Hypoglycemic Effect of Smallanthus sonchifolius (Yacon) Extracts on Animals with Streptozotocin-induced Diabetes (야콘 추출물이 Streptozotocin으로 유도된 당뇨 동물모델에서 혈당강하에 미치는 효과)

  • This study was performed to investigate improvements in diabetes mellitus by extracts of yacon in streptozotocin (STZ)-induced diabetic rats. Fifty rats were divided into a normal group and four experimental groups. STZ (45 mg/kg) was injected intraperitoneally to induce type I diabetes in the four experimental groups. Yacon extracts were administered for 5 weeks. Forty-five ICR mice were also divided into one positive control group and four experimental groups for the oral glucose tolerance test (OGTT) after fed yacon extract. The control group did not eat any yacon extracts, while Group 1 (GI) was fed 125 mg/kg of yacon extracts, Group 2 (GII) was fed 250 mg/kg of yacon extracts, and Group 3 (GIII) was fed 500 mg/kg of yacon extracts. After treatment for 5 weeks, blood glucose in GIII group showed decreased tendency at the 5 week. In OGTT by glucose, the glucose level of yacon treatment group in diabetic rats was significantly decreased compared to the glucose level of the control group, but there was no difference in OGTT by maltose. In ICR mice, the glucose level of the experimental group in OGTT by maltose was significantly decreased compared to the control group. The area of the islets of Langerhans was increased by yacon treatment in a dose-dependent manner on diabetic rats. Insulin concentration of the GIII group was also decreased compared to the control group, while the concentration of fructosamine, total cholesterol, and triglycerides in serum showed no difference. OGTT by glucose or maltose in ICR mice or diabetic rats, area of the Islets of Langerhans, and insulin concentration improved. Yacon treatment may be a useful therapeutic and preventive strategy for diabetes mellitus.

Occurrence of Gray Mold on Yacon Caused by Botrytis cinerea (Botrytis cinerea에 의한 야콘 잿빛곰팡이병의 발생)

  • The gray mold disease occurred on tuberous roots of yacon in storage facilities in Gangneung, Korea, in March 2010. Symptoms typically appeared as in the form of dark brown discoloration on the surface of tuberous roots and water-soaked brown lesions in cross sections of the affected portions. A total of five isolates of Botrytis sp. were obtained from the symptomatic portions. All isolates on potato-dextrose agar (PDA) produced abundant conidia which were pale brown, one-celled, mostly ellipsoid or ovoid in shape and $8.2{\sim}14.8{\times}6.5{\sim}9.9\;{\mu}m$ in size. Large numbers of round to irregular, smooth, black, hard sclerotia were produced on PDA over time. The optimal temperature for mycelial growth and sclerotia formation of the fungal isolates was $20^{\circ}C$. On the basis of morphological and cultural characteristics, all the fungal isolates were identified as Botrytis cinerea. Pathogenicity test on host plants showed that the fungus could infect not only tuberous roots but also leaves and petioles of yacon. This is the first report on gray mold of yacon (Smallanthus sonchifolius) caused by Botrytis cinerea in Korea.

Effect of Yacon (Smallanthus sonchifolius H. Robinson) Extracts on Herbicidal, Fungicidal, and Insecticidal Activities (야콘(Smallanthus sonchifolius H. Robinson) 추출물의 제초, 살균 및 살충활성 효과)

