Investigation of Microbial Contamination the Level in Fish Farms of Jeju East Coast. (제주 동부 연안 넙치 양식장의 미생물학적 오염도 조사)

  • Level of microbial contamination was examined in four fish farms of Jeju east coast for sanitary indication of bacterial contamination such as heterotrophic bacteria, Escherichia coli O157:H7, Salmonella spp., Vibrio parahaemolyticus, Staphylococcus aureus, Bacillus cereus and total coliforms. The samples were collected and investigated from June to October in 2007. Bacterial species of S. aureus, B. cereus and Salmonella spp. were frequently detected from the seawater of above fish farms. Salmonella spp., S. aureus, B. cereus were detected in the range of $0{\sim}4.3{\times}l0^3,\;0{\sim}2.7{\times}l0^4,\;0{\sim}7.6{\times}l0^3$ CFU/ml, respectively, from the fish feed used in all four selected farms. Additionally, Salmonella spp., S. aureus, B. cereus were detected in oliver flounder (Paralichthys olivaceus) of these farms, however total coliforms and E. coli O157:H7 were not detected during our experimental period. For the production of microbiologically safe and healthy oliver flounder, proper quality control of feed, sanitation programs, and continuous monitoring of microorganism are essential practices, which required to include in the farm management system.

Effect of Dietary Supplementation of Fermented Mealworm on the Growth of Juvenile Stone Flounder (Kareius bicoloratus) (사료내 갈색거저리 유산균 발효물 첨가의 돌가자미 치어 성장 효과)

  • In this study, we aimed to develop a feed additive that can promote the growth of stone flounder (Kareius bicoloratus). To this end, we fermented mealworm medium (5%, w/v) with lactic acid bacteria (LAB) with antibacterial function (HeTm_LpWpPa) and evaluated its efficacy as a feed additive. Fish (approximately 5-6 g) were reared in six completely recirculating tanks (100 fish/tank) for 6 weeks. The total length of fish with and without HeTm_LpWpPa supplementation was 8.26 and 8.42 cm and body weight was 19.71 and 20.28 g, respectively, at 6 weeks. The weight gain of the control and experimental groups was 223.26 % and 248.76 %, respectively. The specific growth rate at 6 weeks was 2.79 % (control) and 2.98% (experimental), and the growth rate of the experimental group was slightly higher (0.19 %) than that of the control group. At the end of the experiment (i.e. 6 weeks), the number of fish below the mean weight in the experimental group was 3 (1 %) and that in the control group was 17 (5.7 %). These results indicate that feed supplementation of 10 % HeTm_LpWpPa could be utilized to promote the growth of flounder in farming.

Drug-resistant bacteria isolated from Nakdong river and Carp-ponds (낙동강 잉어와 양식잉어의 장내약제내성균)

  • The intestinal drug-resistant bacteria of carps in three recirculating system ponds in Pusan from May to September 1981 were isolated. Both fishes and waters, sampled from the fish farm in National Fisheries University of Pusan and Nakdong River, were tested in 1982. As results of both experiments, although frequent occurrence of drug-resistant bacteria was low in comparison with the foreign countries, it showed incresing tendency in this experiments, and level of the drug-resistant tolerance was incresing gradually. To decrease drug-resistant bacteria of culturing waters and fishes in our country, it is nessesary that, we should diagnose diseases actually and cure them with a right prescription and dosage. .At the end, it is the best way not to abuse drugs fundamentally.

The Growing and Spawning of tile Catfish, Clarias batrachus in the Aquarium (수조내에서의 열대산 메기, Clarias batrachus의 사육과 산난부화)

  • Fry of a Siamic catfish Clarias batrachus was brought to Korea on August 14,1979 and exper-imentally reared for a whole life cycle in a small water recycling aquarium. After 29 days since start of feeding the fry were measured 5 cm in 1ength and 1.43 g in body weight and after 260 days they were measured 205.7 g average. Chorionic gonadotrophin (2 IU) was injected to a healthy female (224 g) and after 63 hours fertilized eggs were obtained in a small water recycling aquarium. About 1,300 fry were hatched out 20 hours after spawning at $30^{\circ}C$ constant water temperature. The fry began to feed on water fleas 53 hours after hatching.   • Overinflation of cardiac stomach occurred in farmed sterlet sturgeon (Acipenser ruthenus) fingerlings. Affected fish congregated and swam upside-down near the edges of the tanks with abdominal distension. When pressed the dilated abdomen, the fish often exhibited a normal swimming pattern for some time, then resumed their characteristic upside-down swimming patterns. Internally, the fish showed abdominal dropsy and dilated stomach filled with yellowish granular contents. When the contents in the stomach, fish feed, water and excrement in the farm inoculated on Sabouraud destrose agar (SDA) at $25^{\circ}C$ for 48 hr. Candida sp. were isolated except from the feed. Candida sp. in excrement or water in the farm may enter into the stomach with feed and then produce gases in the metabolic process, which in turn may cause the stomach overinflation.

