Antioxidant Activity and Inhibitory Activities of Xanthine Oxidase and Tyrosinase of Yeasts from Wild Flowers in Korea (우리나라 야생화에서 분리한 효모의 항산화 활성과 Xanthine Oxidase 및 Tyrosinase 저해활성)

  • We prepared supernatants and cell-free extracts of yeasts isolated from wild flowers in Korea, and their antioxidant activity and inhibitory activities of xanthine oxidase and tyrosinase were investigated. Among them, cell-free extracts of Kuraishia capsulata UL40-2 and Sporobolomyces ruberrimus 121-Z-3 showed significantly high xanthine oxidase inhibitory activity of 46.4% and 48.3%, respectively. Starmerella bombicola 80-J-1 also showed tyrosinase inhibitory activity of 36.2% in the cell-free extract. Other antioxidant activities and tyrosinase inhibitory activities were not detected or were below 20%. Maximal production of the xanthine oxidase inhibitors were observed when Kuraishia capsulate UL40-2 and Sporobolomyces ruberrimus 121-Z-3 were cultured in the yeast extract-peptone-dextrose media at $30^{\circ}C$ for 24 hour, respectively.

  • 본 연구는 DMZ 및 접경지역에서 생육하고 있는 자생식물에 대하여 조사된 연구자료와 국립수목원 DMZ자생식물원에서 자체 조사로 수행된 미조사지역 연구 등을 집대성한 DMZ 일원의 관속식물상 가운데 행정구역상 고성군, 인제군, 양구군, 화천군에 해당하는 동부지역 관속식물의 유용자원 현황에 대하여 기초자료를 제공하고자 하였다. 동부지역은 고성군의 석호들을 포함한 저층습지 및 해안지역과 설악산(1,708m), 점봉산(1,424m), 화악산(1,468m), 등의 고산지대, 대암산(1,304m) 정상부근에 위치한 고층습지 용늪(1,280m)까지 이르는 다양한 생활환경 특성을 포함하고 있다. DMZ일원의 관속식물상 157과 754속 2,047종 12아종 340변종 105품종 총 2,504분류군 중 40.2%인 111과 438속 873종 4아종 113변종 17품종 1,007분류군이 확인된 동부지역 유용식물의 조사 결과 식용식물 순채, 고려엉겅퀴, 병풍쌈 등 336분류군(33.3%), 사료용식물 산뽕나무, 고광나무, 새 등 334분류군(33.1%), 약용식물 주목, 백부자, 삼지구엽초 등 270분류군(26.8%), 관상용식물 제비동자꽃, 백작약, 만병초 등 118분류군(11.7%), 목재용식물 일본잎갈나무, 소나무, 오동나무 등 38분류군(3.7%), 염료용식물 밤나무, 매발톱나무, 갈매나무 등 26분류군(2.5%) 섬유용 식물 개모시풀, 쐐기풀, 참싸리 등 8분류군(0.7%), 산업용식물 굴참나무, 붉나무, 노박덩굴 3분류군(0.2%), 용도를 알지 못하는 식물 280분류군(27.8%)으로 나타났다.

The Contents of Chlorogenic acid and Eleutheroside E in Eleutherococcus senticosus (Rupr. et Maxim.) Harms (가시오갈피나무의 Eleutheroside E 및 Chlorogenic acid 성분함량(成分含量))

  • Eleutherococcus senticosus (Rupr. et Maxim.) Harms is a tree species that has been traditionally used as a source for oriental folk medicine. It is known to contain both Eeleutheroside E and chlorogenic acid. These two compounds are believed to be effective elements in the tree. In the present study, we analyzed the levels of the two compounds from the plants collected from various locations in Korea to find out whether environment affected the quantity of both compounds in the tissue. When the plants growing in their natural habitats and those growing in plantations were compared, the level of the two compounds showed different pattern. In the case of chlorogenic acid in roots, the plants growing in plantations are found to contain higher levels than did those from natural habitats. On the other hand, higher levels of eleutheroside E were found in the plants from natural habitats. However, when the compounds in the stems were compared, the plants growing in the natural habitats contained higher level in both compounds than did those from plantations. Although the level of both compounds in the leaf tissue varied with the places from which they were originated, it appeared to be less than that found in roots or stems. There exists a tendency that the level of eleutheroside E showed a positive correlation with the diameter of both roots and stems of the plants.

