Development of "Bt-Plus" Biopesticide Using Entomopathogenic Bacterial (Xenorhabdus nematophila, Photorhabdus temperata ssp. temperata) Metabolites (곤충병원세균(Xenorhabdus nematophila, Photorhabdus temperata ssp. temperata)의 대사물질을 이용한 "비티플러스" 생물농약 개발)

  • Bacillus thuringiensis (Bt) is a bacterial biopesticide against insect pests, mainly lepidopterans. Spodoptera exigua and Plutella xylostella exhibit significant decreases in Bt susceptibility in late larval instars. To enhance Bt pathogenicity, we used a mixture treatment of Bt and other bacterial metabolites which possessed significant immunosuppressive activities. Mixtures of Bt with culture broths of Xenorhabdus nematophila (Xn) or Photorhabdus temperata ssp. temperata (Ptt) significantly enhanced the Bt pathogenicity against late larval instars. Different ratios of Bt to bacterial culture broth had significant pathogenicities against last instar P. xylostella and S. exigua. Five compounds identified from the bacterial culture broth also enhanced Bt pathogenicity. After determining the optimal ratios, the mixture was applied to cabbage infested by late instar P. xylostella or S. exigua in greenhouse conditions. A mixture of Bt and Xn culture broth killed 100% of both insect pests when it was sprayed twice, while Bt alone killed less than 80% or 60% of P. xylostella and S. exigua, respectively. Other Bt mixtures, including Ptt culture broth or bacterial metabolites, also significantly increased pathogenicity in the semi-field assays. These results demonstrated that the Bt mixtures collectively names "Bt-Plus" can be developed into potent biopesticides to increase the efficacy of Bt.

Enhanced Pathogenicity of Bacillus thuringiensis Mixed with a Culture Broth of an Entomopathogenic Bacterium, Xenorhabdus sp. (제노랍두스 곤충병원세균 배양액의 비티 미생물 약제 약효증진 효과)

  • The entomopathogenic bacterium, $Xenorhabdus$ sp., was isolated from an entomopathogenic nematode, $Steinernema$ $monticolum$. When these bacteria were injected into the hemocoel of the diamondback moth, $Plutella$ $xylostella$, they caused significant mortality. However, the bacterium was not pathogenic when it was administered orally. This study showed that $Xenorhabdus$ sp. significantly enhanced oral pathogenicity of $Bacillus$ $thuringiensis$ (Bt) against the last instar larvae of $P.$ $xylostella$. Different ratios of culture broth of $Xenorhabdus$ sp. and Bt showed significantly different pathogenicities against $P.$ $xylostella$. In field tests, the optimal bacterial mixture significantly enhanced control efficacy against $P.$ $xylostella$ compared to Bt treatment alone. These results demonstrated that $Xenorhabdus$ sp. culture broth can be developed as a potent biopesticide by enhancing the insecticidal efficacy of Bt.

Control System of Whitefly, Trialeuodes vaporariorum, in Cucumber by the Alternate Application of Insecticides within Each Conventional Group (오이에서 살충제 계열내 교호처리에 의한 온실가루이 방제 체계)

  • In order to establish a whitefly control system using conventional groups of insecticide(carbamate, organophosphorus and pyrethroid insecticides), three alternative application methods were designed on the medium growth stage of cucumber. To discriminate the effectiveness of these sequences observed were the residual activity of insecticides, frequency of insecticide applications, residue of insecticides in cucumber leaves, development of insecticide resistance in whitefly, and yield of fruits. Spraying furathiocarb, a carbamate insecticide, was very effective in reducing the frequency of application for the control of white flies. The effectiveness of furathiocarb was enhanced by the potentiation process to carbofuran, the long residual activity, and the lower development rate of insecticide resistance. Methion, an organophosphorus insecticide, did not show resistance development after successive use but resulted in short residual activity. However, other organophosphates, profenofos and phenthoate, lost their activity by the resistance development. Decreasing activity was common to pyrethroids, deltamethrin and zetacypermethrin due to resistance. From these results it could be drawn a conclusion that furathiocarb, a carbamate insecticide, was the most desirable among conventional insecticide groups for the management of greenhouse whitefly population on the cucumber. To prevent an outbreak of the insect pest by various cause, it was recommended to choose acetamiprid, a nicotinoid, which showed very good control efficacy to the resistance insects to conventional insecticides.