  • This research has been carried out to find out the highest effect on insecticidal, fungicidal and herbicidal activities in leaves, stem and tuber extracts of yacon (Smallanthus sonchifolius) and extraction methods such as water, boiling water and methanol. Characteristics of potential herbicidal components among extraction methods were investigated by solvent fractions such as hexane, chloroform, ethyl acetate, butanol, and water. Generally, methanol extract was the best on inhibition of germination rate, plant height, and root length in cucumber and barley. On the other hand, the inhibition effect on growth in cucumber and barley was the best in tuber among plant parts of yacon. Inhibition of germination rate, plant height, and root length in cucumber and barley in solvent fractions was the best in water fraction, but there were no differences in other fractions. Digitaria sanguinalis L. and Solanum nigrum L. by 5 and 10% extractions of yacon tuber were controlled by more than 70~80% and 95~100%, respectively. However, there was no inhibition effect on foliar treatment in cucumber and barley as affected by 5 and 10% extractions of yacon tuber. Mortality of green peach aphid (Myzus persicae Sulzer) was 50% at 3 days after treatment of 5% extracts of yacon leaves. Mortality of brown plant hopper (Nilaparvata lugens Stal) was only 24% in 5% extracts of stems and leaves with midrib, but was 57% in 5% extracts of leaves without midrib. There was no fungicidal effect on anthracnose (Colletotrichum truncatum), wilt disease (Fusarium oxysporum), verticillium wilt (Verticillium dahliae), bacterial blight (Xanthomonus oryzae) in 5% extracts of yacon leaves.

Effect of Transplanting Date on Growth and Yield of Yacon (정식시기가 야콘의 생육 및 수량에 미치는 영향)

  • Fifty-day-old plug seedlings of yacon (Polymnia sonchiflolia Poeppig & Endlicher) were transplanted on Mar. 23, Apr,8, Apr,23, May 8, May 23, and June 7,2002 at Jeju to evaluate the effect of transplanting date on the growth and yield of yacon in Jeju island. Delaying the transplanting until May 23 greatly decreased plant height stem height, main stem diameter and number of nodes per plant. The number of marketable tuberous roots was about 6.0 per plant with the early transplanting from March 23 to April 231 but decreased to 1.9 per plant with the latest transplanting time, June 7. The yield of marketable tuberous roots of early transplanted (March 23 to April 23) yacon was about 27 MT/ha, and was 32.4 MT/ha at April 23 transplanting, but was remarkably reduced at the three later transplantings, May 8, May 23, and June 7. Dry matter and soluble solid contents of marketable tuberous roots were 14% and 15%, respectively, regardless of transplanting date. The weight of a marketable tuberous root ranged from 153 to 183.9 g at the first three transplantings, but gyeatly reduced with delaying transplanting until May 7. The optimum transplanting date for yacon in Jeju island seems to be from mid-April to late April considering tuberous root yields, the risk of late-spring frost, and cost of seedling raising.

Callus and Micro-Crown Bud Formation in Vitro from Leaf Explant of Yacon (Polymnia sonchifolia Poeppig & Endlicher) (야콘 (Polymnia sonchifolia Poeppig & Endlicher) 잎의 절편체로부터 캘러스 및 기내 소관아 형성)

  • The explants of yacon (Polymnia sonchifolia Poeppig & Endlicher) were cultured to invest th8e dedifferentiation condition, and formative callus from leaf was cultured to find the regeneration and micro-crown bud formation. Basal MS medium was more effective to form callus than 1/2 MS and B$_{5}$ medium. Calli formations from leaf, petiole and lateral bud were more effective on MS medium supplemented with 1.0, 2.0 mg/L 2,4-D and 0.2, 0.4 mg/L kinetin or BA than 1.0, 2.0 mg/L NAA and 0.2, 0.4 mg/L kinetin or BA. Formative callus from leaf was proliferated about 70% on medium supplemented with 1.0 mg/L BA. When callus was proliferated, 63% regeneration rate was shown on medium supplemented with 1.0, 2.0 mg/L BA in case of subculture for 3~4 months but was not shown on medium supplemented with 1.0, 2.0 mg/L kinetin. Micro-crown bud formed as addition of BA at 3~4 months after callus culture and then was obtained many at 5~6 months, it was most formed about 82% on medium supplemented with 5 mg/L BA. Rate of micro-crown bud formation was increased as more over 5 mg/L BA concentration, when this time, however, shoot had thick leaves and short internodes, and then withered before long, Micro-crown bud was formed about 88.0% on medium supplemented with 5% sucrose, that was more increased 28% than with 3% sucrose. The buds of crown bud between harvested in field and formed in vitro were difference only in size, but both were similar in shape according to histological view.