Distribution of Heavy Metals within Forest Soils in Seoul (서울시(市) 산림토양내(山林土壤內) 중금속(重金屬) 분포(分布))

  • Distribution of heavy metal (Zn, Pb, Cu, Cd) concentrations within forest soils was investigated in Mts. Surak and Bulam around the outskirt area, and Mts. Bukak and Inwang around the downtown area of Seoul. Metal concentrations in these soils were higher in the A horizons than in the B horizons. The mean concentrations of Zn, Pb, Cu, and Cd in the A horizons were 4.39 mg/kg, 9.09 mg/kg, 1.67 mg/kg, and 0.03 mg/kg, respectively. The concentrations in the B horizons were 2.18 mg/kg in Zn, 5.74 mg/kg in Pb, 0.55 mg/kg in Cu, and 0.01 mg/kg in Cd, respectively. With positive correlation among the metal concentrations, Zn, Pb, and Cu concentrations in the A horizon were positively correlated with soil organic matter contents (r=0.39~0.83, p<0.01), while Pb and Cu concentrations were negatively correlated with soil pH (r=-0.53~0.59, p<0.01). The results suggest that the metals in forest soils have accumulated more in the downtown than in the outskirt areas.

The Flora of Gwangchiryeong Area Adjacent to the DMZ (DMZ에 인접한 광치령 일대의 식물상)

  • Gwangchiryeong, located adjacent to the Korean DMZ, is a mountain pass connecting Yang-Gu, Gangwon-do with In-je. It reaches a height of 800m (2,624ft) and stands on the flank of Mt. Daeam-san, which has an elevation of 1,304m (4,278ft). It is important to survey the flora of Gwangchiryeong in that not only the area encompassing the waterfall and the valley along with dense old-growth forest has various plant species but also it is a way to make out what kind of plant species exist in the DMZ due to the geographical characteristics of Gwangchiryeong adjacent to the Korean DMZ. Therefore, this study was conducted to elucidate the distribution of vascular plants and their value in Gwangchiryeong. The vascular plants that were collected 9 times from April 2014 to September 2015, and consisted of a total of 513 taxa: 88 families, 297 genera, 441 species, 4 subspecies, 62 varieties and 6 forms. Among those observed species, the rare plant species designated by Korea Forest Service were 22 taxa including Hanabusaya asiatica, Paeonia japonica, Saxifraga octopetala, etc. Endemic species were 22 taxa containing Cirsium setidens, Heloniopsis koreana, Salvia chanryoenica, etc. 18 taxa were recorded as naturalized plants and their naturalized rate and urban index were noted as 3.5% and 5.6%. Among the surveyed 513 taxa, edible, medicinal, pasturing, ornamental, timber, dye, fiber and industrial plants were 197 taxa, 154 taxa, 180 taxa, 49 taxa, 24 taxa, 8 taxa, 4 taxa and 3 taxa, respectively.

Effect of a Combinacon of Mental Activity with Experimental Subway Noise on Hematology (실험적 지하철 소음에서의 지적활동이 혈액생리에 미치는 영향)

  • The present study was sought to clarify whether the combination of mental activity with subway noise affects hematological variables. Fifty-six healthy volunteers participated in this experiment and underwent a stress task consisting of combination_of mental activity (mental arithmetic) with subway noise for 50 min and 60min of recovery after the end of the stress task. Venous blood samples were collected for measuring CBC, prothrombin time (PT), activated partial thromboplastin time (aPTT), erythrocyte sedimentation rate (ESR), fibrinogen concentration, D-dimer and high sensitive C-reactive protein (H-CRP) levels before (baseline), 50min of stress task (S-50m), and 60 min of recovery (R-60m). Total leukocyte, neutrophil and lymphocyte counts significantly increased at R-60m compared with baselines. RBC count at S-50m was higher, while monocyte counts at S-50m and R-60m were lower than those of baselines. aPTTs shortened at S-50m and R-60m, but PT reduced at R-60m as compared with baselines. D-dimer and H-CRP levels at S-50m and R-60m were significantly higher than those of baselines. These findings imply that a combination of mental activity with subway noise nay cause leukocytosis, homo-concentration, shortened PT and aPTT, decreased ESR, and raised D-dimer and H-CRP levels, suggesting possible development of inflammation and prothrombogenic reaction attributable to a subway environment.

Properties of Disconitinuity for the Seoul Granite in the Northeastern Part of Seoul City (서울시 북동부의 서울화강암에 대한 불연속면의 특성)

  • Properties of discontinuity for Seoul Granite in northeastern part of Seoul City were analyzed by dividing structural domains into Surak and Bulam Mtn. areas. Important parameters measured among several engineering properties of a rock during tunnel excavation and road construction are as follows: 1) Orientation of joint, 2) joint spacing, 3) joint density, and 4) uniaxial compressive strength. Orientation, spacing, and density of joints can be directly measured during field investigation using scanline survey, circle-inventory method, and window survey. Uniaxial compressive strength of the rock was calculated by a simple correlation equation although it is originally necessary to prepare core samples in measuring it. Major orientations of joints measured from both areas are 3 sets of joints with different orientations. In other words, they are 2 sets of orthogonal joint and 1 set of sheet joint that is dipping at low angle, and have very similar orientations in both areas. Joint densities in both areas range from 0.039 and 0.066/cm, and average joint length are between 1.30 and 4.52m. Average joint spacing also has values from 10.3cm up to 59.6cm, and shows significant difference along specific orientation of scanlines measured. Values of uniaxial compressive strength calculated on the basis of Schmidt hammer rebound values range from 217 to 335 MPa, which indicates very strong rock type by classification of wall strength.

