Biological Activity of Polyphenol Group Fraction from Korean Pear Peel (한국산 배과피 폴리페놀 분획군의 생리활성효과)

  • Biological activities and anticarcinogenicity of Korean Pear peel were investigated, Electron donating activity and superoxide dismutase (SOD)-like activity of fraction II, III were up to 80% and 50-60% at 50 ppm, respectively, Inhibitory effects on xanthine oxidase were about 80% at 50 ppm, breast adenocarcinoma was about 60% at 2,000 ppm, higher III than II Inhibitory effect on prostate adenocarcinoma was about 23% at 500 ppm. In conclusion, Korean pear peel was expected to use as a functional material.

Effects of Thiosulfinates Isolated from Allium tuberosum L. on the Growth of Human Cancer Cells (부추의 함황화합물이 인체 암세포 증식에 미치는 영향)

  • To develop Allium tuberosum L. as a cancer preventive food material, thiosulfinates and biological active components were isolated from Allium tuberosum L. and the apoptotic effects of thiosulfinates in human cancer cells were examined. Thiosulfinates decreased viable cell numbers in dose- and time-dependent manners. Thiosulfinates at the 20 $\mu g$/mL concentration inhibited more than 60% cell proliferation in HepG2 and A549 human cancer cells, respectively. Also the morphology of cells treated with thiosulfinates of 30 $\mu g$/mL concentration was distorted with shrunken cell mass while the cell number was lower than that of control cells. The $IC_{50}$ values in the HepG2 cells were higher than those of the A549 cells. Thiosulfinates at the 30 $\mu g$/mL concentration showed the formation of apoptotic bodies and a nuclear condensation, and an increase in the cell populations of the sub-G1 phase in the HepG2 cells. These results indicate that thiosulfinates from Allium tuberosum L. inhibited cell proliferation in HepG2 via apoptosis.

Induction of Apoptosis by Combination Treatment with Luteolin and TRAIL in T24 Human Bladder Cancer Cells (T24 방광암세포에서 Luteolin과 TRAIL의 복합 처리에 따른 Apoptosis 유도)

  • Tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) can selectively induce apoptosis by targeting cancer cells. However, some cancer cells are resistant to TRAIL-induced cytotoxicity. One method of overcoming TRAIL resistance is combination treatment with reagents to sensitize cells to TRAIL. Luteolin, a flavonoid, has been shown to have anti-cancer effects by inducing apoptosis and cell cycle arrest in various cancer cell lines in vitro. In this study, we investigated the effects of combination treatment with non-toxic concentration of TRAIL and luteolin in T24 human bladder cancer cells. Combined treatment with luteolin and TRAIL significantly inhibits cell proliferation via activation of caspases by inducing Bid truncation, up-regulation of Bax and down-regulation of X-linked inhibitor of apoptosis protein (XIAP). However, the apoptotic effects of combination treatment with luteolin and TRAIL were significantly inhibited by specific caspases inhibitors. Taken together, these results indicate that combination treatment with TRAIL and luteolin can induce apoptosis in TRAIL-resistant cancer cells through down-regulation of XIAP and modulation of tBid and Bax expression.

Antioxidant effect and inhibitory activities of ethyl acetate fraction from Gardenia jasminoides extract on nitric oxide production and pancreatic cancer cell proliferation (치자 에틸아세테이트 분획의 산화방지, 산화질소 제거 및 암세포증식 억제 활성)

  • To evaluate the radical scavenging activity of phenolic-rich fractions of Gardenia jasminoides, we first measured the levels of total polyphenols in hexane, ethyl acetate, and butanol fractions from the extract of G. jasminoides. The ethyl acetate fraction of G. jasminoides extract (GJ-EA) showed high level of phenolics, potent reducing power, and 2,2-diphenyl-1-picrylhydrazyl/2,2'-azino-bis 3-ethylbenzothiazoline-6-sulfonic acid radical scavenging effect. In addition, GJ-EA inhibited the overproduction of nitric oxide in lipopolysaccharide-activated BV-2 microglia. Furthermore, we found that GJ-EA suppressed $H_2O_2$-induced PANC-1 pancreatic cancer cell proliferation in a concentration-dependent manner and also reduced their migratory ability. These results suggest that GJ-EA may be a good source for functional foods with antioxidant and chemo-preventive activities.