Drug Adsorption Behavior of Polyolefin Infusion Tube Compared to PVC and PU (Non-PVC(폴리올레핀) 수액용 튜브 내면에서의 약물흡착 거동 - PVC 및 PU 수액튜브와의 비교)

  • PVC (polyvinyl chloride) intravenous fluid bags and tubes that contain DEHP (diethylhexyl phthalate) as a plasticizer have several associated disadvantages for intravenous injections. We investigated the drug absorption behaviors on the inner surface of an infusion tube that consisted of commercialized PVC/PU (polyurethane). We developed a non-PVC (polyolefin) tube in order to improve the efficacy of this drug administration method. We prepared four types of non-PVC (polyolefin) infusion tubes using a polyethylene (PE), polypropylene (PP), syndiotactic 1,2-polybutadiene (PB), and styrene-ethylene (SE) copolymer elastomers were prepared using a single screw extruder. The four types of manufactured non-PVC (polyolefin) infusion tubes had good mechanical properties that were equivalent to PVC tube properties. The four types of prepared non-PVC (polyolefin) infusion tubes also prohibited drug absorption when compared to the commercialized PVC and PU tubes. Therefore, based on the results of this study, prepared non-PVC (polyolefin) tubes are good candidates for infusion because they prevent drug absorption and the release of DEHP.

The Contents of Chlorogenic acid and Eleutheroside E in Eleutherococcus senticosus (Rupr. et Maxim.) Harms (가시오갈피나무의 Eleutheroside E 및 Chlorogenic acid 성분함량(成分含量))

  • Eleutherococcus senticosus (Rupr. et Maxim.) Harms is a tree species that has been traditionally used as a source for oriental folk medicine. It is known to contain both Eeleutheroside E and chlorogenic acid. These two compounds are believed to be effective elements in the tree. In the present study, we analyzed the levels of the two compounds from the plants collected from various locations in Korea to find out whether environment affected the quantity of both compounds in the tissue. When the plants growing in their natural habitats and those growing in plantations were compared, the level of the two compounds showed different pattern. In the case of chlorogenic acid in roots, the plants growing in plantations are found to contain higher levels than did those from natural habitats. On the other hand, higher levels of eleutheroside E were found in the plants from natural habitats. However, when the compounds in the stems were compared, the plants growing in the natural habitats contained higher level in both compounds than did those from plantations. Although the level of both compounds in the leaf tissue varied with the places from which they were originated, it appeared to be less than that found in roots or stems. There exists a tendency that the level of eleutheroside E showed a positive correlation with the diameter of both roots and stems of the plants.

Chemical control of Sycamore Lace Bug, Corythucha ciliata(Say) (버즘나무방페벌레의 약제방제(藥劑防除)에 관(關)한 연구(硏究))

  • For the control of Corythucha ciliata, the insecticidal activity of five commercial pesticides (Monocrotophos 25%SL, Phosphamidon 50%SL, Imidacloprid 20%DC, Thiamethoxam 15%DC and Acetamiprid 20%SL) were examined with trunk injection method in Seoul, Sanbon and Chungju. The insecticidal activity of five commercial pesticides was >88%. The insecticidal activity of Monocrotophos maintained 31 days later after trunk injection when treated leaves were supplied to this pest. This result indicates that one application of trunk injection is enough to control this pest, and this method can save the control cost compared with ground application which should be applied two or three times per year to control this pest. To make the model of treatment dosage at each D.B.H class (cm), $1.0m{\ell}/DBH$, $1.5m{\ell}/DBH$ and $2.0m{\ell}/DBH$ pesticide dosage was used. At <20cm, the insecticidal activity of $1.0m{\ell}$ dosage was >95%, and $1.5-2.5m{\ell}$ dosage was needed for the effective control at 30~50cm. More than $2.5m{\ell}$ dosage was effective at >50cm. Spray method was used in laboratory to select pesticides for ground application. Three commercial pesticides (Ethofenprox 20%EC, Ethofenprox 10%WP and Cyfluthrin 1%EC) were used, and all pesticides showed >95% insecticidal activity.

Selection of Systemic Chemicals and Attractiveness of Sunflower to Ricania spp.(Hemiptera: Ricaniidae) Adults (갈색날개매미충 성충에 대한 해바라기의 유인력과 침투이행성 약제 선발)

  • Sunflower, selected as a trap plant that can be controlled by attracting Ricania spp. adults via attraction has the highest attractiveness during the preoviposition period. Considering the ecological characteristics of Ricania spp., adults are distinguished by the preoviposition and oviposition periods and the attractiveness of sunflower to Ricania spp. adults was 91.4~95.2% higher than that of blueberry during the preoviposition period. On August 20, when Ricania spp. adults entered the oviposition season, sunflower attractiveness was low at 9.8~11.6% owing to preference for tree species. Based on the result of the selection of systemic chemicals that could be used concomitantly with sunflower, all chemicals, except etofenprox, showed a high controlling effect of over 90%, and among them, dinotefuran showed the highest insecticidal rate of 95.8%. The systemic chemicals acetamiprid, dinotefuran, thiamethoxam, and imidacloprid persisted for 13 days (survey period). Therefore, the concomitant use of sunflower and systemic chemicals can reduce the density of Ricania spp. entering farmlands and their populations in surrounding habitats, which are expected to help in stabilizing the ecosystem.

