A study on preference for complementary and alternative therapy using Q method (Q방법론을 활용한 보완대체요법 선호에 관한 연구)

  • Today modern people have been searching on managing themselves in easy ways without side-effects to lead a healthy and quality life, accordingly concerns about complementary and alternative medicine have been increasing. The scientific term' complementary and alternative medicine' has been originally used in the US and has become the general term for natural treatment of medical methods, which can be suggested as an alternative solution to modern medical problems. There should be a study based on users' subjectivity to reinvigorate complementary and alternative medicine more than it does now. The purpose of this study is to offer basic materials when assessing practical applications on complementary and alternative medicine using the Q method which is subjective pattern analysis, help understand features of complementary and alternative therapy according to patterns, and find out preferences for patterns of complementary and alternative therapy. As a result patterns of complementary and alternative medicine were classified into three types. Firstly, type I prefers to biological/herbal treatment. Secondly, type II prefers to mixed treatments. Thirdly, type III prefers to manual healing.

Anti-inflammatory and Anti-allergic Effects of Herbal Extracts on Atopic Dermatitis(Part I) (약용식물추출물의 아토피성피부염에 대한 항염증 및 항알레르기효과(제 1보))

  • Atopic dermatitis is a chronic inflammatory skin disease associated with cutaneous hyper-reactivity to environmental triggers. In order to develop effective therapeutic herbal extracts for atopic dermatitis, cytotoxicity, antioxidant, anti-inflammatory and anti-allergic activities were investigated for various herbal extracts. Among candidate extracts, we selected Aloe vera L. (AV), Viola mandshurica W. Becker (VM), Punica granatum L. (PG), Dendrobium nobile L. (DN) and mixture of the above extracts (MX) for further investigations. All of them did not show cytotoxic activities to macrophage RAW264.7 cells below the concentration of 100 ppm. All showed antioxidant, anti-inflammatory and anti-allergic effects, although to various extents. In antioxidant effects, AV showed the highest effect, followed by PG and VM, while DN did the lowest. In evaluation for anti-inflammatory activities in macrophage RAW264.7 cells, AV and DN inhibited almost completely the LPS-induced production of nitric oxide, while AV, DN and VM showed strong inhibitory activities on the LPS-induced production of TNF-${\alpha}$. In anti-allergy effect in mast cell HMC-1, DN showed the highest effect, followed by AV and PG, while VM did the lowest. In the topical allergy reaction induced by compound 48/80 in Sprague-Dawley rat, DN exhibited significant anti-allergic effect, while PG, VM and AV did slight effect. These results suggest that AV, VM, PG and DN have antioxidant, anti-inflammatory and anti-allergic activities, and thus have the potential to reduce and alleviate the symptoms of atopic dermatitis.

Screening of Korean Herbal Medicines with Inhibitory Activity on Advanced Glycation End Products (AGEs) Formation (III) (한국약용식물의 최종당화산물 생성저해활성 검색 (III))

  • Enhanced formation and accumulation of advanced glycation end products (AGEs) have been implicated as a major pathogenesis process leading to diabetic complications, normal aging, atherosclerosis and Alzheimer's disease. In our ongoing project to discover novel treatments for diabetic complications from natural sources, we have investigated on the inhibitory activity of 67 ethanol extracts from 57 Korean herbal medicines against the formation of AGEs in vitro. Of these, 22 extracts were found to have a significant AGEs inhibitory activity ($IC_{50}$<50 ${\mu}g$/ml) compared with aminoguanidine ($IC_{50}$=75.98 ${\mu}g$/ml). Particularly, 6 extracts from 3 herbal medicines, Castanea crenata (flower, leaf, bark-twig), Acer tatarium subsp. ginnala (fruit) and Sapium japonicum (leaf, twig) showed (approximately 8-17 fold) stronger inhibitory activity than that of aminoguanidine.