Induction of Apoptosis in Human Cancer Cells with Extracts of Taraxacum coreanum, Youngia sonchifolia and Ixeris dentate (흰민들레, 고들빼기, 씀바귀 추출물의 암세포 증식 억제 효과)

  • This research examined the effects of Compositae extract on the inhibition of proliferation and apoptosis in human breast and human gastric cancer cells. Compositae extracts which is used in the experiment are Taraxacum coreanum (TC), Youngia sonchifolia (YS) and Ixeris dentata (ID). The proliferation of SK-BR-3, MDA-MB-231 and AGS cells were investigated by MTT assay. ID and YS extracts inhibited proliferation of SK-BR-3, MDA-MB-231 and AGS cells in a dose-dependent manner, but TC have barely affected. In addition, the most effective extract was ID. To assess the apoptosis of ID extract, the nuclei of human cancer cells were stained with DAPI solution respectively. Chromatin condensation, indicated apoptosis, was increased in a dose-dependent manner. We investigated change of ID extract-induced apoptosis proteins on human cancer cells by western blot analysis. The level of Bcl-2 decreased, whereas the level of Bax, cleaved-PARP increased in dose-dependent manner compared with non-treatment. Also Bax/Bcl-2 ratio, which is used in clinical indicator of apoptosis, was increased at ID extract treatment group compared with non-treatment. Moreover the Bax/Bcl-2 ratio of MDA-MB-231 cell was significantly increased as against SK-BR-3, AGS cells. These results indicated that ID extract have anti-proliferation effect better than YS or TC, and induced apoptosis in human breast cancer MDA-MB-231 cell better than human breast cancer SK-BR-3 cell, human gastric cancer. Even if further research is needed, ID can be developed as a chemopreventive or therapeutic agent of breast cancer.

Involvement of Early Growth Response Gene 1 (EGR-1) in Growth Suppression of the Human Colonic Tumor Cells By Apigenin and Its Derivative Isovitexin (Apigenin과 대사물 isovitexin에 의한 인체 대장암세포의 세포활성 억제효과에 있어서의 EGR-1의 역할 연구)

  • It has been previously described that transcription factor early growth response gene product 1 (EGR-1) functions as a tumor suppressor gene. This study was conducted to demonstrate that EGR-1 induction by phytochemical apigenin and its derivative isovitexin can mediate the growth suppression of the intestinal epithelial tumor cells. Apigenin and isovitexin induced EGR-1 gene expression both in the dose and time-dependent manners. Moreover the induction was relatively late around 9-12 hr after treatment of HCT-116 cells, while several anti-inflammatory agent such as NSAIDS and catechins elicit the ECR-1 gene expression at much earlier time about 1-3 hr after treatment. In terms of signal transduction, ERK1/2 was critical for apigenin-induced EGR-1 gene expression and its promoter activation. When EGR-1 gene expression was blocked with EGR-1 small interference RNA, the cytotoxicity of apigenin in the human epithelial cells was attenuated, suggesting the involvement of EGR-1 in the anti-tumoric activity of apigenin. To link the EGR-1 induction to EGR-1-regulated gene products in colon cancer, NSAID-Activated Gene 1 (NAG-1) was demonstrated to be elevated by apigenin and isovitexin at 24-48 hr after treatment. Taken together, apigenin-activated ERK1/2 mediated EGR-1 gene induction, which was associated with suppression of the cellular viability by apigenin compound.

Obovatol Inhibits Inflammation Mediator Generation and Colon Carcinoma SW620, HCT116 Cell Growth Through Induction of Apoptotic Cell Death Via Inactivation of $NF-{\kappa}B$ (Obovatol의 염증매개 생성 억제와 세포자멸사를 통한 Colon Carcinoma SW620, HCT116 세포의 세포증식에 대한 영향)