Chemical Control of Popillia quadriguttata(Coleoptera: Scarabaeidae) and Damage of Zoysia matrella and Newly Recorded Host Plants in Golf Courses (녹색콩풍뎅이(Popillia quadriguttata)의 화학적 방제와 골프장의 금잔디(Zoysia matrella) 및 새로운 기주의 피해)

  • Popillia quqdriguttata is a serious insect pest in Korean golf courses. The adult feeds on leaves of latifoliate trees and herbaceous plants while the larva feeds on roots of turfgrass. Thus, control of p. quadriguttata adults was tried with registered insecticides in golf courses in Korea. The insecticides used in the experiments were carbaryl + phosalone WP, chlorpyrifos + diflubenzuron WP, chlorpyrifos-methyl EC, deltamethrin EC, etofenprox WP, etofenprox + diazinon WP, etofenprox + tebufenozide EC, fenitrothion EC, imidacloprid SC, pyraclofos WP, and tralomethrin EC. Mortalities of P. quadriguttata were over 95% in carbaryl + phosalone WP, chlorpyrifos + diflubenzuron WP, etofenprox + diazinon WP, and fenitrothion EC at petri dish and in greenhouse. In the field test, corrected mortality of P. quadriguttata was 95.2% and 80.9% in carbaryl 40% +phosalone 20% WP and fenitrothion 50% EC, respectively. Spreading agent enhanced toxic effect of used insecticides to p. quadriguttata adults except carbaryl + phosalon 20% EC. p. quadriguttata adult was firstly observed to feed on leaves of Canna generalis at the Yongwon golf club in Jinhae, Gyeongnam and Prunus ameriaca var. ansu and p. salicina at the Anyang Honest golf club in Gunpo, Cyeonggi. Therefure, host plants off quadriguftata became 29 species of 26 genera in 19 families. Zoysia matreila at the fairway and roughs of Anyang Benest golf club was damaged by p. quadriguttata larvae in Octomber, 2002 and not recovered in April, 2003. Damaged turfgrass became yellowish and wilted and that retarded recovering in the following spring.

Study of Antioxidation Action of Lenonuri herba Extract (익모초 추출물의 항산화 작용에 관한 연구)

  • Motherwort (Leonurus sibiricus L), a hemp nettle (Labiatae) plant, grows about 1.5${\~}$2 m high spontaneously to wildness in Korea, China, Japan, and other Asian place. Its medical applications includes women's uterine disease, urination, bloody stool, bloody urine, and hemorrhoids. It's also effective for high blood pressure, heart stimulation, and anti-cancer activity. We first prepared chlroform and methanol extracts of motherwort and then they were fractionated using water, $30\%$ methanol, $60\%$ methanol, and $100\%$ methanol, respectively. Each fractionates is assayed for free radical scavenging activities against DPPH and anti-oxidant activity by TBARS assay measuring lipid peroxidation using LDL. The $30\%$ and $60\%$ methanol fractionates of methanol extracts showed strong anti-oxidant activity compared to vitamin C. They also had more Potent SOD activity using pyrogallol at 250 ppm than that of vitamin C. These results suggest that anti-oxidant activities of motherwort may be applicable to development of natural anti-oxidant cosmetics. Possibility of nature anti-oxidation ability cosmetics is seen possibility low official.

Pharmacological Actions of New Woohwangchungsimwon Pill on Cardiovascular System (신우황청심원의 심혈관계에 대한 약효)

  • In order to investigate the pharmacological properties of New Woohwangehungsimwon Pill (NWCH). Effects of Woohwangehungsimwon Pill (WCH) and NWCH were compared using various experimental models. In isolated rat aorta, NWCH and WCH showed the relaxation of blood vessels in maximum contractile response to phenylephrine ($10^{-6}$M) without regard to endothelium containing or denuded rings of the rat aorta. Furthermore, the presence of the inhibitors of NO synthase and guanylate cyclase did not affect significantly the relaxative effects of NWCH and WCH. NWCH and WCH inhibited the vascular contractions induced by acethylcholine, prostaglandin endoperoxide or peroxide in a dose-dependent manner. In conscious spontaneously hypertensive rats(SHRs), NWCH and WCH decreased significantly heart rate. These, at high doses, had a negative inotropic effect that was a decrease of LVDP and (-dp/dt)/(+dp/dt) in the isolated perfused rat hearts, and also decreased the contractile force and heart rate in the isolated rat right atria. In excised guinea-pig papillary muscle, these had no effects on parameters of action potential at low doses, whereas inhibited the cardiac, contractility at high doses. Furthermore, these had a significant inhibitory effects on heart acceleration in normotensive rats and SHRs. These results suggested that NWCH and WCH have weak cardiovascular effects, and that there is no significant differences between two preparations.