Length of Estrous Cycle and Duration of Estrus following Natural and Induced Estrus in Korean Native Goats (山羊의 自然 및 誘起發情時 發情週期 및 發情持續時間에 관한 硏究)

  • This study was carried out to investigate the length of the estrous cycle and duration of estrus in Korean native goats. The effects of the type of last estrus : induced or natural, and the length of preceeding estrous cycle on the subsequent cycle and duration of estrus in goats were also examined. From 481 observations, the mean length of estrous cycles was 18.1${\pm}$0.5days. The cycle length was significantly(P<0.05) shorter following induced estrus (15.2${\pm}$0.8 d) than natural estrus(19.0${\pm}$0.6 d). Significantly higher(P<0.05) incidence of short estrous cycle was observed following induced estrus(40%) than natural estrus(27%). The frequency distribution in the estrous cycle was 30%, 3%, 56% and 11% for short(3-11 d), medium(12-16 d), normal(17-24 d) and long(${\geqq}$25 d), respectively. The most frequent type of short estrous cycle was 6 days in length. Mean duration of estrus was 34.0${\pm}$0.5 h with a range of 18 to 84 h. Duration of estrus was not significantly different the preceeding natural (33.8${\pm}$0.6 h) and induced estrus (34.4${\pm}$1.1 h), and was not significantly affected by the last cycle length. These results suggest that the short estrus cycles are more frequent following induced estrus than natural one, and the duration of estrus are affected greatly by the various intrinsic and extrinsic factors.

Studies on the Antitumor Activity of Gamisoam-san via Suppressing Angiogenesis and Growth Factor Expression (혈관신생 및 이식암세포증식 억제를 통한 가미소암산의 항암작용연구)

  • Gamisoamsan is a prescription originated in Soamsan which is known as an anti-cancer remedy in the traditional Korean Medicine. To enhance the synergic effects of anti-cancer activity of Soamsan, this study reconstituted the original components of Soamsan with a slight modification and produced a novel herbal remedy, namely Gamisoamsan. To investigate the effects of Gamisoamsan on anti-cancer reaction, I studied the effects of Gamisoamsan on angiogenesis via chorioallantoic membrane (CAM) assay, corneal neovascularization assay and the effects on expression of growth factor which are VEGF, TGF-β, bFGF and IMUP-1. Anti-cancer effects of Gamisoamsan was also abserved through hematological parameters, tumor volume and survival rate in mice. Gamisoamsan inhibited embryonic angiogenesis of blood vessels in CAM assay and inhibited neovascularization of ral cornea. Gamisoamsan reduced cell proliferation in HT1080 cells and IC50 was 2.18 ㎎/㎖ Gamisoamsan reduced the expression of VEGF, TGF-β, bFGF and IMUP-1 which was known as vascular growth factor and this effects of Gamisoamsan was predominant than VP-16. The treatment of Gamisoamsan decreased the CT-26 cell inoculated-tumor volume in mice colon adenocarcinoma and increased mice survival which was inoculated CT-26 cells. The results of the present study suggest that Gamisoamsan extracts has a potential anti-tumor activity and may be an useful remedy to prevent and/or treat cancer.

Relationship Between Conversation Skills, Working Memory and Naming Ability in Aging Adults (노인의 대화기능과 작업기억력 및 이름대기 능력 간의 관련성 연구)

  • For knowing the effects of aging on conversational skills in daily communication, this paper studied for the conversational turn-taking skills, working memory and naming ability on healthy elderly adults over 65 ages. 85 elderly adults participated in this study, which divided into four groups by ages. Speech samples were collected in natural conversation. Memorization of numbers, mental calculation, repetition of words were administered for working memory test. K-BNT was used for the naming ability. One-way ANOVA analysis was used for the comparison of conversational turn-taking skills among four groups. We analyzed the correlation between conversational skills, working memory and naming ability. The results were as follows: first, there were a significant difference in conversational turn-taking skills by age, but not by gender. There was a significant difference in 'Turn-Taking Frequency' and 'Total Utterance Frequency' among four groups. The same results were shown in the scores of females within three groups(exclude groups over 85D)(p<.01). Second, there was a significant correlation between 'rates of maintenance' and 'naming ability'. In addition, it was found that the naming test predicted 'rates of maintenance' skills. The results of this study suggest that word-retrieval ability will be helpful to enhance functional communication skills in aging old adults.