  • 목적 : 후박(厚朴)(Magnolia obovata)에서 추출한 낮은 농도의 obovatol 약침액의 RAW264.7 세포에서 LPS로 유발된 염증, $TNF-{\alpha}$로 유발된 human colon carcinoma SW620 및 HCT116 세포의 세포증식에 대한 영향과 그 기전을 살펴보고자 하였다. 방법 : RAW264.7 세포에서 LPS로 염증을 유발하고 낮은 농도의 obovatol 약침액을 처리한 후 cell viability, NO 생성량, iNOS와 COX-2의 발현, $NF-{\kappa}B$활성, 전사능력을 관찰하기 위해 WST-1 assay, NO determination assay, western blot analysis, EMSA, luciferase activity assay를 시행하였고, HCT116, SW620 세포에 $TNF-{\alpha}$로 증식을 유도하고 낮은 농도의 obovatol 약침액을 처리한 후 cell growth, apoptosis 및 apoptosis와 연관된 $NF-{\kappa}B$의 활성 변화를 관찰하기 위해 WST-1, Cell morphogy test, DAPI staining and TUNEL assay, EMSA, luciferase activity assay를 시행하였다. 결과 : 1. RAW264.7 세포에서 낮은 농도의 obovatol 약침액 처리는 $NF-{\kappa}B$의 활성 및 전사능력을 낮추고 iNOS와 COX-2의 발현과 NO 생성을 감소시켜 LPS로 유발된 염증을 억제하였다. 2. HCT116, SW620 세포에서 낮은 농도의 obovatol 약침액 처리는 $NF-{\kappa}B$의 활성을 낮추어 세포자멸사를 촉진함으로써 $TNF-{\alpha}$로 유발된 암세포의 성장을 억제하였다. 결론 : 이상의 결과는 낮은 농도의 obovatol 약침액이 항염 및 인간 전립선암세포주인 SW620, HCT116에 대한 증식억제 효과가 있음을 입증한 것이며, 향후 이를 바탕으로 한 생체 연구에서의 긍정적인 결과는 obovatol 약침액이 만성염증성 질환 및 대장암의 예방과 치료에 대한 효과적인 치료제 개발에 초석이 될 것으로 기대된다.

Effect of Epigallocatechin Gallate on Inhibition of Cell Proliferation in MDA-MB-231 Human Breast Cancer Cells (Epigallocatechin Gallate가 인체 유방암 세포인 MDA-MB-231의 세포증식억제에 미치는 영향)

  • Epigallocatechin gallate (EGCG), a principal antioxidant derived from green tea, is one of the most extensively investigated chemopreventive phytochemicals. However, the effect of EGCG on proliferation in MDA-MB-231 breast cancer cell is not well known. We investigated the effect of EGCG on protein and mRNA expression related to cell proliferation in MDA-MB-231 human breast cancer cell lines. We cultured MDA-MB-231 cells in the presence of 0, 5, 10 and 20 ${\mu}m$ of EGCG. EGCG significantly inhibited the cancer cell proliferation (p<0.05). In MDA-MB-231 huamn breast cancer cell, EGCG lowered $ErbB_2$ and $ErbB_3$ protein as well as mRNA expression. In addition, protein and mRNA expression of phosphorylated Akt and total Akt were significantly decreased (p<0.05). We suggest that EGCG inhibits cell proliferation through $ErbB_2$, $ErbB_3$ and Akt cell signaling.

Anti-invasive Activity of Human Breast Carcinoma Cells by Genistein through Modulation of Tight Junction Function (인체유방암세포의 tight junction 기능 조절을 통한 genistein의 암세포 침윤 억제 효과)

  • Tight junctions (TJs) that act as paracellular permeability barriers play an essential role in regulating the diffusion of fluid, electrolytes and macromolecules through the paracellular pathway. In this study, we investigated the correlation between the tightening of TJs, permeability and the invasive activity of genistein - a bioactive isoflavone of soybeans - in human breast carcinoma MCF-7 and MDA-MB-231 cells. The inhibitory effects of genistein on cell proliferation, motility and invasiveness were found to be associated with the increased tightness of the TJs, which was demonstrated by an increase in transepithelial electrical resistance and a decrease in paracellular permeability. Additionally, the immunoblotting results indicated that genistein repressed the levels of the proteins that comprise the major components of TJ, claudin-3 and claudin-4, which play a key role in the control and selectivity of paracellular transport. Furthermore, genistein decreased the metastasis-related gene expressions of insulin like growth factor-1 receptor and snail, while concurrently increasing that of thrombospondin-1 and E-cadherin. In addition, we demonstrated that claudins play an important role in the anti-motility and invasiveness of genistein using claudin-3 small interfering RNA. Taken together, our results indicate a possible role for genistein as an inhibitor of cancer cell invasion through the tightening of TJs, which may counteract the up-regulation of claudins. In addition, our results indicate that this may be beneficial for the inhibition of tumor metastasis.
