Effect of Plants Extracts on Lipid Peroxidation of Rat Brain Tissue Induced by Reactive Oxygen Species (활성산소에 의해 유도된 흰쥐 뇌조직의 지질산화에 대한 식물체 추출물의 효과)

  • Abilities of various edible plants and natural antioxidants to protect brain against oxidative damages were evaluated using brain homogenate of perfused Sprague-Dawley rat. Oxidative damage, expressed as lipid peroxidation (LPO), indicating total quantity of malondialdehyde and 4-hydroxyalkenal, increased from 4.1 to 6.9nmol/mg protein by treatment of $2.5{\mu}M$ ferrous sulfate and 7.5mM hydrogen peroxide as source of reactive oxygen species (ROS) on brain homogenate for 10min at $37^{\circ}C$ Mallow(88%) in leafy vegetables, small potato (93%) in root vegetables, green red pepper (76%) in fruit vegetables, and avocado (96%) in fruits showed highest LPO inhibition capacities. Ability of mushrooms decreased in order of nameko, shiitake, pine mushroom, oyster mushroom, and new type pine mushroom. Among natural antioxidants tested, (+)catechin (91%), (-)epigallocatechin gallate (85%), (-)epicatechin gallate (83%), and kaempferol(83%) showed high LPO inhibition capacities.

Quality Characteristics of Cookies Prepared with Button Mushroom(Agaricus bisporous) Powder (양송이버섯을 첨가한 쿠키의 품질 특성)

  • This study examined the effects of adding four different amounts (0%: control, 4%: M-4, 8%: M-8, 12%: M-12) of button mushroom (Agaricus bisporous) powder on the quality characteristics of cookies. The bulk density and pH of the dough, and firmness, color, spread factor, consumer acceptability of cookies, and total phenol compound content and free radical scavenging activity of mushroom powder and cookies were measured. While the bulk density and pH of the dough as well as spread factor of the cookies significantly decreased, the firmness, L values and total phenol compound contents and free radical scavenging activity of the cookies significantly increased with increasing mushroom powder content(p<.05). The consumer acceptability scores for the button mushroom cookie groups ranked significantly(p<.05) higher than those of the control group in general acceptability, appearance, flavor, color, taste, and texture. This study suggests that button mushroom powder is a good ingredient for increasing the acceptability and functionality of cookies.

Effect of Letinus edodes, Agaricus bisporus and Pear Powder on Tenderization of Beef Eye of Round (표고버섯, 양송이버섯, 배를 첨가한 홍두깨살의 연육 효과)

  • The purpose of this study was to investigate the effects of protease in Letinus edodes, Agaricus bisporus and Pear powder after freeze drying which has the ratio of 3% on the Tenderness of the Beef Eye of Round muscle. It were marinated in distilled water (Control), 3% Letinus edodes powder (L3), 3% Agaricus bisporus powder (A3), and 3% Pear powder (P3). As a result, enzyme activities were highest in Agaricus bisporus (p<0.001). There are significant difference in pH (p<0.001), color of the beef were slightly different between the C (control) group and the sample groups. The cooking loss showed the lowest value in the control and the highest value in the water holding capacity of Agaricus bisporus. In addition, Agaricus bisporus showed the lowest shear force values than the other sample groups(p<0.001).

Optimal height of installing sticky traps for monitoring fungus gnats (버섯파리류 예찰을 위한 끈끈이트랩의 최적 설치위치)

  • Agricultural sticky traps are used for forecasting flying insects, including fungus gnat adults. Forecasting using sticky trap is an important measure to determine pesticides or environmentally friendly control means. Sticky traps of various colors can be installed easily and quickly. The yellow sticky trap is most effective for forecasting fungus gnat adults. In addition, they are used for control purposes by mass trapping. We tested the optimum installation height of yellow sticky traps to forecast and control the fungus gnat adults effectively in the cultivation of mushroom and horticultural plants. The number of captured fungus gnat adults was highest on the second floor for button mushroom and oyster mushroom, 100 cm above the floor for sawdust shiitake mushroom, 20 cm under the floor for hydroponic strawberry, and 20 cm above the floor for hydroponic tomato. This suggests that sticky trap height should be installed differently depending on the cultivation type or environment for effective forecasting and control of fungus gnat adults.