Application of a Pond System to Korea for Treatment and Recycling of Wastewater (하수 처리 및 재활용 연못시스템의 국내 응용)

  • The applicability to Korea is examined of a pond system which treats and recycles wastewaters. Air temperature and solar radiation of the pond systems at Corinne, Utah, and Eudora, Kansas, which are located in temperate regions of the U.S., are compared with those of Kimpo lying in the mid-western part of Korea. Analyzed are also $BOD_5$ and SS concentrations, algal concentrations, pH levels, and water temperature of the two systems. Air temperature of Kimpo is quite similar to that of the two systems, and solar radiation of Kimpo is more conducive than that of the systems to the growth of algae during summer. Analysis of $BOD_5$ and SS concentrations in the final effluent of the systems shows that they meet the secondary treatment standards. The study demonstrates that wastewater treatment pond system which is similar in design to the systems can be reliably utilized at Kimpo, Korea. A model is proposed which can integrate a pond system with aquaculture and agriculture.

Removal of Ammonia-N by Immobilized Nitrifier Consortium (고정화된 질화 세균군에 의한 암모니아성 질소 제거)

  • Nitrifier consortium immobilized in Ca and Ba-alginate beads were packed into two bioreactors and the performances of bioreactors were evaluated for the removal of ammonia nitrogen from synthetic aquaculture water. The total ammonia nitrogen (TAN) concentration of the influent was continually kept about 2g TAN/㎥. At the HRT of 0.6hr, ammonia nitrogen removal rate of two bioreactors were about 52.6 and 51.0g TAN/$\textrm{m}^3$/day, respectively. At the respect of ammonia nitrogen removal, two bioreactor showed the similar abilities. The second trial with nitrifier consortium immobilized in Ca-alginate bead was carried out to evaluate the ammonia nitrogen removal rate for 35 days. The highest ammonia nitrogen removal rate was 82g TAN/$\textrm{m}^3$ when HRT was about 0.3hr.

  • A new bacterial infection occurred among the cultured Korean catfish, Silurus asotus, in Chunbuk prefecture, Korea, 1993. This disease produced about 30% mortality in the fish for 4 months. The diseased fish was swimming listless at the water surface with head up and tail down, sometimes spinnig in circles. The most outstanding clinical sign was ulceration on the skull and at the base of the pectoral fins. The causative organism was isolated from the brain, kidney, spleen and liver of diseased fish, and identified as Edwardsiella icaluri by the biochemical and biophysical characteristics. After intraperitoneal innoculaton of the isolate, the pathogenicity was proved positive for Korean catfish, S. asotus, and channel catfish, Ictalurus punctatus, but negative for carp, Cyprinus carpio.

Effect of Dietary Lipids on Growth on Nile Tilapia, Oreochromis niloticus (지질 종류에 따른 나일틸라피아의 성장 효과)

  • Experiments were conducted to find out suitable dietary lipids in the practical feed for Nile tilapia, Oreochromis nilotius. Soybean oil, corn oil, fish oil or cuttlefish liver oil was mixed in the diet and tested. The best weight gain and feed efficiency were obtained from the fish fed soybean oil or corn oil, both rich in 18 : 2n-6. But the growth rate of fish fed diet with fish oil or cuttlefish liver oil were inferior. The levels of 20 : 3n-9 in the total lipid were not different in all tested lipids, but that in the polar lipid of the fish oil or cuttlefish liver oil were lower than that in the soybean oil or corn oil. Thus, vegetable oil such as soybean oil or corn oil, rich in 18 : 2 n-6, were suitable as dietary lipid for Nile tilapia. Unlike other fish species, fish oil or cuttlefish liver oil showed no essential dietary value for this species.

Treatment of Recycling Wastewater of Aquaculture Using DynaSand Filter( I ) (상향류식 연속역세여과를 이용한 양어장 순환수 재이용(I))

  • To improve the efficiency of biological filters used for treating recycled wastewater of aquaculture, the conventional and advanced DynaSand Filters were tested. Applying the fluid mechanics and physical chemistry theories to wastewater treatment plant, the advanced filter focuses on detection of wastewater characteristics and automatic operation of DynaSand Filter to improve backwashing, oxygen transport, turbidity removal and continuous head loss. The Filter removes COD, ammonia, and suspended solids (SS), the major pollutants in aquaculture wastewater. The total volume of the DynaSand Filter is 70 L, while the working volume is 35 L per cycle. The removal efficiency of the advanced DynaSand Filter is 71 % for SS (as against 55 % for the conventional filter), 84 % for ammonia, 85 % for T-P, and 88 % for SRP.