The Effects on Antimicrobial and Anticarcinogenic Activity of Momordica Charantia L. (메탄올로 추출한 여주 분획성분의 항균 및 항발암 효과)

  • This study was performed to determine the antimicrobial and anticarcinogenic activities of the Momordica charantia L. (MC) on several microorganisms and human cancer cell lines. In the paper disk test, its antimicrobial activity was increased in proportion to its concentration. Among the various solvent fractions of Momordica charantia L., the ethylether partition layer (MCMEE) showed the strongest antimicrobial activity. Also, the ethylacetate partition layer (MCMEA) and the butanol partition layer (MCMB) showed antimicrobial activity. We also determined the cytotoxicity and chemopreventive effect of Momordica charantia L. extract and fractions on human cancer cells. The experiment was conducted to determine the cytotoxicity of Momordica charantia L. partition layers on HepG2, HeLa and MCF-7 cells by MTT assay. Among the various partition layers of Momordica charantia L., MCMEE and MCMEA showed strong cytotoxic effects on all cancer cell lines. The chemopreventive effect of the quinone reductase induced activities of HepG2 cell, the hexane partition layer (MCMH) at a dose of 50 $\mu\textrm{g}$/mL was 3.62 times more effective compared with the control values of 1.0. Therefore, based on these studies, Momordica charantia L. may be developed into a potentially useful cancer chemopreventive agent.

Antioxidant Properties in Water and 70% Ethanol Extracts of Houttuynia Cordata Thunb (어성초의 추출물에 따른 항산화력 연구)

  • Houttuynia cordata Thunb is a famous traditional Chinese medicine. Also it has rich in functional and bioactive substances with outstanding immunopotentiation, anti-cancer, antimicrobial, and anti-aging effects. This study was evaluated water and 70% ethanol extracts of Houttuynia cordata Thunb using in vitro system antioxidant properties methods. The scavenging DPPH and ABTS radicals of 2 mg/mL 70% ethanol extract were 91.8 and 54.8%, respectively. Also, The FRAP assay of this showed increase in activity. In results, the antioxidant properties of 70% ethanol extract increased significantly compared to water extract(p < 0.05). Therefore, 70% ethanol extract of Houttuynia cordata Thunb may contribute to antioxidant property.

Development of Functional Misutkaru Drink Added with Several Medicinal Herbs Extracts (약용식물추출물을 첨가한 건강미숫가루음료 개발)

  • In order to develop of health-oriented convenience foods for rice consumption, functional Misutkaru drink was made by adding several medicinal herbs extracts to fundamental Misutkaru based cereal powder(rice, barley and soybean). Fundamental Misutkaru drink was prepared with the ratio of cereal mixed rice(5g), barley(5g) and soybean(5g) on water (100 mL) and several medicinal herbs extracts. Liriopis Tuber(50%), Omija(15%), Ginseng(10%) and Jujube(25%) were mixed to fundamental Misutkaru as functional Misutkaru. And then it was extracted to added water of 10times during 36hrs at 75$^{\circ}C$. For instant Misutkaru drink of health-oriented convenience foods, mixture of them was made with fundamental Misutkaru drink(80%), extract of several medicinal herbs(20%), and it's sensory score was high. Autoclaved functional Misutkaru drink for long storage period was changed rheological type badly. Overall acceptability and storage period of nonautoclaved functional Misutkaru drink to added extract of several medicinal herbs was higher than fundamental Misutkaru drink.

Antioxidative effects of traditional medicinal plants on lipid peroxidation (지질 과산화에 대한 전통약용 식물의 항산화 효과)

  • To assess the antioxidative activity of 12 medicinal plants on lipid peroxidation, twelves traditional medicinal plants extracted with 95% methanol were investigated the antioxidative activity using DPPH, thiocyanate acid method, and thiobarbituric acid (TBA) methods. Out of 12 medicinal plants extracted with methanol, the extraction yields of Sedum kamtschaticum was the highest values (49.46%) among them and Geranicum sibiricum, Saururus chinensis root (R), Agrimonia pilosa leaf (L), Agrimonia pilosa root was the lowest value (9.97%). Radical scavenging effect of the selected traditional medicinal plants extracted from different extract solution were examined by 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical method. Antioxidative activity of methanolic extracts was higher than those of ethanol and n-hexane extracts. Scavenging effects in Sedum kamtaschaticum (R) determined by DPPH radical showed the highest among the 12 plants. The antioxidative effects of the first four medicinal plants were similar to those of butylated hydroxyanisole (BHA), and butylated hydroxytoluene (BHT), but higher than that of tocopherol, which was used as a handled control. Antioxidative effects of each indicated concentration of the methanolic extracts on linoleic acid by thiocyanate method was the highest in Sedum kamtschaticum and followed by Geum japonicum and Agrimonia pilosa and their antioxidative effect were similar to those of BHA, and BHT, but higher than that of tocopherol. Antioxidative effects of the selected medicinal methanolic extract on linoleic acid by thiocyanate acid method were examined for 15 days. Peroxidation of control and tocopherol group occurred on days 5 and 9, respectively, but BHA, BHT, selected medicinal methanolic extract group did not occur until on day 15. Antioxidative effects of the selected medicinal methanolic extract on linoleic acid by TBA method were examined for 15 days. Antioxidative activity was similar to those obtained by thiocyanate acid method.

