L$. The concentrations of anatoxin-a in the algal and water samples were highest in July. The relationships between tile aquatic environmental factors and the anatoxin-a concentration were also analyzed to identify the crucial elements for toxin production. The anatoxin-a concentrations in the algal samples exhibited a high correlation with nitrate, the TN/TP ratio, TDN (P<0.05), and TPN/TPP ratio (P<0.01), whereas the anatoxin-a concentrations in the water samples were highly related to the water temperature, conductivity (P<0.01) , pH, phycocyanin, and phycocyanin/chlorophyll a ratio (P<0.05).

  • Our previous research demonstrated the essential role of the xenB gene in stress response to RDX by using Pseudomonas sp. HK-6 xenB knockout. We have extended this work to examine the cellular responses and altered proteomic profiles of the HK-6 xenA knockout mutant under RDX stress. The xenA mutant degraded RDX about 2-fold more slowly and its growth and survival rates were several-fold lower than the wild-type HK-6 strain. SEM revealed more severe morphological damages on the surface of the xenA mutant cells under RDX stress. The wild-type cells expressed proportionally-increased two stress shock proteins, DnaK and GroEL from the initial incubation time point or the relatively low RDX concentrations, but slightly less expressed at prolonged incubation period or higher RDX. However the xenA mutant did not produced DnaK and GroEL as RDX concentrations were gradually increased. The wild-type cells well maintained transcription levels of dnaA and groEL under increased RDX stress while those in the xenA mutant were decreased and eventually disappeared. The altered proteome profiles of xenA mutant cells under RDX stress also observed so that the 27 down-regulated plus the 3 up-regulated expression proteins were detected in 2-DE PAGE. These all results indicated that the intact xenA gene is necessary for maintaining cell integrity under the xenobiotic stress as well as performing an efficient RDX degradation process.

  • As a result of population growth and economic growth, household and industrial wastes continue to rapidly increase every year. Especially, sewage sludge produced at final stage is increasing with the constant construction and putting in good order of the sewage plant. In addition to the government's prohibition for filling up the sludge, it became more and more difficult to discharge wastes to the sea as London Dumping Convention '96 came into effect. And sewage sludge and the livestock wastes are expected to be thoroughly prohibited from discharging to the sea from 2012. So we need desperately economical and useful alternatives to compact and reuse these wastes. The purpose of this study is to evaluate the utilization of solidified sludge-soil mixture as an enhancement and covering material. To determine the proper mixed ratio of solidified sludge, this study conducted basic physical properties tests, compaction tests, uniaxial compression tests, and permeability test. It was found that the higher the ratio of solidified sludge, the lower the coefficient of permeability. Upon the results of particle size distribution, the mixed ratio of solidified sludge that meet the enhancement material condition was 59% or lower for SP granite soil and 48% or lower for SM granite soil respectively.

  • The blower erosion phenomenon was investigated to develop a long-life blower with a volume flow rate of 10,000 ㎥/min with the required total pressure efficiency of 83% or more. The blower performance and blower erosion were predicted through numerical analysis by computational fluid dynamics(CFD). The conditions used for numerical analysis were an air volume of 16,200 ㎥/min, a rotation speed of 893 rpm, and a temperature of 330℃. The specific gravity, particle size, and amount of the dust was 3.15, 90 ㎛~212 ㎛, and is 265 kg/min, respectively. To examine the effects of a dust deflector on erosion, erosion analysis was performed by comparing the models with and without a dust deflector. Numerical analysis showed that when the dust deflector is installed, the average tended to decrease by 167% in the impeller and 133% in the boss. CFD using the Finne's model for erosion revealed a parallel restitution coefficient of 1 and a perpendicular restitution coefficient of 0.1. The blower performance of case 5 was 691.7 mmAq, and the efficiency was 83.3% when the rotation speed and the air volume flow rate were 880 rpm and 16,200 ㎥/min, respectively.

The Effects of Live Yeast(Saaccharomyces cerevisiae) Supplementation on the Performance of Laying Hens (활성효모 첨가가 산란계의 생산성에 미치는 영향)