Development of the Waxed Rice Straw Pulp Tray Reduced in Moisture Absorption (흡습억제기능을 보유한 왁스처리 볏짚 트레이의 개발)

  • The purpose of this study was to development the waxed rice straw pulp tray(WRSPT) which had low water sensitivity. The quality changes of mushrooms(Agaricus bisporus) packaged respectively in the WRSPT, non-treated rice straw pulp tray(NRSPT) and expanded polystyrene tray(EPST) were observed during storage. The weight losses of mushrooms in the WRSPTs were less than in the NRSPTs by maintaining below 3% at $0^{\circ}C$ and the applications of WRSPTs were more effective at $20^{\circ}C$. The weight increases of WRSPTs were more lowered than those of NRSPTs by marking below 15% during storage at $0^{\circ}C$. There were no significant differences in the Hunter L values and hardness values of mushrooms in the three types of trays at $0^{\circ}C$. The cap openings of mushrooms were more proceeded in NRSPTs than in WRSPTs and EPSTs at $20^{\circ}C$. As a result, it may be favorable to control excessive moisture absorption of the NRSPT mainly composed of cellulose pulp by introducing such chemical treatments as wax treatment.

Browning end Color Characteristics in Mushrooms (Agaricus bisporus) As Influenced by ionizing Energy (버섯의 갈변 및 색도에 대한 전리에너지의 영향)

  • Browning and color characteristics of stored mushrooms (Agaricus bisporus)following ionizing irradiation were investigated in connection with quality deterioration. The phenolic compounds of stored mushrooms showed a gradual decreasing tendency, while extractable browning pigments apparently rose from around 3days of storage under the conditions of $9{\pm}1^{\circ}C,\;80{\pm}7%$ RH and packaging with a corrugated paper box wrapped up in PE. ${\gamma}-irradiation$ at 2 to 3 kGy resulted in a significant reduction of their changes. Immediately after treatment, irradiated mushrooms were more discolored, i.e. a lower Hunter L value and higher Hunter a and b values than control. However, the subsequent storage for 15 days resulted in a preventive influence of ionizing energy on mushroom discoloration. This beneficial effect of ionizing energy was somewhat higher in the pilei than in the stipes of mushrooms and was found to increase lineally with increasing doses up to 3 kGy.

Isolation and characterization of Bacillus subtilis NO12 from button mushroom substrates (양송이 배지로부터 분리된 Bacillus subtilis NO12의 특성)

  • Twelve strains of bacteria with cellulase and xylanase activities were isolated from spent mushroom substrates collected from button mushroom cultivation farm, Buye, Chungcheongnam-do in Korea. Among them, one strain, designated NO12, with higher cellulase and xylanase activities was selected by agar diffusion method. The strain NO12 was identified to be a Bacillus sp. by biochemical characteristics using Bacillus ID kit and MicroLog system. Comparative 16S rDNA gene sequence analysis showed that strain NO12 formed a distinct phylogenetic tree within the genus Bacillus and was most closely related to Bacillus subtilis with 16S rDNA gene sequence similarity of 99.2%. Based on its physiological properties, biochemical characteristics, and phylogenetic distinctiveness, strain NO12 was classified within the genus Bacillus, for which the name Bacillus subtilis NO12 was proposed. The cellulase and xylanase activities of B. subtilis NO12 were slightly increased according to bacterial population from exponential phase to stationary phase in the growth curve for B. subtilis NO12. The xylanase activity continuously increased from the beginning of the exponential phase and exhibited maximum activity in the middle of the exponential phase.