Antioxidant and Antihemolytic Activity of Ethanol Extracts of Rubus coreanus Miquel (복분자 에탄올 추출물의 항산화 및 항용혈 효능)

  • To develop a natural antioxidant and anti-hemolytic agents, we investigated the effects of ethanol extracts of Rubus coreanus Miquel (crude extracts). Crude extract was extracted with ethanol. Antioxidant activity of crude extracts was evaluated by employing two different assays, 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical scavenging activity and super oxide dismutase(SOD)-like activity. Also, anti-hemolytic activity of crude extract was determined using $H_2O_2$-induced hemolysis in normal rat red blood cells (RBC) or plasma. The extracts obtained from crude extract dose-dependently increased the scavenging activity on DPPH-induced radicals and SOD-like activity. RBC oxidative hemolysis induced by $H_2O_2$ was significantly suppressed by the extracts of Rubus coreanus Miquel in a dose-dependent manner. These results suggest that crude extracts may have value as the potential antioxidant and anti-hemolytic medicinal plant.

Evaluation of Physicochemical and Fermentation Qualities of Moru Wines Supplemented with Pine Needles or Medicinal Herbs (약용 식물을 첨가한 머루주의 이화학적 특성과 발효 품질 평가)

  • For the development of Moru wine with enhanced sensory qualities, Moru (Vitis amurensis) wines were fermented with pine needles from Pinus densiflora Siebold et Zuccarini to make Moru-pine wine or medicinal herbs from Astragali Radix and Viscum album to make Moru-herb wine. Moru without pine needles or medicinal herbs was included as a control. Pine needles and Astragali Radix/Viscum album delayed the fermentation of Moru wine, but after 40 days of fermentation and aging, final ethanol contents, pH, acidity, and the sugar/organic acid content in these kinds of Moru wines had no differences. The final ethanol level and acidity of Moru wines were 11.5~12.9% and 1.1~1.2%, respectively, but any sugars (glucose, fructose and sucrose) were not detected in all Moru wines. These results are consistent with the general characteristics of Moru wines, which have a high acidity and low sugar contents. Relatively low browness and antocyanins were detected in Moru-herb wine decreasing its chromaticity in a sensory test. Overall, the supplements of Astragali Radix/Viscum album reduced the color of normal Moru wine, which might be applied toward the development of conventional Moru wines.

Screening of Natural Antioxidant from Plant and Their Antioxidative Effect (식물성 천연 항산화물질의 검색과 그 항산화력 비교)

  • Certain parts of 95 species of edible and medical plants were extracted with water and 75% of ethyl alcohol. After addition of those extracts to palm oil, lard and soybean oil at different level, their antioxidative activities were compared by Rancimat test. Six species among them seemed to have rather strong antioxidative activity and high extracting yields(i.e. Taraxacum platycarpum, Plantago asiatica, Rhus javanica L., Lycopus lucidus, Astragalus membranaceus, Taraxacum platycarpumH). Among them, the Rhus javanica L. ethanol extract retarded greatly the induction period of palm oil and lard. When 600 ppm of Rhus javanica L. extract were added to palm oil and lard, AI(antioxidant index was expressed as induction period of oil containing various plant extracts/induction period of control oil) of each was 1.35 and 3.03 respectively. This result indicated that the Rhus javanica L. extract was more effective on lard than the other oils.