  • In order to study the effects of supplementation of live yeast(Saccharomyces cerevisiae) on the performance of laying hens, five experiments were conducted. Two experiment were conducted during summer period, one with 37 wk old Dekalb-Delta strain laying birds(Exp. 1) and the other one with 100 wk old molted Nick Chick Brown laying birds(Esp.2) . In each experiment, 240 birds were divided into 12 groups of 20 birds each and randomly distributed. Each of the two experimental diets(Control 71 and 0.05% live yeast supplemented 72) was fed to 6 groups for 4 wks in Exp.1 and 3 wks in Exp.2. Three experiments were conducted during winter period, Exp.2 with 54 wk old Hy-Line strain laying birds, Exp.4 with 52 wk old Hy-Line strain laying birds, and Exp.5 with 36 wk old broiler breeder(Indian River strain). In each experiment, 540 birds were divided into 18 groups of 30 birds each and randomly distributed. Each of the 3 experimental diets(Control:$T_1$0.05% live yeast supplemented:$T_2$ and 0.1% live yeast supplemented : T$_3$) was fed to 9 groups for 6 wks in Exp.3, 9 wks in Exp.4 and 4 wks in Exp.5. In Exp.4, Latin Square design was employed to determine the effects of switching feeds at 3 wk intervals. All hens were housed in cages of commercial farm and experimental diets were made with commercial layer feeds. In Experiment 1, egg production was significantly(P<0.05) higher in $T_2$. Feed intake was significantly (P<higher in 72 at 1st wk but 4 wk average was not significantly different. Feed efficiency was significantly(P<0.01) better in 72 at End wk but 4 wk average was not significantly different. Other parameters, such as weight, soft egg production, cracked egg production and mortality were not significantly different. In Experiment 2, egg production was significantly(P<0.05) higher in $T_2$. Feed efficiency was significantly (P<0.05 and P<0.01) better at End wk and 3rd wk but 3 wk average was not significantly different. Soft egg production was significantly(P<0.05) higher in 72. Other parameters were not significantly different. In Experiment 3, egg productions were significantly(P<0.05) different among treatments : $T_3$ was higher than $T_1$ and $T_2$ was higher than $T_1$. Egg weight of $T_1$ and $T_2$was significantly(P<0.05) heavier than $T_3$. Feed intake of $T_2$ and $T_3$ was significantly(P<0.05) higher than $T_1$ at 6th wk but overall average was not significantly different. Soft egg production were significantly(P<0.01) different among treatments:$T_1$ was higher than $T_3$ was higher than $T_2$. Feed efficiency cracked e99 Production and mortality were not signifcantly different. In Experiment 4, egg production tended to increase as the level of live yeast supplementation increased but they were not statistically different. In Experiment 5, egg production of broiler breeders of $T_3$ was significantly(P<0.01) higher than $T_1$. Feed intake of $T_3$ was significantly(P<0.05) greater than $T_1$ and $T_2$ at 3rd wk but overall average was not significantly different. Fertility and hatchability tended to be higher in the supplemented groups than in the control.

A study of the arch length discrepancy and the diagnostic analysis (치열궁내 공간 부조화와 진단적 평가에 관한 연구)

  • Predicting the arch length discrepancy by simply comparing the available arch perimeter with tooth materials is merely a 2-dimensional analysis of the teeth movement. However, the real teeth movement takes place 3-dimensionally and is affected by various factors such as, the arch fen the curve of Spee and the axis of the incisors. The purpose of this study is to clarify the relationship between the decrease in the arch perimeter and the horizontal positional change of the incisors after extraction of the 1st bicuspids, for more analytic evaluation of the arch length discrepancy at pre-treatment model analysis stage. In addition to that to evaluate the effect of the curve of Spee, teeth axis to the basal plane, and the incisional crowding to the treatment outcome. All patients were treated at the department of orthodontics, dental hospital, Yonsei university. Inclusion criteria for patients selection were as follows. $\cdot$ Angle classification I malocclusion with bialveolar protrusion $\cdot$ Extraction of 4 1st bicuspids $\cdot$ No tooth anomaly or prosthesis $\cdot$ No abnormal attrition $\cdot$ No ectopically erupted teeth $\cdot$ Angle classification I canine and molar relationship $\cdot$ Less than 3mm of crowding Model analysis of the above patients was performed and the following conclusions were obtained. 1. When the intercanine distance was maintained, the available space for the distal movement of the mandibular incisors after the extraction of the 4 1st bicuspids was larger than the space provided by the extraction of the 4 1st bicuspids. However the difference was less than 1mm. The more tapered the anterior arch form, the larger the difference. 2. Compared to the situation in which the intercanine distance was maintained, when the intercanine distance was expanded to meet the width of the Posterior teeth, the incisors could move about 3mm more distally. 3. The positional difference of the incisal tip was insignificant whether the central incisors were moved by tipping or bodily movement. 4. When the anterior crowding was solved without changing the intercanine distance, the larger the anterior arch length was, the more the anterior movement of the incisors. 5. When the curve of Spee was levelled, the increase in the arch perimeter was less than half of the deepest curve of Spee.