A Study on the properties of mushroom compost produced by different methods for the development of a self-propelled compost turner (자주식(自走式)퇴비교반기 개발을 위한 제조방법별 양송이 퇴비배지의 특성에 관한 연구)

  • A self-propelled turner was developed to produce mushroom compost at low costs and high efficiency, and the uniformity of compost produced by an excavator, compost mixer, and self-propelled turner was compared. The material of the compost was mixed with rice straw and chicken manure at a dry weight ratio of 3:1. After the final turning, internal temperature distribution, water content of the compost pile, ash ratio, and uniformity of the compost pile were examined. After the compost was completed, the water content was $69.9{\pm}0.54%$, $72.1{\pm}0.15%$, and $74.5{\pm}0.82%$, respectively. The length of rice straw was $47.5{\pm}15.4cm$, $24.9{\pm}10.1cm$, and $31.0{\pm}10.6cm$, respectively. The ash content in the dry weight of each compost was $25.0{\pm}6.2%$, $33.6{\pm}4.2%$, and $28.2{\pm}1.1%$, respectively. The deviation in the length of rice straw was affected by the linear velocity of the spinner in the compost mixer and the self-propelled turner, which were 21.5 m/sec and 9 m/sec, respectively. As a result, the most uniform mushroom compost was produced by the self-propelled turner.

Plant growth promotion effect of Arthrobacter enclensis Yangsong-1 isolated from a button mushroom bed (양송이배지로부터 분리한 Arthrobacter enclensis Yangsong-1의 식물생장촉진효과)

  • An auxin-producing bacterium Yangsong-1 was isolated from a button mushroom bed in Chung cheongnam-do. The strain Yangsong-1 was classified as a novel strain of Arthrobacter enclensis based on a chemotaxonomic and phylogenetic analysis. The isolated A. enclensis Yangsong-1 was confirmed to produce indole-3-acetic acid (IAA), which is one of the auxin hormones. When the concentration of IAA was assessed by HPLC quantity analysis, the maximum concentration of IAA, $152.903mg\;L^{-1}$, was detected from the culture broth incubated in R2A medium containing 0.2% L-tryptophan for 48 h at $35^{\circ}C$. A negative relationship between IAA production and pH was estimated to show that the increase in IAA caused pH acidification of the culture. The effect of the supplement on L-tryptophan, a known precursor of IAA production, appeared to be at maximal production at 0.2% concentration and was rather reduced at concentration above 0.4%. To investigate the growth-promoting effects on the crops, the culture broth of A. enclensis Yangsong-1 was placed in water cultures and seed pots of mung beans and lettuce. In consequence, the adventitious root induction and root growth of mung beans and lettuce were 1.5 and 1.9 times higher, respectively, than those of the control.

Field research of cultivation technique for stable production of common mushroom(Agaricus bisporus) (양송이 안정생산을 위한 생산기술 현장연구)

  • Common mushroom production per area has been decreased and are up to less than 50% of the 1980 production. To determine the main reasons for the decrement, we performed this study. Two main reasons, which are mushroom disease and the low compost quality because of mechanized compost making, were assessed. In mechanized mushroom farms, nitrogen concentration in compost was lower than recommended and total compost quantity was about 100-150 $kg/3.3m^2$, which was also lower than usual. Our study revealed that higher nitrogen concentration (about 1.5%) in compost gave better production. Also, use of large amount of compost appeared to increase the mushroom production, although more insects and disease problems were observed. The relationships between the presences of microorganisms and occurrence of diseases were assessed by monitoring the microorganism densities near the mushroom farms. Higher number of microorganisms were observed near the mushroom farm area, compared to control region, Daechon beach. Most contaminating molds were found in the circulating fans, tunnel and culture room floor. The bacterial isolates were collected from the air in mushroom culture room and killed with 0.005% Benzalkonium solution, indicating treatment of Benzalkonium are the effective methods to sanitize the mushroom culture room.