Large scale splitter-less FFD-SPLITT fractionation: effect of flow rate and channel thickness on fractionation efficiency (대용량 중력장 SPLITT Fractionation: 분획효율에 미치는 채널 두께와 유속의 영향)

  • SPLITT fractionation (SF) allows continuous (and thus a preparative scale) separation of micronsized particles into two size fractions ('fraction-a' and 'fraction-b'). SF is usually carried out in a thin rectangular channel with two inlets and two outlets, which is equipped with flow stream splitters at the inlet and the outlet of the channel, respectively. A new large scale splitter-less gravitational SF (GSF) system had been assembled, which was designed to eliminate the flow stream splitters and thus is operated by the full feed depletion (FFD) mode (FFD-GSF). In the FFD mode, there is only one inlet through which the sample is fed. There is no carrier liquid fed into the channel, and thus prevents the sample dilution. The effects of the sample-feeding flow rate, the channel thickness on the fractionation efficiency (FE, number % of particles that have the size predicted by theory) of FFD-GSF was investigated using industrial polyurethane (PU) latex beads. The carrier liquid was water containing 0.1% FL-70 (particle dispersing agent) and 0.02% sodium azide (used as bactericide). The sample loading rate was varied from about 4 to 7 L/hr with the sample concentration fixed at 0.01%. The GSF channel thickness was varied from 900 to $1300{\mu}m$. Particles exiting the GSF channel were collected and monitored by optical microscopy (OM). Sample recovery was monitored by collecting the fractionated particles on a $0.45{\mu}m$ membrane filter. It was found that FE of fraction-a was increased as the channel thickness increases, and FE of fraction-b was increased as the flow rate was increased. In all cases, the sample recovery has higher than 95%. It seems the new splitter-less FFD GSF system could become a useful tool for large scale separations of various types of micron-sized particles.

Photosynthesis and Formation of UV-absorbing Substances in Antarctic Macroalgae Under Different Levels of UV-B Radiation (중파자외선에 대한 남극산 해조류의 광합성 및 자외선 흡수물질 형성 반응)

  • Effects of artificial and solar W-B radiation on five rhodophytes (Curdiea racovitzae, Gigaytina skottsbergii, Mazzaella obovata, Myriogramme manginii, Palmaria decipiens) from Antarctica have been investigated using PAM fluorescence in laboratory and in the field. Laboratory studies showed that there were significant differences in the UV sensitivity between different species, and that the differences appeared to be correlated with the depth of collection of the specimens. It was apparent from the observations that the samples such as M. manginii and P. decipiens collected from 20-30 m depths were move sensitive to W-B radiation compared with those collected from shallower depths, The present study confirmed that an acclimation to the surrounding light regime could be an important factor to determine the UV-sensitivity of a species or individuals and that PAM measurements are rapid and non-destructive methods to evaluate UV influences. From field studies on M. manginii and P. decipiens it was observed that both plants exhibited changes in the effective quantum yield, with the minimum values nt noon followed by n recovery in the evening. Photoinhibition occurred in these species could therefore be accounted for by so- called dynamic photoinhibition. It seems likely that this protective mechanism may contribute to survival of the species in shallow water where they may encounter intense solar radiation. The presence or absence of the W- B component under solar radiation differently affected the photosynthetic recovery process, and the rate of recovery was much stoney in UV- present than in W- absent conditions. Functional role of W- B appears to delay the recovery of photosynthesis in the studied macroalgae. Differential sensitivity to UV-B recognised between M. manginii and P. decipiens seemed to correspond well with the amount of UV-absorbing substances (UVAS) contained in the respective species. Higher tolerance to solar radiation by the latter species may be due to the higher amount of UVAS. There were variations of UVAS concentrations in algal thalli depending on the season and depth of collection.

A Study on Long-term Variations of BOD and COD as Indicators of Organic Matter Pollution in the Han River (한강 본류에서 유기물 오염도 지표인 BOD와 COD에 대한 장기변동 특성)

  • This study was performed to investigate the degree of long-term pollution at the mainstream of the Han River by comparing the concentration of BOD and COD from 1975 to 2011. The long-term annual average BOD and COD concentration at the mainstream of the Han River showed an increasing trend as it flowed downstream from Paldang Dam to Gayang. The concentration of BOD ($r^2$=0.646) and COD ($r^2$=0.260) showed a consistent decreasing trend for 37 years. In the case of Paldang Dam, BOD has maintained a decreasing trend, whereas the COD value showed an increasing trend after the 1990s. Therefore, a control of non-biodegradable materials in areas around Paldang Dam is required. The result of the seasonal variations of BOD and COD is as follows: spring>winter>summer and fall (p<0.001). The time series analysis revealed a strong correlation for every 12-month period. Also, the amount of water discharge at Paldang Dam has to be systematically controlled because the amount of water discharge from the dam influences the water quality at the mainstream of the Han River.
